Pulmonary capillary endothelial cells (ECs) consist of two populations, CAP1 and CAP2; how each population reacts to diverse tissue injury is incompletely understood. Using single-cell multiome and mouse genetics, we characterize the induction and function of a truncated isoform of Ntrk2, Ntrk2-T1, in multiple lung injury models. Upon Sendai parainfluenza infection, Ntrk2-T1 is broadly induced in CAP1s after the initial interferon response, associated with increased intronic chromatin accessibility, and persists for weeks. Ntrk2-T1 ECs arise from CAP1s but not CAP2s-traced by

Type I interferons are secreted in response to various stimuli and are used as a treatment for many diseases, including infections with the hepatitis B virus (HBV) and its satellite virus, hepatitis delta (HDV). HDV significantly aggravates HBV-mediated liver damage and is - in contrast to HBV - a strong inducer of interferon responses, including IFN-β. As the role of IFN- β in liver metabolism is so far ill explored, we studied its impact on hepatocyte metabolism in HDV-infected cultures. Transcriptome analysis, isotope tracing and functional tests on differentiated, HDV-infected hepatocytes showed reduction of mitochondrial TCA cycle and respiratory activity …

Atopy-Related Immune Profiles Are Subject To Genetic Influence As Evaluated Using School-Aged Twin Pairs, Jonatan Leffler, James F. Read, Danny Mok, Michael Serralha, Anthony Bosco, Patrick G. Holt, Richard Saffery, Deborah H. Strickland

Research outputs 2022 to 2026

The interaction of genetic and environmental contributions to immunological traits and their association with atopic disease remain unclear. Flow cytometry and in vitro cytokine responses were used to characterize immune profiles from 93 school-aged twin pairs. Using an established twin pair analytical strategy, the genetic and environmental influences on immunological traits were evaluated, along with their association with atopy. Our findings suggest strong genetic influence on several traits, particularly B cell abundance. In contrast, cytokine responses from in vitro stimulations appeared mainly shaped by environmental exposures. Regarding associations with atopy, greater abundance of both B cells and basophils were observed …

University students often face significant academic, social, and environmental pressures, making them particularly vulnerable to psychological distress. Despite this, there is limited evidence regarding the prevalence and associated factors of mental health problems among university students in Iraq. Therefore, this study aimed to assess the prevalence and severity of depression, anxiety, and stress in this population and to examine their associations with sociodemographic characteristics, insomnia, internet use, and physical activity. A cross-sectional study was conducted among Iraqi university students using a structured questionnaire, including the DASS-21, Insomnia Severity Index, and Internet Addiction Test. Descriptive and inferential statistics were used to …

BACKGROUND AND OBJECTIVE: NRG/RTOG 0521 randomized men with high-risk localized prostate cancer (PC) to androgen suppression (AS) and definitive radiotherapy (RT) ± docetaxel-based chemotherapy (CT). The overall survival (OS) benefit with CT initially reported was lost on longer follow-up. The Decipher genomic classifier (GC) measures multiple transcripts relevant to docetaxel action. Basal/luminal differentiation portends differential response to AS and CT for high-risk localized and metastatic hormone-sensitive PC. We validated the Decipher GC in pretreatment biopsy samples for risk stratification and examined basal-luminal subtyping to predict docetaxel response.

METHODS: Decipher GC scores and basal-luminal cellular subtypes were generated for specimens from …

In this perspective, we highlight the relevance of the FA-Hippo signaling pathway and its regulation of the Yes-associated protein (YAP) and the transcriptional coactivator with a PDZ-binding domain (TAZ) as main players in the process of implants integration. The modulation and responses of YAP/TAZ triggered by substrate and ECM stiffness are of particular interest in the construction of materials used for medical implants. YAP/TAZ nuclear localization and activity respond to the substrate stiffness by several mechanisms that involve the canonical and non-canonical Hippo signaling and independently of the Hippo cascade. YAP/TAZ regulate the expression of genes involved in several mechanisms …

Primary testicular (PT) diffuse large B-cell lymphoma (DLBCL) is a rare and aggressive lymphoma with distinct clinical and molecular characteristics. To identify prognostic biomarkers in PT-DLBCL, in this study we analyzed DNA and RNA samples of PT-DLBCL tumors from 206 patients using next-generation sequencing platforms and assays. Genetic alteration analysis found that multiple chromosomal copy number variations (CNVs), TP53 transcript mutations with high variant allele frequency, and MCD subtype had significantly adverse prognostic effects, whereas elevated microsatellite instability had a significantly favorable prognostic effect in PT-DLBCL. Targeted RNA-seq analysis identified a PTL gene expression signature by comparing PT-DLBCL with systemic …

Background: Limited ancestral diversity has impaired our ability to detect risk variants more prevalent in ancestry groups of predominantly non-European ancestral background in genome-wide association studies (GWAS). We construct and analyze a multi-ancestry GWAS dataset in the Alzheimer's Disease Genetics Consortium (ADGC) to test for novel shared and population-specific late-onset Alzheimer's disease (LOAD) susceptibility loci and evaluate underlying genetic architecture in 37,382 non-Hispanic White (NHW), 6728 African American, 8899 Hispanic (HIS), and 3232 East Asian individuals, performing within ancestry fixed-effects meta-analysis followed by a cross-ancestry random-effects meta-analysis.

Results: We identify 13 loci with cross-population associations including known loci at/near CR1, …

Racial disparities in the clinical outcomes of triple-negative breast cancer (TNBC) have been well-documented, but the underlying biological mechanisms remain poorly understood. To investigate these disparities, we employed a multi-omic approach integrating imaging mass cytometry and spatial transcriptomics to characterize the tumor microenvironment (TME) in self-identified Black American (BA) and White American (WA) TNBC patients. Our analysis revealed that the TME in BA patients is marked by a network of endothelial cells, macrophages, and mesenchymal-like cells, which correlates with reduced patient survival. In contrast, the WA TNBC microenvironment is enriched in T-cells and neutrophils, indicative of T-cell exhaustion and suppressed …

Neuroactive steroids (NAS) have long been recognized for their hypnotic and anesthetic properties in both clinical and preclinical settings. While sex differences in NAS sensitivity are acknowledged, the underlying mechanisms remain poorly understood. Here, we examined sex-specific responses to an endogenous NAS epipregnanolone (EpiP) in wild-type mice using behavioral assessment of hypnosis (loss of righting reflex, LORR) and in vivo electrophysiological recordings. Specifically, local field potentials (LFPs) were recorded from the central medial thalamus (CMT) and electroencephalogram (EEG) signals were recorded from the barrel cortex. We found that EpiP-induced LORR exhibited clear sex differences, with females showing increased sensitivity. Spectral …

The Attitudes And Knowledge Of Organ Donation And The ‘Opt Out’ Consent System Among Irish Third Level Students., Rachel Webster, Emma Flynn, Tara O'Brien

International Undergraduate Journal of Health Sciences

Currently there are approximately 500 people in Ireland waiting for an organ donation while only approximately 280 transplants are carried out per year. While close to 80% of Irish people support organ donation it is estimated that only one-third of those are registered donors. This is the basis for the new ‘Opt Out’ policy within the ‘Human Tissue (Transplantation, Post-Mortem, Anatomical Examination and Public Display) Act 2024’ that has recently been signed into law (as of 28th February 2024). This study aimed to investigate third-level students’ awareness of this new legislation and to determine if there would be an …

A Meta-Analysis Of The Relationship Between Estradiol, C-Reactive Protein, Interleukin-6 And Tumour Necrosis Factor-Alpha, And Their Association With Breast Cancer Prognosis, Keelin M. Crowley

International Undergraduate Journal of Health Sciences

This meta-analysis reviewed interleukin 6 (IL-6) levels across various tumour stages, compared transdermal and oral estradiol levels on IL-6 and C-reactive protein (CRP), and examined the IL-6 and tumour necrosis factor alpha (TNF-a) levels in healthy versus breast cancer patients. This analysis covers studies from 2003 to 2020.

Results: IL-6 can be used as a prognostic biomarker for breast cancer, as levels are seen to increase as the cancer stage advances. While transdermal estradiol does not have any significant effects on IL-6 or CRP, oral estradiol seemed to display an increase in. The baseline IL-6 levels differ between control subjects …

Audit Of The Impact Of The Pause In The National Screening Service For Breast Cancer During The Covid-19 Pandemic, Ava Coakley

International Undergraduate Journal of Health Sciences

In Ireland there is a free national screening service for breast cancer called BreastCheck. BreastCheck invites women aged fifty to sixty-nine to have screening mammograms once every two years to detect any densities in their breast mammogram which could indicate breast cancer. If densities are seen on a mammogram a breast core is sampled and sent to a histopathology laboratory where a part of the tissue is examined by a consultant pathologist using a microscope. Further tests can then be performed on this sample if it is diagnosed as breast cancer. Tests include immunohistochemistry and dual in-situ hybridisation (DISH). BreastCheck …

A Review Of Breast Implant Associated Anaplastic Large Cell Lymphoma (Bia-Alcl) In The Histopathology Laboratory In Cork University Hospital (Cuh), Ruth Purcell

International Undergraduate Journal of Health Sciences

Breast implantation is a common procedure for either breast reconstruction, following a mastectomy or for cosmetics. Approximately 10 million individuals have breast implants worldwide. Breast implants are composed of an outer silicone layer that are filled with either silicone gel or saline and have a lifespan of approximately 16 years (Quesada et al., 2018). When the implantation occurs, a fibrous capsule is naturally formed around the implant. However, it has become apparent in recent years that there is a link between the use of textured implants and a rare type of lymphoma. Breast Implant Associated- Anaplastic Large Cell Lymphoma (BIA-ALCL) …

Implementation Of Quantitative Blood Loss To Detect Postpartum Hemorrhage After Vaginal Birth, Raven S. Simpson Jul 2025

Implementation Of Quantitative Blood Loss To Detect Postpartum Hemorrhage After Vaginal Birth, Raven S. Simpson

Dissertations

Problem: In the U.S., postpartum hemorrhage (PPH) is attributed to maternal morbidity and mortality, resulting in long-lasting effects of extended recoveries, surgical interventions, birth-related trauma, interruptions in mother-infant bonding, or death. Performing estimated blood loss (EBL) measurement methods postpartum is subjective and often associated with over- and underestimations of blood volume. The delay in recognition interferes with timely, efficient, and appropriate interventions, yielding a PPH (≥1000ml).

Methods: A quality improvement (QI) project was initiated with a descriptive, pre-implementation/post-implementation focus on a high-risk Labor and Delivery unit to explore the effects of Quantitative Blood Loss (QBL) measurements on the identification of …

Ldt Fda Ruling: A Conundrum For Laboratories Across The Usa, Charma Henry

MSMLS Capstone Projects

Abstract

In the article “The Future of Laboratory Medicine Understanding the New Pressures," author Mauro Panteghini expresses that the clinical laboratory has always undergone significant changes due to technological advances and external economic pressures. “Laboratories are indeed an easy target for economic restrictions and limitations due to their technological characteristics.” (Panteghini, 2004, pg. 205) Though these words were written over 20 years ago, no more valid words have been spoken in light of the newest pressure all laboratories face with the new LDT regulation set by the FDA.

This regulation places drastic economic pressure on laboratories to pay the FDA …

Myb And Hif1Α Crosstalk Drives Hypoxia-Induced Transcriptional Reprogramming And Adaptive Signaling Alterations In Pancreatic Cancer, Shashi Anand, Kunwar Somesh Vikramdeo, Mohammad Aslam Khan, Ajay Pratap Singh

Cell and Molecular Biology Publications

Pancreatic cancer is an aggressive malignancy, characterized by extensive desmoplasia and a hypoxic tumor microenvironment that contributes to therapy resistance. MYB, a proto-oncogene encoding a transcription factor, plays a crucial role in pancreatic tumor growth and metastasis. Recently, we also revealed a role of MYB in hypoxic survival of pancreatic cancer cells by promoting metabolic reprogramming through interaction with HIF1α, modulating its expression and binding to glycolytic gene promoters. In this study, we investigated how hypoxia influences the genome-wide occupancy of MYB using chromatin immunoprecipitation sequencing (ChIP-seq), and whether this effect is modulated by its interaction with HIF1α. In addition, …

Bowel Regimen Protocol Implementation For Pediatric Patients Following Cardiac Surgery, Eryn Watson Jul 2025

Bowel Regimen Protocol Implementation For Pediatric Patients Following Cardiac Surgery, Eryn Watson

Dissertations

Problem: Pediatric patients with congenital heart disease (CHD) are at an increased risk of constipation following cardiac surgery, which contributes to complications and delays in recovery. Standardized protocols are effective tools for prophylactic management of constipation. However, there is no current standardized protocol in place for postoperative bowel management in a pediatric cardiac intensive care unit (CICU).

Method: This quality improvement project (QI) used a descriptive design with pre- and post-implementation data collection to evaluate the effects of a postoperative bowel regimen protocol on constipation rates and compliance with ordering prophylactic bowel regimens. Convenience sampling was used to identify patients …

Maternal Hyperleptinemia Alters Placental Transcriptome And Progesterone Regulation Without Affecting Early Equine Fetal Growth, Lucinda L. Thorburn Jul 2025

Maternal Hyperleptinemia Alters Placental Transcriptome And Progesterone Regulation Without Affecting Early Equine Fetal Growth, Lucinda L. Thorburn

LSU Master's Theses

While consequences of equine obesity are well defined, the extent to which persistent hyperleptinemia and accompanying leptin (Lep) resistance affect the fetal-maternal environment warrants further exploration. This study compared endocrine, fetal, placental, and uterine parameters during early pregnancy (through day 70) in mares with elevated leptin versus normal leptin (NL) concentrations. Fourteen mares were classified as either NL (n = 6; Lep 0.9 ± 0.3 ng/mL), moderate leptin (ML; n = 4; Lep 3.0 ± 0.3 ng/mL), or high leptin (HL; n = 4; Lep 5.8 ± 0.7 ng/mL) based on annual plasma leptin. Mares were artificially inseminated and induced …

Dose Characterization Of Galactic Cosmic Rays In The Human Brain: A Phits-Based Study With A Segmented Anatomical Phantom, Shanice N. Manning

LSU Master's Theses

Astronauts aboard the International Space Station (ISS) are chronically exposed to a complex space radiation environment composed of galactic cosmic rays (GCR), solar particles, and Earth's trapped radiation. These high-energy particles interact with spacecraft shielding, producing secondary particles that can penetrate inside and deposit energy in sensitive tissues. Although animal studies have been useful in assessing space radiation risks, anatomical and physiological differences limit their applicability to human anatomy especially neuroanatomy. To address this gap, we developed a computational framework to model radiation dose distribution within the human brain using Monte Carlo simulations in Particle and Heavy Ion Transport code …

Role And Electrophysiological Properties Of Sexually Dimorphic Oxytocin Receptor Expressing Neurons In The Anteroventral Periventricular Nucleus Of The Female Mice, Armita Abdollahi Govar Jul 2025

Role And Electrophysiological Properties Of Sexually Dimorphic Oxytocin Receptor Expressing Neurons In The Anteroventral Periventricular Nucleus Of The Female Mice, Armita Abdollahi Govar

LSU Doctoral Dissertations

Oxytocin is a neurohypophyseal hormone that plays a crucial role in regulating mother-infant bonding and maternal behavior. Oxytocin modulates sex-specific parental behaviors by binding to the oxytocin receptor (OXTR)-expressing neurons in the brain. This study investigates the sexually dimorphic OXTR neurons in the anteroventral periventricular nucleus (AVPV) to understand how oxytocin modulates behavior in females. Specifically, we explored the behavioral roles, neural projections, and intrinsic electrophysiological characteristics of these neurons in the female mice. Using immunohistochemistry revealed that the number of OXTR-expressing neurons was enhanced in postpartum females compared with virgin females, suggesting that these neurons are involved in female-specific …

Pathological events often impact tissue regions in a spatial variable manner, making it challenging to identify therapeutic targets. Spatial transcriptomics (ST) is a powerful technology to map spatially variable molecular mechanisms, yet suitable analytical methods have been lacking. We introduce SPaSE (Spatially-resolved Pathology ScorE), an optimal transport-based algorithm to compare ST data from diseased and control tissues. SPaSE computes a “pathology score” for each spot in the diseased sample, quantifying the pathological impact at that spot. In post-MI (myocardial infarction) mouse hearts, these scores delineated zones that matched independent expert annotations. Modeling pathology scores from gene expression revealed signatures predictive …
