Artificial intelligence is increasingly being utilized in pharmaceutical research and development to speed up drug discovery, enhance clinical trial design, and support regulatory decision-making. However, the increasingly rapid adoption of AI in the pharma industry has outpaced the development of consistent ethical governance, raising concerns related to data privacy, transparency, regulatory compliance, and stakeholder trust. This study explored the methods by which project managers could develop and utilize ethical decision-making frameworks to facilitate responsible AI implementation in pharmaceutical research and development. Chronic neuropathic pain severely diminishes quality of life, yet the supraspinal neural circuits that sustain pain perception and mediate analgesic relief remain incompletely characterized. While peripheral and spinal pain pathways are well established, relatively few studies have mapped pain- or opioid-related neuronal activation across the entire brain. This dissertation employed activity-dependent genetic labeling (TRAP2/Ai9), whole-brain tissue clearing, light sheet microscopy, and graph-theoretic network analysis to characterize brain-wide activation patterns across chronic pain and analgesic conditions in mice.

Modern lifestyles reduce sunlight exposure and increase artificial light at night, potentially promoting vitamin D deficiency (VDD) and melatonin (MEL) suppression. The two hormones regulate immunity, erythropoiesis, and platelet (PLT) indices, but their combined hematological effects are not well characterized. We investigated the independent and combined effects of VDD and MEL deficiency on leukocyte profiles, erythrocyte indices, and PLT parameters in albino rats using two 12-week models (n = 8/group): continuous light (CL)-induced MEL suppression (Control, CL, VDD, and CL + VDD) and pinealectomy (Px)-induced MEL deficiency (Sham. Lymphedema is a chronic condition that can significantly impact physical function, psychosocial well-being, and participation in meaningful occupations. Although traditional management primarily focuses on physical symptom reduction, gaps remain in addressing long-term participation and quality of life, particularly among underserved populations. The purpose of this capstone project was to develop an occupation-based support group manual designed to support both the physical and psychosocial needs of adults living with lymphedema.

The Role of the "Indulgent Mindset" as a Modulator for Neuropsychiatric Biomarkers and Self-Reported Mental Wellbeing
Objective: Limited understanding persists regarding how an "indulgent mindset" about food shapes neuropsychiatric outcomes. Building on the "milkshake study" demonstrating that mindset modulates ghrelin response independent of nutritional content, this pilot study examines whether an indulgent mindset intervention affects neuropsychiatric biomarkers and mental wellbeing.

SACAD: Scholarly Activities
Traditional metabolism instruction often leaves students struggling with pathway interconnectedness and metabolite flux. To foster systems thinking, we are developing an educational board game where students manipulate tokens to navigate physiological constraints and achieve target energy goals. Grounded in Design-Based Research (DBR), the game undergoes iterative refinement based on player feedback. The design utilizes constructivism by allowing students to "fail safely" and revise strategies in real-time, while leveraging embodied cognition to externalize abstract flux into the physical trading of resource tokens.

Septal ring assembly protein ZapB (EMR18431.1) is involved in Z-ring formation, and nucleoid segregation during the bacterial cell division . It promotes cell division in the initial stages of the cell cycle through direct interaction with FtsZ, thus stimulating Z-ring assembly. The ZapB inhibition can make bacterial cell susceptible to primary antibiotics, and therefore considered as an important approach for adjuvent therapy against MDR bacterial infections. Hence in the current study, ZapB protein of multidrug resistant Klebsiella pneumoniae was cloned, and expressed in E. coli system. Traumatic brain injury (TBI) is a major public health concern associated with long-term disability and high healthcare utilization. Evidence guiding post-acute rehabilitation for medically complex TBI populations remains limited, particularly in long-term acute care hospital (LTACH) settings serving medically unstable patients. This study examined clinical and system-level factors associated with functional rehabilitation outcomes among adults with moderate-to-severe TBI in LTACHs. Guided by the Socioecological Model, recovery was conceptualized as interactions across individual, clinical, and organizational levels. Musculoskeletal ultrasound (MSK-US) is a dynamic, cost-effective imaging modality increasingly integrated into Doctor of Physical Therapy (DPT) curricula. Despite growing interest, variability exists in the integration and implementation of MSK-US in entry-level DPT programs. This study aimed to assess the current status of MSK-US education in accredited DPT programs across the United States. A cross-sectional survey was distributed to all 294 accredited DPT programs in the U.S. via email. The survey consisted of 32 questions that addressed curriculum content, instructional methods, faculty qualifications, and barriers to implementation. Participation was voluntary, and 44 programs responded. No experimental interventions were applied. The …

Glucagon-like peptide-1 (GLP-1) therapies are under investigation for a growing number of neuropsychiatric conditions, including substance use disorders. Cigarette smoking accounts for the largest proportion of substance use-related morbidity and mortality, in part reflecting increased risk for cardiometabolic disease among people who smoke. Given modest quit rates with approved smoking cessation therapies, medications with novel mechanisms of action are needed to expand the available monotherapy and combination treatment options. Treatments that prevent post-cessation weight gain could further reduce relapse and cardiometabolic risks following smoking cessation. ZNRF3 and RNF43 are closely related transmembrane E3 ubiquitin ligases with significant roles in development and cancer. Conventionally, their biological functions have been associated with regulating WNT signaling receptor ubiquitination and degradation. However, our proteogenomic studies have revealed EGFR as the protein most negatively correlated with ZNRF3/RNF43 mRNA levels in multiple human cancers. Through biochemical investigations, we demonstrate that ZNRF3/RNF43 interact with EGFR via their extracellular domains, leading to EGFR ubiquitination and subsequent degradation facilitated by the E3 ligase RING domain. Hepatocellular carcinoma (HCC) is a major cause of cancer-related mortality and is largely driven by metabolic disorders such as obesity and type 2 diabetes. The AMP-activated protein kinase (AMPK) is a master regulator of metabolism, and its activation has been proposed as a therapeutic strategy for treating metabolic disorders. However, although AMPK activity is down-regulated in HCC, the precise role of AMPK in HCC development has not been clearly delineated. Current spatial T cell receptor (TCR) profiling approaches lack the resolution needed to link clonal identity, transcriptional state, and spatial positioning of individual T cells in the tumor microenvironment. Here, we introduce a spatial TCR profiling strategy that resolves individual T cell clones together with their transcriptional states at single-cell resolution and applied the method to human head and neck squamous cell carcinoma. Chimeric antigen receptor T-cell (CAR-T) therapy has emerged as a great advancement in the field of oncology, offering a targeted and adaptable cancer treatment. Traditional therapies, including surgery, chemotherapy, and radiation often have limitations, such as relapse and resistance, highlighting the need for more precise therapeutic options. CAR-T therapy addresses many of these challenges through engineering a patient’s own T-cells to recognize and destroy cancerous cells with specificity and long-lasting immune activity. Potassium is an essential macronutrient for plant growth and development, yet excessive potassium fertilization can induce salt stress with detrimental consequences for crop productivity and nutritional quality. Despite its agricultural relevance, potassium chloride induced stress remains significantly understudied compared to classical sodium-based salinity. This thesis investigates the physiological, biochemical, and molecular responses of Raphanus sativus to KCl stress using an integrated approach that combines germination assays, mineral profiling, and gene expression analysis.

Background: Zebrafish (Danio rerio) models are gaining popularity in biomedical research; however, the high infrastructure cost, at approximately $20,000 per unit, is a significant hurdle for primary undergraduate institutions (PUIs) establishing zebrafish facilities. BACKGROUND: Since the inception of spinal radiosurgery decades ago, the factors influencing spinal cord recovery have remained unclear. Elucidating these factors would aid in reirradiation planning and delivery. We describe modeling reirradiation normal tissue complication probabilities (NTCP) from compiled published data, akin to the collaborative Quantitative Analysis of Normal Tissue Effects in the Clinic (QUANTEC) and High Dose per Fraction, Hypofractionated Treatment Effects in the Clinic (HyTEC) projects.
