Background: The negative impact of adverse childhood experiences (ACEs) on child development is documented. The parent-child relationship protects against ACEs and improves healthy child development, playing a crucial role in preventing and mitigating ACEs by strengthening parental resilience. However, there is a gap in the literature on our understanding of parental resilience's impact on the parent-child relationship within the social-ecological model (SEM) (i.e., intra- and interpersonal, community, and societal levels). Type 2 diabetes mellitus (T2DM) is a global metabolic disorder characterized by insulin resistance and impaired glucose uptake. Despite the availability of pharmacological therapies, limitations such as adverse effects and high costs highlight the need for alternative therapeutic candidates. Tinospora cordifolia has been widely reported to contain bioactive compounds with antidiabetic potential; however, comparative evaluation of their interaction with glucose transporter type 4 (GLUT4) remains limited.

A significant proportion of individuals who are heavily exposed to infectious tuberculosis patients do not acquire Mycobacterium tuberculosis (Mtb) infection, as detected by an interferon gamma release assay (IGRA). We examined circulating metabolite profiles and metabolic genotypes in 199 heavily exposed IGRA-negative tuberculosis household contacts in Indonesia. Based on differentially abundant metabolites, activity of several pathways including arachidonic acid, arginine and proline, glutathione, and tryptophan metabolism, correlated with a negative IGRA at three months. This study aims to reduce the incubation period (commercial sterility test) of tomato paste cans before the bacteriological inspection in quality control laboratories. 62 cans of tomato paste were incubated under temperatures (mesophilic 25,37°C and thermophilic 55°C) to accelerate potential spoilage. Chemical indicators(pH, titratable acidity, and lactic acid) and microbial indicators (Total plate count and yeast/molds) were monitored daily to identify the earliest reliable spoilage signals. Incubation at elevated temperatures accelerated acid buildup. D+L lactic acid levels measured using a rapid enzyme assay increased with titratable acidity, while pH decreased. Background: Diabetes mellitus is a progressive metabolic condition marked by hyperglycemia due to irregularities in insulin production or activity, leading to poor carbohydrate, protein, and fat metabolism. This study aimed to evaluate the relationship between exposure to environmental pollutants, particularly benzo[a]anthracins (BaA), and the severity of the inflammatory response and liver dysfunction in COVID-19 patients, using a combination of statistical analysis and molecular modeling. Molecular docking studies revealed stable and robust interactions between benzo[a]anthracins and cytochrome P450 (CYP450) enzymes, suggesting that these interactions may contribute to increased oxidative stress and hepatocyte damage. The statistical analysis included 70 COVID-19 patients, stratified by benzo[a] anthracene exposure. Allopurinol is a well-established xanthine oxidase inhibitor widely used in the management of hyperuricemia, gout, tumor lysis syndrome, and uric acid nephrolithiasis. This review provides a comprehensive overview of its pharmacology, mechanism of action, pharmacokinetics, clinical efficacy, safety profile, and emerging insights from recent clinical and translational studies. Despite being in clinical use for over five decades, allopurinol remains the first-line urate-lowering therapy worldwide due to its proven efficacy, favorable safety profile, and cost-effectiveness. This honors thesis examines the biochemical, ethical, and public health consequences of insufficient post-operative follow-up care for undocumented immigrants injured in border falls. Discussing pathways of inflammation resolution, wound healing, and bone remodeling, this thesis argues that recovery depends on tightly regulated molecular and cellular processes that are highly vulnerable to disruption without continued monitoring and rehabilitation (Loi et al., 2016; Maruyama et al., 2020). Meningiomas are the most common primary central nervous system tumors, and while most are benign, atypical and anaplastic variants frequently recur and have limited effective treatment options. Recent trials of PD-1 blockade in unselected meningioma cohorts have demonstrated modest activity, with low objective response rates. These findings highlight the need to identify molecular biomarkers that may predict immunotherapy sensitivity in this disease. MCU, originally known as CCDC109A, is widely recognized as the gene responsible for encoding a pore-forming subunit of a Ca2+-selective channel, mitochondrial Ca2+ uniporter complex (mtCUC). While MCU expression is typically highly mitochondrial-specific, we report here a protein variant derived from the MCU gene, termed MCU-S, which lacks the mitochondria-targeting sequence (MTS) and forms a Ca2+-permeable channel outside of mitochondria. The mRNA of MCU-S was ubiquitously expressed in all cell types/tissues tested, with particularly high expression in human platelets. MCU-S protein formed Ca2+ channels at the plasma membrane, which exhibited similar channel properties to those observed in mtCUC. MCU-S channels …

The purpose of this qualitative descriptive phenomenological study was to understand how mentorship assisted a new Emergency Medical Services (EMS) clinician in navigating the challenges of the first year at an EMS agency in Central Texas. Guiding the study was Lev Vygotsky’s theory of social constructivism, which focuses on knowledge gained through human activities, social interactions, and behavior modeling. Early-onset colorectal cancer (EOCRC) has increased among adults younger than 50 years, raising concerns regarding risk identification and screening practices in younger populations. This study examined the association between colorectal adenomas and EOCRC among adults aged 20–44 years in the United States and evaluated whether this relationship was influenced by selected demographic and behavioral factors. A cross-sectional analytic design was used to examine medical claims data from the Optum Partner Health database. The study population included 186,790 individuals aged 20–44 years who underwent colonoscopy procedures. Demographic variables included age group, sex, race/ethnicity, and body mass index (BMI). Background: Bipolar Disorder (BD) is a severe mental illness, often emerging during adolescence, that is associated with significant psychosocial impairment. BD risk is elevated in offspring of affected parents, particularly those with antecedent symptoms (e.g., mood lability, subthreshold manic symptoms). However, robust, replicable brain-based markers of risk in this population are lacking. We assessed the degree to which the Regional Vulnerability Index for BD (RVI-BD), quantified as an individual's brain-wide similarity to BD patterns derived from meta-analytic studies, is elevated in youth at familial risk and correlates with symptom predictors of BD.

This mixed methods hermeneutic phenomenological study focused on an investigation of the perceptions of missile operations alert duty and the effects on health and sleep hygiene among Air Force missile operations officers. Questionnaires, interviews, and surveys were all used to gather data on subjects’ experiences and perspectives, including methods to mitigate the effects of poor sleep. Driving questions of research included how missile alert schedules affect sleep habits, as well as how missileers perceived alert duties may impact their health. Electronic cigarette use among university students is a growing public health concern, with approximately 20-25% reporting current use. Despite extensive research on vaping initiation, factors associated with vaping frequency among established users remain understudied. This study examined stress-coping mechanisms, peer influence, and health risk perceptions as predictors of vaping frequency, guided by Bandura's Social Cognitive Theory (SCT). Cisplatin (DDP)-based chemotherapy serves as a foundational cornerstone in the clinical management of esophageal squamous cell carcinoma (ESCC). However, intrinsic or acquired resistance to DDP persists as a significant clinical challenge, severely undermining therapeutic efficacy and contributing to unfavorable patient prognoses. Through functional screening with a CRISPR/Cas9 knockout library, transcriptomic profiling of cisplatin-resistant cell lines and clinical specimens, and in vitro/in vivo validation assays, we identified PHKG2 as a key mediator of cisplatin resistance in ESCC. Mechanistically, PHKG2-mediated cisplatin resistance is driven by the phosphorylation of IGF2BP3 at residues T225 and T306. Local dendritic computations are thought to critically influence neuronal signaling and plasticity yet remain largely unexplored in vivo due to challenges in stably imaging small structures at ultrafast timescales. We developed a 3D real-time motion correction platform for movement-stabilized, ultrafast two-photon voltage imaging. By co-labeling CA1 pyramidal neurons with voltage and calcium indicators, we simultaneously measured somato-dendritic and electro-calcium coupling at multiple dendritic sites. We characterized isolated dendritic spikes and distance-dependent backpropagation of naturally occurring and photostimulation-evoked bursts and single spikes. The aim of this study was to investigate hematological and biochemical alterations associated with gram-negative bacterial lower urinary tract infections (UTIs) in dogs and cats. For this purpose, 56 dogs and 36 cats were examined between October 2022 and May 2023. Twenty-eight dogs and twenty-two cats with clinical signs of lower UTIs were confirmed using ultrasonography, urinalysis, hematobiochemical analysis, and molecular identification of recovered pathogen. In dogs, UTIs were significantly associated with pyuria (82%), crystalluria (85.7%), and proteinuria. The intratumoral microbiome is increasingly recognized as a regulator of cancer biology, yet sex-specific patterns and their relevance to cancer disparities remain poorly understood. We perform a multi-kingdom analysis of more than 5,000 tumors from seven datasets to identify sex-differential microbial taxa across aerodigestive and gastrointestinal cancers. We identify and validate 22 taxa with consistent sex-biased abundance, including in real-world cohort. These microbes show cancer-type- and microbe-specific associations with tumor transcriptomes, oncogenic pathways, and immune cell infiltration. Female-enriched microbes are linked to increased estrogen signaling and interferon responses, whereas male-enriched taxa show opposing patterns. In gastric cancer, intratumoral Epstein-Barr virus …
