Understanding The Role Of Toxs As A Bile Sensing Component Of The Toxrs Virulence Regulatory System In Vibrio Spp., Minje Kim
Dartmouth College Ph.D Dissertations
Pathogenic Vibrio bacteria, such as V. cholerae and V. parahaemolyticus, colonize in the human small intestine to cause severe gastrointestinal disease. During the infection, these bacteria encounter bile salts, antimicrobial cholesterol metabolites secreted into the intestine. Pathogenic Vibrio species have evolved to utilize bile salts as signals to regulate virulence. The signaling depends in part on ToxRS, a conserved co-component transmembrane transcription regulator. ToxRS consists of the transcription factor ToxR and its membrane-tethered binding partner ToxS. ToxS is required for ToxR stability and full transcriptional activity. Although bile salts are known to influence ToxRS-dependent virulence gene expression, the molecular …

Division of Pulmonary, Allergy, and Critical Care Medicine Faculty Papers
BACKGROUND: Numerous in vivo studies have demonstrated beta-2-adrenoceptor (β2AR) -agonism as permissive in the development of allergic lung inflammation, and have implicated the arrestin-dependent signaling arm of the β2AR in mediating this effect. However, the specific cell type(s) mediating β2AR regulation of allergic lung inflammation remain unestablished.
METHODS: To explore the potential contribution of airway epithelia in this phenomenon, we compared the ability of ractopamine (RP), recently identified as a Gs-biased beta-agonist, to that of the unbiased/balanced beta-agonist albuterol (ALB), on IL-13-stimulated mucin and cytokine production in human airway epithelia cultures in air-liquid interface (HAE).
RESULTS: ALB, which activates both …

Challenging Myths In Speech-Language Pathology: A Scientific Review Of Augmentative And Alternative Communication (Aac) Use, Bilingualism, And Late Talking, Mariah Morris
Undergraduate Honors Capstone Projects
Persistent myths within the field of speech-language pathology continue to influence clinical decision-making and contribute to biases experienced by both providers and clients. This scientific review examines common misconceptions surrounding late talking, bilingualism, and augmentative and alternative communication (AAC) by evaluating current evidence-based research. Specifically, this review addresses three central questions: Do children outgrow late talking? Does bilingualism cause language delay in children? Does AAC hinder speech development?
An analysis of multiple peer-reviewed studies revealed little statistical support for the belief that bilingualism or AAC use causes language delays. Instead, research indicates that factors such as limited access to language-rich …
Determining Zap-70 Signaling Pathways Mediated By Ccr7 In Tcr T Cell Activation, Vivian Torres
Determining Zap-70 Signaling Pathways Mediated By Ccr7 In Tcr T Cell Activation, Vivian Torres
Open Access Theses & Dissertations
Activation of T lymphocytes is the main event that occurs in the adaptive immune response after engagement of the T cell receptor (TCR) with the peptide of the major histocompatibility complex (pMHC) molecules that are demonstrated by antigen presenting cells [1]. A key step in this process is the recruitment and activation of the zeta chain associated protein of 70 kDa (ZAP-70). ZAP-70 couples TCR engagement to downstream intracellular signaling pathways through phosphorylation of tyrosine residues within intracellular tyrosine activation motifs (ITAMs) [2]. Although there have been extensive studies on the signaling response through TCR activation, the regulatory influence of …
Molecular Mechanisms Of Pfas Induced Protein Dysfunction: Implications For Neurodegenerative Risk, Daisy Lee Wilson
Molecular Mechanisms Of Pfas Induced Protein Dysfunction: Implications For Neurodegenerative Risk, Daisy Lee Wilson
Open Access Theses & Dissertations
Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants with widespread human exposure and well-established links to adverse health outcomes. However, despite increasing epidemiological and experimental evidence, the molecular mechanisms underlying PFAS-induced neurotoxicity remain incompletely defined. This dissertation investigates PFAS toxicity across biological scales, integrating protein biophysics with neuronal cell models to elucidate pathways relevant to neurodegeneration. At the molecular level, we demonstrate that PFAS disrupts the structure and function of globular proteins, including the impairment of α-lactalbumin's calcium-binding capacity and of β-lactoglobulin's retinol binding, through concentration-dependent conformational destabilization. By applying these findings to a neuronal model, we show that …

Investigating The Influence Of Dietary Phytogenics And Postbiotics On Horizontal Transmission Of Histomonas Meleagridis In Commercial Turkey Poult Hens, Diego Fernando Cortes Velez
Graduate Theses and Dissertations
Histomonas meleagridis is a parabasalid protozoan and the causative agent of histomonosis, or blackhead disease, in turkeys. Withdrawal of the last remaining in-feed arsenical from the market in 2015 led to the re-emergence of the disease. As a result, there is a need to investigate alternatives, such as dietary inclusion of postbiotic and phytogenic compounds, to reduce H. meleagridis transmission and disease severity in commercial turkey flocks. The purpose of this project was to evaluate the influence of dietary inclusion of three different combinations of postbiotic and phytogenic compounds on H. meleagridis transmission in turkey poult hens. Phase-change nanodroplets (PCNDs) continue to generate significant research interest due to their potential to extravasate into tissue, to be targeted for molecular imaging and drug delivery, and to undergo an induced phase-change to "activated" microbubbles (MBs) for ultrasound (US) imaging. To accurately quantify molecular markers, however, one assumes a consistent proportion of PCNDs in a region of interest (ROI) are stably activated and imaged. Herein we present a framework for developing a diagnostic sequence that is optimized for PCND activation uniformity, contrast, and acquisition time. To develop this framework, activation was examined at three scales of increasing complexity: single, adjacent, …

Personality Characteristics Of Emergency Nurses And Their Subjective Fit In The Emergency Department, Jose Michael Maria
Personality Characteristics Of Emergency Nurses And Their Subjective Fit In The Emergency Department, Jose Michael Maria
Theses & Dissertations
Nursing workforce challenges in the United States persist despite widespread recruitment and retention efforts, and the COVID-19 pandemic has further exacerbated shortages, turnover, and workforce instability. Despite interventions such as residency programs, financial incentives, and organizational initiatives, long-term retention remains inconsistent, particularly in high-demand settings like emergency departments. Ongoing issues such as overcrowding, rising patient acuity, and workforce attrition contribute to burnout, compassion fatigue, and compromised patient outcomes, perpetuating a cycle of staffing strain. Grounded in Person–Environment Fit theory, this research proposes that aligning emergency nurses’ personality traits with the specialty practice environment, the emergency department, may offer a more …
Optimisation Of Photosensitive Recording Materials For Broadband Holographic Optical Elements, Michael Murray
Optimisation Of Photosensitive Recording Materials For Broadband Holographic Optical Elements, Michael Murray
Doctoral
The introduction of broadband (white) LED outdoor lighting has led to significant energy savings. However, this has come at the cost of increased light pollution which has negative impacts both on ecological systems and human health. This light pollution is largely the result of the lack of control measures for the directionality of the light emitted by outdoor LED lighting. The lack of directionality also results in higher energy consumption in order to compensate for the light scattered to the atmosphere and sufficiently illuminate the target area. In this thesis holographic optical elements (HOEs) are proposed as a complementary technology …
