Overlap Bias In The Case-Crossover Design, With Application To Air Pollution Exposures, Holly Janes, Lianne Sheppard, Thomas Lumley

UW Biostatistics Working Paper Series

The case-crossover design uses cases only, and compares exposures just prior to the event times to exposures at comparable control, or “referent” times, in order to assess the effect of short-term exposure on the risk of a rare event. It has commonly been used to study the effect of air pollution on the risk of various adverse health events. Proper selection of referents is crucial, especially with air pollution exposures, which are shared, highly seasonal, and often have a long term time trend. Hence, careful referent selection is important to control for time-varying confounders, and in order to ensure that …

Calcium And Glycolysis Mediate Multiple Bursting Modes In Pancreatic Islets, Richard Bertram, Leslie S. Satin, Min Zhang, Paul Smolen, Arthur Sherman Jan 2004

Calcium And Glycolysis Mediate Multiple Bursting Modes In Pancreatic Islets, Richard Bertram, Leslie S. Satin, Min Zhang, Paul Smolen, Arthur Sherman

Pharmacology and Toxicology Publications

Abstract

Pancreatic islets of Langerhans produce bursts of electrical activity when exposed to stimulatory glucose levels. These bursts often have a regular repeating pattern, with a period of 10–60 s. In some cases, however, the bursts are episodic, clustered into bursts of bursts, which we call compound bursting. Consistent with this are recordings of free Ca2+ concentration, oxygen consumption, mitochondrial membrane potential, and intraislet glucose levels that exhibit very slow oscillations, with faster oscillations superimposed. We describe a new mathematical model of the pancreatic β-cell that can account for these multimodal patterns. The model includes the feedback of …

Aminoglycosides For Intra-Abdominal Infection: Equal To The Challenge?, John A. Bailey, Katherine S. Virgo, Joseph T. Dipiro, Avery B. Nathens, Robert G. Sawyer, John E. Mazuski

Publications from the Office of the Dean

Background: Aminoglycosides, combined with antianaerobic agents, have been used widely for the treatment of intra-abdominal infection. However, some prospective randomized controlled trials and other data suggested that aminoglycosides were less efficacious than newer comparators for the treatment of these infections. We therefore performed a meta-analysis of all prospective randomized controlled trials utilizing aminoglycosides to reevaluate the efficacy of these agents for the treatment of intra-abdominal infection.

Methods: Published English-language prospective randomized controlled trials comparing aminoglycosides with other agents for treatment of intra-abdominal infection were identified by MEDLINE search. For each study, data were collected regarding the number of …

I Don't Care If I Ever Get Back: Marathons Lasting 20 Or More Innings, Phil Lowry, Darren B. Glass

Math Faculty Publications

This article looks at marathon games of baseball. For purposes of this article, a marathon is defined as a game lasting 20 or more innings. In my research I have discovered 341 marathons. These games are hard to find. Leagues either keep no records, or keep track only of their longest game; only the Texas League keeps records on all marathons. Nobody has ever before explored such questions as: What is the probability a game will go x number of innings? How often should we expect a marathon of 20 or more innings, or 40 or more innings? What is …

Medical Training Using Simulation: Toward Fewer Animals And Safer Patients, Jonathan Balcombe Jan 2004

Medical Training Using Simulation: Toward Fewer Animals And Safer Patients, Jonathan Balcombe

Experimentation Collection

This paper presents the current status of computer-based simulation in medicine. Recent technological advances have enabled this field to emerge from esoteric explorations in academic laboratories to commercially available simulators designed to train users to perform medical procedures from start to finish. Today, more than a dozen companies are producing virtual reality simulators and interactive manikins for training in endoscopy, laparoscopy, anaesthesia, trauma management, angiography, and needle insertion. For many of these procedures, thousands of animals are still being used in training. Yet simulation has many advantages that can transcend scientific, ethical, economic and logistical problems that arise when using …

Graphical Technique For Identifying A Monotonic Variance Stabilizing Transformation For Absolute Gene Intensity Signals, Kellie J. Archer, Catherine I. Dumur, Viswanathan Ramakrishnan Jan 2004

Graphical Technique For Identifying A Monotonic Variance Stabilizing Transformation For Absolute Gene Intensity Signals, Kellie J. Archer, Catherine I. Dumur, Viswanathan Ramakrishnan

Biostatistics Publications

Background

The usefulness of log2 transformation for cDNA microarray data has led to its widespread application to Affymetrix data. For Affymetrix data, where absolute intensities are indicative of number of transcripts, there is a systematic relationship between variance and magnitude of measurements. Application of the log2 transformation expands the scale of genes with low intensities while compressing the scale of genes with higher intensities thus reversing the mean by variance relationship. The usefulness of these transformations needs to be examined.

Results

Using an Affymetrix GeneChip® dataset, problems associated with applying the log2 transformation to absolute intensity …

When The Advantaged Become Disadvantaged: Men’S And Women’S Actions Against Gender Discrimination, Mindi D. Foster, Stacey Arnt, Jill Honloka Jan 2004

When The Advantaged Become Disadvantaged: Men’S And Women’S Actions Against Gender Discrimination, Mindi D. Foster, Stacey Arnt, Jill Honloka

Psychology Faculty Publications

Intergroup theories suggest that different social identities will either discourage or encourage the taking of action against discrimination (Bartky, 1977; Jost & Banaji, 1994). However, research (e.g., Branscombe, 1998) has shown that discrimination is a less negative experience for men than for women. As such, it is possible that men may take greater action than women, regardless of identity. However, men’s responses to their perceived disadvantage has not yet been tested. Among those induced to ascribe to a gendered stereotype identity, men endorsed more action than women did.Among those induced to ascribe to an identity based on a gendered social …

Translation Research: From Accurate Diagnosis To Appropriate Treatment, Craig P. Webb, Harvey I. Pass

Wayne State University Associated BioMed Central Scholarship

Abstract

This review article focuses on the various aspects of translational research, where research on human subjects can ultimately enhance the diagnosis and treatment of future patients. While we will use specific examples relating to the asbestos related cancer mesothelioma, it should be stressed that the general approach outlined throughout this review is readily applicable to other diseases with an underlying molecular basis. Through the integration of molecular-based technologies, systematic tissue procurement and medical informatics, we now have the ability to identify clinically applicable "genotype"-"phenotype" associations across cohorts of patients that can rapidly be translated into useful diagnostic and treatment …

Meta-Analysis, Spatial, And Temporal Models To Analyze The Possible Association Between Childhood Leukemia In Proximity To Nuclear Facilities, Peter Baker

MUSC Theses and Dissertations

The purpose of this dissertation was to gain a better understanding of the possible association between childhood leukemia and nuclear facilities by conducting a meta-analysis and by the use of spatial and temporal models with respect to Pickering Nuclear Generator (PNG) in Ontario, Canada. The meta-analysis was conducted to combine and statistically analyze the many studies of childhood leukemia in the vicinity of nuclear facilities. Our focus was on studies that calculated SMRs or SIRs for individual nuclear sites. Due to variability between studies in defining age and geographic zones, eight separate analyses were performed based on age and zone …

Response Of Dark-Adapted Retinal Rod Photoreceptors, H. Khanal, V. Alexiades, E. Dibenedetto

Publications

The process of phototransduction, whereby light is converted into an electrical response, in rod and cone photoreceptors in the retina, involves as a key setp, the diffusion of the cytoplasmic, signaling molecules cGMP (cyclic guanosime monophosphate) and Ca2+ diffuse in the cytoplasm (the fluid surrounding the discs). the complex geometry of the rod creates computational difficulties. We present spatio-temporal compuational models for interacctions and diffusion of cGMP and Ca2+ in the cytoplasm of vertebrate rod photoreceptors, as well as numerical simulations fo the response to light of dark-adapted Salamander rods.

Development Of Dose Coefficients For Radionuclides Produced In Spallation Targets, Phillip W. Patton, Mark Rudin Jan 2004

Development Of Dose Coefficients For Radionuclides Produced In Spallation Targets, Phillip W. Patton, Mark Rudin

Transmutation Sciences Physics (TRP)

Dose coefficients permit simple determination of radiation dose associated with various exposure scenarios, and ultimately permit radiation safety personnel to assess the health risks to workers in a nuclear facility. Specifically, radiation safety personnel use dose coefficients to determine the radiation dose incurred to a tissue or organ system from a given exposure. These parameters are often expressed in terms of Annual Limits on Intake (ALIs) and Derived Air Concentrations (DACs).

Results from this study will be used to produce ALIs and DACs for these rare radionuclides created by spallation target systems that are not included in Federal Guidance Report …

The Mare Reproductive Loss Syndrome And The Eastern Tent Caterpillar Ii: A Toxicokinetic/Clinical Evaluation And A Proposed Pathogenesis: Septic Penetrating Setae, Thomas Tobin, J. Daniel Harkins, John Ford Roberts, Patricia W. Vanmeter, Tara A. Fuller Jan 2004

The Mare Reproductive Loss Syndrome And The Eastern Tent Caterpillar Ii: A Toxicokinetic/Clinical Evaluation And A Proposed Pathogenesis: Septic Penetrating Setae, Thomas Tobin, J. Daniel Harkins, John Ford Roberts, Patricia W. Vanmeter, Tara A. Fuller

Maxwell H. Gluck Equine Research Center Faculty Publications

Reviewing the mare reproductive loss syndrome (MRLS), it is proposed that the fundamental mechanism of this syndrome, which includes early fetal loss, late fetal loss, uveitis, pericarditis, and encephalitis, is tissue penetration by septic barbed setal fragments (septic penetrating setae) from Eastern tent caterpillars (Malacosoma americanum). Once ingested, these barbed setal fragments migrate through moving tissues, followed by rapid hematogenous spread of bacteria, bacterial emboli, and/or septic fragments of setae (septic penetrating setal emboli), collectively referred to as septic materials. Pathogenic bacteria, therefore, enter the horse as hitchhikers on or in the caterpillar setal fragments, and MRLS is caused by …

Long-Range Trends In Adult Mortality: Models And Projection Methods, John Bongaarts Jan 2004

Long-Range Trends In Adult Mortality: Models And Projection Methods, John Bongaarts

Poverty, Gender, and Youth

This Population Council working paper has two objectives: (1) to test a new version of the logistic model for the pattern of change over time in age-specific adult mortality rates, and (2) to develop a new method for projecting future trends in adult mortality. A test of the goodness-of-fit of the logistic model for the force of mortality indicates that its slope parameter is nearly constant over time. This finding suggests a variant of the model that is called the shifting logistic model. A new projection method based on the shifting mortality model is proposed and compared with the widely …

Incremental Genetic K-Means Algorithm And Its Application In Gene Expression Data Analysis, Yi Lu, Shiyong Lu, Farshad Fotouhi, Youping Deng, Susan J. Brown

Wayne State University Associated BioMed Central Scholarship

Abstract

Background

In recent years, clustering algorithms have been effectively applied in molecular biology for gene expression data analysis. With the help of clustering algorithms such as K-means, hierarchical clustering, SOM, etc, genes are partitioned into groups based on the similarity between their expression profiles. In this way, functionally related genes are identified. As the amount of laboratory data in molecular biology grows exponentially each year due to advanced technologies such as Microarray, new efficient and effective methods for clustering must be developed to process this growing amount of biological data.

Results

In this paper, we propose a new clustering …
