Over a decade of advances in Clustered Regularly Interspersed Short Palindromic Repeats (CRISPR) and CRISPR-associated protein 9 (Cas9)-based technologies have culminated in the first-ever FDA-approved CRISPR/Cas-based therapy. Aside from this approved therapy for sickle cell anemia, several CRISPR/Cas-based therapies are currently under development or testing for a range of chronic diseases, including viral diseases like human immunodeficiency virus type 1 (HIV-1) infection, genetic diseases like familial hypercholesterolemia, and cancer. PURPOSE: To provide a descriptive summary and to quantitatively examine racial and ethnic disparities in the use of continuous glucose monitoring (CGM) and continuous subcutaneous insulin infusion (CSII) among people living with diabetes in the United States.
RESEARCH DESIGN AND METHODS: We followed the Cochrane and PRISMA guidelines (PROSPERO: CRD420251124678). Search was conducted on PubMed, Embase, and Cochrane up to August 2025. Background: Graves' disease is an autoimmune disorder that affects the thyroid gland, a small butterfly-shaped organ located in the anterior of the neck. In this case, the immune system mistakenly attacks the thyroid, leading to the overproduction of thyroid hormones. The most common result of Graves' disease is an overactive thyroid gland, which produces an excess of thyroid hormones (thyroxine or T4 and triiodothyronine or T3). This can lead to symptoms such as weight loss, rapid heartbeat, anxiety, irritability, heat intolerance, and increased sweating. Endophytic actinobacteria are a diverse group of Gram-positive bacteria with high G+C content, which inhabit plants’ internal tissues, and represent a significant yet underexplored reservoir of bioactive secondary metabolites. In host plants, a remarkable repertoire of metabolites, including broad-spectrum antibiotics, anticancer, antiviral, antifungal, antioxidant, and immunomodulatory compounds, is synthesized by these microbial allies, carrying significant medicinal and agricultural potential. Present study was conducted to evaluate the various liver enzymes in experimentally induced enterotoxemic goat inorder to ensure the therapeutic potential of probiotics and antibiotics. Background: Bovine babesiosis is a protozoan disease that is transmitted by ticks and has significant veterinary and economic impacts on tropical livestock systems. Prior to this study, no epidemiological data was available of Sindh, Pakistan.
Purpose: To estimate the Babesia spp. prevalence and estimate the risk factors with respect to cattle and buffaloes in five tehsils of District Naushahro Feroze, Sindh, Pakistan.

Chlamydia trachomatis is one of the most common sexually transmitted infections, often asymptomatic, yet a major cause of infertility. In clinical practice, vaginal discharge and cervical ectropion are frequently used as syndromic indicators. The complement system is a central component of innate immunity that coordinates host defense, immune surveillance, and inflammatory responses through tightly regulated proteolytic cascades. Genetic variation within complement genes contributes substantially to interindividual differences in complement activity and disease susceptibility. While coding variants directly alter complement protein structure and function, the microRNA (miRNA)-mediated control and post-transcriptional regulation is not fully understood in shaping complement gene expression across immune and inflammatory mechanisms. Women do not always have the same opportunities as men, particularly in cultures where more traditional gender norms are embraced. Male allyship can positively impact women’s lives by increasing opportunities (e.g., safety, education attainment, career development, financial security). In addition, understanding and embracing gender differences can optimize women’s participation and leadership contributions. By intentionally removing barriers and encouraging full participation, women’s talents and perspectives can be leveraged. This both expands opportunities for women and enhances innovation, improves decision-making, and contributes to stronger communities.

Every year, over 700,000 people, particularly children under five, die from vector-borne diseases worldwide. Effectively controlling endemics and preventing new outbreaks requires an integrated approach that can lead to the elimination of both vectors and diseases. In the last two decades, integrating medical interventions and vector control has significantly reduced the incidence of Gambian Human African Trypanosomiasis (g-HAT), with the World Health Organization validating eight countries as having eliminated the disease as a public health problem. Introduction: Healthcare professionals often implement quality improvement (QI) initiatives but lack training to implement and sustain QI initiatives. Leading Healthcare Improvement (LHI) is a course that combines principles from change management, team science, and leadership effectiveness into an implementation strategy to enable clinicians to lead sustainable change.
Methods: LHI is a 6-session, 4-month interactive training. Learners work on an active QI project throughout the course. A Wilcoxon signed-rank test was conducted to evaluate pre- and post-course changes in learners' self-reported confidence across five QI competencies.

BACKGROUND: Early identification of worsening heart failure (HF) may improve outcomes.
OBJECTIVES: This study aims to assess if insertable cardiac monitor (ICM)-based high-risk detection combined with centrally managed, nurse-facilitated, individually protocolized diuretic interventions is safe and improves HF outcomes.
METHODS: A Reveal LINQ (Medtronic) ICM with an investigational HF risk-status software was implanted in participants with HF who were randomized 1:1 to an intervention arm (high-risk HF alert triggering protocolized diuretic regimen) or an observation arm (standard care). Adolescent overweight/obesity is an increasing public health concern and contributes to the risk of non-communicable diseases later in life. Physical activity plays an important role in weight regulation. However, its association with overweight/obesity may vary according to residential setting. This study aimed to examine the association between physical activity and overweight/obesity among adolescents living in rural and urban areas of Indonesia. A cross-sectional study was conducted using data from the 2023 Indonesian Health Survey. The study included 115,372 adolescents aged 10–18 years. Despite advances in therapies, cardiogenic shock (CS) remains associated with high mortality rates, often necessitating mechanical circulatory support (MCS) to preserve end-organ perfusion. Despite increasing rates of MCS utilization, less is known about sex-related differences in clinical outcomes to guide practice. Additionally, standardized management protocols for CS patients requiring MCS that account for sex differences are lacking. BACKGROUND: Arrhythmia burden in ambulatory patients with symptomatic heart failure (HF) without cardiac implantable electronic devices (CIEDs) is not well defined, and it remains uncertain whether device-guided remote congestion management modifies arrhythmia occurrence.

Background: The preferred treatment for hypogonadism, testosterone replacement therapy (TRT), has a varying efficacy that is currently not fully understood. This observation may be explained by polymorphism of the androgen receptor (AR), a ligand-binding transcription factor whose polyglutamine tract length affects various health conditions and androgen sensitivity. It has been theorized that this polymorphism may also affect the severity of hypogonadism. This study sought to determine a correlation between hypogonadism severity, TRT efficacy, and number of CAG trinucleotide repeats in the AR gene.

Sinonasal squamous cell carcinoma (SNSCC) comprises a heterogeneous group of tumors with emerging molecular subtypes. Human papillomavirus (HPV)-associated SNSCC is a distinct entity, although actionable genomic alterations beyond HPV-driven pathways remain underexplored. Distinguishing HPV-associated SNSCC from carcinoma ex inverted papilloma can be particularly challenging in limited biopsy specimens, as these lesions may appear morphologically similar. We report a case of HPV-associated SNSCC with concurrent FGFR3::TACC3 fusion and NOTCH1 p.C1416 stop-gain mutation.

Intraductal carcinoma (IDC) of the salivary gland is an uncommon epithelial neoplasm characterized by a predominantly intraductal or intracystic proliferation rimmed by a myoepithelial cell layer. Advances in molecular profiling have revealed recurrent gene fusions in IDC, most frequently involving RET, although fusions implicating other kinase genes have also been described. Among these, ALK rearrangements are rare and remain poorly characterized. Herein, we report a case of oncocytic IDC of the parotid gland harboring a STRN::ALK fusion, further expanding the molecular landscape of this tumor. Static and dynamic balance are vital for performance and injury prevention in roller skate athletes. While proprioceptive and strengthening exercises are extensively used, evidence of their combined impact in young athletes is rare. This study evaluated the effects of an eight-week proprioceptive and strengthening program on postural and lower-limb balance parameters in roller skaters aged 11–15 years. The study randomly assigned 30 athletes to an intervention group (n=15) or a control group (n=15). This research aimed to analogize the resistance patterns against carbapenem antibiotics in Enterobacterales uropathogens between two healthcare facilities. The authors performed this study at the Basic Medical Sciences Institute, Jinnah Postgraduate Medical Center, and the National Medical Center. A total of 374 UTI patients were enrolled, equally distributed in the two centers (187 each hospital). Molecular assays supplemented phenotypic susceptibility testing to detect carbapenem resistance. Statistical analysis, including multivariate logistic regression, was performed to identify associations between hospital type and carbapenem resistance. More than half of caregivers of children with chronic kidney disease (CKD) report experiencing moderate depression (53.3%), while 30% and 16.7% report mild and severe depression, respectively. As primary caregivers, mothers require strong self-efficacy to manage psychological burden associated with caring for children with CKD. Self-hypnosis may enhance self-efficacy and alleviate anxiety and depression in this population.
