Loss Of Αt-Catenin Alters The Hybrid Adhering Junctions In The Heart And Leads To Dilated Cardiomyopathy And Ventricular Arrhythmia Following Acute Ischemia., Jifen Li, Steven Goossens, Jolanda Van Hengel, Erhe Gao, Lan Cheng, Koen Tyberghein, Xiying Shang, Riet De Rycke, Frans Van Roy, Glenn L Radice Feb 2012

Loss Of Αt-Catenin Alters The Hybrid Adhering Junctions In The Heart And Leads To Dilated Cardiomyopathy And Ventricular Arrhythmia Following Acute Ischemia., Jifen Li, Steven Goossens, Jolanda Van Hengel, Erhe Gao, Lan Cheng, Koen Tyberghein, Xiying Shang, Riet De Rycke, Frans Van Roy, Glenn L Radice

Center for Translational Medicine Faculty Papers

It is generally accepted that the intercalated disc (ICD) required for mechano-electrical coupling in the heart consists of three distinct junctional complexes: adherens junctions, desmosomes and gap junctions. However, recent morphological and molecular data indicate a mixing of adherens junctional and desmosomal components, resulting in a 'hybrid adhering junction' or 'area composita'. The α-catenin family member αT-catenin, part of the N-cadherin-catenin adhesion complex in the heart, is the only α-catenin that interacts with the desmosomal protein plakophilin-2 (PKP2). Thus, it has been postulated that αT-catenin might serve as a molecular integrator of the two adhesion complexes in the area composita. …

The Role Of Calcineurin In Dendritic Remodeling And Epileptogenesis In A Rat Model Of Traumatic Brain Injury, John Campbell Feb 2012

The Role Of Calcineurin In Dendritic Remodeling And Epileptogenesis In A Rat Model Of Traumatic Brain Injury, John Campbell

Theses and Dissertations

Traumatic brain injury (TBI), a leading cause of death and disability in the United States, causes potentially preventable damage in part through the dysregulation of neural calcium levels. This dysregulation likely affects the activity of the calcium-sensitive phosphatase, calcineurin, with serious implications for neural function. To test this possibility, the present study characterized the role of calcineurin in a rat model of brain trauma, the lateral fluid percussion injury model. Golgi-Cox histochemistry revealed an acute post-TBI loss and delayed overgrowth of dendritic spines on principal cortical cells. The spine loss appeared to require calcineurin activity, since administering a calcineurin inhibitor, …

Exercise Is Medicine: Some Cautionary Remarks In Principle As Well As In Practice, Ross Neville Jan 2012

Exercise Is Medicine: Some Cautionary Remarks In Principle As Well As In Practice, Ross Neville

Articles

On the basis of extensive research on the relationship between physical activity, exercise and health, as well as strong support from policymakers and practitioners, the “Exercise is Medicine” initiative has become something of a linchpin in the agenda for modern healthcare reform and reflects a broader acceptance that the philosophy of health politics must shift from social engineering to performativity. However, in spite of the avowed commitment to encouraging individuals to take on a more reflexive relation to their health, it remains unclear as to whether an initiative such as this is, unambiguously, a good thing. In this paper, a …

Considering A Complemental Model Of Health And Fitness, Ross Neville Jan 2012

Considering A Complemental Model Of Health And Fitness, Ross Neville

Articles

This article examines the concept of fitness, which, in spite of its much avowed cultural significance, has become the subject of much critical attention. In particular, it considers the now contested relation of fitness to health; the fact that, although there appears to be a clear consensus on a simple causal relation between the two, this has been deemed illusory outside the medico-scientific context of its production. In response to the problems with both of these positions, this article examines the potential for reconfiguring the relation between fitness and health on new terms. A complemental model of health and fitness …

It's A Birth Not A Procedure: An Ethnographic Study Of Intrauterine Fetal Death In A Labor And Delivery Unit Of An American Hospital Setting, Catherine Mcleod Griffin Jan 2012

It's A Birth Not A Procedure: An Ethnographic Study Of Intrauterine Fetal Death In A Labor And Delivery Unit Of An American Hospital Setting, Catherine Mcleod Griffin

Wayne State University Dissertations

Life transitions such as birth and death constitute a significant area within anthropological studies of ritual. It is important to investigate how individuals, groups, and communities organize around these events. Birth and death can be considered as rites of passage that mark key life transitions (van Gennep 1909/1960). Thus birth and death related rituals need to be investigated within the social and cultural context of American hospital settings to better understand the social organization of life, death, and personhood. In the American hospital setting, a reproductive loss at any gestational age receives the medical diagnostic label of an intrauterine fetal …

Cellular Plasticity In White Adipose Tissue: In Vivo Identification Of Bipotent Adipocyte Progenitors In Adult White Adipose Tissue, Yun-Hee Lee Jan 2012

Cellular Plasticity In White Adipose Tissue: In Vivo Identification Of Bipotent Adipocyte Progenitors In Adult White Adipose Tissue, Yun-Hee Lee

Wayne State University Dissertations

Nutritional and pharmacological stimuli can dramatically alter the cellular composition and phenotype of white adipose tissue (WAT). Nonetheless, the identity of progenitors that contribute to this cellular plasticity in vivo remains poorly understood. Utilizing genetic lineage tracing techniques in combination with in situ imunohistochemical analysis, we demonstrate that brown adipocytes (BA) that are induced by beta3-adrenergic receptor activation (ADRB3) in WAT arise from the proliferation and differentiation of cells that express platelet-derived growth factor receptor alpha (PDGFRA), CD34 and Sca1 (PDGFRA+ cells). PDGFRA+ cells have a unique morphology in which extended processes contact multiple cells in the tissue microenvironment. Surprisingly, …

Anthropological Data Regarding The Adaptiveness Of Hebephilia, Raymond B. Hames, Ray Blanchard

Department of Anthropology: Faculty Publications

Hebephilia is the sexual attraction to early pubertal children in Tanner Stages 2 and 3, generally ages 11 through 14.

The second author of this letter (R.B.), a member of the Paraphilias Subworkgroup of the Work Group on Sexual and Gender Identity Disorders for DSM-5, proposed a diagnostic entity for DSM-5 that resembles the ICD-10 model, but with a different name: Pedohebephilic Disorder. In the original proposal (Blanchard, 2010a), this disorder would have three subtypes: pedophilic, hebephilic, and pedohebephilic. In the current version of the proposal, which is still under consideration, the name has been changed to Pedophilic Disorder, in …

Coplanar Pcb77 And Angii Induced Vascular Disorders, Madhura Parulkar Jan 2012

Coplanar Pcb77 And Angii Induced Vascular Disorders, Madhura Parulkar

Theses and Dissertations--Nutritional Sciences

Previous studies demonstrated that coplanar PCBs promote inflammation by release of pro-inflammatory cytokines like TNF, MCP-1, and VCAM-1 from endothelial cells as well as adipocytes. Also these PCBs at small doses may contribute to the development of obesity by inducing adipocyte differentiation. Obesity is a known risk factor that promotes cardiovascular disorders like atherosclerosis and AAAs. Evidence shows Ang II, a component of the RAS, leads to the formation of atherosclerosis and AAAs in both normal as well as hyperlipidemic mice. Earlier studies in our laboratory have also shown that coplanar PCB-77 promotes atherosclerotic lesion formation in ApoE-/- mice. The …

Research outputs 2012

Problem: This paper addresses the implications of working with vulnerable groups in the context of an online health community when members move from co-presence online to co-presence offline. Theoretical Approach: The case study presented in this paper challenges the expectation that self-revelations are usually more common in online environments because of the anonymity and comfort experienced there. Methodology: Taking as its example the events in a research project designed to investigate the relative features of online communities and social network sites by using a ‘netnographic’ approach, this paper examines the introduction of live chat sessions with a view to building …

Nanocrystals Of Chemotherapeutic Agents For Cancer Theranostics: Development And In Vitro And In Vivo Evaluation, Christin P. Hollis Jan 2012

Nanocrystals Of Chemotherapeutic Agents For Cancer Theranostics: Development And In Vitro And In Vivo Evaluation, Christin P. Hollis

Theses and Dissertations--Pharmacy

The majority of pharmacologically active chemotherapeutics are poorly water soluble. Solubilization enhancement by the utilization of organic solvents often leads to adverse side effects. Nanoparticle-based cancer therapy, which is passively targeted to the tumor tissue via the enhanced permeation and retention effect, has been vastly developed in recent years. Nanocrystals, which exist as crystalline and carry nearly 100% drug loading, has been explored for delivering antineoplastic agents. Additionally, the hybrid nanocrystal concept offers a novel and simple way to integrate imaging agents into the drug crystals, enabling the achievement of theranostics. The overall objective of this dissertation is to formulate …

Because of an increase in food insecurity over the past decade, the U.S. and many developing countries are focusing efforts on issues relating to child hunger. School feeding programs have been in existence for roughly 220 years but modifications have been made over the years to improve the quality of food they serve. Little research has been conducted about adult perceptions of school feeding programs and the impact it has on the community in the U.S. as well as developing countries such as Ghana, West Africa. Because of the gap in literature on school feeding programs and adult perceptions about …

A Role For Reactive Oxygen Species In Photodynamic Therapy, Michael Price Jan 2012

A Role For Reactive Oxygen Species In Photodynamic Therapy, Michael Price

Wayne State University Dissertations

Photodynamic therapy (PDT) is based on the ability of certain photosensitizing agents to selectively localize in neoplastic cells and their vasculature. Subsequent irradiation at a wavelength corresponding to a photosensitizer absorbance band excites the photosensitizer molecules, leading to energy transfer reactions and fluorescence. It was initially concluded that the phototoxic effect occurred when energy from the excited state of the photosensitizer was transferred to dissolved oxygen to form singlet oxygen. This product has a very brief half-life and will cause cellular damage only in the immediate vicinity of its formation. But an excited-state photosensitizer can also interact with oxygen to …

School of Veterinary and Biomedical Sciences: Faculty Publications

Using a recombinant vesicular stomatitis virus encoding eGFP fused in-frame with an essential viral replication protein, the phosphoprotein P, we show that during passage in culture, the virus mutates the nucleotide C289 within eGFP of the fusion protein PeGFP to A or T, resulting in R97S/C amino acid substitution and loss of fluorescence. The resultant non-fluorescent virus exhibits increased fitness and growth advantage over its fluorescent counterpart. The growth advantage of the non-fluorescent virus appears to be due to increased transcription and replication activities of the PeGFP protein carrying the R97S/C substitution. Further, our results show that the R97S/C mutation …

Gender, Gender Roles, And Anxiety: Perceived Confirmability Of Self Report, Behavioral Avoidance, And Physiological Reactivity, Milena Stoyanova, Debra Anne Hope Jan 2012

Gender, Gender Roles, And Anxiety: Perceived Confirmability Of Self Report, Behavioral Avoidance, And Physiological Reactivity, Milena Stoyanova, Debra Anne Hope

Department of Psychology: Faculty Publications

Despite the well-documented gender effect in anxiety, less is known about contributing factors to women’s greater risk for anxiety and fears. The present study examined the relationship between gender, gender role orientation (i.e., expressivity/instrumentality) and fear of harmless insects (tarantula), using a multimodal approach of self-report measures, a Behavioral Approach Test (BAT), and physiological reactivity. Participants (144 college students; 67 women, 77 men) completed a questionnaire packet and then were instructed to approach a tarantula. We were unable to replicate Pierce and Kirkpatrick’s (1992) findings that men underreport anxiety. Consistent with the literature, women in the study experienced greater anxiety …

To promote the vaccine's release, Merck launched Gardasil's "One Less" advertising campaign that included television commercials, print ads and a consumerfocused website (www.Gardasil.com), each promoting the message that "you" could now be "one less woman" affected by cervical cancer ("One Less" campaign). The vaccine, tested and approved only for females age 9-26, was …

Suppression Of A Nonsense Mutation In A Mouse Model Of Hurler Syndrome, Dan Wang Jan 2012

Suppression Of A Nonsense Mutation In A Mouse Model Of Hurler Syndrome, Dan Wang

All ETDs from UAB

Nonsense suppression therapy utilizes compounds that suppress translation termination at disease-causing premature termination codons (PTCs) to restore full-length, functional protein synthesis. This approach may provide a treatment for many diseases caused by nonsense mutations including Hurler syndrome (mucopolysaccharidosis type I - Hurler, MPS I-H). MPS I-H is a lysosomal storage disease caused by severe &alpha-L-iduronidase deficiency and subsequent lysosomal glycosaminoglycan (GAG) accumulation. In this work, we investigated the effectiveness of nonsense suppression therapy as a treatment for MPS I-H. We first generated a targeted knock-in mouse model that carries the Idua-W392X nonsense mutation homologous to the human IDUA-W402X mutation frequently …

Motivation, Need Support And Need Satisfaction In Youth Soccer Players, Erica Lippitt

College of Graduate Studies: Theses & Dissertations

Author's abstract: Physical activity levels and obesity rates vary greatly across ethnic groups in the United States (CDC, 2011). One popular way to increase physical activity levels in youth is through youth sport participation, which may be influenced by athlete motivation. Numerous studies have determined motivation levels in youth sport participants, but very little done in comparing levels across different ethnic groups. Identifying differences in motivation levels based on ethnicity can allow coaches and sport administrators to adjust practices and program setups to increase participation and meet needs of player competence. The purpose of the study was to determine if …

Biotin Affects The Immune Response Of Piglets Inoculated With Porcine Circovirus Type 2, Chen Hong, Zhang Keying, Ding Xuemei, Chen Daiwen

Turkish Journal of Veterinary & Animal Sciences

Weanling crossbred pigs were used in a 5-week trial to evaluate the effects of biotin and porcine circovirus type 2 (PCV2) challenge on serum cytokine concentrations and humoral immunoresponse. Although not significant, there was a trend towards reduced interferon-\gamma concentration in the challenged pigs compared to the controls, especially at 14 and 21 days postinoculation (dpi). Biotin supplementation with 50 or 200 µg/kg biotin improved these levels, with higher doses producing an earlier and stronger response. Although the magnitude of the immune response was small and inconsistent, diet supplementation with 50 or 200 µg/kg biotin appeared to increase serum immunoglobulin …

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Transitional cell carcinoma (TCC) of the urinary bladder is the most common cancer of the urinary tract. Most of the TCC cases are of the superficial type and are treated with transurethral resection (TUR). However, the recurrence rate is high and the current treatments have the drawback of inducing strong systemic toxicity or cause painful cystitis. Therefore, it would be of therapeutic value to develop novel concepts and identify novel drugs for the treatment of bladder cancer. Ki-67 is a large nucleolar phosphoprotein whose expression is tightly linked to cell proliferation, and curcumin, a phytochemical derived from the rhizome Curcuma …

Mechanisms By Which Exercise Training Attenuates Blood Pressure In Animals: Roles Of Cytokines, Oxidative Stress, And Glycogen Synthase Kinase-3Â, Deepmala Agarwal

LSU Doctoral Dissertations

Hypertension is a chronic multifactorial condition with high morbidity and mortality rates, currently affecting about one billion people worldwide. Currently available anti-hypertensive medications are found to be effective in reducing blood pressure (BP), but still more than 50% of those diagnosed with hypertension fail to respond to these anti-hypertensive regimens. Although hypertension has multiple etiologies, physical inactivity has been found to have strong correlation with the disease, so exercise has recently been recommended as a part of lifestyle modifications for all hypertensive patients. Therefore, the present series of in vivo and in vitro studies were undertaken to gain more insight …
