A recombinant baculovirus encoding a single-chain murine major histocompatibility complex class I molecule in which the first three domains of H-2Kd are fused to beta 2-microglobulin (beta 2-m) via a 15-amino acid linker has been isolated and used to infect lepidopteran cells. A soluble, 391-amino acid single-chain H-2Kd (SC-Kd) molecule of 48 kDa was synthesized and glycosylated in insect cells and could be purified in the absence of detergents by affinity chromatography using the anti-H-2Kd monoclonal antibody SF1.1.1.1. We tested the ability of SC-Kd to bind antigenic peptides using a direct binding assay based on photoaffinity labeling. The photoreactive derivative …

This project was designed to determine if significant differences existed between the level of psychological differentiation, as determined by the Group Embedded Figures Test (GEFT), of individuals entering a treatment center for chemical dependency and the length of stay at the facility.
The subjects consisted of 43 white males who were seeking treatment for addiction to mood altering chemicals at a regional treatment facility in Bowling Green, Kentucky. Each subject was interviewed to collect pertinent background information pertaining to past substance abuse and was evaluated to determine psychological differentiation, utilizing the GEFT.

Evidence For The Alternative Pathway Of Complement Activation In The Nurse Shark, Lieneke Cecile Culbreath

Complement is activated via two pathways: classical (CCP) and alternative (ACP). The CCP has been demonstrated in the nurse shark. The ACP has not been demonstrated in any cartilagenous fish. Nurse shark serum was evaluated for complement activity by its ability to lyse heterologous erythrocytes. As CCP activity requires calcium and magnesium, activity of shark serum chelated with EGTA (a selective calcium chelator) or EDTA (a chelator of calcium and magnesium) was assessed. Activity remained in serum chelated with EGTA but not EDTA. Furthermore, activity of chelated serum was enhanced by added magnesium. Activation of shark complement by activators of …

This study examined the relationship between chronic low back pain (CLBP) patients' perceived (stated) levels of function and their measured (observed) performance in squatting and stair climbing activities as compared to healthy volunteers. Twenty patients with CLBP and 20 healthy subjects were asked through an interview to self-assess their ability to comfortably perform stair climbing and squatting as well as other tolerances. The subjects were then asked to perform the activities and their performance levels were recorded. Results of the t-tests and Analysis of Variance (ANOVA) procedures revealed that patients' estimate of squatting and stair climbing abilities as well as …

Method And Apparatus For Detecting Microorganisms Within A Liquid Product In A Sealed Vial, Robert A. Lodder

An apparatus and method are provided for the noninvasive and nondestructive detection of microorganisms within a liquid product contained within a sealed vial. The apparatus includes a near-IR light source that produces both incident and reference beams having a wavelength between 800 and 2500 nm and, more preferably, 1100 and 1360 nm. The apparatus also includes an integrating sphere having incident and reference beam ports and a sample window opposite the incident beam port. A detector is mounted in the integrating sphere substantially adjacent the sample window. A substantially U-shaped mirror is provided to hold the vial. The U-shaped mirror …

It is usually expected that if we are overwhelmed by an episode of crisis or loss that we may temporarily cope ineffectively, whereas when weare exposed to a sequence of challenges and stressors that are managable, we learn gradually to become better able to cope with such events. The "toughness" concept relates to analogous processes at the physiological level. The concept is based upon a wide variety of research with both animals and humans that is reviewed in detail elsewhere (Dienslbier, 1989). The focus of this chapter will instead be upon how toughness influences both physiological and psychological responses to …

A comprehensive analysis of precompetition dietary beliefs and behaviors of competitive bodybuilders has yet to be documented in the literature. The purposes of this study were (a) to quantify and document types of supplements competitive male and female bodybuilders consume, (b) to document how much money they spend on them each month, (c) to describe what dietary modifications and behaviors they follow in preparation for a contest and (d) to describe their nutrition beliefs associated with these habits. Three hundred and nine competitive male and female bodybuilders completed questionnaires regarding their precompetition dietary practices. It was found that bodybuilders consume …

Pharmaceutical compositions and methods of inhibiting the enzyme elastase and increasing the biological half-life and/or potency in terms of inhibitory activity of the enzyme elastase of peptide compounds is achieved by use of a polymer of the formula P--(L--R)q wherein P is a polymer containing at least one unit of the formula (An Bn) wherein An Bn is substantially nonbiodegradable, and has an average molecular weight of about 1,000 to 5,000 daltons, L is a covalent bond or a linker group and R is a peptide.

Biodegradable Microspheres As A Carrier For Macromolecules, Patrick P. Deluca, Frantisek Rypacek

A method for preparing biodegradable microspheres having a three-dimensional network in which biologically active macromolecular agents are physically entrapped therein. The microsphere is able to degrade and release the macromolecular agent at a controlled rate. The method involves emulsifying a vinyl derivative of a biodegradable hydrophilic polymer, a water-soluble monovinyl monomer and a biologically active macromolecule in water, and copolymerizing the biodegradable hydrophilic polymer and the water-soluble monovinyl monomer such that the biologically active macromolecule is entrapped therein.

Occupational therapists treating older people with Alzheimer disease know that they must also consider the others who are affected by the disease, the informal caregivers. Intervention is most effective when it enables both the impaired person and the primary caregiver to manage the secondary symptoms of dementia. Unfortunately, little is understood about how caregivers approach and carry out their tasks and about why male and female caregivers respond differently to their caregiving role in terms of depression, burden, stress, and substance abuse. We have previously shown that the delta E3 site is an essential element for transcriptional activation by the human T-cell receptor (TCR) delta enhancer and identified two factors, NF-delta E3A and NF-delta E3C, that bound to overlapping core (TGTGGTTT) and E-box motifs within delta E3. In this study, we show that protein binding to the core motif is necessary but not sufficient for transcriptional activation by the delta E3 element. In contrast, protein binding to the E-box motif does not contribute significantly to enhancer activity. A similar core motif present within the enhancers of T-cell-tropic murine retroviruses has been shown …

Objective: To examine the literature for evidence that psychological factors predispose certain individuals to development of reflex sympathetic dystrophy (RSD).
Data Sources: English-language journal articles that described psychological data on patients diagnosed with RSD, identified through Medline search and bibliography reviews.
Study Selection: All studies reporting data on psychological factors in adult, adolescent, or child patients with RSD were included.
Data Extraction: We extracted data regarding psychological factors in patients with RSD and assessed validity of these studies through evaluation against seven basic research criteria.

Field tests of upper body muscular strength and endurance (UBMSE) are often administered to children, but little is known about the determinants of performance on these tests. Therefore the purpose of this investigation was to examine potential determinants of performance on several common field tests of UBMSE including pull-ups, flexed-arm hang, push-ups, and two types of modified pull-ups. Subjects were 56 girls, and 38 boys, ages 9 to 11 years. Potential determinants assessed were age, height, weight, gender, % fat, physical activity, and laboratory measures of muscular strength and endurance. While the history books refer to hemophilia as the disease of kings because it was once common among European royalty, the disorder now makes headlines for a completely different reason. It is estimated that at least 70% of the hemophiliacs whoo received blood products between 1978 and 1985 have been infected with HIV.

Several of the new reproductive technologies have been cultivated from our current understanding of the genetic programming and cellular processes that are involved in the major morphogenetic events of mammalian preimplantation development. Research directed at characterizing the patterns of gene expression during early development has shown that the embryo is initially under maternal control and later superseded by new transcriptional activity provided by the activation of the embryonic genome. Several embryonic transcripts encoding: (i) growth factors, (ii) cell junctions, (iii) plasma membrane ion transporters, and (iv) cell adhesion molecules have been identified as contributing directly to the progression of the …
