Short-chain fatty acids (SCFAs) produced by gut microbial fermentation influence host metabolism and neuroinflammatory processes implicated in Alzheimer’s disease (AD). However, the relationship between fecal SCFAs, microbial taxa, and cerebral amyloid-β (Aβ) burden in cognitively unimpaired individuals remains unclear. Fecal SCFAs were quantified using GC-MS, and microbial species were profiled by shotgun metagenomics in 87 participants. Associations between SCFAs, demographics, APOE ε4 status, and Aβ burden were tested using nonparametric statistics and multivariable regression. Microbial–SCFA links were evaluated using Spearman correlations and multivariate ordinations, with mediation analysis exploring potential indirect pathways. Acetate was the predominant SCFA and demonstrated the most …

Sarcopenia and obesity, characterised respectively by reduced muscle mass and function and increased adiposity, may co-occur as sarcopenic obesity (SO). This systematic review summarises cohort evidence linking these body composition phenotypes to dementia risk. MEDLINE, EMBASE, Cochrane Database and PsycINFO were searched from inception to February 2026. Outcomes included incident all-cause dementia (ACD) and subtypes. Pooled hazard ratios were calculated for each phenotype using random-effects models. Fourteen publications were included for sarcopenia, 72 for obesity, and four for SO. Late-life obesity (≥65 years) was protective for ACD (HR 0.83, 95% CI 0.76–0.89), whereas mid-life obesity (<65 years) modestly increased risk (HR 1.09, 95% CI 1.01–1.16). Sarcopenia was associated with increased risk (HR 1.42, 95% CI 1.26–1.58). SO showed no significant association (HR 1.43, 95% CI 0.87–1.99). These findings highlight age-dependent effects of obesity and an elevated dementia risk with sarcopenia; the predictive role of SO requires further research.

Background Type 2 diabetes is associated with increased Alzheimer’s disease risk and brain beta amyloid (Aβ) burden, suggesting an underlying mechanistic relationship between Alzheimer’s disease and type 2 diabetes. Animal studies show exercise reduces levels of brain Aβ and tau, and while human studies are somewhat limited, some studies have reported physical activity is associated with lower brain Aβ and tau levels. Exercise has well established links to reductions in insulin resistance; thus, as physical activity can impact both insulin resistance and Alzheimer’s disease pathology and/or biomarkers, it is reasonable to hypothesise that a mediating relationship may exist. The objective …

Alzheimer’s disease (AD), a complex neurodegenerative disorder characterised by progressive cognitive decline, affects millions globally. With no current cure, treatment focuses on symptom management and slowing disease progression. Early identification is therefore crucial. Fecal microRNA (miRNA) analysis is emerging as a promising non-invasive diagnostic tool. Evidence suggests that miRNAs present in fecal samples may regulate gene expression linked to AD pathology, impacting cellular functions and disease progression. Dietary factors critically influence gut microbiota composition and diversity, affecting brain health via the gut brain axis. Macronutrients such as carbohydrates, proteins, and fats modulate miRNA expression and gut microbiota, influencing AD risk. …

Research outputs 2022 to 2026
Previous studies have demonstrated that the immunoglobulin G (IgG) N-glycome and transcriptome are potential biochemical signatures of chronological and biological ages, and several aging clocks have been developed. By integrating the IgG N-glycome and transcriptome, we propose a novel aging clock, gtAge. We developed a deep reinforcement learning-based multiomics integration method called AlphaSnake. The results showed that AlphaSnake achieved a predicted coefficient of determination (R2) value of 0.853, outperforming the concatenation-based integration method (R2 = 0.820). The gtAge estimated by AlphaSnake explained up to 85.3% of the variance in chronological age, which was higher than that in …

Chronic alcohol consumption (CAC) is associated with an enhanced risk of bone fracture, reduced bone density, and osteoporosis. In a rhesus macaque model of voluntary ethanol consumption, CAC induces functional, transcriptomic, and epigenomic changes in hematopoietic stem and progenitor cells (HSPCs) and their resultant monocytes/macrophages, skewing them toward a hyper-inflammatory response. In the present study, those studies were extended to investigate alterations in osteoclast development, which, in postnatal life, differentiate from HSPCs and play a critical role in maintaining bone homeostasis. Spectral flow cytometry revealed a skewing of HSPCs toward granulocyte-monocyte progenitors in the CAC group, consistent with an increased …

Immunotherapies have greatly improved outcomes for patients with renal cell carcinoma (RCC), yet response rates remain suboptimal and the factors promoting therapy resistance versus sensitivity are incompletely understood. Currently, no preclinical model of orthotopic renal cancer exists that permits evaluation of tumor antigen–specific (TAS) CD8+ tumor-infiltrating lymphocytes (TILs). To address this deficiency, we developed a mouse renal cancer model that permits tracking of adoptively transferred TAS CD8+ T cells. Renca-LUC tumor cells were transduced to express tumor ERK (tERK), a model antigen expressing a one-amino-acid change from wild-type ERK, resulting in recognition by tERK/H-2Kd-specific DUC Thy1.1 …

Emerging evidence suggests altered circulating metabolite levels in individuals with Alzheimer’s disease (AD) and/or mild cognitive impairment (MCI) compared to healthy controls; how these metabolites alter with early cortical amyloidosis—before symptom onset—remains unclear. This cross-sectional study aimed to investigate plasma metabolites obtained through targeted profiling using the Biocrates AbsoluteIDQ® p400 HR kit. The study cohort comprises 100 cognitively unimpaired (CU) older adults, stratified into CU Aβ− (negative positron emission tomography [PET]-amyloid-beta [Aβ]) and CU Aβ+ (positive PET-Aβ) groups. Significantly altered metabolites were further analysed for associations across amyloid plaque deposition, cognitive performance and plasma tau phosphorylated at threonine 181 (P-tau181) …

Epigenetic changes, in particular DNA methylation, accumulate with age across different tissues, but whether these changes follow consistent patterns across different organs remains poorly understood. Here we show, through a meta-analysis of more than 15,000 human methylation profiles spanning 17 tissues, that aging produces both conserved and tissue-specific epigenetic signatures. We identify systemic shifts in methylation levels, increases in methylation variability, and growing molecular disorder across tissues. Network analysis revealed tightly connected gene clusters that are not modified by beneficial interventions, alongside a more modifiable cluster linked to NAD+ metabolism, supporting NAD+ as a potential therapeutic target in …

Background Encopresis, fecal soiling is often secondary to functional constipation and retention. It is significantly impact a child's psychological health as it can result in feelings of shame, social isolation, and low self-esteem and negatively affect their quality of life and their families. Objective To evaluate and compare the efficacy and safety of interferential stimulation (IF) applied on sigmoid colon and anorectal neuromuscular electrical stimulation (NMES), in combination with behavioral therapy and pelvic floor muscle training (PFMT), in improving continence outcomes in children with secondary encopresis refractory to laxatives. Methods In this comparative randomized controlled clinical trial, 30 children (5–10 …

Title Facial expression with the type of eye opening associates with multi-organ failure: an Observational prospective ICU cohort study Background Accurate prognostication in the intensive care unit (ICU) remains a challenging task. Conventional severity scores such as the Acute Physiology and Chronic Health Evaluation II (APACHE II) quantify physiological derangement but overlook real-time neurological and affective responsiveness. Behavioral indicators—particularly facial expressivity and eye-opening response—may reflect cortical and brainstem integrity, offering a simple, non-invasive adjunct for bedside mortality prediction. Methods In this single-center, prospective observational study, 210 adult patients in the ICU were assessed within 24 hours of admission. Two blinded …

Background Necrotizing soft tissue infections (NSTIs) are rapidly progressive infections that are dangerous to life, with high morbidity and mortality. Prompt surgical and medical management is necessary and stands mostly on precise identification as well as risk stratification. In order to enhance diagnostic as well as prognostic accuracy, prognostic scoring systems like the Laboratory Risk Indicator for Necrotizing Fasciitis (LRINEC) as well as the most recent Site, Immunosuppression, Age, Renal impairment, and inflammatory markers (SIARI) score, were introduced. Methods A prospective observational cohort study was carried out during June-August 2024 in Shebin Elkom Teaching Hospital among 60 patients who had …
