To investigate the serum AMH and various biochemical parameter levels in patients with cryptorchidism, oligospermia, or varicocele. Materials and methods: This study included 100 participants, divided into 5 groups: cryptorchidism (n = 20), varicocele (n = 20), oligospermia (n = 20), and 2 control groups [control 1, 8-12 years old (n = 20) and control 2, 18-24 years old (n= 20)]. Using the blood samples drawn from both patient and control groups, AMH, testosterone, FSH, LH, and prolactin parameters were investigated. Aim: Hyperthermia is an effective therapy for cancer and tumor angiogenesis. However, the development of thermotolerance is an important problem. We aimed to investigate the effects of KNK437 and quercetin on angiogenesis and thermotolerance in endothelial cells (ECs). Materials and methods: ECs were grown in EGM-2 medium. ECs were preincubated with KNK437 or quercetin and then treated with single and fractionated hyperthermia. Aim: To investigate the histology and function of the kidney in mice exposed to different concentrations of phenol through the gavage method for 10 consecutive days. Materials and methods: Forty female BALB/c mice were selected and randomly divided into 1 control group and 3 experimental groups. The control group was administered distilled water through the gavage method, while the experimental groups received daily doses of 80, 180, and 320 mg/kg of phenol, respectively. This study was performed in order to evaluate the effects of depression and life factors on the social networking scores of elderly people. Effect Of Prenatal Exposure To Cigarette Smoke On The Conduction Of Sensory Nerve Fibers In The Rat, Jumah Shakhanbeh

Aim: To investigate the effect of prenatal exposure to cigarette smoke (CS) on the conduction of sensory nerve fibers in the adult rat offspring. Materials and methods: Pregnant rats were exposed to CS by inhalation during the gestation period. The saphenous nerve in the adult offspring was exposed in vivo, and the conduction parameters of the threshold, maximum strength of stimulation, conduction velocity, and amplitude of compound action potentials (CAPs) of A\alpha\beta-, A\delta-, and C-fibers were examined. Results: Prenatal exposure to CS induced significant reduction (P < 0.005) in the peak amplitude of CAPs of A\alpha\beta- and A\delta-fibers. The conduction velocity of A\alpha\beta-fibers and C-fibers was significantly (P < 0.025-0.05) increased after prenatal exposure to CS. Conclusion: Prenatal exposure to CS attenuates the conduction of sensory nerve fibers in the rat saphenous nerve.

To investigate whether cytokines are effective in predicting relapses among patients with acute brucellosis. Materials and methods: This trial was conducted in 42 patients who were being followed-up with diagnosis of acute brucellosis. Serum samples were obtained on days 0 and 45. Since brucellosis has been called a chronic illness and treatment of patients infected with this bacterium has not been successful, the present study aim ed to evaluate the effects of several medicinal plant extracts on clinical Brucella strains. Materials and methods: First, Brucella strains were isolated from brucellosis patients. Then an antibiotic resistance assay was performed for these strains, while the antibacterial activity of the above-mentioned extracts was evaluated. We developed a new liquid-liquid extraction (LLE) and reversed-phase HPLC method that uses a postextraction spiked internal standard for the determination of plasma warfarin. Anemia is common in the general population and one of the most frequently observed nutritional deficiency diseases in the world today. The aim of this study was to investigate the prevalence and etiology of anemias in a large cohort representing the Turkish nation. Materials and methods: The study population comprised 2187 subjects. Serum iron, iron binding capacity, ferritin, vitamin B12, and folic acid analyses were conducted with autoanalyzers using commercial kits. To determine possible therapeutic effects of oral lycopene supplementation on plasma insulin levels, lipid peroxidation, blood glucose levels, and the antioxidant defense system of streptozotocin-induced diabetic rats. Materials and methods: Classical biochemical methods were used to determine plasma malondialdehyde (MDA), nitric oxide (NO), glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD), and glutathione (GSH) levels. This study investigated the value of HbA1c as a screening and diagnostic test for diabetes mellitus (DM) in high-risk Turkish individuals. Materials and methods: A total of 295 participants were successfully screened. Patients were divided into 4 groups based on their oral glucose tolerance test results, according to criteria put forth by the American Diabetes Association; 120 (40.7%) had normoglycemia, 44 (14.9%) had DM, 62 (21%) had impaired fasting glucose, and 69 (23.4%) had impaired glucose tolerance. Adult Attention-Deficit/Hyperactivity Disorder (ADHD) affects~ 4.4% of the adult population, and patients with this disorder frequently have comorbid substance abuse disorders. Methylphenidate (MPH) is considered a first line pharmacotherapy agent in the treatment of ADHD, and is commonly used or co-abused with alcohol. Hydrolysis of oral racemic methylphenidate (dl-MPH) by carboxylesterase 1 (CES1) limits the bioavailability of the pharmacologically active d-MPH isomer to approximately 30% and that of the inactive 1-MPH isomer to only 1-2%. Co-administration of dl-MPH with ethanol results in elevated d-MPH plasma concentrations, as well as CES1 mediated enantioselective transesterification of 1-MPH to 1-ethylphenidate (EPH). Purpose: To explore (i) the potential of polyethylenimine (PEI)-DNA nanoparticles as a vector for delivering genes into human corneal fibroblasts, and (ii) whether the nanoparticle-mediated soluble extracellular domain of the transforming growth factor–β type II receptor (sTGFβRII) gene therapy could be used to reduce myofibroblasts and fibrosis in the cornea using an in vitro model.

Nuclear receptor (NR) protein retinoid X receptor (RXR) is a ligand-inducible transcription factor that binds retinoids for regulation of target genes. Agonist binding induces a major structural rearrangement in the ligand binding domain (LBD) and formation of the coactivator binding interface. RXR-signaling pathways control proliferation, differentiation, and growth in epithelial tissues. Dynamic changes to the actin cytoskeleton are required for synaptic plasticity and long-term memory formation. However, the molecular mechanisms that mediate filamentous actin (F-actin) dynamics during both activity-dependent synaptic potentiation and long-term memory encoding are poorly understood. Myosin II motor proteins are highly expressed in actin-rich growth structures in neurons, including dendritic spines. Recent work demonstrates that these molecular machines mobilize F-actin in response to synaptic stimulation and are required for memory encoding in CA1 hippocampus of rodents. DNA double-strand breaks (DSB) with chemically modified end-termini are the most significant lesions resulting from radio/chemotherapeutic intervention of cancer and non homologous end-joining (NHEJ) factor Artemis nuclease has been implicated in the repair of such breaks. To examine whether the resolution of terminally blocked DNA DSBs is the biologically relevant function of Artemis, Artemis deficient fibroblasts were stably complemented with wild type or an endonuclease deficient D165N mutant Artemis. Physiological levels of wild type (WT) Artemis completely restored DSB repair proficiency and resistance to γ-radiation, bleomycin, and neocarzinostatin. Tobacco dependence is high in women who suffer from anxiety disorders yet little is known about the contributions of nicotinic acetylcholine receptors (nAChRs) on anxiety-like behavior. β2*nAChRs (*denotes assembly with other subunits) are the most abundantly expressed nAChRs in the brain yet little is known about the contributions of β2*nAChRs on anxiety-like behavior in female mice. In this study, antagonism and nicotine effects on anxiety-like behavior was investigated across the life span in 6, 12 and 24-month-old drug-naïve knockout (KO), heterozygous (HET) and a gain of function α6L9S mice and wild type (WT). As a category A agent in the Center for Disease Control bioterrorism list, Rift Valley fever (RVF) is considered a major threat to the United States (USA). Should the pathogen be intentionally or unintentionally introduced to the continental USA, there is tremendous potential for economic damages due to loss of livestock, trade restrictions, and subsequent food supply chain disruptions. We have incorporated the effects of space into a mathematical model of RVF in order to study the dynamics of the pathogen spread as affected by the movement of humans, livestock, and mosquitoes. Velo-cardio-facial syndrome/DiGeorge syndrome, also known as 22q11.2 deletion syndrome (22q11DS) is the most common microdeletion syndrome, with an estimated incidence of 1/2,000 – 1/4,000 live births. Approximately 9–11% of patients with this disorder have an overt cleft palate (CP), but the genetic factors responsible for CP in the 22q11DS subset are unknown. The TBX1 gene, a member of the T-box transcription factor gene family, lies within the 22q11.2 region that is hemizygous in patients with 22q11DS. Inactivation of one allele of Tbx1 in the mouse does not result in CP, but inactivation of both alleles does. Based on these data, …

The CAG trinucleotide repeat mutation in the Huntington's disease gene (HTT) exhibits age-dependent tissue-specific expansion that correlates with disease onset in patients, implicating somatic expansion as a disease modifier and potential therapeutic target. Somatic HTT CAG expansion is critically dependent on proteins in the mismatch repair (MMR) pathway. To gain further insight into mechanisms of somatic expansion and the relationship of somatic expansion to the disease process in selectively vulnerable MSNs we have crossed HTT CAG knock-in mice (HdhQ111) with mice carrying a conditional (floxed) Msh2 allele and D9-Cre transgenic mice, in which Cre recombinase is expressed specifically in MSNs …

BACKGROUND: One-fifth of patients with seemingly 'curable' pancreatic ductal adenocarcinoma (PDA) experience an early recurrence and death, receiving no definable benefit from a major operation. Some patients with advanced stage tumors are deemed 'unresectable' by conventional staging criteria (e.g. liver metastasis), yet progress slowly. Effective biomarkers that stratify PDA based on biologic behavior are needed. To help researchers sort through the maze of biomarker data, a compendium of ∼2500 published candidate biomarkers in PDA was compiled (PLoS Med, 2009. 6(4) p. e1000046).

Embryos of the annual killifish Austrofundulus limnaeus can enter into dormancy associated with diapause and anoxia-induced quiescence. Dormant embryos are composed primarily of cells arrested in the G1/G0 phase of the cell cycle based on flow cytometry analysis of DNA content. In fact, most cells in developing embryos contain only a diploid complement of DNA, with very few cells found in the S, G2, or M phases of the cell cycle. Diapause II embryos appear to be in a G0-like state with low levels of cyclin D1 and p53. However, the active form of pAKT is high during diapause II. …
