Lipocalin-2 Ameliorates The Signs And Outcomes Of Diabetes Mellitus In An Animal Model, Saeeda Mohammed Al Jaberi Jun 2021

Lipocalin-2 Ameliorates The Signs And Outcomes Of Diabetes Mellitus In An Animal Model, Saeeda Mohammed Al Jaberi

Theses

Lipocalin-2 (LCN2) is a new adipocytokine consisting of 198 amino acids. It is also referred to as neutrophil gelatinase-associated lipocalin, siderocalin, uterocalin, α1-microglobulin related protein, or 24p3. LCN2 belongs to a large group of transport proteins that are capable of carrying small and lipid soluble molecules in blood circulation. It has two membrane receptors, megalin/glycoprotein GP330, which binds human LCN2 and SLC22A17 or 24p3R, which forms complexes with mouse Lcn2 protein. LCN2 is encoded by a gene located at chromosome locus 9q34.11. LCN2 was initially isolated from neutrophil granules released at site of infection and inflammation in human and from …

Delayed Modulation Of Glutamate Receptors By Anti-Epileptic Drugs After Traumatic Brain Injury In Rats, Edgar Rodriguez Jun 2021

Delayed Modulation Of Glutamate Receptors By Anti-Epileptic Drugs After Traumatic Brain Injury In Rats, Edgar Rodriguez

Dissertations, Theses, and Capstone Projects

Traumatic brain injury (TBI) is a significant health concern. Around 74 million people sustain a traumatic brain injury worldwide. The damage caused by TBI produces two types of injury; primary and secondary injuries. Primary injury is caused within milliseconds and is irreversible. Secondary brain injury is delayed and produced by molecular, cellular, and structural disruption after the initial injury. One of the most devastating dysfunction after TBI is glutamate neurotransmitter overactivation that could lead to neurotoxic levels of glutamate in the brain (i.e., excitotoxicity). Excitotoxicity has been linked with the development of epilepsy after TBI, also known as post-traumatic epilepsy …

Chromosomal fragile sites are implicated in promoting genome instability, which drives cancers and neurological diseases. Yet, the causes and mechanisms of chromosome fragility remain speculative. Here, we identify three spontaneous fragile sites in the Escherichia coli genome and define their DNA damage and repair intermediates at high resolution. We find that all three sites, all in the region of replication termination, display recurrent four-way DNA or Holliday junctions (HJs) and recurrent DNA breaks. Homology-directed double-strand break repair generates the recurrent HJs at all of these sites; however, distinct mechanisms of DNA breakage are implicated: replication fork collapse at natural replication …

BACKGROUND: 16S rRNA amplicon sequencing is an accurate method of detecting microbial infection without culture. It is unclear if sequencing has additional benefits over routine diagnostic methods for Helicobacter pylori testing.

METHODS: We enrolled Mongolian volunteers with dyspepsia. Using routine diagnostic methods, positive H. pylori was defined as positive results on histology/immunohistochemistry, culture, rapid urease test, or serology; negative H. pylori was defined by negative results from all these tests. We performed 16S rRNA sequencing on gastric biopsy specimens and calculated cutoffs for operational taxonomic units (OTUs) and relative abundance (RA) to define positive results using ROC curves.

RESULTS: We …

Novel biomarkers for hepatocellular carcinoma (HCC) surveillance in patients with cirrhosis are urgently needed. We previously identified osteopontin (OPN) as a promising biomarker for the early detection of HCC. This study is to further validate the performance of OPN and identify fatty acids (FA) that could improve OPN's performance in HCC risk assessment in patients with cirrhosis. To that end, we selected 103 patients with cirrhosis under surveillance. Among them, 40 patients developed HCC during follow-up. We investigated in these 103 patients, the association between HCC incidence and prediagnostic serum levels of AFP, OPN, and 46 FAs. OPN performance was …

OBJECTIVE: White adipose tissue (WAT) expansion regulates energy balance and overall metabolic homeostasis. The absence or loss of WAT occurring through lipodystrophy and lipoatrophy contributes to the development of hepatic steatosis and insulin resistance. We previously demonstrated that sole small ubiquitin-like modifier (SUMO) E2-conjugating enzyme Ube2i represses human adipocyte differentiation. The role of Ube2i during WAT development remains unknown.

METHODS: To determine how Ube2i impacts body composition and energy balance, we generated adipocyte-specific Ube2i knockout mice (Ube2i

RESULTS: Surprisingly, Ube2i

CONCLUSIONS: Our results demonstrate that Ube2i expression in mature adipocytes allows WAT expansion during postnatal growth. Deletion of Ube2i in …

Amplification of MYCN is a poor prognostic feature in neuroblastoma (NBL) indicating aggressive disease. We and others have shown BET bromodomain inhibitors (BETi) target MYCN indirectly by downregulating its transcription. Here we sought to identify agents that synergize with BETi and to identify biomarkers of resistance. We previously performed a viability screen of ∼1,900 oncology-focused compounds combined with BET bromodomain inhibitors against MYCN-amplified NBL cell lines. Reanalysis of our screening results prominently identified inhibitors of aurora kinase A (AURKAi) to be highly synergistic with BETi. We confirmed the anti-proliferative effects of several BETi+AURKAi combinations in MYCN-amplified NBL cell lines. Compared …

Atypical Structural Neuroconnectivity Associated With Emotional Face Processing And Clinical Affect In Children With Chromosome 22q11.2 Deletion Syndrome, David Dodd Stephenson Jr

LSU New Orleans Theses and Dissertations

Schizophrenia spectrum disorder (SCZ) is a debilitating psychiatric disorder that affects approximately 1% of the general population. The etiopathology of this disorder is not fully understood with genetic and environmental factors underpinning vulnerability and disease progression. Children with chromosome 22q11.2 deletion syndrome (22q11.2DS) are at ultra-high risk for developing SCZ with 30% developing the disorder in adolescence or young adulthood. To investigate associations with psychosis vulnerability, much work has been performed looking at neurological underpinnings between schizophrenia and 22q11.2DS to assess similarities between both genetic and idiopathic forms of schizophrenia in adults. However, many past studies have assessed this using …

Library Philosophy and Practice (e-journal)

The retrieval of superior-quality photographs with a minimal exposure is an interesting subject in radiography. Pre-processing is a key phase in signal and image processing such as healthcare, telecommunications, and satellite, and it focuses on reducing or eliminating the extent of noise in the image. Denoising aids in the recovery of finer data and relevant material. Medical images including ECG, Ultrasound, CT-scan, and X-ray provide incredibly fine data that has to be precise and noise-free in order for the knowledge and aspects of interest to be retained during diagnosis. Various noise reduction technologies for medical images, such as wavelet transform, …

Because there is no effective treatment for late-stage prostate cancer (PCa) at this moment, identifying novel targets for therapy of advanced PCa is urgently needed. A new network-based systems biology approach, XDeath, is developed to detect crosstalk of signaling pathways associated with PCa progression. This unique integrated network merges gene causal regulation networks and protein-protein interactions to identify novel co-targets for PCa treatment. The results show that polo-like kinase 1 (Plk1) and DNA methyltransferase 3A (DNMT3a)-related signaling pathways are robustly enhanced during PCa progression and together they regulate autophagy as a common death mode. Mechanistically, it is shown that Plk1 …

Effect Of Serine-Dipeptide Bacterial Lipid Treatment On Gene Expression In Hepg2 Liver Cells, Shaharia Ferdus May 2021

Effect Of Serine-Dipeptide Bacterial Lipid Treatment On Gene Expression In Hepg2 Liver Cells, Shaharia Ferdus

Honors Scholar Theses

While greater abundances of Bacteroidetes are associated with better health outcomes, this phylum is a major producer of serine-dipeptide lipids, which are thought to promote inflammation via Tolllike receptor (TLR) stimulation. Lipid 654 (L-654) is one class of serine-dipeptide lipids that has been isolated from arterial plaques and is thought to contribute to atherogenesis. Recent evidence shows that not only does long-term L-654 exposure attenuate atherosclerosis, it also significantly reduces hepatic lipid levels and markers of inflammation in mice. To better understand the impact on the human liver, L-654 was administered to HepG2 hepatocytes at concentrations of 0.5 μg/mL or …

Glun2d Nmda Receptors Gate Fear Extinction Learning And Interneuron Plasticity, Christophe J. Dubois, Siqiong June Liu May 2021

Glun2d Nmda Receptors Gate Fear Extinction Learning And Interneuron Plasticity, Christophe J. Dubois, Siqiong June Liu

School of Medicine Faculty Publications

The cerebellum is critically involved in the formation of associative fear memory and in subsequent extinction learning. Fear conditioning is associated with a long-term potentiation at both excitatory and inhibitory synapses onto Purkinje cells. We therefore tested whether fear conditioning unmasks novel forms of synaptic plasticity, which enable subsequent extinction learning to reset cerebellar circuitry. We found that fear learning enhanced GABA release from molecular layer interneurons and this was reversed after fear extinction learning. Importantly an extinction-like stimulation of parallel fibers after fear learning is sufficient to induce a lasting decrease in inhibitory transmission (I-LTDstim) in the cerebellar cortex, …

Characterizing The Gene Regulatory Network Required To Specify The Excitatory Neurotransmitter Phenotype Of V0v Spinal Interneurons, Amber Kellie Woodard

Theses - ALL

The majority of neurons within the spinal cord are interneurons. Interneurons make important connections with other cells that allow signals to be relayed between the brain and the rest of the body, and aid in functions such as locomotion. One key functional characteristic of interneurons is the neurotransmitter that they use to communicate with other cells. Interneurons that use GABA or glycine neurotransmitters to communicate have an inhibitory phenotype, while interneurons that use glutamate neurotransmitters have an excitatory neurotransmitter phenotype. Without the correct neurotransmitter phenotype, connections within the central nervous system will malfunction. It is therefore important to understand how …

Targeting androgen receptor (AR) has been shown to be promising in treating glioblastoma (GBM) in cell culture and flank implant models but the mechanisms remain unclear. AR antagonists including enzalutamide are available for treating prostate cancer patients in clinic and can pass the blood-brain barrier, thus are potentially good candidates for GBM treatment but have not been tested in GBM orthotopically. Our current studies confirmed that in patients, a majority of GBM tumors overexpress AR in both genders. Enzalutamide inhibited the proliferation of GBM cells both in vitro and in vivo. Although confocal microscopy demonstrated that AR is expressed …

Genotype-phenotype relationships shape health and population fitness but remain difficult to predict and interpret. Here, we apply an evolutionary action method in mutational landscapes to unravel genes and pathways connected to autism spectrum disorder (ASD). Evolutionary action predicts the impact of missense variants on protein function by measuring motions in fitness landscapes, based on phylogenetic distances and substitution odds in homologous sequences. By examining 368 pathways across 2,384 individuals with ASD (probands), we found that 23 pathways, a total of 398 genes, had de novo missense variants biased to higher evolutionary action scores than expected by random chance, including axonogenesis, …

INTRODUCTION: Idiopathic pulmonary fibrosis (IPF) is an orphan disease characterized by progressive loss of lung function resulting in shortness of breath and often death within 3-4 years of diagnosis. Repetitive lung injury in susceptible individuals is believed to promote chronic oxidative stress, inflammation, and uncontrolled collagen deposition. Several preclinical and retrospective clinical studies in IPF have reported beneficial outcomes associated with the use of proton pump inhibitors (PPIs) such as esomeprazole. Accordingly, we sought to investigate molecular mechanism(s) by which PPIs favorably regulate the disease process.

METHODS: We stimulated oxidative stress, pro-inflammatory and profibrotic phenotypes in primary human lung epithelial …

Recent work has shown that bilirubin has a hormonal function by binding to the peroxisome proliferator-activated receptor-α (PPARα), a nuclear receptor that drives the transcription of genes to control adiposity. Our previous in silico work predicted three potential amino acids that bilirubin may interact with by hydrogen bonding in the PPARα ligand-binding domain (LBD), which could be responsible for the ligand-induced function. To further reveal the amino acids that bilirubin interacts with in the PPARα LBD, we harnessed bilirubin’s known fluorescent properties when bound to proteins such as albumin. Our work here revealed that bilirubin interacts with threonine 283 (T283) …

Examining Perceptions Of Anorexia Nervosa, Polly Mcgonigle

Undergraduate Honors Theses

Anorexia nervosa (AN) is an eating disorder characterized by a restriction of energy intake, an intense fear of gaining weight, and often distorted body image. AN has the second highest mortality rate of all psychiatric disorders, due to high suicide rates and medical complications associated with malnutrition. An estimated 10% of those who have AN die because of the disorder (Insel, 2012). Interacting factors—genetic, biological, environmental, and psychosocial—contribute to the etiology and maintenance of AN. However, outside of research settings, AN is misunderstood as having primarily environmental roots (Salafia, et. al). Blame is placed on societal expectations and the disorder …
