Differential Effect Of Glutathione Depletion On Glycogenolysis In Isolated Rat Hepatocytes Mediated By A-Adrenoceptor Agonists And Glucagon, Mukadder Atmaca, Jeffrey R. Fry
Turkish Journal of Medical Sciences
Glutathione (GSH) exerts a variety of cytoprotecive effects, but is readily depleted from cells under a variety of stressful stimuli. The impact of GSH depletion on receptor-mediated activity in rat hepatocytes has been studied with regard to glycogenolysis stimulated with a-adrenoceptor agonists or glucagon, which exert their actions through different signalling pathways. Depletion of GSH content (70%) by diethyl maleate had no effect on the redox status of the cells or on basal or glucagon-stimulated glycogenolysis, but significantly reduced the response to the a-adrenoceptor agonists adrenalin and phenylephrine. These results highlight the possible modulatory effects of GSH on receptor-mediated activity …

Turkish Journal of Medical Sciences
Sister chromatid exchange (SCE) and chromosomal aberration (CA) in peripheral lymphocytes have been widely used in assessing exposure to mutagens and carcinogens. One of the extensively studied genotoxins is benzo[a]pyrene (BaP). The aim of the present study was to examine the ability of BaP to induce different individual cytogenetic response measured by SCE and CA frequency. The possible influence of genetic polymorphism was also taken into account, by including donors representing positive or null glutathione S-transferase M1 (GSTM1) genotypes. SCEs and CAs were analyzed from 72-h whole-blood lymphocyte cultures of 16 GSTM1 positive and 15 GSTM1 null donors after treatment …

Turkish Journal of Medical Sciences
Recent technology enables the use of cell-free fetal DNA in maternal plasma and serum for noninvasive prenatal genetic diagnosis. This study was designed to evaluate factors most likely to influence the success of a simple, cost efficient, reliable and replicable conventional PCR technique in the clinical routine of prenatal genetic diagnosis of selected cases. The results strongly suggest that DNA extraction and PCR cycle optimization are 2 major success-limiting steps and the maternal plasma is a better choice over serum for DNA extraction for such prenatal genetic diagnosis. In addition, the use of a ready-to-use PCR mixture containing heat-activated Taq …

Polycyclic aromatic hydrocarbons (PAHs) are one of the main environmental pollutants in urban areas. Cytogenetic analysis of chromosomal aberrations is of great concern as they are involved in the mechanism of carcinogenesis. The aim of this study was to assess the genotoxic effect of occupational exposure to PAHs in a group of 30 Bulgarian traffic policemen compared to 30 office clerks. Structural and numerical chromosome aberrations were analyzed by the conventional method in 100 to 300 cells per person. The exposure assessment was performed by personal sampling of air in the respiratory area. PAHs were identified by high performance liquid …

Polarized Mueller Matrix Analytical Model For Glucose Measurement In Vitro, Shamaraz Firdous, Masroor Ikram
Turkish Journal of Medical Sciences
In this study we present a noninvasive and nonradioactive glucose in vitro measurement technique using the Mueller polarization analytical model. The glucose solution and aqueous humor phantoms are illuminated with a He-Ne (632.8 nm) laser. This in vitro study provides a base and reference data for in vivo eye glucose monitoring. The 16 Mueller polarization matrix element images give information about eye absorption and scattering, and changes in polarization patterns to measure glucose concentration. The interior part of the eye (aqueous humor) contains a birefringent scattering medium for glucose measurement. The Mueller matrix polarimetric technique for simultaneous extraction of rotating …

Quality Control Studies On Conventional Carbamazepine Tablets Available On The Turkish Drug Market, Tansel Çomoğlu, Nurşi̇n Gönül
Turkish Journal of Medical Sciences
Carbamazepine (CBZ) is a widely used antiepileptic drug. It is used to control grandmal seizure as well as in the treatment of neuralgia. The drug is characterised by slow and irregular gastrointestinal absorption due to its low water solubility. In clinics, for conventional dosage forms of CBZ, single daily dosing is insufficient; effective CBZ level is provided by multiple drug administration. Multiplicity of dosage causes inconsistent plasma CBZ levels leading to side effects because of narrow therapeutic and toxic levels. In this study, quality controls of different conventional CBZ tablets which are commercially available on the Turkish drug market were …

Does Early Enteral Feeding Affect Clinical Outcome Or Cytokine Profiles After Elective Surgery For Colorectal Cancer?, Elizabeth Anne Baker, Lynne Williams, Sanaa El-Gaddal, Fintan Bergin, David John Leaper
Turkish Journal of Medical Sciences
Patients who have intestinal cancer are prone to malnutrition, which can result in deterioration of physiological function, poor wound healing, increased risk of infection and prolonged hospital stay. These complications are particularly relevant after colorectal surgery. Early enteral feeding, to enhance nutritional status, has been reported to reduce post-operative complications, length of hospital stay and morbidity and mortality after upper gastro-intestinal cancer surgery. Inflammatory cytokines (IL-6, TNF-a (\alpha)), clinical outcomes and postoperative complications were measured in 147 patients undergoing elective surgery for colorectal cancer (84 standard care; 64 patients had early enteral feeding in addition). There were no significant differences …

Nucleic acid amplification methods to detect Mycobacterium tuberculosis complex (MTBC) in clinical specimens are increasingly used to diagnose tuberculosis. A number of nucleic acids amplification assays have been developed to detect MTBC DNA in clinical material. These assays differ in their requirements for sample volume and sample preparation, methods of amplification, and methods of detection. There are advantages and disadvantages of all assays; thus, there is probably no assay which is best suited to all situations. Quality control programs for molecular diagnostics (QCMD) based on nucleic acid amplification have not been widely implemented in clinical laboratories and remain limited to …
