Polymorphisms in the genes encoding cytochrome p450 (CYP) and thiopurine S-methyl transferase (TPMT) enzymes catalyze the metabolic reactions of several drugs. These polymorphisms might be responsible for adverse drug reactions. Turkish population data for these genes still needs to be elucidated. We aimed to detect the allele frequencies of thiopurine S-methyl transferase (TPMT), cytochrome p4503A4*1B (CYP3A4*1B) and cytochrome p4503A4*3 (CYP3A5*3) gene variants in the Turkish population. The encapsulated follicular variant of papillary thyroid carcinoma constitutes the most common source of consultation material in thyroid pathology. We present our five consecutive cases to illustrate this issue. The diagnosis of the encapsulated follicular variant of papillary thyroid carcinoma is considerably controversial. These tumors are encapsulated neoplasms and have the nuclear features of papillary thyroid carcinoma. Although the criteria were described for the diagnosis, a considerable subjectivity is present among the pathologists. Therefore, the categories of "well differentiated carcinoma" and "well differentiated tumor of uncertain malignant potential" were included in the diagnostic spectrum of these neoplasms. We prospectively studied the characteristics of 401 patients with first-ever-in-a lifetime stroke who have been admitted consecutively into stroke care unit. During the evaluation of distribution according to age, patients were divided into 7 age groups with 10-year intervals. It was determined that ischemic stroke was mostly seen in the age group 71-80 (32.2%) and the age group 61-70(28.2%). The major risk factor of ischemic stroke was hypertension, followed by diabetes mellitus, ischemic heart disease, smoking and hypercholesterolemia. The three most frequent etiologic categories were small-vessel disease, atherosclerotic large vessel disease and cardioembolism. The anatomical distribution of ischemic stroke were …

The aim of this study is to evaluate factors influencing utilization of health services by women aged 15 years and older having any health symptoms within the previous 15 days in a city center located in the eastern part of Turkey. In this cross-sectional study, subjects (1,000 households) were sampled from the "city-information-system" based on household satellite photographs of the city. Nine hundred and thirty-five households out of 1,000 were included in the study (response rate: 93.5%), and 1,613 subjects were interviewed. Aim: Dukes' staging system is a simple system used widely in the staging of colorectal cancer. This study was designed to analyze the applicability of the modified Dukes' classification system in gastric cancer cases. Methods: The prognostic factors affecting survival in 139 gastric cancer cases who had had at least 15 lymph nodes removed were analyzed. Modified Dukes' and TNM classifications were investigated to correlate statistically significant prognostic factors. Aim: In immunohistochemistry, antibodies are used to detect and localize antigens in cellular compartments in sections taken from frozen or paraffin blocks. Currently, computerized image processing techniques are widely available to analyze immunohistochemically stained tissues; however, the image analysis programs are generally highly specific and expensive. We aimed to show the feasibility of computer assisted image analysis (CAIA) by using two computer programs currently in widespread use. Pleural effusions are very common in general population. There are more than 50 reasons for the etiology. There is limited data on the prevalence of pleural effusions in a population of pregnant women. We prospectively screened 45 pregnant women between May and October 2006. All the pregnant women underwent thoracic ultrasonography during their routine check. Five out of 45 subjects (11%) had minimal and bilateral pleural effusion. None of the subjects was symptomatic. All of the effusions regressed without any intervention. In conclusion, the prevalence of pleural effusions is not rare during pregnancy and physicians should be aware of the …

Aim: The aim of this study was to define aphasia frequency and its clinical types together with the relationship between the clinical types and age, gender, hemiplegic side, etiology and educational level of the hemiplegic patients. Methods: All hemiplegic patients who admitted to Ankara Physical Therapy Training and Research Hospital (APTTRH) from January 2002 to January 2003 were included in this study. During this period, 478 hemiplegic patients were hospitalized for rehabilitation. Aim: Owing to the widespread use of antibiotic drugs in both human and animals, we thought it would be important to determine the effect of these on lactoperoxidase (LPO; E.C. 1.11.1.7) activity during lactation. The in vitro effects of gentamicin sulfate and ampicillin sodium drugs on bovine LPO enzyme activity in breast-milk were investigated. Materials and Methods: LPO was purified with Amberlite CG 50 resin, CM Sephadex C-50 ion-exchange chromatography, and Sephadex G-100 gel filtration chromatography from skimmed bovine milk. Aim: The aim of this study was to investigate the carriage of b-lactamase genes in ampicillin-resistant (Amp^r) Escherichia coli (E. coli) isolates from human normal fecal flora. Methods: Ten Amp^r E. coli strains isolated from the stool samples of 21 healthy persons with no antibiotic use during at least three months were screened for TEM-, SHV-, or OXA-type \beta-lactamase genes by polymerase chain reaction (PCR). The susceptibility of the strains to antibiotics was determined by disk diffusion method, and minimum inhibitory concentration (MIC) of ampicillin to the strains was determined by agar dilution method. 3,4-Methylenedioxymethamphetamine (MDMA), also known as "ecstasy", is a synthetic, psychoactive drug chemically similar to the stimulant methamphetamine and the hallucinogen mescaline. It can cause cognitive, physical, psychological and neurological side effects. Ecstasy-induced acute renal failure, liver damage and brain edema are reported in the literature. We report an MDMA-induced toxic hepatitis case which was thought to have viral origin.

The goal of this study was to elucidate the molecular mechanism by which type I IFN inhibits assembly and release of HIV-1 virions. Our study revealed that the IFN-induced ubiquitin-like protein ISG15 mimics the IFN effect and inhibits release of HIV-1 virions without having any effect on the synthesis of HIV-1 proteins in the cells. ISG15 expression specifically inhibited ubiquitination of Gag and Tsg101 and disrupted the interaction of the Gag L domain with Tsg101, but conjugation of ISG15 to Gag or Tsg101 was not detected. BACKGROUND: The structural maintenance of chromosome 3 (SMC3) protein is a constituent of a number of nuclear multimeric protein complexes that are involved in DNA recombination and repair in addition to chromosomal segregation. Overexpression of SMC3 activates a tumorigenic cascade through which mammalian cells acquire a transformed phenotype. This has led us to examine in depth how SMC3 level affects cell growth and genomic stability. In this paper the effect of SMC3 knockdown has been investigated. INTRODUCTION: Predicting the clinical course of breast cancer is often difficult because it is a diverse disease comprised of many biological subtypes. Gene expression profiling by microarray analysis has identified breast cancer signatures that are important for prognosis and treatment. In the current article, we use microarray analysis and a real-time quantitative reverse-transcription (qRT)-PCR assay to risk-stratify breast cancers based on biological 'intrinsic' subtypes and proliferation. Enormous effort has been devoted to the development of a vaccine against human immunodeficiency virus (HIV). But it is proving to be an unprecedented challenge to create an effective vaccine mainly due to the high genetic variability of the virus and the necessity of cytotoxic T lymphocytes (CTL) for containing the infection. Currently pursued vaccine strategies appear to induce CTL in nonhuman primate models but in the early clinical trials, these strategies fail to fully control the viral infection. New strategies that can cover the vast genetic diversity of HIV are needed for the development of a potent vaccine.

Porphyromonas gingivalis, an important periodontal pathogen, is an effective colonizer of oral tissues. The organism successfully invades, multiplies in, and survives for extended periods in primary gingival epithelial cells (GECs). It is unknown whether P. gingivalis resides in the cytoplasm of infected cells throughout the infection or can spread to adjacent cells over time. We developed a technique based on flow cytofluorometry and fluorescence microscopy to study propagation of the organism at different stages of infection of GECs. The genetic diversity among globally circulating human immunodeficiency virus type 1 (HIV-1) strains is a serious challenge for HIV-1 vaccine design. We have generated a synthetic group M consensus env gene (CON6) for induction of cross-subtype immune responses and report here a comparative study of T-cell responses to this and natural strain env immunogens in a murine model. FoxP3 is a lineage specific marker for regulatory T-cells (Treg). We have generated FoxP3 knock-in (KI) mice by introducing a bicistronic GFP reporter into the endogenous FoxP3 locus, allowing us to faithfully track T-reg in vivo. Recently, we have also generated a MOG 35-55/IAb-tetramer. The combination of these two technologies enables us to study the in vivo behavior of myelin specific T-reg and effector T-cells (T-eff) during EAE. Polymorphisms of TIM-1 have been associated with susceptibility to asthma; however, its role in regulating autoimmunity has not been studied. Several questions need to be addressed to evaluate whether Tannerella forsythia is to be considered a periodontal pathogen. T. forsythia has been detected in periodontal health and disease, so could it be a pathogen? The species was not detected in many studies despite finding other putative pathogens, so could it be important in pathogenicity? The challenges of working with T. forsythia include its fastidious and anaerobic growth requirements for cultural detection. True aneurysms of the PFA are uncommon. Rupture risk in true PFA aneurysms are extremely high (42%). Therefore a rapid evaluation and treatment planning is essential. Ultrasonography and color Doppler ultrasonography have priority in diagnosis of these cases. Angiography, CT and CT angiography are performed in conjunction with ultrasonography for planning the procedure before surgical treatment and follow-up after treatment.

Central nervous system (CNS) involvement has been recognized as the second leading cause of death in patients with systemic lupus erythematosus (SLE), although hydrocephalus is seen very rarely. We report a case of communicating hydrocephalus in a 24-year-old woman having previously diagnosed SLE without antiphospholipid antibody syndrome or cerebral venous angiographic abnormality. We propose direct damage-thrombosis of small sized venous structures or immune complex deposition within arachnoid villi, which impair cerebrospinal fluid flow, as a possible, yet unproven, pathophysiologic mechanism for hydrocephalus in SLE.

Increased levels of protein-associated O-linked N-acetylglucosamine (O-GlcNAc) have been correlated with increased tolerance to stress. Therefore the goal of this study was to determine whether enhanced flux through the hexosamine biosynthesis pathway (HBP), which leads to elevated levels of O-GlcNAc, increased the tolerance of the heart to ischemia and reperfusion (I/R). Hearts from male rats were isolated and perfused with Krebs-Henseliet buffer containing 5mM glucose, and global, no-flow ischemia was induced for 20min followed by 60 min reperfusion. 20 min pre-treatment with 10 mM glucosamine, which enters the HBP directly, significantly improved functional recovery and decreased cardiac Troponin I release …
