Chulalongkorn University Theses and Dissertations (Chula ETD)
Macrophage plasticity is a process that allows macrophages to switch between two opposing phenotypes based on differential stimuli. Functional plasticity of macrophages contributes to disease pathogenesis such as cancer, metabolic diseases, autoimmune diseases and systemic infections. Epigenetics plays an important role in regulating this process. Macrophages stimulated with lipopolysaccharide (LPS) (M(LPS)) produce high level of pro-inflammatory cytokines such as IL-12, TNFα and IL-6 and low level of anti-inflammatory cytokine IL-10, while macrophages stimulated with LPS in the presence of immune complex (IC) (M(IC)) produce high level of IL-10 and low level of IL-12. In this study, we investigated the plasticity …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Interferon (IFN)-λ is a relatively unexplored, yet promising anti-viral agent. IFN-λ has recently been tested in clinical trials of chronic hepatitis B, with the advantage that side effects may be limited compared with IFN-α, as IFN-λ receptors are found only in epithelial cells. To date, IFN-λ downstream signaling pathway remains largely unelucidated, particularly via proteomics methods. Here, we report that IFN-λ3 inhibits HBV replication in HepG2.2.15 cells, reducing levels of both HBV transcripts and intracellular HBV DNA. Quantitative proteomic analysis of HBV-transfected cells was performed following 24-hour IFN-λ3 treatment, with parallel IFN-α2a and PBS treatments for comparison using a dimethyl …

Phosphoproteomic Analysis Of Fc Gamma Receptor Iib Deficient Macrophages In Enhancing The Severity Of Endotoxin Tolerance, Thunnicha Ondee
Chulalongkorn University Theses and Dissertations (Chula ETD)
Sepsis is a severe response to systemic infection and one of the leading causes of death in patients world-wide. Bacterial endotoxin (lipopolysaccharide; LPS), is a potent inducer of inflammation that has been associated with the pathophysiology of sepsis. The repeated LPS exposure can induce a suppressive effect as refer to endotoxin tolerance. Fc gamma receptor IIb (CD32b), the only inhibitory receptor among FcGR family, associates with systemic lupus erythematosus (FcGRIIb defunctioning polymorphisms) which are commonly found in Asian populations. Because of an inhibitory nature of the signaling, endotoxin tolerance in lupus with FcGRIIb deficient (FcGRIIb-/-) is interesting and might be …

Migraine headache generates in trigeminal nociceptive system which is activated by cortical spreading depression (CSD) phenomenon. There are also some neurotransmitter changes relating to pathogenesis of migraine, one of representative examples is serotonin (5-HT) depletion that may predispose to migraine development. Trigeminal ganglion (TG) is a structure containing cell body of neurons that undertakes a role as first order neuron in trigeminal nociceptive system. Moreover, the alteration of glutamate may contribute to hyperactivity of its specific receptors, for instance N-methyl-D-aspartate (NMDA) receptor. Endogenous biomolecule, such as kynurenic acid (KYNA) which is derived form of 5-HT can inhibit NMDA receptor. Basically, …

Background/aim: Multitarget therapy for lupus nephritis (LN) remains in its exploratory phrase and the recent evidence is insufficient. This study aimed to evaluate the efficacy and safety of mycophenolate mofetil (MMF), tacrolimus (TAC), and steroids (multitarget therapy) versus intravenous cyclophosphamide (IVC) and steroids in induction treatment of LN. Materials and methods: We searched for randomized controlled trials of MMF plus TAC versus IVC in LN using PubMed, EMBASE, the Cochrane Central Register of Controlled Trials, the China Biology Medicine Database, and the China National Knowledge Infrastructure Database. We assessed the retrieved citations and selected studies according to predefined inclusion and …

Background/aim: Early warning scores (EWS), widely used around the world but not yet in Turkey, are composed of physiological parameters designed to determine potentially worsening patients to perform necessary interventions in time. The aim is to determine the national EWS (NEWS) of the patients to assess the relation between this score and length of hospital stay (LOHS), transfer to the ICU, 24-h and 28-day mortality rates, and the frequency of in-hospital cardiac arrest (IHCA). Materials and methods: NEWS of all the patients in the internal medicine clinics were calculated via a point prevalence study. The LOHS, transfer to the ICU, …

Background/aim: The Mann Assessment of Swallowing Ability (MASA) is an efficient tool that allows physicians to determine the alertness, cooperation, and respiration of patients, which are important factors when assessing swallowing . This study aimed to translate the MASA into Turkish (T-MASA) and to assess its reliability and validity in patients during the early period after a stroke . Materials and methods: The scale was administered to 141 patients in the early period after a stroke. For reliability, both internal consistency (Cronbach's alpha and corrected item-to-total correlations) and interrater reliability were analyzed. The procedures were scored by two blinded independent …
