Detection Of Nonrandom Association Of Alleles From The Distribution Of The Number Of Heterozygous Loci In A Sample, R Chakraborty

The distribution of the number of heterozygous loci in two randomly chosen gametes or in a random diploid zygote provides information regarding the nonrandom association of alleles among different genetic loci. Two alternative statistics may be employed for detection of nonrandom association of genes of different loci when observations are made on these distributions: observed variance of the number of heterozygous loci (s2k) and a goodness-of-fit criterion (X2) to contrast the observed distribution with that expected under the hypothesis of random association of genes. Genetics And Epidemiology Of Gallbladder Disease In New World Native Peoples, K M Weiss, R E Ferrell, C L Hanis, P N Styne

Native peoples of the New World, including Amerindians and admixed Latin Americans such as Mexican-Americans, are highly susceptible to diseases of the gallbladder. These include cholesterol cholelithiasis (gallstones) and its complications, as well as cancer of the gallbladder. Although there is clearly some necessary dietary or other environmental risk factor involved, the pattern of disease prevalence is geographically associated with the distribution of genes of aboriginal Amerindian origin, and levels of risk generally correspond to the degree of Amerindian admixture. This pattern differs from that generally associated with Westernization, which suggests a gene-environment interaction, and that within an admixed population …

The existence of extrathyroidal monoiodotyrosine (MIT) has been debated because of uncertain results from measuring serum MIT. The reason is that the isotopic and chromatographic assays used have not been sensitive enough. This investigation undertook to establish the existence of MIT in serum utilizing a radioimmunoassay (RIA), to study the physiology of serum MIT, and to discover the pharmacologic effects of exogenous MIT.

The present experiment was conducted to determine the relationship between the human menstrual cycle and gustatory hedonic response. Hedonic response was defined as the perceived pleasantness or unpleasantness of a taste stimulus. Gender differences in taste hedonic have been observed; these may be due to differences in endocrine systems. Animal taste preferences are altered when reproductive hormonal status is experimentally manipulated. Previous researchers report that human females' hedonic responses to sucrose vary systematically with phase of menstrual cycle.

The ability of preimplantation mouse embryos to utilize glucose oxidatively is controlled, in part at least, at the level of glycolysis. Various experimental observations are reviewed that indicate the regulatory mechanism in delayed implanting blastocysts involves the classic negative allosteric feedback of high levels of ATP on phosphofructokinase while the situation in 2-cell embryos appears to be more complicated. That is, in addition to the usual negative effect of ATP and citrate on phosphofructokinase, there appears to be a modification of hexokinase that prevents phosphorylation of adequate amounts of glucose and results in low levels of fructose-6-phosphate at the 2-cell …

Effect Of Membrane Filtration Of Antimalarial Drug Solutions On In Vitro Activity Against Plasmodium Falciparum, J. Kevin Baird, C. Lambros

Antimalarial activities ofchloroquine, mefloquine, amodiaquine, and quinine in vitro against Plasmodium falciparum were diminished as a consequence ofmembranefiltration. Filtered drug solutions gave ID50 values up to 25-fold greater than those of non-filtered (ethanol-sterilized) drug solutions. Loss ofactivity byfiltration was overcome by increasing the drug concentration prior to filtration. Water solutions filtered through Millex-GSfilter units consistently showed an absorbance maximum at 277 nm, accompanied by a lesser peak at 225 nm. Water filtrates from Nucleopore and Millex-G Vfilters showed no absorbance at 277 nm and only slight absorbance was evidentfor the Gelmanfilter unit. Development Of A Mathematical Model Of Renal Function For Clinical Application, Barbara A. Holshouser

This model describes the orthoiodohippurate (OIH) distribution and clearance in the renovascular system. The model is described by five compartments: the blood, left and right kidneys, bladder and the red blood cell compartment. Data for these compartments, except the RBC compartment, are collected with scintillation detectors monitoring OIH tagged with 1-131 as the radiopharmaceutical passes through each compartment. Time/ activity curves are plotted for the data and used as inputs to the model parameter estimation routine. The compartments are described by a set of first order ordinary differential equations solved using the Adam's methods. For parameter estimation, an iterative predictor-corrector …

The present studies were undertaken in order to a) verify the presence of a human Thy 1 antigen, b) demonstrate its similarity to murine Thy 1, and c) biochemically characterize the molecule. Previously, a 25,000 molecular weight molecule had been isolated from a human T lymphoblastoid cell line, Molt 3, using isolation procedures most frequently used for the isolation of rodent Thy 1. The human Thy 1 (p25) was demonstrated to be biochemically similar to murine Thy 1.2 by molecular weight and ability to bind Lens Culinaris lentil lectin. Linear regression procedures are described which give an unbiased estimate of the percent functional viability of rat polymorphonuclear leukocytes (PMN's). These statistical techniques allow discrimination between variables which result in increased PMN survival after freezing. A review of current response surface techniques was made as they relate to optimizing the functional viability of PMN's after freezing. Experimental designs based on the steepest ascent and simplex methodologies were considered. Since the hill climbing efficiency of the steepest ascent and simplex methods are comparable, the steepest ascent procedure was selected as it is more readily adaptable to a batch mode analysis.

Genetically Controlled Variation Of “Acid” Beta-Galactosidase Detected In Rattus Norvegicus By Isoelectric Focusing, T C Douglas, K A Kimmel, P E Dawson

Two genetically variant forms of rat "acid" beta-galactosidase were found to differ in isoelectric point and pH dependence, but not in thermostability or sensitivity to inhibition by p-mercuribenzoate (PMB). The results of two backcrosses and an intercross indicated that the isoelectric focusing phenotypes are controlled by two codominant alleles at a single autosomal locus, for which we propose the name Glb-1. No significant linkage between Glb-1 and albino (LG I), brown (LG II), or hooded (LG VI) was observed. Strain-specific differences in total levels of kidney beta-galactosidase were detected, but it is not yet known whether the variation is controlled …

T Cell Growth Factor Receptors. Quantitation, Specificity, And Biological Relevance, Richard J. Robb, Allen Munck, Kendall A. Smith

To examine directly the hypothesis that T cell growth factor (TCGF) interacts with target cells in a fashion similar to polypeptide hormones, the binding of radiolabeled TCGF to various cell populations was investigated. The results indicate that TCGF interacts with activated T cells via a receptor through which it initiates the T cell proliferative response. Internally radiolabeled TCGF, prepared from a human T leukemia cell line and purified by gel filtration and isoelectric focusing, retained biological activity and was uniform with respect to size and charge. A computer model of the left ventricular (LV) epicardial surface was developed from Jang’s truncated ellipsoidal model, which has a minor to major axis ratio of 6:10. The model was applied in calculating the area of a segment of the myocardium, jeopardized by an impending occlusion of its associated artery. The endangered segment of the LV muscle was also visualized in different projections by rotations in three space and graphical displays simulating the three dimensions.

The present study was designed to examine the effects of gradual DPH withdrawal on the matching to sample performance of three mentally retarded persons. The percentage of correct responses per session served as the dependent variable and the sample and comparison stimuli were red, green, and blue illumination of translucent response windows. The results showed that doses of diphenylhydantoin considerably lower than the suggested optimim therapeutic level can impair the performance of mentally retarded individuals on a simple discrimination task. These studies explore the distribution of blood flow among small samples of placental tissue. Labeled microspheres (14 ± 1 pm) were Injected Into the left ventricle of six un-anesthetized ewes and Into the jugular vein of the fetus In utero. A total of 3,576 samples weighing 31. +11. (S.D.) mg were taken from seventeen cotyledons and counted for flow labels. Maternal and fetal flow within the cotyledons were both normally distributed with standard deviations of 44%. Consecutive medical records of 10,622 births were viewed, January, 1962, through December, 1972, from McAllen General Hospital, McAllen, Texas. The overall incidence of birth defects was consistent with rates for the United States. Birth defects detected in Spanish surnamed were significantly higher than for non-Spanish surnamed. Spanish surnamed bore eight times more defects of the central nervous system than non Spanish surnamed. Males had significantly more defects than females, and had almost seven times more defects of the urogenital system. The most distinctive feature of this survey was the high incidence of atelectasis. It has been previously demonstrated that 7,12-dimethylbenz(a) anthracene (DMBA) is an initiator of ovarian granulosa cell tumors. It is also known that DMBA leads to destruction of primordial oocytes, and that tumor induction may involve the interaction of pituitary gonadotropins. However, the effects of DMBA on ovarian steroidogenesis and uterine growth in relation to the tumorigenic process have not been investigated. Therefore, the present study was conducted to examine these parameters using adult C57Bl/6N mice treated in proestrus and diestrus II with 0.1 ml corn oil with or without 1.5 mg DMBA and killed 44 hours later. Plasma samples were obtained from seven hypertriglyceridemic (>150 mg/dl) chronic renal failure patients before and after regular hemodialysis. The effect of the plasma on the lipolytic activity of bovine milk lipoprotein lipase was investigated. The lipoprotein lipase activity, as measured by release of C14-labeled free fatty acid from triglyceride, was decreased 8-67% when incubated with uremic plasma as compared to the control plasma (44 mg/dl triglyceride). The inhibition noted prior to hemodialysis did not appear to be alleviated by the dialysis but subsequent treatment with an isolated ultrafiltration technique did appear to remove an inhibitory substance as …
