To combat the growing issue of petroleum plastic waste, alternative bio-based polymers are being developed. Many of these biopolymers are made from bio-derived materials, or are biodegradable, but the most promising polymers fall in both categories. Polyhydroxyalkanoates (PHAs) are one such class of polymers, and poly-3-hydroxybutyrate (P3HB), the most popular PHA, has shown great potential. This study utilized two types of cotton-derived glucose, alongside commercial glucose, as a feedstock for the biosynthesis of P3HB by Cupriavidus necator (also known as Ralstonia eutropha). Evaluation Of Anti-Bacterial Efficacy Of Ginger, Resin Gum, And Nigella Sativa Irrigants Against Enterococcus Faecalis: An In Vitro Study, Iman El Ghazzawy, Leen Imad Hasbini, Reem Chamseddine, Wael Betbout, Roula Abiad

International Arab Journal of Dentistry

Objectives: To evaluate the antibacterial efficacy of Ginger, Resin Gum, and Nigella Sativa, against Enterococcus Faecalis.

Methods: Thirty-eight extracted single-rooted premolars were inoculated with E.faecalis, except the negative control, and incubated for 4 weeks. After instrumentation samples were divided into 4 groups (n=7) according to final irrigation: NaOCl+EDTA (Group A), Nigella sativa (B), Resin Gum (C), Ginger (D), plus 2 control groups (n=5): Saline (E), and Saline without bacterial inoculation (F). Cultures were collected at baseline and after irrigation.

An Examination Of The Experiences Of An African American Male Diver On A Power Four Swim Team: An Elite Interview, Steven N. Waller Ph.D., Tiffany M. Quash Ph.D.

International Journal of Aquatic Research and Education

Despite the incremental increase in the number of Black people who are learning to swim, the number of Black divers has not kept pace. In the NCAA Database, swimmers and divers were not segmented; thus, they all appear in the category of “swimming” by race and sex. When viewing the 2024 Division I NCAA Demographics Database, 89 (.02%) of 4307 student-athletes participating in swimming were Black women. For men, the data is equally dismal, revealing that 81 (.03%) of 2,608 student-athletes who were identified as swimmers were Black. Neoadjuvant chemotherapy (NAC) is a critical component of breast cancer treatment, but the molecular mechanisms underlying resistance remain poorly understood. This study aimed to identify transcriptomic changes associated with NAC resistance across four breast cancer subtypes: Luminal A, Luminal B/HER2-positive, Luminal B/HER2-negative, and Triple-Negative Breast Cancer (TNBC). RNA-seq analysis was performed on paired pre- and post-NAC breast cancer samples from 32 nonresponders. Differentially expressed genes (DEGs) were identified, and functional enrichment analyses were conducted. Protein-protein interaction (PPI) networks were constructed to identify hub genes. Nongenetic transcription evolution has been increasingly explored and recognized to drive tumor cell progression and therapeutic resistance. As the regulation hub of transcription machinery, cyclin-dependent kinase 9 (CDK9) is the gatekeeper of RNA polymerase II transcription, and CDK9 dysfunction results in transcriptomic reprogramming and tumor cell progression. We recently reported that the heat shock protein 90 (HSP90)-MYC-CDK9 network drives therapeutic resistance in mantle cell lymphoma (MCL) through transcriptomic reprogramming. RAS pathway (RASp) mutations induce proliferative features, and promote transformation in chronic myelomonocytic leukemia (CMML). However, the unique clonal landscape and hierarchy of distinct RASp mutations remain unexplored. To characterize the landscape, architecture, and implications of unique RASp mutations in CMML, we evaluated a cohort of 814 patients with CMML. We identified 461 RASp mutations among 342 patients (42%). N/KRAS and CBL mutations were the most common, frequently involved the P-loop or RING domains, respectively, and frequently appeared as dominant events (63% and 65%, respectively). Background: Genetic ancestry is an important factor to account for in DNA methylation studies because genetic variation influences DNA methylation patterns. One approach uses principal components (PCs) calculated from CpG sites that overlap with common SNPs to adjust for ancestry when genotyping data is not available. However, this method does not remove technical and biological variations, such as sex and age, prior to calculating the PCs. The first PC is therefore often associated with factors other than ancestry.

The public is paying increased attention to the environmental impacts associated with cattle production systems in the US. Thus, cattle producers are increasingly exploring ways to communicate their efforts to reduce those environmental and climate impacts. This study uses online and social media data from 2018 to 2022 to examine public interest in and sentiment about beef and dairy cattle production when discussed within the context of greenhouse gas (GHG) emissions. This research seeks to evaluate the efficacy of the Teaching at the Right Level (TaRL) strategy in conjunction with the Problem-Based Learning (PBL) model on the scientific learning outcomes of eighth-grade students at middle school Islam Terpadu Khairul Imam. This research addresses the inadequate scientific problem-solving abilities and science learning outcomes of Indonesian students, necessitating a more interactive and individualized instructional technique. This study experimentally investigates the dynamic damping performance of passive tuned liquid column vibration absorbers (TLCVAs), focusing on the influence of vertical column inclination angle and orifice disc configuration. Free vibration tests were conducted on a primary system equipped with a TLCVA using a custom-designed test rig. Ripple et al. (2025) recently argued that large carnivore recovery in Yellowstone National Park triggered one of the world’s strongest trophic cascades, citing a ∼1500 % increase in willow crown volume derived from plant height data. In this comment, we show that their conclusion is invalid due to fundamental methodological flaws. These include use of a tautological volume model, violations of key modeling assumptions, comparisons across unmatched plots, and the misapplication of equilibrium-based metrics in a non-equilibrium system. We report that in humans, mice, fruit flies, and worms, the ribosomal RNAs and the transcribed spacers of 45S are densely packed with organism-specific sequence motifs that are primarily shared with nervous system genes. The human ribosomal RNAs and 45S spacers contain 1,723 such motifs. Specific combinations of these motifs are predominantly found in 3,430 human nervous system genes, of which 1,046 are genes associated with brain disorders, including autism spectrum disorder and schizophrenia. The sequences of the 1,723 motifs and their locations in the introns and exons of nervous system genes are unique to primates. Experimental evidence indicates that …

Background: Prostate cancer (PCa) will affect approximately 12% of US men in their lifetime and remains the 2nd most common cause of cancer-specific death. However, there is a broad spectrum of severity of PCa; some require aggressive treatment and others can "watch-and-wait". Distinction of clinically significant prostate cancer (csPCa) from indolent PCA with current imaging methods remains challenging.

Background: Osteosarcoma (OS) lung metastases remain a significant therapeutic challenge. Innate immune activation is a promising therapeutic approach. Innate immune agonists can modulate the tumor immune microenvironment and improve therapeutic response.

Methods: Using an experimental syngeneic OS lung metastasis BALB/c mouse model with K7M3-luc OS cells, we evaluated the antitumor effects of yeast-derived particulate β-glucan in prevention and therapeutic settings. We then assessed whether the CD40 agonist (CD40a) in combination with β-glucan increased therapeutic response in two different immune-competent mouse models of OS lung tumor burden.

Genetic Engineering Of Esophageal Organoids: Crispr-Based Knock-In For Cell Lineage Tracing, Kyung-Pil Ko, Jae-Il Park

Esophageal organoids serve as powerful systems to study epithelial lineage hierarchies and cancer biology. Gene manipulation in these organoids has traditionally involved overexpression or knockout strategies. However, CRISPR-/Cas9-based knock-in (KI) approaches now enable precise cell lineage tracing and live imaging. Here, we describe protocols to generate fluorescent KI organoids from murine esophageal epithelium by tagging Krt13 (BFP) and Sox2 (mNeon). These dual-reporter organoids allow direct monitoring of growth dynamics and differentiation trajectories. We outline CRISPR/Cas9 design, donor construction using homology-independent approaches (CRISPaint), delivery into organoid cells, enrichment and single-clone isolation, and validation by fluorescence. Blastocystis is the most ubiquitous intestinal protozoan parasite found in humans and animals worldwide. Commonly, ST7 and ST6 are the major Blastocystis subtypes reported in birds, and their frequencies fluctuate greatly with various factors. However, little literature is available on the prevalence and subtype characteristics of this parasite in free-living wild birds in the same city. For this purpose, a total of 114 fresh fecal specimens were collected from wild free-living birds in urban districts of Xinxiang city and tested for Blastocystis using PCR of the barcoding region of the small subunit ribosomal RNA (SSU rRNA) gene. This paper compares various codes and standards for the behavior of “Concrete-Filled Steel Tubes” (CFST) columns under fire and axial load. The codes evaluated include European standards (EC4.2 and EC3.2), the American standard (AISC 360), and the Australian standard (AS/NZS 2327). The fire resistance of Concrete-Filled Steel Tubular (CFST) and steel hollow (SH) columns was assessed using a computer program developed with Visual Studio .NET. The program's results were compared with previous experimental data to evaluate the accuracy of various codes and standards in predicting the behavior of these columns under fire conditions. This paper will give an overview of primary hyperaldosteronism, which is a condition characterized by the excess production of aldosterone from the adrenal glands. Since its discovery in 1954 by James Conn, primary hyperaldosteronism has been discovered to have a variety of causes, with the more common ones being either benign tumors or inherited disorders such as congenital adrenal hyperplasia. Affected patients display conditions such as hypertension or hypokalemia, which may lead to symptoms including headaches, blurred vision, or dizziness. Crayfishes Of Kansas: List Of Taxa, Synonyms, And References, Mark E. Eberle

There are 14 species of crayfishes (Decapoda: Cambaridae) reported from Kansas, although there are questions about the systematics of some taxa. These species are listed by their current names and the synonyms under which they have been reported since 1885, along with their conservation status. Published references that include each species in Kansas are also noted. In addition, information about species with type localities in Kansas and unresolved taxonomic issues are summarized. The list of species has been updated in 2025 to include two species introduced to Kansas and other new information.

The BCL2 family of proteins plays a pivotal role in regulating apoptosis and cellular homeostasis, making them critical therapeutic targets in cancer and other diseases characterized by pathological cell survival. BH3 mimetics, small molecules that selectively inhibit anti-apoptotic BCL2 family members, have achieved significant clinical success, particularly in hematologic malignancies. However, several challenges remain, including resistance mechanisms, toxicity (such as MCL1 inhibitor-associated cardiotoxicity), and the intricate balance between apoptotic and non-apoptotic functions.
