Red lentils are protein-rich sources that are highly regarded in the plant-based food industry for their color. Appropriate drying methods are essential for maintaining the physicochemical and nutritional properties of the proteins. This study investigates the impact of three drying methods, namely oven drying, freeze-drying, and supercritical carbon dioxide (SC-CO₂) drying, on the physicochemical and functional properties of red lentil proteins (RLP). SC-CO₂ drying was optimized at varying temperatures (35–55 °C), pressures (10–20 MPa), and CO₂ flow rates (2–3 L/min), with optimal drying observed at 55 °C, 20 MPa, and 3 L/min, resulting in significantly reduced moisture content (5.7 %). …

Cyclotides are plant-derived cyclic peptides recognized for their crucial role in host defense against insects and fungal pathogens. They are characterized by an ultra-stable head-to-tail cyclic backbone and a cystine knot formed by three conserved disulfide bonds, and are considered to be a promising group of molecules for a diverse array of agricultural and pharmaceutical applications. Cyclotides have been identified in five major plant families, with the Violaceae being a particularly rich source. Wheat is the most widely cultivated crop in the world, with over 215 million hectares grown annually. The 10+ Wheat Genomes Project recently sequenced and assembled to chromosome-level the genomes of nine wheat cultivars, uncovering genetic diversity and selection within the pan-genome of wheat. Here, we provide a wheat pan-transcriptome with de novo annotation and differential expression analysis for these wheat cultivars across multiple tissues. Using the de novo annotations we identify cultivar-specific genes and define the core and dispensable genomes. High-mountain lakes were historically fishless due to natural barriers, but human introductions have led to widespread fish presence. Although particularly intensive during the last decades, historical documents indicate introductions in European high mountains already during the 14th and 15th centuries, but they could have occurred before, provided the intensive land use of the high mountain had started earlier. We used ancient environmental DNA from lake sediments (sedDNA) to investigate this hypothesis. Pacific herring (Clupea pallasii) is a critical forage fish to the marine environment, economy, and culture of Alaska. However, despite the importance of Pacific herring in Alaska and throughout the North Pacific, knowledge gaps exist for foundational genomic characteristics of the species, including genetic population structure and the basis of genetic sex determination. In this thesis, I addressed these knowledge gaps using low coverage, whole genome sequencing. Glioblastoma (GB), acute myeloid leukemia (AML), chronic lymphocytic leukemia (CLL), cholangiocarcinoma, and chondrosarcoma (CS) cancers all contain mutations in the gene isocitrate dehydrogenase 2 (IDH2). The mutant IDH2 enzyme exhibits transformation of alpha-ketoglutarate (αKG) into the oncometabolite D-2-hydroxyglutarate (D2HG) in the mitochondria of these cancers. Mitochondrial-mediated transfer between cancer cells and recipient cells is a significant event that impacts the physiology of the receiving cell, specifically the epigenetic landscape. Dissolved organic matter (DOM) is a major reservoir of organic carbon in aquatic ecosystems and plays a central role in global carbon cycling through its transformation and remineralization to carbon dioxide (CO₂). As DOM moves from terrestrial environments into streams and rivers, its chemical composition is altered by a combination of microbial and photochemical oxidation processes that regulate both its reactivity and its persistence during transport. In the past decade, a surge in the amount of electronic health record (EHR) data in the United States occurred, driven by a favorable policy environment created by the Health Information Technology for Economic and Clinical Health (HITECH) Act of 2009 and the 21st Century Cures Act of 2016. Clinical notes for patients’ assessments, diagnoses, and treatments are captured in these EHRs in free-form text by physicians, who spend a considerable amount of time entering them. Oceanographic lidar systems remotely characterize the vertical structure of the upper ocean by recording the light backscattered from a laser pulse as it propagates through the water column. Historically these systems have been constrained by limited detection ranges, often capturing only a portion of the illuminated water column, typically fading to noise around the 10% isolume. A primary constraint has been the limited dynamic range of digitizers, which inhibits the ability to resolve both intense near-field and faint far-field backscatter signals within a single acquisition. Margalefidinium polykrikoides is a harmful cosmopolitan dinoflagellate that blooms in coastal waters. The effect of temperature and salinity on the growth of M. polykrikoides VA strain was examined using microscopy and growth models. M. polykrikoides Group III VA strain grew better at warmer temperatures and lower salinities than M. polykrikoides Group III NY strain, Group I Korea strain, and Japan strain (unknown group). Modelers need to use the temperature and salinity growth responses from M. polykrikoides Group III VA strain to better simulate and predict M. polykrikoides blooms in the Chesapeake Bay.

New strategies are urgently needed against antimicrobial resistance, a major health threat, and the different mechanisms regulating the bacterial cell division machinery offer multiple opportunities for developing novel therapeutics. FtsZ, an essential protein of this process, is targeted by multiple small molecules, and benzodioxane-benzamides (BDOBs) are among the most potent inhibitors in several bacterial species. BDOBs mechanisms are however poorly understood, particularly their impact on FtsZ's interplay with partners and ability to assemble phase-separated biomolecular condensates potentially involved in stress sensing. The BREAKWATER Phase III study investigated encorafenib and cetuximab plus mFOLFOX6 versus chemotherapy with or without bevacizumab for the treatment of patients with previously untreated BRAF V600E – mutant metastatic colorectal cancer. The study showed significantly improved objective response rate by blinded independent central review, and significantly longer progression-free survival by blinded independent central review and overall survival in patients treated with first-line encorafenib and cetuximab plus mFOLFOX6 compared with chemotherapy with or without bevacizumab. Dysfunction or dysregulation of deubiquitination is closely related to the initiation and development of multiple cancers. Targeted regulation of deubiquitination has been recognized as an important strategy in tumor therapy. However, the mechanism by which drugs regulate deubiquitinase is not clear. Here, we identified ubiquitin-specific peptidase 48 (USP48), a member of the ubiquitin-specific protease family highly expressed in various tumors, as a specific substrate for the activated caspase-3. During drug induced apoptosis of AML cells, activated caspase-3 cleaves USP48 through recognizing the conservative motif DEQD located at 611-614 sites of human USP48. Circadian organization is involved in many gastrointestinal tract (GIT) functions such as the maintenance of intestinal barrier integrity. There is compelling evidence that perturbation of the circadian clock decreases intestinal epithelial cells' resiliency to alcohol-induced injury. One of the most common causes of circadian misalignment is wrong-time eating (largest meal at dinner) in modern societies. Yet, few studies have examined the importance of peripheral circadian rhythms of the GIT to alcohol consumption. Introduction: Immunotherapy is firmly established as a treatment regimen in various solid tumors, driven by its exceptional benefits in a selected group of patients. Despite widespread adoption of immune checkpoint blockade (ICB) across diverse solid tumors, the quest for a clinically informative biomarker for long-term benefit remains unmet.

Methods: A total of 49 patients with metastatic NSCLC treated with ICB were included. Long-term (LTR) and short-term responders (STR) were defined as those with a response to ICB lasting more than 24 months or less than 6 months, respectively. Background: Meniscal preservation has been demonstrated to contribute to long-term knee health and has been a successful intervention in isolation and in patients with anterior cruciate ligament reconstruction (ACLR). The long-term results of meniscal repair in the setting of revision ACLR have yet to be documented.

Purpose: To report the incidence of meniscal repair failures at the 6-year follow-up in a cohort of patients who underwent concurrent revision ACLR and primary meniscal repair.

The small, tubulin-binding protein STMN2 is highly expressed in neurons and is implicated in amyotrophic lateral sclerosis. STMN2 degrades rapidly and accumulates at axotomy sites, suggesting fast turnover is crucial for its neuroprotective function. We show that STMN2 was primarily degraded by the ubiquitin-proteasome system. Its membrane-targeting N-terminal domain promoted fast turnover, whereas its tubulin-binding domain promoted stabilization. Proximity labeling and imaging showed that tubulin binding reduced STMN2 targeting to trans-Golgi network membranes. Pull-down assays showed that tubulin binds preferentially to soluble over membrane-bound STMN2. Background: Sensitivity and specificity are foundational metrics for cancer detection tools. However, most machine learning algorithms prioritize overall accuracy during optimization, which fails to align with clinical priorities of early detection. We aim to develop a feature selection machine learning algorithm while maximizing sensitivity at a given specificity.

Methods: We developed SMAGS-LASSO, a machine learning algorithm that combines our developed Sensitivity Maximization at a Given Specificity (SMAGS) framework with L1 regularization for feature selection. Cnidarians have become valuable models for understanding many aspects of developmental biology including the evolution of body plan diversity, novel cell type specification, and regeneration. Most of our understanding of gene function during early development in cnidarians comes from a small number of experimental systems including Hydra and the sea anemone, Nematostella vectensis. Objectives: Onconephrology is an expanding subspecialty focused on the management of cancer patients with renal injury. This study used a comprehensive bibliometric analysis to emphasize the need for cooperation between oncologists and nephrologists, exploring current trends and future research areas in onconephrology.

Methods: Relevant literature on onconephrology published between 1 January 2000 and 27 April 2024 was retrieved from the Science Citation Index Expanded of the Web of Science Core Collection, followed by manual screening. Bibliometric analyses were performed using CiteSpace, VOSviewer, and Bibliometrix software.

Fusobacterium nucleatum (Fn) is commonly enriched in colorectal cancer (CRC) and associated with poor outcomes, though its mechanisms remain unclear. Our study investigated how Fn affects the tumor microenvironment through single-cell transcriptomic analyses of 42 CRC patient tissues, comparing Fn-positive and Fn-negative tumors. We discovered that Fn impairs IgA plasma cell development and secretory IgA (sIgA) production by disrupting communication with tumor-associated macrophages. Statistical methods in clinical research tend to become entrenched. Innovations threaten the status quo. The "right way" becomes frozen in lore. This is so even when the "right way" is not best. "Statistical significance" and the associated requirement of "high power" is an example. This attitude is an impediment to efficient design. Willingness to address some design issues with moderate power enables building highly informative and highly efficient clinical trials. Background: Almost all mammals rely on the thymus and bone marrow to generate and differentiate B and T cells essential for adaptive immunity. A few members of the family Soricidae, or true shrews within Eulipotyphla, have also evolved an enlarged pancreas of Aselli, a kidney-sized organ hypothesized to serve this primary immune role, and whose gene expression profile is unknown. Results: Here we introduce transcriptomes of juvenile Eurasian common shrews (Sorex araneus, family Soricidae) pancreas of Aselli. The present investigation deals with the phytochemical, anticancer, antioxidant, anti-inflammatory, antidiabetic, and Anti-constipation activities of Ficus carica (FC) fruit. The phytochemical analysis of FC fruit has revealed a rich array of bioactive compounds, highlighting the fruit's complex and diverse natural chemistry together with some secondary metabolites that contribute to its high antioxidant activity which was evident from 1-Diphenyl-2-picrylhydrazyl (DPPH), 2,2′-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and Ferric reducing power (FRAP) assays. Fruit extracts of FC demonstrated anticancer activity against MCF-7 breast cancer, U2OS osteosarcoma, HepG2 liver cancer, and Huh7it liver cancer cell lines. Seagrass meadows play a key role in the global carbon cycle through storing carbon in their biomass and soils. However, the lack of global assessments of carbon stocks in seagrass biomass and net primary production (NPP) limits our understanding of their role in the carbon cycle. Here, we provide global estimates of biomass carbon stocks and NPP encompassing seagrasses with different life-history strategies, across bioregions and 66 countries. Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited treatment options due to the absence of estrogen (ER), progesterone (PR), and human epidermal growth factor receptor 2 (HER2) receptors. We previously developed highly adaptable polyamidoamine (PAMAM) dendrimer-based gel nanoparticles (GDP-uPA/GTI) with dual-targeting capabilities against urokinase-type plasminogen activator receptor (uPAR) and ribonucleotide reductase R2 (R2) to achieve selective antitumor effects. In this study, we investigated the underlying mechanisms of GDP-uPA/GTI-induced tumor inhibition. In vivo, GDP-uPA/GTI significantly downregulated R2 expression and activated caspase 8 and caspase 3, indicating caspase-dependent apoptosis. We present draft genome sequences of four Gram-negative phosphate-solubilizing microbes isolated from Pea Ridge Mine tailings in Missouri, USA. Belonging to the family Burkholderiaceae, these isolates exhibit a phosphate-mineral-dissolving phenotype in liquid and solid cultures. Genome sizes span 5.6–9.7 Mb, with GC contents of 61.6%–67.0%.

Increased interest and investigation into the sustainability of broiler production has led to a need for reliable and repeatable means to measure water intake and its conversion rate into salable meat. Therefore, a study was conducted to characterize the water intake and water conversion ratio (WCR) of male and female broilers from four modern broiler strains. Labor challenges are underpinning large multinational farm machine manufacturers' development of autonomy solutions for their large-scale machine offerings. This study simulates a linear optimization model to examine the economics of large-scale autonomous machines for a rotational maize and soybean farm in the Midwest US. Results support the hypothesis that autonomous machines can be economically viable for farms facing severe labor shortages. However, under current technology and pricing structures, conventional mechanization remains the most profitable option for farms with reliable labor.
