Mutations in the tumor suppressor gene TP53 have been identified in breast cancer-associated fibroblasts and are associated with poor patient prognosis. However, the functional impact of fibroblastic mutant p53 on breast cancer development remains unclear. To investigate this, we compared female mice harboring HER2-driven mammary tumors with a fibroblast-specific Trp53 mutation (NP) to those with wild-type fibroblastic Trp53 (N). NP mice exhibited significantly shorter median tumor-free survival than N mice. RNA sequencing of NP and N tumors and mammary glands revealed numerous differentially expressed genes (DEGs) between tumors and the corresponding glands in both genotypes. Escalated doses of radiotherapy associate with improved local control and overall survival (OS) in intrahepatic cholangiocarcinoma (iCCA), but personalization remains limited because conventional size-based CT criteria correlate poorly with outcomes. We hypothesized that quantitative enhancement measurements would better predict clinical outcomes and guide individualized RT optimization. In a retrospective cohort of 154 patients, we analyzed pre- and post-RT CT scans using quantitative European Association for Study of Liver (qEASL) to derive viable tumor volumes, comparing enhancement-based metrics with size-based RECIST and linking them to outcomes via survival and mathematical modeling. We present a narrative review focusing on pulses’ geographical origin and distribution, their impact on human evolution and history, and their influence on human health. Pulses, including dry peas, beans, and lentils, are renowned for their richness in chemical antioxidants. Despite containing antinutrients, processing techniques preserve their health advantages. Epidemiological research has consistently demonstrated that the consumption of pulses is associated with favorable effects on metabolism. This evidence is further supported by molecular and clinical research, which has elucidated potential nutrigenomic mechanisms and effects on gut microbiota composition underlying their health benefits. Tea (Camellia sinensis (L.) O. Kuntze) is a major economic crop in Indonesia, yet its productivity is strongly influenced by environmental variability, particularly rainfall seasonality. Fluctuating yields under dry and wet conditions pose challenges for sustainable tea production. This study aimed to assess the effects of contrasting seasonal rainfall on seed-derived tea plants' yield and yield components. The Galápagos archipelago is known for its unique endemic fauna shaped by isolation and geological dynamism. This thesis explores the evolutionary history of Galápagos iguanas, a monophyletic group comprising the marine iguana (Amblyrhynchus cristatus) and three land iguanas: Conolophus marthae, C. pallidus, and C. subcristatus. These species differ in distribu- tion, population size, and conservation status, with C. marthae classified as Critically Endan- gered due to its restricted range and small population. Insect frass, a by-product of insect rearing, represents a rich organic carbon source with diverse microbes that can enhance soil health and promote beneficial plant-microbe interactions. The genus Melanoplus represents one of the most diverse groups of acridid grasshoppers in North America, comprising over 430 described species within the subfamily Melanoplinae. Among its various species groups, the Tribulus group consists of flightless grasshoppers typically found in open woodlands and forest margins, primarily in the eastern United States. The Melanoplus tribulus group was first established by Blatchley (1920) as Obovatipennis Group. However, since species groups are informal taxonomic units not governed by nomenclatural rules, the group later became known as the Tribulus group. It currently includes 11 taxa. Studies were conducted in Oktibbeha and Clay Counties in Mississippi to determine the response of green antelopehorn milkweed and hemp dogbane to triclopyr and other auxin herbicides in non-cropland areas. These studies were conducted at four locations with well established populations for each species throughout 2020 to 2023, with applications repeated at each location the following year if populations persisted. Treatments included a high and low rate of each herbicide studied, with all four triclopyr formulations included for both species. Beef cattle routinely undergo a variety of stressors within the preconditioning and feedlot receiving phases which may impair animal performance and health. Trace mineral supplementation, either through dietary or injectable sources, is proposed to mitigate these effects through metabolic and immune system support. Two systematic reviews and meta-analyses were conducted to evaluate the efficacy of two different oral supplementation methods, organic (OTM) and inorganic (INR), and injectable trace mineral (ITM) solutions on average daily gain (ADG) and morbidity. Across 20 studies, OTM supplementation increased ADG (P = 0.01) when compared to INR, but did not affect morbidity (P = 0.92). …

Mycobacterium ulcerans, the causative agent of Buruli ulcer, produces mycolactone, a polyketide toxin traditionally studied for its role in human pathogenesis. This dissertation presents a comprehensive spatiotemporal analysis of M. ulcerans and mycolactone producing mycobacteria (MPM) ecology in the Americas and also reveals mycolactone's role in the environment as a microbial community regulator. Through comparative studies in French Guiana (endemic) and the southeastern United States (non-endemic), combined with experimental microcosm investigations, this research contributes to environmental pathogen ecology understanding. Apple peel color is one determining factor of apple quality and is a highly sought after trait by both growers and consumers. Through-hikes on long distance backpacking routes have the potential to both improve physical fitness and pose health risks. PURPOSE: To quantify changes in cardiorespiratory fitness and body composition, while recording dietary intake and sleep habits in a sample of through-hikers. METHODS: Participants completed two lab visits consisting of a maximal graded treadmill test and body composition assessments. This case study was intended to analyze the impact of a knee injury prevention program (KIPP) for a young female athlete in a short recreational softball season. Temporal partitioning is one method that wildlife may employ to persist among the increased predation, competition, and human presence pressures of urban spaces. Food safety strategies are being developed in the hopes of combating foodborne pathogens and their risks. Previous techniques and technologies are struggling to meet the needs of the public, leading to the demand for new, innovative methods. Mild high-pressure processing (HPP) of 350 MPa was used in a synergetic combination with mild heat of 60°C and the antimicrobial carvacrol for the decontamination of non-typhoidal Salmonella serovars that resulted in a >5 log CFU/mL reduction. Chickpea proteins offer significant nutritional and functional benefits in food products; however, their utilization is often restricted by off-flavors, primarily for lipoxygenase (LOX) and lipase activity. This study aims to develop a novel extraction method by combining acid (pH 2.0) extraction with the conventional alkali (pH 9.0) method to inactivate enzymes and enhance protein quality. Acid-extracted proteins exhibit the highest purity (83.4%), albumin content, and structural flexibility (46.65% random coil). Both acid and sequential acid-alkali-extracted proteins significantly reduced LOX (0.75 and 0.88 U/g) and lipase (0.10 and 0.08 U/g) activity. The endoplasmic reticulum (ER) stress sensor inositol-requiring enzyme 1α (IRE1α) is an important regulator of innate immune cells. However, its role in pulmonary innate host defenses remains poorly understood. This dissertation elucidates the dual, context-dependent role of IRE1α in host defense against methicillin-resistant Staphylococcus aureus (MRSA) pneumonia. We demonstrate IRE1α’s function as both a detrimental mediator of acute pathology and an essential facilitator of innate immune memory. Our work revealed that acute IRE1α activation during primary MRSA infection is detrimental. In lungs and alveolar macrophages (AMs), IRE1α is activated in response to MRSA infection. Tributyltin (TBT), Dibutyltin (DBT), and Pentachlorophenol (PCP) are persistent environmental contaminants that have been shown to increase immune-cell production of pro-inflammatory cytokines and thus have the capacity to cause chronic inflammation and its associated pathologies such as cancer, cardiovascular disorders, and autoimmune conditions. TBT has been detected in human blood at concentrations up to 260 nM, DBT up to 0.3 µM, and PCP between 0.15 and 5 µM. Tumor necrosis factor-alpha (TNF-α) is a pro-inflammatory cytokine produced by immune cells in response to pathogen- or damage-associated molecular patterns (PAMPs/DAMPs) via Toll-like receptors (TLRs). Cowpea (Vigna unguiculata L. Walp) is an annual legume of immense importance across the world. However, knowledge of its ability to tolerate aluminum (Al) at toxic levels is not as comprehensive. This stress is an issue globally and especially in the southeastern United States, which has many Al-toxic soils. Here we determined multiple cultivars both tolerant and susceptible to Al toxicity. This was expounded upon by employing RNA sequencing on two cultivars from the United States with significantly different tolerance levels to characterize genes and pathways associated with Al toxicity and response. Cover cropping has emerged as a climate adaptation strategy in sustainable agriculture to enhance agroecosystem resilience against weather extremes such as floods, droughts, and erratic rainfall patterns. But how well cover crops (CCs) can buffer crop yields against weather variability depends on their ability to regulate improvements in soil hydrology. Through a global meta-analysis of 1007 pairwise observations from 146 studies spanning across 34 countries, we investigated the overall effects of CCs on six key soil hydraulic properties and further examined how environmental and management factors moderate these overall responses. The farm bill of 2014 and 2018 legalized hemp (Cannabis sativa L.) production for cannabidiol (CBD), grain, and fiber uses in the U.S. Agronomic management practices for floral hemp for CBD production were not well studied. To optimize the agronomic management practices for high CBD content and THC concentration below the compliance level of 0.3% on a dry weight basis requires specific management and adaptability. This study estimates the economic contributions of the greenhouse and nursery industry, commonly known as the "green industry," in Tennessee for 2023. Utilizing data on employment, labor income, value added, and industry output, alongside economic multipliers generated from the Input-Output modeling software, IMPLAN (Impact Analysis for Planning), we provide a comprehensive analysis of the green industry's impact on the state's agricultural economy. The findings are presented in 2025 dollars, highlighting the industry's significance for informing future policy decisions. Overexploitation of antibiotics in the animal industry has driven the need for reduced usage, but this reduction has intensified the challenge posed by Salmonella enterica and Campylobacter jejuni to livestock systems and public health. This situation underlines the need for sustainable antimicrobial alternatives. Although cyanobacteria are promising in this respect, since they may produce a variety of bioactive metabolites, the amounts of these metabolites in their natural state are insufficient to elicit potent antibacterial action. Purinergic signaling relies on ATP release through exocytosis and large-pore channels. Large-pore channels permeate both small anions like chloride and large signaling molecules like ATP, but how this broad cargo selectivity is structurally controlled remains elusive. Here we investigate PANX1, a prototypical large-pore channel, and uncover structural plasticity at the extracellular entrance formed by seven tryptophan (W74) residues. The W74 sidechains are flexible, sampling conformations that range from a constricted state permissive only to chloride to a dilated state compatible with ATP. These states are coupled to variable cation-π interactions between W74 and arginine 75 (R75), suggesting a mechanism for …

Falls are a leading cause of injury and loss of independence in older adults, often linked to deficits in lower-limb muscle function and gait mechanics. Eccentric exercise can improve muscular resilience, while aquatic walking offers a safe, supportive environment to retrain gait; however, little is known about how these modalities interact at the neuromuscular level. Microcrystalline tests are chemical tests that involve the mixing of a substance with a specific reagent resulting in the formation of a crystal structure. Currently, microcrystalline tests are used as presumptive tests in forensic drug chemistry laboratories to provide the suspected identity of unknown substances. There is, however, a limited amount of research on this use of microcrystalline structures. Additionally, this existing research is outdated and does not include some of the newer commonly encountered controlled substances including fentanyl, clonazepam, and diazepam.
