The gut microbiota has significant effects on the performance, health status, and immune systems of chickens. The complex microbial community structure begins to form after chicks hatch with the intake of feed, water, and air into the digestive system. Disrupting the balance of this system can lead to the proliferation of pathogenic microorganisms. Until recently, studies on microbiota relied on traditional microbiological techniques that cultured only a small fraction of the microbial diversity. However, significant advancements in DNA sequencing and bioinformatics have enabled the identification of diverse bacterial populations in environmental samples. In this study, a total of 2000 broiler …

Bacterial Chondronecrosis with Osteomyelitis (BCO) lameness is an infection of opportunistic bacteria in the structural skeletal bones impacting multiple animal species, particularly poultry species. BCO lameness results in significant financial losses to industrial poultry production and increases the risk of foodborne illnesses, posing a major threat to consumers' food safety. As BCO lameness is an inherent risk of fast body weight gain in poultry species, especially broiler chickens, abundant studies have been conducted in broilers and turkeys. Nevertheless, BCO lameness incidence in ducks remains elusive. Thus, this is the first survey investigating the prevalence of BCO lameness cases in ducks. …

Best Integrated Writing
This paper examines the role of eco-feedback systems in promoting pro-environmental behaviors by providing users with easily accessible information about their resource consumption. While these systems have shown potential to reduce consumption, they can face difficulties in maintaining user engagement and achieving long-term behavior change. There are several key factors related to the design of feedback interfaces that influence the effectiveness of eco-feedback systems. Gamification has been proposed as an effective way to enhance user engagement but requires careful design to connect behaviors from the digital to the real space for learning transfer. Research on the long-term usage of these …

Algal biofuels have been investigated as an alternative to fossil fuels and first-generation biofuels for transportation in the United States since the 1970s. Yet after five decades of development, scalability and implementation remain limited—largely due to persistent barriers such as low biomass productivity, modest lipid yields, and energy-intensive processing methods. These technical challenges significantly constrain the feasibility of large-scale commercialization despite substantial research and investment. To evaluate progress toward commercial viability, this study harmonized energy inputs and outputs across 508 observations on the production of algal biofuel energy return on energy investment (EROEI) in the United States. While bioethanol achieves …

Selective autophagy of mitochondria is known to promote cancer cell survival and progression, including in triple-negative breast cancer (TNBC). Here, we apply an integrated multi-omics approach together with functional experimental analyses to investigate metabolic adaptations that support mitochondrial quality control in TNBC. We detail a mitochondrial quality control mechanism, complementary to mitophagy, that is enabled by a program of heightened extracellular sphingomyelin salvaging in TNBC coupled with extracellular vesicle-mediated intracellular clearance of mitochondrial damage. Targeting of this onco-metabolic pathway via repurposing of eliglustat, a selective small molecule inhibitor of glucosylceramide synthase, results in ceramide-mediated compensatory mitophagy and cancer cell death …

Mapping the entire transcriptome at single-cell resolution under its natural spatial context is essential for investigating the oncogenesis and progression of diseases. The recently emerged targeted in-situ technologies retain the spatial organization of cells at high resolution, although they remain limited in the number of genes that can be simultaneously measured. To overcome this obstacle, numerous computational methods have been developed to predict unmeasured gene expression in spatial transcriptomics data by leveraging scRNA-seq data. Most of these methods focus on the expression of individual genes and usually generate highly variable predictions. In this study, we introduce PASTA (PAthway-oriented Spatial gene …

Treatment of lung adenocarcinomas (LUADs) that exhibit activated epidermal growth factor receptor (EGFR) with EGFR tyrosine kinase inhibitors (TKIs) has limited efficacy. Assessment of the impact of EGFR TKI on the LUAD surfaceome remodeling reveals potential therapeutic targets. We identify placental type alkaline phosphatase (ALPP), which has restricted expression in normal tissues, among upregulated surface proteins following EGFR TKI treatment of both TKI sensitive as well as resistant cells. EGF treatment represses ALPP expression, whereas EGFR TKIs upregulate its expression through dephosphorylation and activation of FoxO3a, a transcriptional regulator that binds to the promoter region of ALPP. The combination of …

Immunological intratumor heterogeneity (ImTH) describes the variability in the types, spatial distribution, and functional states of immune cells within tumors. While evidence suggests that ImTH influences tumor progression and therapeutic response, few studies have provided a quantitative characterization of ImTH. Here, we present Scoring Immunological Intratumor Heterogeneity (ScImTH), a novel algorithm that quantifies ImTH by calculating the Shannon entropy of immune cell type proportions within the tumor microenvironment. Using bulk, single-cell, and spatial transcriptomic datasets, we show that reduced ScImTH scores are associated with unfavorable survival outcomes, tumor progression-related molecular and phenotypic features, immunosuppressive states, and resistance to immunotherapy across …

Leydig cells are anatomically located in the testicular interstitial tissue, and their main function is to produce and secrete testosterone and indirectly support spermatogenesis. We previously reported that the combination treatment of cordycepin and radiation can effectively induce Leydig tumor cell apoptosis through cell cycle arrest, caspase activation, endoplasmic reticulum (ER) stress, reactive oxygen species (ROS) accumulation, and DNA damage. However, there is still a lack of scientific evidence for the susceptibility of normal Leydig cells to the combination treatment. In the present study, mouse TM3 Leydig progenitor cells were used as a model to evaluate the effects and mechanisms …

Gut microbiota has recently gained attention for its role in regulating multiple host pathways and contributing to disease developments. Fecal metabolomics using liquid chromatography—mass spectrometry (LC—MS) offers a promising approach to study gut microbial metabolites; however, it remains technically challenging due to the complex, heterogeneous nature of fecal samples and the lack of standardized protocols. This study aimed to establish a robust and reproducible untargeted fecal metabolomics workflow. We systematically evaluated sample preparation parameters―including sample amount, extraction solvent, numbers of extraction, and sample-to-solvent ratio―and assessed method reproducibility. Additionally, we compared three LC—MS data acquisition workflows using 10 samples from inflammatory …

In this study, we investigated the impact of the SGLT2 Inhibitor empagliflozin (EMPA) on diabetic hearts and its regulatory mechanisms in high-fat-diet (HFD)- and streptozotocin (STZ)-treated rats. Male rats orally administered HFD/STZ treatment for eight weeks, with or without EMPA (10 mg/kg), were used as our in vivo model. Hematoxylin and eosin (H&E) staining was used for histological examination. Western blot analysis and immunohistochemistry were used to analyze the expression of …

Lung cancer is the leading cause of cancer-related deaths worldwide, driven by carcinogens such as tobacco-derived nitrosamines, dioxins, and urethane. This study evaluated the chemopreventive effects of two monoterpenes, α-pinene and D-limonene, using NNK/TCDD- and urethane-induced lung tumor models and A549 lung cancer cells. Both compounds significantly reduce tumor number and size, with enhanced effects when combined. Mechanistic studies revealed suppression of PRC1, β-catenin, and c-Myc, alongside activation of p53, Bax, and caspase-3, indicating inhibition of the PRC1—Wnt/β-catenin pathway and induction of apoptosis. These findings suggest α-pinene and D-limonene as safe, promising agents for lung cancer chemoprevention.
