Analyzing The Effects Of Message Testing On The Public's Opinion About Gene Editing In Agriculture And Medicine, Luke Sanford Wimpy

This paper will present results from the final survey of a multi-stage project examining factors that influence opinions about gene editing in agriculture and medicine. In this final survey, 3,200 U.S. respondents were recruited and randomized to view one of eight videos that communicated the safety of gene editing to determine which factors are most influential in changing opinions. The eight videos were designed based on data collected from focus groups and three previous surveys in this multi-stage project. The eight videos varied by three attributes: messenger, message, and application. The messenger in the videos was either a researcher or …

Over the past several decades, cold- and cool-water fish populations across the Upper Midwest, including Minnesota and Wisconsin, have shown troubling signs of decline. Among the most affected is the walleye/Ogaa (Sander vitreus), a species deeply embedded in the ecological balance, cultural identity, and economic vitality of the region, and central to the lifeways, treaty-reserved rights, and resource stewardship of Tribal Nations, including Ojibwe communities across the Upper Midwest. Increasing land costs and labor constraints continue to challenge beef producers. More intensive practices can increase harvest efficiency; however, many producers are deterred from implementing them due to increased labor and management considerations. Virtual fencing (VF) may provide a reduced labor alternative to traditional fencing for managing intensive grazing. This dissertation evaluated management strategies and VF technology to improve grazing efficiency of stocker cattle systems. In Experiment 1, VF was used to effectively manage groups of steers grazing in adjacent paddocks separated by only virtual boundaries. Heifers represent a large proportion of cattle placed in feedlots, making pregnancy management an important consideration for maintaining performance and carcass value. Melengestrol acetate (MGA) is commonly included in feedlot diets to suppress estrus and prevent pregnancy in intact heifers during the finishing period. Alternatively, spaying has historically been utilized as a management practice to allow heifers to graze alongside breeding cattle without the risk of pregnancy. Phase-change nanodroplets (PCNDs) continue to generate significant research interest due to their potential to extravasate into tissue, to be targeted for molecular imaging and drug delivery, and to undergo an induced phase-change to "activated" microbubbles (MBs) for ultrasound (US) imaging. To accurately quantify molecular markers, however, one assumes a consistent proportion of PCNDs in a region of interest (ROI) are stably activated and imaged. Herein we present a framework for developing a diagnostic sequence that is optimized for PCND activation uniformity, contrast, and acquisition time. Purpose: Radiation therapy (RT) has emerged as an effective local therapy option for patients with unresectable hepatocellular carcinoma (HCC) and is associated with improved overall survival (OS). However, some patients still have poor outcomes after RT, and biomarkers are needed to stratify prognostic groups. High circulating growth hormone (GH) levels promote HCC proliferation and survival and are associated with aggressive disease. In this study, we evaluated whether pretreatment GH levels are associated with OS and progression-free survival (PFS) in patients who underwent RT for unresectable HCC.

When an individual is found deceased, one of the tools used in the identification process is estimating the post-mortem interval (PMI). This estimate is used to determine how long an individual has been deceased once their remains are found. To estimate an accurate PMI, knowing how decomposition is affected by external factors is key. Over the last thirty years the mortality rate of the undocumented population at the US-Mexico border has risen, requiring methods distinguishing US-born individuals from immigrants to improve the likelihood of identification and repatriation through foreign consulates. Forensic anthropologists with the Pima County Office of the Medical Examiner (PCOME) have advocated for the use of the biocultural profile, combining the biological profile with factors including oral health, osteological stress, and personal effects. Mitochondria are dynamic organelles that can fragment or fuse to support different bioenergetic demands and cellular processes, although the role of mitochondrial fission and fusion during embryonic development is not well understood. Fluorescence lifetime imaging (FLIM) of the mitochondrial cofactor NADH can be used to visualize mitochondrial networks and infer aspects of cellular bioenergetics in a label-free manner. We used NADH FLIM to test whether germband cells undergo changes in cellular metabolism during Drosophila convergent extension (CE)––a process in which hundreds of epithelial cells undergo coordinated intercalation to elongate the embryo. Campylobacter jejuni is a leading cause of foodborne illness and a major trigger of Guillain–Barré Syndrome (GBS), an autoimmune neuropathy caused by molecular mimicry between bacterial lipooligosaccharides (LOS) and host nerve gangliosides. While prior genomic studies have focused primarily on human clinical isolates, the prevalence and distribution of GBS-associated genes in non-human reservoirs remain poorly characterized. This study used genomic datasets derived from analyses of human and non-human C. jejuni isolates to assess the distribution of GBS-associated genes, with a focus on the neuABC sialylation pathway. Abstract (also on poster): Previous studies in our lab show that an alternate isoform of Bruton’s tyrosine kinase, BTK-C, is required for the survival of several epithelial-derived cancer cells. BTK-C is transcribed from an alternative promoter and includes a 34-amino-acid N- terminal extension with a palmitoylation sequence that alters its subcellular localization. Nine other human kinases have similar alternative N-termini (ANT), often producing palmitoylated or non-palmitoylated isoforms from different transcriptional start sites. Among these, ABL1 is a known oncogenic kinase and key drug target. Translocation mutations at the 5′ end of ABL1 are a major cause of adult leukemias. This …

This study compared five solvents - urea, carbonate-bicarbonate, citrate, phosphate, and water - for their ability to extract proteins from beech (BM) (Hypsizygus marmoreus) and Swiss brown (SB) (Agaricus bisporus) mushrooms. Using mass spectrometry-based proteomics and bioinformatics, we found that urea produced the highest total protein identifications by data-dependent acquisition at 1% false discovery rate (FDR): BM: 1739, SB: 1907; and by data-independent acquisition: BM: 1969, SB: 2172 proteins, especially enriching cytosolic and ribosomal proteins. Carbonate-bicarbonate favoured the extraction of metabolic and translational proteins, while citrate targeted extracellular and membrane-associated proteins related to cell wall dynamics. Clostridioides difficile is a major cause of healthcare-associated diarrhea. Infection typically occurs after antibiotic treatment disrupts the gut microbiome, allowing C. difficile to colonize and produce toxins leading to disease (CDI). Standard antibiotic treatments can resolve acute infection but frequently lead to recurrence, highlighting the need for alternative strategies that restore the gut ecosystem. Fecal microbiota transplantation (FMT), the transfer of stool from a healthy donor, has emerged as an effective therapy for recurrent C. difficile infection (rCDI). Understanding the mechanisms underlying the success of FMT for treatment of rCDI could lead to safer, more defined microbial mixtures.

Seagrass meadows, though covering a small percentage of the ocean floor, play a critical role in marine carbon sequestration. However, the mechanisms behind this role remain uncertain due to challenges in mapping seagrass extent and flux, accounting for spatial variation in carbon storage, and incorporating site-specific histories. This study investigates the effects of short-term (≤10 years) seagrass loss on sediment carbon stocks within a large seagrass meadow in the Great Barrier Reef region. Habitat condition and area shape global species distributions, with shallow-water reefs hosting a disproportional share of marine biodiversity. Although reef area is a well-established predictor of marine species richness, its historical context is less well understood. Here, we show that the rise of tropical marine biodiversity is closely tied to reef expansion in space and time. During the Early/Mid Miocene (23 to 11.6 million years ago), Indo-Pacific reefs reached unprecedented size and thickness, surpassing any reef systems in the past 66 million years. Growth anomalies (GAs) are coral diseases characterised by tumour-like skeletal lesions reported globally, yet their causes remain poorly understood. Microorganisms are integral to coral health, but the role of bacterial communities in GAs remains unclear. We investigated an outbreak of GAs in Isopora palifera at the Cocos (Keeling) Islands using 16S rRNA amplicon sequencing to compare bacterial communities of GA-affected and asymptomatic corals, surrounding water and potential pollution sources. After harvesting the animal, pork muscle continues to undergo changes as it turns from muscle into meat. During this early period, the muscle uses up energy and produces acid, which lowers pH and can affect important quality traits such as color and how well the meat holds water. A key driver of these early changes is calcium, a natural mineral inside muscle cells that helps control contraction and energy use. In this study, we tested whether mitochondria (the cell’s “powerhouse”) help control calcium after harvest and, in turn, influence early postmortem changes in pork. In water limited environments, alfalfa is often criticized for its high water use, prompting implementation of new irrigation technologies, deficit irrigation, and adapted crop genetics. Cumulative potential benefits from combining, or stacking, these water-optimizing practices have not been identified. Serum-containing media has been shown to increase incidence of large offspring syndrome and to alter gene expression, protein synthesis, and cell signaling. These factors together can result in increased pregnancy loss when combined with assisted reproduction technologies. However, there are very few direct comparisons of commercial serum-free media (IVF Bioscience™) and serum-containing media on both IVF and cloned embryo development in cattle. The aim of this study was to determine the role of serum in oocyte maturation and subsequent embryo development. Sheep producers in the Intermountain West region of the US rely on productive rangeland ecosystems that vary widely in forage composition and nutrient availability. As such, supplementation is often necessary to offset dietary deficiencies. Soils and forages grown on rangelands are known to be highly variable in mineral content. Even so, a third to half of the producers in the Intermountain West choose not to provide a complete mineral supplement. Cetaceans, despite their ability to cross wide distances, enabling mostly high levels of gene flow, can develop unique genetic lineages driven by variations in life-history traits, diverse migratory routes and local adaptation, complicating taxonomy and delineation of conservation units. Comprehensive genetic data are essential to overcome the challenges of obtaining suitable sample sizes from wild cetaceans while untangling cryptic species diversity. The Bryde's whale (Balaenoptera edeni) has a complicated discovery and taxonomic history that hinders accurate conservation assessments. Cheese ripening is a complex process that shapes the flavor, texture, and overall quality of cheese, yet many of the factors that control how these changes occur are still not fully understood. One factor that may play an important role is redox potential, which describes the balance between oxidation and reduction reactions happening inside the cheese. These reactions can influence both the activity of bacteria and the chemical processes that create flavor. Past efforts to control redox using chemical additives often harmed beneficial bacteria, limiting their usefulness.
