Sequencing Of Chenopodium A-Genome Diploids To Identify Potential Mechanisms Of Speciation: A Preliminary Study, Riley Roser Jun 2026

Sequencing Of Chenopodium A-Genome Diploids To Identify Potential Mechanisms Of Speciation: A Preliminary Study, Riley Roser

Theses and Dissertations

The genus Chenopodium is of increasing significance, mainly due to allotetraploid quinoa (C. quinoa Willd.,2x = 4x =36, AABB genome) becoming globally recognized for its high nutritional value and tolerance for stress. Diploid A-genome relatives of quinoa in the Western Hemisphere have shown speciation patterns congruent to adaptive radiation, particularly in the North American continent, where a number inhabit ecological niches while others form mixed populations where they are sympatric. Characterization of the genetic mechanism(s) underlying this rapid speciation, could clarify their potential usefulness for improving quinoa . Previous whole-genome assemblies of AA diploids C. watsonii A. Watson and C. …

Abstract – Bactrocera dorsalis is a major horticultural pest. Biocontrol using parasitoids, such as Fopius arisanus, is a sustainable tool for managing this pest. However, the mechanisms enabling the successful parasitism of B. dorsalis are poorly understood. Using transcriptomic profiling, cellular bioassays, and targeted metabolite analyses, we investigated how F. arisanus modulates the immunity and metabolic activity of B. dorsalis to achieve a high parasitism rate. Our results revealed a weak and late encapsulation response of B. dorsalis towards F. arisanus. Additionally, parasitism reduced the number of circulating haemocytes by 35%, lowered constitutive granulocyte counts and inhibited phenoloxidase activity in …

Proteomic Evidence Indicates Ultrasound-Assisted Extraction Enhances Recovery Of Membrane-Associated Proteins In Hericium Erinaceus, Tanvir Ahmed, Angéla Juhász, Utpal Bose, Netsanet Shiferaw Terefe, Michelle L. Colgrave

Research outputs 2022 to 2026

Membrane-associated fungal proteins mediate essential functions and contain bioactive proteins with therapeutic potential but are underrepresented in aqueous extracts due to hydrophobicity and lipid associations. Ultrasound-assisted aqueous extraction was applied to Lion’s mane (Hericium erinaceus), and the proteins were fractionated by molecular weight (≥30 kDa, 10–30 kDa, 3–10 kDa). Proteomic profiling using data-dependent (DDA) and data-independent (DIA) mass spectrometry identified 1500 and 1702 proteins, respectively (1% global FDR). Ultrasonication induced surface irregularity and a less compact, heterogeneous microstructure. Topology predictions showed higher proportions of transmembrane and secretory proteins in ultrasonicated fractions. Extraction preserved protein integrity (82% molecular-weight retention, 97% tryptic …

Examining The Relationship Between Civic Agriculture Participation And Civic Engagement In South Texas, Allison Kaika, Dongkyu Kim, William Donner, Alexis Racelis, Owen Temby

School of Earth, Environmental, & Marine Sciences Faculty Publications

Civic agriculture is often credited with strengthening democratic life by encouraging civic and political engagement, yet this claim has scarcely been tested in socioeconomically marginalized regions. Using original survey data collected through a combination of random and purposive sampling, this study assesses that relationship in the Lower Rio Grande Valley of Texas, a majority-Hispanic agricultural region marked by high poverty and low civic participation. Phone and online surveys were administered in English and Spanish to reach both the general population and individuals involved in civic agriculture initiatives, yielding over 400 responses. We examine whether civic agriculture participation predicts greater community …

Agrivoltaic Shading Reduces Soil Greenhouse Gas Fluxes In A Semi-Arid Vineyard, Marcelo Gomes Da Silva, Rafael M. Almeida, Carine Cocco, Engil Pereira

School of Earth, Environmental, & Marine Sciences Faculty Publications

Agrivoltaic systems are expanding in semi-arid regions, raising questions about how panel driven shading alters soil greenhouse gas exchange. We used automated chambers to measure high frequency soil CO2, N2O, and CH4 fluxes in a newly established vineyard agrivoltaic system in Texas across three shading intensities (0%, 35%, 88%) that represent the light environment beneath panels as shaped by panel tilt and solar trajectory. Shading primarily cooled the soil, reducing mean soil temperature from 35.9 °C (0%) to 32.5 °C (35%) and 30.5 °C (88%), while mean soil water content shifted modestly (0.125, 0.106, and 0.130 m3 m−3, respectively). Over …

Cartilage extracellular matrix (ECM), a hydrated collagen II-aggrecan composite, undergoes dynamic turnover during both normal homeostasis and disease-associated remodeling. This study elucidates a crucial role for decorin in promoting the retention and stability of nascent aggrecan within this matrix. By applying bio-orthogonal click-labeling, we demonstrate that loss of decorin accelerates the release of nascent aggrecan under both physiological and inflammatory conditions, without affecting its preferential localization to the pericellular matrix. Conversely, supplementation with exogenous decorin mitigates inflammation-induced loss of nascent aggrecan, supporting its potential as a therapeutic target. At the molecular level, decorin exhibits strong binding affinity for aggrecan, and …

Why Communicating Minimum Drawdown Elevations For Lake Powell And Lake Mead Can Mitigate Risks Of Volatile Colorado River Flow., Brittany Fager, David Rosenberg, Erik Porse Jun 2026

Why Communicating Minimum Drawdown Elevations For Lake Powell And Lake Mead Can Mitigate Risks Of Volatile Colorado River Flow., Brittany Fager, David Rosenberg, Erik Porse

Reports

There is consensus across Reclamation’s hydrologic ensembles that volatile Colorado River flow will continue, including annual drops of 6 million acre-feet of natural flow above Lake Powell. This volatility poses risk that water managers cannot respond fast enough with the consequence that Lake Powell and Lake Mead drawdown so they no longer fulfill their Congressionally legislated purposes: deliver water, generate hydropower, protect native fish, or sustain infrastructure. In this post we suggest four reservoir operational strategies to reduce all those risks: (1) Identify Catastrophic Elevations where we lose the ability to operate reservoirs for one or more legislated purposes, (2) …

Effects Of Chronic Experimental N Deposition On Northern Hardwood Forests Fade After Cessation Of N Additions, Eileen C. Reeves, Andrew Burton, Katherine E. Higdon, Jennifer R. Eikenberry Jun 2026

Effects Of Chronic Experimental N Deposition On Northern Hardwood Forests Fade After Cessation Of N Additions, Eileen C. Reeves, Andrew Burton, Katherine E. Higdon, Jennifer R. Eikenberry

Michigan Tech Publications

Background: Anthropogenic nitrogen (N) deposition has the potential to affect forest ecosystem carbon (C) storage and sequestration both positively and negatively. Effects on aboveground biomass increment seem to be largely positive, but the impact of elevated N inputs on soil microbial systems in forests is often negative, reducing microbial biomass and soil respiration rates. Twenty-four years of experimental N deposition at a rate of 3 g N m−2 y−1 broadly suppressed soil microorganisms across four northern hardwood forest sites. To assess responses to reductions in chronic N inputs, soil and sugar maple root samples from the study sites …

Validated Crispr/Cas9 Guide Rnas Targeting Neurodevelopmental Genes In The Tunicate Ciona Robusta, Sydney Popsuj, Tenzin Kalsang, Eduardo D. Gigante

Faculty Articles

The tunicate Ciona robusta provides a powerful and simplified model for dissecting the genetic control of developmental and cell biology. With a larval CNS composed of just over 200 neurons and sensory cells, it has also emerged as a model organism for neurobiology and the development of the nervous system. Although CRISPR/ Cas9-mediated mutagenesis is now routinely used in Ciona as an important technique used to interrogate gene function in diverse biological processes, validated single-guide RNAs (sgRNAs) have yet to be validated for several key neural genes. Here, we report the design and experimental validation of 25 novel sgRNAs targeting …

Intramuscular Nad⁺ Therapy And Neuroenergetic Symptom Modulation: An Exploratory Retrospective Human Pilot Study, Zachary J. Dickey Do, Kirtan Patel Bs, Randy Aldret Edd, Cscs, Atc, Navneet Sharma Md, Glenn Goodwin Do

Journal for Sports Neuroscience

Abstract

Background: Nicotinamide adenine dinucleotide (NAD⁺) is a central regulator of neuronal energy metabolism, mitochondrial function, and axonal integrity. Following mild traumatic brain injury (mTBI) and repetitive subconcussive exposure, neurons undergo a neurometabolic crisis characterized by impaired oxidative phosphorylation, increased ATP demand, and depletion of intracellular NAD⁺ pools. Pre clinical models demonstrate that NAD⁺ depletion contributes to central energy dysregulation, cognitive dysfunction, and axonal and neuronal degeneration through disruption of NAD⁺-dependent pathways including SIRT1-mediated mitochondrial resilience, PARP1-driven NAD⁺ consumption, and SARM1-associated axonal breakdown. Despite growing preclinical evidence supporting NAD⁺based neuroprotection, human clinical data remains absent.

Methods: A real-world clinical pilot …

Scientific Note: Here The Whole Time: A Revised Understanding Of White Alumroot (Heuchera Alba, Saxifragaceae) In The Susquehanna River Valley., Anais Barnes, Madeline Wickers, Scott Schuette, Tanisha M. Williams, Ryan Folk, Christopher T. Martine

Faculty Journal Articles

Heuchera alba and H. pubescens (Saxifragaceae) are closely related species of alumroot endemic to the Appalachian region of eastern North America that can be difficult to distinguish morphologically. Heuchera pubescens has been understood to occupy a range from Kentucky to Pennsylvania, with the distribution of H. alba restricted to Virginia and West Virginia—plus a recently-recorded extension into Pennsylvania discovered with the help of social media. The uncovering of H. alba in Pennsylvania has challenged historical perceptions of Heuchera distributions in the state, particularly as this relates to the current concept of H. pubescens which may be in need of revision. …

Rape Pregnancies And Consequent Ptsd Resulting From The Overturn Of Roe V. Wade, Emma Lindenfelser, Yulia Garaeva, Nenette Nti-Agyemang, Ellen Davis, Anna Kabwa, Alexandra M Calaman

Binghamton University Undergraduate Journal

Prior to the 1973 Roe v. Wade ruling, access to safe legal abortions was limited and often restricted along racial, economic, and geographic lines. Nearly fifty years later, women’s health policy evolved again following the 2022 Dobbs v. Jackson Women’s Health Organization decision overturning Roe v. Wade. This court ruling decimated federal protections of abortion. This paper presents the research that the 2022 Dobbs judicial decision may contribute to and exacerbate rape-related PTSD on 1 out of 20 American who are unable to access a safe abortion after a rape-related pregnancy. Pregnant women living in conservative states that have outlawed …

Exploring The Effects Of Pest-Induced Hemlock Mortality On Growth In Neighboring Trees, Sidney L. Booher Jun 2026

Exploring The Effects Of Pest-Induced Hemlock Mortality On Growth In Neighboring Trees, Sidney L. Booher

McNair Culminating Research Projects

Trees provide many important products and services to humans. In particular, tree growth is critical to carbon storage, air and water quality, and helps determine demographic processes like population growth and reproductive success. Importantly, tree growth is sensitive to both abiotic (environmental) and biotic factors and is vulnerable to ecological disturbances. Understanding how tree populations respond to disturbances, like invasive pests, is therefore crucial to formulating timely and effective forest management practices.

Here, I used tree ring analysis to investigate how competition for water between neighboring trees changes following the decline of Eastern and Carolina Hemlock(ECH) trees due to the …

Exploring The Therapeutic Potential Of Kinetin In Allergic Contact Dermatitis, Hamna Muzaffar Jun 2026

Exploring The Therapeutic Potential Of Kinetin In Allergic Contact Dermatitis, Hamna Muzaffar

Thesis/ Dissertation Defenses

Allergic contact dermatitis is T cell mediated inflammatory skin disorder commonly studied using contact hypersensitivity mouse model. It induces local inflammation and further evidence suggests that gut microbiome also contributes to disease progression. However, kinetin, a natural plant hormone modulates these immune processes. After TNCB successfully induces CHS, kinetin treatment resulted in significant reduction in serum IgE levels while its effect on ear swelling and histological changes were modest. Microbiome analysis revealed no major difference in alpha diversity instead beta diversity demonstrated increased microbial heterogeneity in TNCB-treated mice group. Conversely, Kinetin-treated groups exhibited a microbial profile similar to wild type …

Shared Premotor Resources Underlie Dual-Task Interference Between Speech And Balance, Weina Lu, Sasha Lifton Lewis, Zoey Wire, Ingo Helmich Jun 2026

Shared Premotor Resources Underlie Dual-Task Interference Between Speech And Balance, Weina Lu, Sasha Lifton Lewis, Zoey Wire, Ingo Helmich

Exercise and Sport Studies: Faculty Publications

Humans frequently speak while standing, potentially engaging overlapping motor-cognitive resources. Dual-task paradigms with increased motor-cognitive demands reliably impair postural control, presumably due to competition for shared neural resources. The premotor cortex is critically involved in balance stabilization and in resolving competing motor plans and is also engaged during processing of action-related language. We therefore hypothesized that a central processing bottleneck emerges in the PMC when speaking and controlling posture simultaneously. Forty-one right-handed healthy adults (25 female, 16 male) maintained an upright stance while either verbally describing or mentally imagining navigation through a visually presented maze from an egocentric or allocentric …

Chromatin Insulators Homie And Nhomie Can Interact With Distant Copies Either Together Or Separately, With Distinct Outcomes For Enhancer-Promoter Interactions, Miki Fujioka, Wenfan Ke, Paul Schedl, James B. Jaynes Jun 2026

Chromatin Insulators Homie And Nhomie Can Interact With Distant Copies Either Together Or Separately, With Distinct Outcomes For Enhancer-Promoter Interactions, Miki Fujioka, Wenfan Ke, Paul Schedl, James B. Jaynes

Department of Biochemistry and Molecular Biology Faculty Papers

Chromatin insulators, a.k.a. boundary elements, separate regions of the chromosome with distinct chromatin characteristics, including distinct histone modifications. This activity affects gene expression by allowing chromatin domains to be stably regulated and maintained. Insulators also block enhancer-promoter interactions and, somewhat paradoxically, facilitate other interactions, particularly when they stitch together distant regions of the chromosome by pairing with specific partners. Here we explore how long-range interactions facilitated by insulator pairing are affected by the presence of two potentially competing partners. Our results show that when two partners are present, they can reduce each other's effects on distant gene expression, suggesting that …

Uncovering Dkk1 As A Therapeutically Relevant Target In Aggressive Er+ Breast Cancer Using A Novel Antiestrogen, Kristen Young Jun 2026

Uncovering Dkk1 As A Therapeutically Relevant Target In Aggressive Er+ Breast Cancer Using A Novel Antiestrogen, Kristen Young

Dissertations

Over 70 percent of breast cancers are estrogen receptor-positive (ERα+), meaning that the hormone estrogen drives tumor growth. Although first-line targeted therapies that directly block ERα from hormone-dependent pathologies are initially effective, almost half of patients ultimately progress. These patients show high mutational rates to the ESR1 gene that encodes for ERα. A hotspot missense tyrosine to serine mutation at position 537 (Y537S) initiates hormone-independent, allele-specific transcriptional programs that increase metastasis. While next-generation ERα-targeted therapies show promise, benefits are often limited compared to standard-of-care. Understanding the mechanisms of action used by the Y537S ESR1 breast cancer cells to evade therapeutic …

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Mitochondria undergo fusion and fission. While DRP1 regulates fission, fusion is controlled by OPA1, MFN1, and MFN2. The balance between these processes and the crosstalk between machineries remains poorly understood. MFN2 mutations cause Charcot-Marie-Tooth disease type 2 A (CMT2A), affecting mitochondrial fusion and morphology. However, their role in fission is unclear. Using skin fibroblasts from CMT2A patients (L248H and M376V MFN2 mutations) and wild-type mouse embryonic fibroblasts expressing these variants, we studied how MFN2 mutations impact mitochondrial dynamics beyond fusion. We analyzed mitochondrial morphology and dynamics by live-cell confocal microscopy and tested fusion/fission protein levels, oxygen consumption rate (OCR), extracellular …
