Optimizing Marination Methodology, Holding Temperature, And Holding Time To Upgrade The Physicochemical And Sensorial Attributes Of Japanese Quail (Coturnix Japonica) Breast Fillets, Naveen Uduwawithana, Ruvini Mutucumarana
Turkish Journal of Veterinary & Animal Sciences
The aim was to optimize the effect of meat marination method (unmarinated, tumbling, injection, and immersion), holding temperature (4 oC and 8 oC), and holding time (4 h, 8 h, and 12 h) on the physicochemical and sensorial properties of Japanese quail (Coturnix japonica) breast fillets. Ninety-six deboned breast fillets (mean weight ± SD; 10 g ± 2) from 42-day-old unsexed Japanese quails were marinated. Injection marination showed the highest uptake (p < 0.05). The method × temperature × time interaction (p < 0.05) was significant for cooking yield, meat pH, hardness, cooking loss, and surface color lightness (L*). The highest pH (p < 0.05) was reported when the unmarinated control was retained for 8 h. Tumbling marinated meat when held for 12 h at 8 oC yielded the maximum cooking yield (60.3%). The cooking loss was minimal in unmarinated, tumbled, and immersion-marinated meat when held for 12 h. Irrespective of the temperature, 4-h holding time caused the minimum meat hardness in all four treatments. Tumble-marinated meat when held for 12 h at 8 oC resulted in the minimum cooking loss, hardness, and lightness (L*), as well as the maximum cooking yield. Meat when held at 4 oC for 4 h following injection marination had the highest scores for color, aroma, marinade penetration, flavor, and overall acceptability. In conclusion, quail breast fillets when tumble-marinated and held at 8 oC for 12 h were the best in terms of overall meat quality. Quail breast fillets held for 4 h at 4 oC following injection marination were rated highest by the panelists.

Diabetic retinopathy (DR) is a leading cause of vision loss, which is driven in part by oxidative stress-induced dysfunction of ocular tissues. We previously showed that high glucose conditions trigger senescence pathways and damage the retinal pigment epithelium (RPE). Here, we identify glycitin, a natural isoflavone, as a candidate cytoprotectant and define its mechanism of action in high glucose-exposed ARPE-19 cells. Using RNA-seq followed by Ingenuity Pathway Analysis, we mapped glycitin-responsive programs and validated key nodes experimentally. Glycitin partially restored cell proliferation, suppressed oxidative stress, and reversed high glucose-induced premature senescence, primarily through modulation of the PI3K/AKT pathway. These findings …

Twenty-two percent of groundwater wells tested nationwide have one or more contaminants above a health threshold, yet many private well owners are unaware of these risks because they have not tested their water. Researchers piloted a community well testing clinic for education/outreach and surveyed participants about outcomes. Two months later, 75% of participants with high or intermediate health risk water quality results had taken action or intended to, indicating that community private well testing clinics can be effective in mitigating human health risks from contaminated drinking water.

Educating Livestock Producers And Monitoring The Ecological Health Of Grazing Spaces, Morgan Mathisonslee, Matt R. Raven, Crista Derry
Journal of Extension
Addressing the climate catastrophe requires a systems approach with a focus on regenerating agricultural landscapes. To address these complex problems, we offered a monitoring training focused on a systems approach to livestock stewardship using the Savory Institute's Ecological Outcomes Verification protocol to monitor ecosystem health. The training encouraged producers to use systems-based management practices to promote ecological regeneration. We found that despite producers not adopting the monitoring protocol, there were increases in the ecological health of their farms. We suggest Extension professionals consider teaching similar ecological monitoring protocols to benefit producers and support claims about regenerative agriculture.

Pi3kδ Inhibition Alters Cd8 T Cell Differentiation And Reprograms The Tumor Microenvironment Following Adoptive Immunotherapy, Alexandrea Joy Turnquist
Dartmouth College Ph.D Dissertations
T cell exhaustion remains a significant barrier to effective adoptive cell therapy in solid tumors. Here, we demonstrate that in vitro treatment with the PI3Kδ inhibitor CAL-101 generates T cells with enhanced stemness and metabolic fitness, though the stemness of these cells is not dependent on HIF-1α -dependent mechanisms. These cells show increased mitochondrial dependence and spare respiratory capacity while maintaining normal basal metabolism. Under chronic antigen stimulation, CAL-101-treated T cells, including human T cells, resist terminal exhaustion and maintain stem-like properties. Using single-cell RNA sequencing and spatial transcriptomics of B16 melanoma tumors, we found that CAL-101-treated T cells preferentially …
