Quantification Of Blue Carbon Within The Marine Animal Forest On The Rio Grande Valley Artificial Reef, Michelle Kathryn King May 2025

Quantification Of Blue Carbon Within The Marine Animal Forest On The Rio Grande Valley Artificial Reef, Michelle Kathryn King

Theses and Dissertations

The protection and enhancement of coastal ecosystems provides a number of benefits such as heightened biodiversity, increased fisheries, and enhanced carbon capture (Yeemin et al., 2024). The study of blue carbon ecosystems has expanded rapidly in the last two decades since the term was coined in 2009 (Lovelock & Duarte, 2019). The most well-known blue carbon ecosystems include vegetation-dominated ecosystems such as seagrass meadows, salt marshes, and mangrove forests (Howard et. al., 2014). Prior studies have overlooked Marine Animal Forests (MAF) ability to capture carbon, until recent studies on soft coral (Coppari et al., 2019) and sponge (Rix et al., …

Kusurkar, Sherry Lee, Wei Jiang, Avinoam Nevler, Matthew Mccoy, Michael J. Pishvaian, Rosalie C. Sears, Jonathan R. Brody, Charles J. Yeo, Aditi Jain

Department of Surgery Faculty Papers

Pancreatic ductal adenocarcinoma (PDAC) remains one of the deadliest cancers demanding better and more effective therapies. BARD1 or BRCA1-Associated -Ring Domain-1 plays a pivotal role in homologous recombination repair (HRR). However, its function and the underlying molecular mechanisms in PDAC are still not fully elucidated. Here, we demonstrate that BARD1 is overexpressed in PDAC and its genetic inhibition suppresses c-Myc and disrupts c-Myc dependent transcriptional program. Mechanistically, BARD1 stabilizes c-Myc through ubiquitin-proteasome system by regulating FBXW7. Importantly, targeting BARD1 using either siRNAs or CRISPR/Cas9 deletion blocks PDAC growth in vitro and in vivo, without any signs of toxicity to mice. …

Assessment Of Dose-Dependent Cytotoxicity Of Caffeine On Human Colorectal Adenocarcinoma And Foreskin Fibroblast Cell Lines, Sanjay Srinivasan

Theses/Capstones/Creative Projects

Caffeine is a stimulant well-known for its alertness enhancement and energy functions. Beyond its neurological effects, caffeine also influences cellular qualities, including viability and membrane integrity. This study aimed to investigate the cytotoxic effects of caffeine on two distinct human cell lines: Human Colorectal Adenocarcinoma (HT-29) and Foreskin Fibroblasts (HFF). Cells were exposed to varying concentrations of caffeine to evaluate whether its effects were dose dependent. Cell viability was assessed using the Trypan Blue exclusion and Alamar Blue assays. Results showed that HFF cell death was more evident than the HT-29 Cells, displaying a potential therapeutic for cancer treatment.

The Effect Of Calorie Restriction On Humoral Responses Towards The Influenza Vaccine, Grayson Kobluk May 2025

The Effect Of Calorie Restriction On Humoral Responses Towards The Influenza Vaccine, Grayson Kobluk

Celebrating Scholarship and Creativity Day (2018-)

Calorie restriction (CR) - a reduction in caloric intake without malnutrition - has been widely studied for its impact on lifespan and the onset of age-related diseases. Emerging research indicates that CR also influences immune function, though its effects can vary depending on the degree of restriction, duration, and individual characteristics. This study examined how CR effects vaccine-specific antibody responses following influenza vaccination. Male and female mice were maintained on either a control diet or a calorie-restricted diet and vaccinated with an inactivated influenza virus vaccine. Antibody titers were measured 28 days post-vaccination to evaluate the magnitude of the humoral …

Effect Of Glycation On The Binding Interactions Of Serum Albumin And Silver Nanoparticles, Megan Kirk May 2025

Effect Of Glycation On The Binding Interactions Of Serum Albumin And Silver Nanoparticles, Megan Kirk

Celebrating Scholarship and Creativity Day (2018-)

Nanoparticles are particles ranging from 1-100 nanometers in size. Most nanoparticles, upon contact with biological matrices, are immediately coated with proteins, forming the protein corona, which can alter the structure and function of proteins. Despite the tremendous advancement of nanotechnology over the last decade, the interactions of nanoparticles with proteins are not well characterized. This project investigates the effect of glycation on the binding interactions of HSA, the most abundant protein in human blood, with silver nanoparticles using various spectroscopic techniques. This is important because in diabetic patients, HSA is exposed to high concentrations of glucose over extended periods of …

Celebrating Scholarship and Creativity Day (2018-)

Creatine kinase (CK) is an enzyme that's found in the skeletal muscle, heart muscle and brain. When any of these tissues are damaged, they leak creatine kinase into your bloodstream. Elevated CK levels may indicate muscle injuries, cramping, or disease1. There are different types of CK, which are also known as isozymes for the cytoplasm, CK-MM, CK-MB, and CK-BB. CK-MM focuses on musculoskeletal, CK-MB focuses on heart muscles, and CK-BB focuses on the brain tissue. There are also two isozymes in the mitochondria15. Cramping is common problem amongst runners and can be described as “painful, spasmodic contraction of the skeletal …

Walleye And Sauger Movement And Entrainment In Lewis And Clark Lake, William Joseph Radigan May 2025

Walleye And Sauger Movement And Entrainment In Lewis And Clark Lake, William Joseph Radigan

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Walleye Sander vitreus and Sauger Sander canadensis are both socioeconomically important sportfish species in Lewis and Clark Lake, an interjurisdictionally managed mainstem Missouri River reservoir fishery. Since 2011, adult catch per unit effort (CPUE) of both Walleye and Sauger has remained at approximately 50% of pre-2011 levels. A presumed reason for the suppressed CPUE of adult Walleye and Sauger is substantial entrainment of larval and adult fish resulting from the reservoir’s high turnover rate (7.2 d). Acoustic telemetry was used to quantify adult movement and entrainment and ichthyoplankton trawls were used to assess larval entrainment. Relative importance of factors driving …

Illustrating Insects: Using Art To Encourage Experiences In Entomology, Nia Kaufmann May 2025

Illustrating Insects: Using Art To Encourage Experiences In Entomology, Nia Kaufmann

Honors Program: Senior Projects (Public)

The goal of this creative project is to use art to highlight the diversity of our insect neighbors and encourage others to explore their local environments. In order to accomplish this, I created a poster-sized illustration of five insects I’ve encountered on the University of Nebraska-Lincoln’s City Campus. All insects are posted within the silhouette of Memorial Stadium, each with an ecological and personal significance. The contrast of a well-known building with less popular insects invokes the audience to reconsider how much attention they pay to the wilderness around them.

Landscape Effects On The Beneficial Arthropod Community In Agriculturally Dominated Environments, Hannah E. Stowe May 2025

Landscape Effects On The Beneficial Arthropod Community In Agriculturally Dominated Environments, Hannah E. Stowe

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Ecosystem services in agriculturally dominated regions rely, at least in part, on arthropods. Environment and biodiversity influence beneficial arthropods in population size, community structure, diversity, and ecosystem service provisioning. Predation is among the most important services provided by arthropods in agroecosystems. Understanding what alters predation and predator activity can support the successful application of biological control and, more broadly, integrated pest management. Land use and edge adjacency, field characteristics, landscape, seasonality, and diel period all alter predatory behavior. Crop fields dominate Nebraska agroecosystems, although interspersed perennial grasslands persist through the Conservation Reserve Program (CRP). Such grasslands supply crucial resources and …

Gene Editing Approaches To Improving Grain Amino Acid Profiles In Maize And Sorghum, Joseph Preston Hurst May 2025

Gene Editing Approaches To Improving Grain Amino Acid Profiles In Maize And Sorghum, Joseph Preston Hurst

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Maize grain is deficient in lysine. While the opaque2 mutation increases grain lysine, o2 is a transcription factor that regulates a wide network of genes beyond zeins, which leads to pleiotropic and often negative effects. Additionally, the drastic reduction in 19kDa and 22kDa alpha-zeins causes a floury kernel, unsuitable for agricultural use. Sorghum, closely related to maize, has a similar lack of lysine that can be corrected with prolamin alteration and also subject to the same deleterious kernel texture. CRISPR/cas9 gives researchers the ability to directly target genes of interest. In this research, gene editing was used to specifically target …

Constructing An Attractive Targeted Sugar Bait For Reducing Mosquito Populations, Xi Xian Ng May 2025

Constructing An Attractive Targeted Sugar Bait For Reducing Mosquito Populations, Xi Xian Ng

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Mosquito-borne pathogen transmission continues to pose a significant burden to global public health, well-being, and economic productivity. In the absence of universally available vaccines and amidst rising insecticide resistance that leads to product failures, there is an urgent need to develop new tools to mitigate mosquito-borne disease risk. Two general strategies exist to reduce transmission risk: personal protective measures and area-wide chemical interventions. Attractive Targeted Sugar Bait (ATSB) systems leverage the sugar-feeding behavior of mosquitoes—critical for sustaining their flight, metabolism, development, and fecundity—by delivering oral toxicants through attractively formulated sugar baits. This study aimed to develop an optimized ATSB formulation …

Implications Of Acetylation And Phosphorylation On Metabolic Enzymes, Nour Fatema May 2025

Implications Of Acetylation And Phosphorylation On Metabolic Enzymes, Nour Fatema

Graduate Theses and Dissertations

All living things depend on glycolysis and the TCA cycle for energy production and metabolic control. Post-translational modifications (PTMs) such as acetylation and phosphorylation, which can change enzyme activity, substrate binding, and overall metabolic balance, tightly regulate these pathways. Diseases like cancer and metabolic problems are associated with disruptions in these alterations.

Serine phosphorylation and lysine acetylation are two of the many PTMs that are evolutionarily conserved processes that alter protein function. Previously thought to be a static histone alteration, acetylation is now understood to be a dynamic regulator of metabolic enzymes that modifies contacts, charge, and conformation. In response …

Effects Of Plant Growth-Promoting Microorganisms On Microbial Community Dynamics And Plant Growth Responses Of Aeroponically Cultivated Spinach, Veronica Snow May 2025

Effects Of Plant Growth-Promoting Microorganisms On Microbial Community Dynamics And Plant Growth Responses Of Aeroponically Cultivated Spinach, Veronica Snow

All NMU Master's Theses

Regional food insecurity caused by the inability to use conventional farming techniques year-round in cold climates increases the need for access to quality, locally sourced, and affordable foods year-round. Shifting from a soil-based system to aeroponics can increase yields, as aeroponic systems are not weather-reliant and can operate year-round. Aeroponics, a subgroup of hydroponics, induces growth by misting nutrient-rich solutions onto the plant roots. These systems use space and water more efficiently and rely less on fertilizers and pesticides but are vulnerable to errors in nutrient application, power loss, and rapid pathogen spread. Adding Vermicompost (VCT) and plant growth-promoting microorganisms …

A Multi-Tiered Approach To Improving Vaccination: Adjuvanticity, Alternate Routes Of Delivery, And Science Communication, Haley Agnes Bridgewater

Boise State University Theses and Dissertations

Vaccines have been used to prevent infectious disease for over 200 years. While they are an incredible way to prevent loss of life, they can be improved upon. Researchers around the globe are working to improve the immune response and ultimately the protection provided by existing and new vaccines. In this dissertation, I investigate three ways to improve vaccine immune responses: using an adjuvant, using a prime-boost intramammary delivery route in cows, and using microneedle delivery route. Cholera toxin (CT) is a bacterial toxin that has a non-toxic B subunit (CTB) used as a vaccine adjuvant. However, little is known …

Characterizing Successional Dynamics And Host Impacts On The Big Sagebrush Leaf Microbiome, Jacob A. Heil

Boise State University Theses and Dissertations

The leaf microbiome contributes to the functioning of Earth’s ecosystems through interactions with host plants, modifying their health, for better or for worse. The leaf microbiome contains bacteria, fungi, and other microorganisms that interact with each other and are subject to the processes governing community assembly. Microbial community assembly on the leaf is influenced by host plant characteristics, interactions between the different community members, the selective and dispersal-mediated effects of the weather, and time. To study the effects of a leaf microbiome on its host plant it is first important to characterize it; observing the species present and how they …

Culturing And Characterization Of Arbuscular Mycorrhizal Fungi Native To The Sagebrush Steppe, Jacob Venable May 2025

Culturing And Characterization Of Arbuscular Mycorrhizal Fungi Native To The Sagebrush Steppe, Jacob Venable

Boise State University Theses and Dissertations

Arbuscular mycorrhizal fungi (AMF) are obligate biotrophs that form symbiotic associations with the roots of over 70% of terrestrial plants. These symbioses are crucial in plant nutrient acquisition, stress tolerance, and soil health. AMF inoculum could be valuable for ecological restoration, but challenges in culturing and multiplying AMF limit their use in natural habitats. In this project, I investigated methods to characterize and culture AMF native to the sagebrush steppe. For comparison, I also grew a non-native strain of the AMF fungus Rhizophagus clarus. First, I used trap cultures to multiply AMF present in roots and field soil collected …

Minimizing Secondary Spinal Cord Injuries With Hypertonic Saline., Francisco A. Cortez-Thomas May 2025

Minimizing Secondary Spinal Cord Injuries With Hypertonic Saline., Francisco A. Cortez-Thomas

Master of Engineering Theses

Spinal cord injury (SCI) disrupts white matter, causing periaxonal swelling, axonal degeneration, and impaired function. To examine vascular edema’s role, we developed a 3Dprinted dual-compartment imaging chamber to mimic and manipulate interstitial and vascular fluid dynamics in real time. We hypothesized that hypertonic saline (HTS) in the "vascular" chamber would osmotically shift fluid from the periaxonal space and preserve myelinated fibers after SCI. Using two-photon excitation microscopy, we imaged myelin and axons ex vivo in Thy1YFP+ transgenic mice following C5 contusive SCI and assessed three clinically relevant HTS concentrations versus normal saline for their effectiveness in mitigating periaxonal swelling …

Dental Microwear Of Miocene Primates From The Carpathian Basin, Rudabánya, Hungary, Anna K. Wilcox

Biological Sciences Undergraduate Honors Theses

This study reconstructs and compares the dietary habits of the late Miocene primates Anapithecus hernyaki and Rudapithecus hungaricus by analyzing dental microwear patterns on fossilized teeth recovered from Rudabánya, Hungary. Previous research suggested that these sympatric species were both primarily frugivores but were unable to separate them by diet. However, earlier studies were limited by smaller sample sizes and less advanced methodologies. This study used an expanded sample and modern microwear texture analysis techniques to compare molar and incisor microwear patterns between the two species. Results revealed that Rudapithecus molars exhibited higher complexity compared to Anapithecus molar complexity, suggesting a …

Assessment Of Nitrogen Source Utilization During Nitrogen Fixation And Nitrogenase Regulation By Methanosarcina Acetivorans, Avery Carter May 2025

Assessment Of Nitrogen Source Utilization During Nitrogen Fixation And Nitrogenase Regulation By Methanosarcina Acetivorans, Avery Carter

Biological Sciences Undergraduate Honors Theses

Nitrogen is a crucial element in biomolecules such as proteins and DNA, essential for all cellular life. Biological nitrogen fixation, a process utilized by certain bacteria and archaea, reduces dinitrogen (N₂) to ammonia (NH₃), enabling growth in the absence of fixed nitrogen sources like NH₃. This process is driven by the metalloenzyme nitrogenase, which catalyzes the reaction: N₂ + 16ATP + 8e⁻ + 8H⁺ → 2NH₃ + H₂ + 16ADP + 16Pi. Due to its high energy demand, nitrogenase production and activity are tightly regulated. Among archaea, nitrogenase is found exclusively in methanogens, where its regulation remains poorly understood. We …

The goal of this study was to determine the conditions that would yield the highest amount of SpoIIE. The density of the cultures of bacteria …

The Role Of Cis-Acting Super-Enhancer Long Noncoding Rnas In Triple Negative Breast Cancer Progression, Sakthi Muthukrishnan May 2025

The Role Of Cis-Acting Super-Enhancer Long Noncoding Rnas In Triple Negative Breast Cancer Progression, Sakthi Muthukrishnan

Biological Sciences

Breast cancer encompasses a diverse range of diseases characterized by the uncontrolled proliferation of cells within the mammary gland. This condition results in an accumulation of a large number of genetic mutations within an individual, altering the complex internal signaling system of a cell. Millions of women are affected by breast cancer, which remains a significant threat despite medical advancements. Ductal carcinoma in situ (DCIS) acts as a precursor to invasive ductal carcinoma (IDC), however, not all cases progress to invasive cancer. Triple-negative breast cancer (TNBC) is identified to be one aggressive subtype of IDC, where the cells lack sufficient …
