Background: Photodynamic Therapy (PDT) utilizes specific wavelengths of light to activate photosensitizing chemical compounds, known as photosensitizers, which induce the generation of cytotoxic reactive oxygen species (ROS) for the targeted destruction of cancer cells. Among various photosensitizers for PDT, Protoporphyrin IX (PpIX) is widely employed in oncology and dermatology due to its natural in situ generation via the metabolic conversion of 5- aminolevulinic acid (ALA), a non-phototoxic prodrug. Systemic administration of ALA after 3-6 hr drug delay leads to peak PpIX accumulation in tissues, facilitating therapeutic and diagnostic applications. Indeterminate pulmonary nodules (IPNs), which are nodules that cannot be classified as definitively benign or malignant at the time of detection, are now diagnosed on the order of millions per year. Management of IPNs remains heavily debated, and routine practice ultimately involves some balance of overall clinical risk assessment and additional diagnostic tests or procedures which may generate significant risk, cost, and worry. Biomarkers are biologically based tests or indicators capable of accurately characterizing the physiologic properties of homeostasis and disease that are not otherwise easily evaluated by the clinician. Biomarker discovery for complex diseases, such as cancer, hinges on uncovering molecular signatures that capture intricate, interconnected relationships within biological data-a challenge that traditional statistical and machine learning methods often fail to meet due to the complexity of high-dimensional gene expression profiles. To overcome this, we introduce the expression graph network framework (EGNF). This cutting-edge graph-based approach integrates graph neural networks with network-based feature engineering to enhance the predictive identification of biomarkers. In silico perturbation models, computational methods that can predict cellular responses to perturbations, present an opportunity to reduce the need for costly and time-intensive in vitro experiments. Many recently proposed models predict high-dimensional cellular responses, such as gene or protein expression to perturbations such as gene knockout or drugs. However, evaluating in silico performance has largely relied on metrics such as $R^{2}$, which assess overall prediction accuracy but fail to capture biologically significant outcomes like the identification of differentially expressed (DE) genes. Polygenic risk scores (PRS) are widely used to assess genetic susceptibility in Alzheimer's disease (AD) research. However, the rapid expansion of PRS studies has led to dataset-specific biases-stemming from factors like population makeup, genotyping methods, and analysis pipelines-that result in inconsistent variant prioritization and limit generalizability and reproducibility. To address these challenges, we propose a transductive learning framework that integrates multiple PRS datasets for more robust risk variant prioritization, incorporating genome-wide association study (GWAS) priority scores as biologically informed priors. Despite increasing research on the Neotropical underground environment, anurans remain poorly understood in this habitat. It was analyzed scientific literature records of anurans in Brazilian natural caves and performed a redundancy analysis (RDA) to identify the environmental, taxonomic, geographic, geological and land-use variables that influence their cave occupancy. It was examined a total of 247 literature records, encompassing 83 species/morphospecies, which revealed distinct cave-dwelling tendencies. Some anuran species occupy caves at altitudes between 5 to 21 meters above sea level (m asl), in areas characterized by rocky outcrops and grassland. Maternal underweight continues to rise and, if not properly managed, will lead to increased mortality. This contradicts the United Nations Sustainable Development Goal of reducing maternal mortality by 2050. Therefore, this study aimed to determine the probable effect of dietary supplements on maternal underweight in Nigeria. This study employed probit estimation using the maximum likelihood technique in 2022, with the 2018 Nigeria Demographic Health Survey data as the basis for analysis. The results showed a statistically significant effect of dietary supplements on maternal underweight in Nigeria. Antarctic mosses play foundational roles in terrestrial ecosystem processes, yet their functional ecology remains poorly understood. This study developed a novel functional trait-based framework for dominant moss species across glacier retreat gradients in the South Shetland Islands, addressing a critical gap in Antarctic bryophyte ecology. Nutrient- and water-related traits were measured for four moss species--Polytrichastrum alpinum, Bryum pseudotriquetrum, Sanionia uncinata, and Syntrichia saxicola--along with Deschampsia antarctica, across nine sites spanning early to late successional stages. Results revealed strong interspecific trait differentiation: P. alpinum exhibited conservative nutrient-use and rapid water-uptake strategies; S. uncinata and B. pseudotriquetrum …

Background: The aim of this cohort study was to investigate the association between hyperthyroidism and the likelihood of developing herpes zoster in Taiwan.

Methods: Using the National Health Insurance Research Database (NHIRD) in Taiwan, we selected individuals aged 20-84 who were newly diagnosed with hyperthyroidism between 2013 and 2020 as the hyperthyroidism group. These individuals were then matched with a control group without hyperthyroidism in a 1:1 propensity score matching for sex, age, and baseline comorbidities. Purpose: The objective of this study is to formulate a nanoemulgel using Punica granatum (P. granatum) seed oil and study the antibacterial, anticancer, and anti-inflammatory properties. Methods: The process to formulate a nanoemulsion from P. granatum seed oil involved using the self-nanoemulsifying technique, with Span 80 and Tween 80 serving as the emulsifying agents. Carbopol hydrogel was combined with the nanoemulsion to produce the nanoemulgel. The particle size, polydispersity index (PDI), rheological behavior, antibacterial, cytotoxic, and anti-inflammatory properties were subsequently examined.

Rising ocean temperatures and increasingly frequent marine heatwaves threaten the physiological performance and survival of marine larvae. This study quantifies the thermal tolerance, respiration, and swimming behavior of Chthamalus fissus cyprid larvae to establish a baseline for understanding their vulnerability under future climate scenarios. Larvae were collected from the Scripps Pier in La Jolla, CA and exposed to a range of temperatures (12–50°C, dependent on the experiment) to assess oxygen consumption, mortality, and swimming speeds. Oxygen consumption rates increased significantly with temperature, peaking at 30°C, with cyprids exposed to temperatures above 26°C showing significantly higher respiration rates than those below …

Membrane association is fundamental to Rat sarcoma (RAS) function, driving both its physiologic signaling and oncogenic transformation. This review consolidates recent advances in the study of RAS-membrane interactions, emphasizing the molecular mechanisms underlying its membrane engagement and oligomerization. We first discuss the roles of RAS lipid modification and conformational diversity of its intrinsically disordered C-terminus in these processes, and we then examine the debate surrounding RAS dimerization and its potential role in the formation of higher-order oligomers. Introduction: Blood flow restoration to ischemic cardiac tissue is a critical and necessary intervention that has an associated downside: reperfusion injury. Reperfusion injury is a complicated process mediated in part by a pathological increase in reactive oxygen species (ROS). Although extremely elevated levels of ROS can cause cell damage, a minimum level of ROS is necessary for activation of Hydroxylated-HIF-1a (HIF-1a) and inhibition of Peroxisome Proliferator-Activated Receptor-alpha (PPARa) during myocardial ischemic reperfusion injury (MIRI). Turtles worldwide are declining from habitat loss and fragmentation, predation, and illegal collection, and their slow life histories and sensitivity to environmental change make reliable abundance and demographic data critical for conservation. Wood turtles (Glyptemys insculpta) are declining across much of their range, yet baseline information is lacking in Michigan’s Lower Peninsula. To fill that gap, we performed standardized visual mark–recapture surveys at 27 one-kilometer river reaches in two distinct watershed systems during springs of 2022 and 2023. Aims: An intrinsic feature of gene transcription is the formation of DNA superhelices near the transcription bubble, which are resolved upon induction of transient double-stranded breaks (DSBs) by topoisomerases. Unrepaired DSBs are pathogenic as they lead to cell cycle arrest, senescence, inflammation, and organ dysfunction. We posit that DSBs would be more prevalent at the genomic sites that are associated with gene expression. The objectives were to identify and characterize genome-wide DSBs at the nucleotide resolution and determine the association of DSBs with transcription in cardiac myocytes.

Acute Lymphoblastic Leukemia (ALL) is a hematologic malignancy that primarily affects children and young adults. While advancements in treatment—including chemotherapy and cranial irradiation—have significantly improved 5-year survival rates in children to nearly 90%, radiation-induced cognitive impairment (RICI) remains a persistent and debilitating side effect. The brain, particularly the hippocampus, is highly susceptible to radiation-induced oxidative stress and inflammation. In the treatment of ALL, the combined use of chemotherapy and radiotherapy contributes to a heightened oxidative burden, exacerbating the risk of cognitive dysfunction. Ratios between two metabolites are sensitive indicators of metabolic changes. Lipidomic profiling studies have revealed that plasma ether lipids, a class of glycero- and glycerophospho-lipids with reported health benefits, are negatively associated with obesity. Here, we utilized lipid ratios as surrogate markers of lipid metabolism to explore the processes underlying the inverse relationship between ether lipid metabolism and obesity. Plasma lipidomics data from two independent human cohorts (n = 10,339 and n = 4,492) were integrated to assess the associations between 82 lipid ratios and obesity-related markers in males and females. To investigate the impacts of dietary Bacillus-based probiotics and yeast-derived prebiotics on the hepatic transcriptome profile, 500 Hisex White laying hens were randomly allotted into five dietary treatments from 37 to 52 weeks of age: control; control + Bacillus subtilis; control + Bacillus subtilis and Bacillus licheniformis; control + Bacillus coagulans; and control + Saccharomyces cerevisiae yeast cell wall. The concept of internal models dominates contemporary theories of sensorimotor control, with researchers across neurosciences, specifically motor control, routinely explaining observed behaviors through computational representations that supposedly exist within the nervous system. In this perspective, I present a critical examination of internal model frameworks in sensorimotor control. I argue that representational approaches mischaracterize biological systems for several fundamental reasons: (1) Internal models require homuncular interpreters, creating infinite regress problems; (2) The purported neural implementations of internal models remain empirically elusive despite decades of research; (3) Biological movement systems exhibit multiscale, nonlinear, and non-Gaussian dynamics that fundamentally defy reduction to conventional …

Concurrent use of medications can modulate the effectiveness of immunotherapy. Although this interaction is well documented for immune checkpoint inhibitors, whether this occurs with new experimental compounds has not been evaluated. A computerized data extraction tool was used to collect clinical data and identify the prescription of a predefined set of medications within 30 days of immunotherapy infusion in the Department of Investigational Cancer Therapeutics at The University of Texas MD Anderson Cancer Center. The primary endpoints were median overall survival (OS) and progression-free survival. Tumor responses were addressed using RECIST. We identified 897 patients. The most prevalent tumor types …

Treatment failure in newborns may result from antibiotic resistance, a significant challenge in the management of infectious illnesses worldwide. So the main objective of this research was to develop a newborn-specific antibiogram at the University of Science and Technology hospital (USTH) in Sana’a, Yemen. This work investigates the efficacy of fenugreek seed powder (FSP), an agricultural product, as an economical and sustainable biosorbent for the extraction of crystal violet (CV) dye from aqueous environments. The biosorbent was analyzed using XRD, FTIR, and SEM-EDX, validating its amorphous structure, surface functional groups, and porous shape. Batch biosorption tests were adjusted utilizing a Box–Behnken design (BBD), incorporating three independent variables: FSP dose (0.02–0.1 g/100 mL), pH (4–10), and time (5–60 min). The model exhibited exceptional prediction accuracy (R² = 0.99, CV = 2.12%) and recognized all three variables as statistically significant (p < 0.05). The maximum CV removal attained was 91.8% efficiency at an FSP dose of 0.1 g/100 mL, pH 7, and a time of 60 minutes. FTIR examination indicated changes in the –OH, C=O, and C–O functional groups, signifying their participation in dye interaction, whilst SEM pictures validated dye deposition on the biosorbent surface. The biosorption process was ascribed to electrostatic forces, hydrogen bonding, π–π stacking, and pore filling. These findings underscore FSP as a viable, economical biosorbent for the removal of cationic dyes in wastewater treatment applications.

This study analyzes mollusk remains collected during the 2007 excavation season at the Knights’ Hotel site in ‘Akko, which was part of a Crusader-period residential neighborhood. A total of 538 shells were identified, including marine shells from the Mediterranean and Red Seas, freshwater shells and land snails. The analysis of these shells sheds light on the lives of the Crusader-period residents of ‘Akko, including their trade connections and use of shells for construction, ornamentation, pigment containers and raw material for the inlay industry.

Three Intermediate Bronze Age wicks discovered in the Newe Efrayim cemetery constitute some of the earliest known archaeological evidence of wick use, an essential object for lighting, yet lacking distinctive features, as it was intended to be burned, making it a rare find. The exceptional preservation of the wicks sheds light on both preservation issues and the technological aspects of wick production in antiquity. The wicks were made from textiles, indicating the maximal reuse of a valuable raw material after it had served its original purpose. Oocyte vitrification success is highly dependent on cryoprotectant agent (CPA) choice as well as possible additives that could help preserve mitochondrial membrane potential (mMP), reactive oxygen species (ROS) production, and calcium homeostasis within the cell. It is a delicate balance between levels needed for proper oocyte function and activation and levels needed to induce apoptotic pathways. Cardiac allograft vasculopathy remains a major cause of late morbidity following heart transplantation. While accumulating evidence implicates innate macrophages in the inflammatory progression of CAV, the underlying mechanisms remain incompletely understood. In murine models of CAV, we identified proteolytic cleavage of MERTK, a key anti-inflammatory receptor on macrophages, as a contributing factor to CAV progression. In a model of CAV, MERTK deficiency accelerated allograft rejection and increased intimal leukocyte infiltration. In contrast, mice expressing a genetically cleavage-resistant form of MERTK (MertkCR/CR) exhibited prolonged graft survival, reduced intimal thickening, diminished immune cell infiltration, and decreased circulating effector T cells. …
