Importance: Initiation of postoperative radiation therapy (PORT) within 6 weeks of surgery is associated with improved outcomes among patients with head and neck squamous cell carcinoma. However, the relationship of barriers to care with timely PORT is unknown.

Objective: To categorize barriers to timely PORT, evaluate the association of barriers to care with initiation of timely PORT, and describe the primary reason for delay among patients without timely PORT.

Design, setting, and participants: This prospective cohort study at a US academic medical center included adults with head and neck squamous cell carcinoma undergoing curative-intent surgery with an indication for PORT. …

The conflation of sex and gender in sport and exercise science research has led to gaps in representation and understanding of gender-inclusive outcomes. This invited editorial presents a framework to guide researchers in conducting statistical analyses that account for multiple gender identities beyond the traditional binary classification. The invited editorial guides deliberation on whether to test for sex or gender differences in sport or exercise science research. This study explores the cost functions of the Fixation & Uptake of Nutrients (FUN) model, which aims to optimize the amount of carbon used when plants acquire their necessary nutrients from the soil to maximize nitrogen (N) and phosphorus (P) uptake while minimizing the carbon cost. Specifically, we aimed to quantify the sensitivity of FUN outputs to the parameters belonging to the cost of nitrogen and phosphorus uptake functions by arbuscular mycorrhizal and ectomycorrhizal fungi. Background: While highly efficacious for numerous cancers, immune checkpoint inhibitors (ICIs) can cause unpredictable and potentially severe immune-related adverse events (irAEs), underscoring the need to understand irAE biology.

Methods: We used a multidimensional approach incorporating single-cell RNA sequencing, mass cytometry, multiplex cytokine assay, and antinuclear antibody (ANA) profiling to characterize the peripheral immune landscape of patients receiving ICI therapy according to irAE development.

The β-barrel assembly machinery (Bam) is essential for assembling all transmembrane β-barrel outer membrane proteins in gram-negative bacteria. The Bam complex consists of the central β-barrel protein BamA and accessory Bam lipoproteins, including the widely conserved and essential BamD. BamD is assumed to be an essential regulator of OMP assembly, as its absence causes a global defect in OMP biogenesis. Here, we challenge this view by demonstrating that BamD essentiality is both conditional and substrate specific. The UFD-1 (ubiquitin fusion degradation 1)-NPL-4 (nuclear protein localization homolog 4) heterodimer is involved in extracting ubiquitinated proteins from several plasma membrane locations, including the endoplasmic reticulum. This heterodimer complex helps in the degradation of ubiquitinated proteins via the proteasome with the help of the AAA+ATPase CDC-48. While the ubiquitin-proteasome system is known to have important roles in maintaining innate immune responses, the role of the UFD-1-NPL-4 complex in regulating immunity remains elusive. This series of soundscapes represents dawn and dusk. Morning Cacophony was recorded at 6:00 AM, May 2023 on the Tympanuchus Reserve near Crookston, MN — a rare remnant prairie never plowed. Voices of the prairie: sedge wren, bobolink, greater prairie chicken, sandhill crane, red-winged blackbird, clay-colored sparrow. Night Chorus was captured at 2:00 AM, June 2024 on restored grassland at Bluestem Preserve near Moorhead, MN — once farmland, now thriving prairie. International Journal of Exercise Science 18(3): 984-994, 2025. The purpose of this study was to assess the effect of a KN95 mask on the performance, physiological response, and perception of high-intensity interval training (HIIT). Twenty college-aged participants (Male: 10, Female: 10) completed a two-visit, crossover, counterbalanced study. The HIIT workout included four 1-minute “all-out” intervals at 3.5% of body mass with 4-minute rest intervals, performed with or without a KN95 mask. Following HIIT, participants remained on the cycle ergometer for a 15-minute recovery period. For each interval, peak power and average power were recorded. This study evaluates the effects of ZOA Energy Drink, a commercially available energy beverage, on muscular strength, sustained attention, and mood in exercise-trained individuals. Using a randomized, blinded, two-arm design, 24 participants (15 males, 9 females; age = 31.8 ± 8.4 years) were assigned to consume either ZOA Energy Drink or a caffeine-matched placebo. The primary outcomes included psychomotor vigilance (PVT), mood state (POMS), handgrip strength, and push-up performance. Baseline body composition was also assessed using bioelectrical impedance.

The taxonomic position of Androctonus populations in Oman was investigated by analyzing morphological variation in a comprehensive sample representing 150 records gathered from 95 localities spread throughout the country. Two new allopatric species are described, separated by multivariate morphometric analysis and meristic characters: (1) A. omanensis sp. n., distributed across most of Oman, is characterized by wider, deeper metasomal segments IV–V, shorter legs, more robust pedipalps, more granules on lateral inframedian carinae of metasoma II–III, and sparser setation on basitarsal spurs; and (2) A. ammophilus sp. n., distributed in the dunes of the Rub’ al-Khali (Empty Quarter) of …

The vertebrate eye is one of the most metabolically active organs in the body and is particularly sensitive to environmental stressors such as oxygen deprivation. Understanding how eye development proceeds under such conditions can provide critical insights into mechanisms of cellular resilience, tissue organization, and developmental plasticity. The annual killifish Austrofundulus limnaeus represents a unique model for investigating these processes due to its remarkable tolerance to anoxia during embryogenesis. Expression of genes is dependent on promoter sequences in DNA, which direct transcription by RNA polymerase. Transcription factors interact with promoters and RNAP and are often conserved and are essential for proper gene expression. In this thesis, a functional region of archaeal TFEα, homologous to eukaryotic TFIIEα, has been identified, and specific amino acids in these regions have been determined to have significant importance in TFEα's ability to promote transcription in early initiation. Though there has been substantial progress in the development of anti-human epidermal growth factor receptor 2 (HER2) therapies to treat HER2-positive metastatic breast cancer (MBC) within the past two decades, most patients still experience disease progression and cancer-related death. HER2-directed tyrosine kinase inhibitors can be highly effective therapies for patients with HER2-positive MBC; however, an understanding of resistance mechanisms is needed to better inform treatment approaches. We performed whole-exome sequencing on 111 patients with 73 tumor biopsies and 120 cell-free DNA samples to assess mechanisms of resistance. Plant functional traits typically show strong covariation, e.g. as in the worldwide leaf economics spectrum (LES). Covariation is thought to arise from selective forces and physical constraints. However, processes shaping covariation at interspecific scales may differ from those at intraspecific scales. Potential sources of intraspecific trait variation include variation in genetics, abiotic environment, and biotic environment; any of these factors may cause divergence from interspecific patterns. Cyclic diguanylate monophosphate (c-di-GMP) is a second messenger that is responsible for attachment and motility in many prokaryotic systems and controls the signaling pathway for the motile to sessile transition in many bacteria. Many c-di-GMP-metabolizing proteins are diguanylate cyclases (DGCs), responsible for the synthesis of c-di-GMP and characterized by a c-di-GMP synthesizing domain, e.g., GGDEF domain. Other c-di-GMP-metabolizing proteins contain c-di-GMP degradative domains, e.g., EAL, characterized by phosphodiesterase activity (PDE). In Agrobacterium tumefaciens, out of 31 predicted proteins regulating the c-di-GMP level, this project focuses on the predicted dual-function DGC/PDE, DcpB. Introduction: Meridianin C (MC) is a marine-derived indole alkaloid that has demonstrated kinase inhibitory and anti-tumor activities. Despite its diverse biological properties, no previous reports have systematically evaluated the in vivo quantitative analysis of MC and its metabolites. Methods: In this study, MC was synthesized following a previously reported procedure with slight modifications. A sensitive, accurate, and reliable ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS) method was developed to simultaneously detect MC and its five major metabolites (MC-1-N-O-GluA, MC-1-N-O-SO3H, MC-2′-N-O-GluA, MC-2′-N-O-SO3H, and MC-O-GluA-didehydration) in rat plasma. A non-indigenous species (NIS) of benthic foraminifera was first identified in a core collected in 1993 in San Francisco Bay, California, USA, and subsequently identified as Trochammina hadai Uchio, 1962. Archived samples and literature reviews were used to determine that the species, which is native to Asia, arrived in San Francisco Bay between the early 1960s and 1983. Through molecular analyses of specimens, archived samples and literature reviews from 1930-1983, and site surveys of harbors and estuaries along the western North American seaboard in 1994-2024, in total more than 2500 samples, we documented the presence of T. hadai at 73 …

Single-cell studies on breast tissue have contributed to a change in our understanding of breast epithelial diversity that has, in turn, precipitated a lack of consensus on breast cell types. The confusion surrounding this issue highlights a possible challenge for advancing breast atlas efforts. In this perspective, we present our consensus on the identities, properties, and naming conventions for breast epithelial cell types and propose goals for future atlas endeavors. Our proposals and their underlying thought processes aim to catalyze the adoption of a shared model for this tissue and to serve as guidance for other investigators facing similar challenges.

Dinitrogen (N) fixation provides bioavailable nitrogen to the biosphere. However, in some habitats (e.g., sediments), the metabolic pathways of organisms carrying out N fixation are unclear. We present metabolic models representing various chemotrophic N fixers, which simulate potential pathways of electron transport and energy flow, resulting in predictions of whole-cell stoichiometries. By balancing mass, electrons, and energy for metabolic half-reactions, we quantify the electron usage for nine N fixers. Our results demonstrate that all modeled organisms fix sufficient N for growth. Aerobic organisms allocate more electrons to N fixation and growth, yielding more biomass and fixing more N, while methanogens …

Introduction: Type 2 diabetes (T2D) is a major risk factor for developing tuberculosis (TB). However, understanding the role of defective T cell responses in T2D and TB has been difficult, largely due to inconsistencies across studies. Chronic urinary tract infections are persistent bacterial infections with the potential to drive antibiotic resistance. Like other persistent bacterial infections, intracellular bacterial reservoirs and biofilm formation hinder the clearance of pathogens despite long courses of antibiotic therapy. New strategies for treatment of these persistent infections are needed. Here we describe the results of an open-label individual patient study using bacteriophage therapy to treat a chronic urinary tract infection in an immunocompetent 72-year-old woman without underlying urolithiasis or indwelling devices. We co-administered HP3.1, HP3, and ES19 bacteriophages with demonstrated in vitro activity via bladder instillation, orally, and as a topical formulation. …

As work continues unabated in the study of noncovalent bonding, particularly σ-hole bonds, new challenges have emerged as the participation of transition metals in interactions of this sort is fast becoming appreciated. While there are certain similarities with the halogen, chalcogen, etc, bonds, in which the main group elements participate, there are certain unique properties of these metal atoms that must be analyzed before a complete understanding can be attained.
