Stem cell niches are essential for regulating stem cell self-renewal and differentiation during tissue repair and regeneration. However, the mechanisms supporting stem cell function in highly regenerative organisms, such as the freshwater planarian Schmidtea mediterranea, remain unclear. Using spatial transcriptomics, we identified two cell types associated with planarian stem cells: secretory cells we term "hecatonoblasts" and intestinal cells. Surprisingly, while hecatonoblasts were in close physical proximity to stem cells in the mesenchyme, they were dispensable for regeneration. In contrast, intestinal cells, despite lacking direct contact with stem cells, regulated both their position and function during regeneration. Electron microscopy revealed diverse …

Biological nitrogen fixation (BNF) is the primary input of new reactive nitrogen to natural terrestrial ecosystems. However, this flux is poorly constrained due to its unclear drivers and associated control mechanisms. Here, we extend the existing theory of nitrogen (N) isotope mass balance to estimate BNF rates and then use a Bayesian approach to constrain the BNF rates in natural terrestrial ecosystems by using measurements of natural N-isotope ratios (δ15N) in plants (δP) and soil (δS). Together with pairwise δP and δS measurements from 18 forest sites covering diverse climates and thousands …

Dissecting The Intrinsic And Extrinsic Regulators Of Prostate Cancer Dormancy In The Bone Microenvironment, Mostafa M. Nasr
Dissecting The Intrinsic And Extrinsic Regulators Of Prostate Cancer Dormancy In The Bone Microenvironment, Mostafa M. Nasr
USF Tampa Graduate Theses and Dissertations
Metastatic disease is the primary driver of cancer-related mortality. In prostate cancer (PCa), patient survival decreases dramatically once the disease spreads to distant organs, with over 80% of metastases occurring in the bone. This dissemination is often followed by a clinically silent period of tumor dormancy that can last for years, creating a critical reservoir of cells poised for lethal relapse. A significant barrier to understanding and targeting this dormant phase has been the lack of robust experimental models. This thesis first addresses this challenge by establishing a novel stress-induced model of PCa dormancy, validated across multiple murine and human …

Turkish Journal of Agriculture and Forestry
This study identifies salt-tolerant durum wheat genotypes for breeding programmes by integrating phenotypic, genotypic, and computational analyses of 27 Mediterranean varieties. Cluster analysis and principal coordinate analysis (PCoA) revealed two distinct genetic groups: modern genotypes (high-yielding Tunisian/Italian cultivars) and ancient genotypes (traditional Tunisian landraces). Among tested cultivars, JnehKhottifa and Grécale exhibited exceptional salinity tolerance, maintaining >80% germination at 200 mM NaCl, while Om Rabiaa and Agili showed sensitivity (<50% germination). Bayesian network modelling was used to explore the probabilistic relationships between simple sequence repeat (SSR) markers (e.g., CFD40 and GWM182, PIC = 0.561) and important traits such as root length and germination rate under stress. Genetic diversity analysis (average PIC = 0.407) highlighted the potential of SSR markers for trait-marker associations. These findings establish a valuable platform for enhancing marker-assisted selection salt- tolerant durum wheat varieties, essential for securing yields in saline-prone Mediterranean regions.

Pre- and postharvest techniques play a key role in improving the quality and shelf life of fresh horticultural products. The aim of this 2-year study was to evaluate the postharvest phenolic and organic acid content in eggplants under different fertilizers and postharvest treatments. Before planting, all plots were treated with 2000 cc da−1 of humic acid through a drip irrigation system. Plots treated only with humic acid were considered the control group. Fifteen days after planting, the other plots were treated with 2000 cc da−1 of liquid vermicompost fertilizer or 2000 cc da−1 of organomineral fertilizer via drip irrigation. Postharvest, …

The increasing demand for sustainable and functional foods has driven interest in utilising agro-industrial by-products, such as elderberry pomace, to enhance the nutritional and antioxidant properties of food products. This study aimed to develop a functional cottage cheese enriched with elderberry pomace powder (EPP) and evaluate its nutritional composition, antioxidant potential, sensory acceptability, and textural properties. Cottage cheese samples were formulated with EPP at concentrations of 3% and 5%, alongside a control sample devoid of EPP. The results showed that the inclusion of EPP significantly increased the levels of anthocyanins, flavonoids, and polyphenols, along with enhanced antioxidant activity, compared to …

This study aimed to address the issue of suction force attenuation in the air-suction jujube (red date) harvester due to pipeline pressure loss. The active flow control method based on magnetohydrodynamics was used to optimize the pipeline flow field and reduce pressure loss. Through multiphysics field coupling theory modeling and numerical simulation, the regulatory effects of different electromagnetic field parameters on the pipeline flow field were characterized. An experimental platform was established to verify that the optimized electrode tip density, electrode voltage, and magnetic induction intensity could significantly increase the outflow gas velocity, pressure difference, and harvesting efficiency of the …
Genetic Characterization And High-Resolution Total Mrna Sequencing Of The Millipede Cherokia Georgiana Bollman, 1889, Elena Cruz
Biology Theses
There are approximately 12,000 described species within the class Diplopoda. Only six species, falling within four of sixteen orders, have fully sequenced genomes. No whole genomes are available for incredibly diverse families like Xystodesmidae. Many interesting characteristics in this group are poorly defined at the genetic level, such as the production of a defensive hydrogen cyanide secretion and UV fluorescence in the order Polydesmida. Here, we present a genetic characterization of the polydesmid millipede Cherokia georgiana Bollman, 1889. We include tissue-specific sequencing metrics, alignment and assembly of mitochondrial DNA consensus sequence according to tissue type, and phylogenetic tree construction using …

Allometric And Mobile Terrestrial Lidar Modeling Of Above-Ground Woody Biomass Of Populus In Coppice Production, Heidi Renninger, Krishna P. Poudel
Research Data
Poplars (Populus spp.) and their hybrids are increasingly being grown in coppice production to generate bioenergy feedstocks at frequent intervals. Allometric equations are required to predict aboveground biomass (AGB) of coppiced individuals with minimal field measurements. Likewise, remote sensing tools like LiDAR (light detection and ranging) can be used if models are available to predict AGB from point cloud data. Therefore, this study sought to develop equations to predict dry woody AGB from field measurements and LiDAR data from coppiced poplar field trials containing eastern cottonwood (P. deltoides) and hybrid poplar taxa. We found that taxa-specific allometric …
