Chagas disease remains one of the most persistent and neglected parasitic infections in the Americas, advancing silently through the intracellular replication of Trypanosoma cruzi amastigotes and the invasive spread of trypomastigotes. The survival of the parasite is critically reliant on protein N-myristoylation, a lipid modification catalyzed by N-myristoyltransferase (NMT). This dissertation aims to advance beyond prior studies by systematically investigating NMT inhibition across multiple biological levels, spanning from the characterization of the purified enzyme, through cellular infection models, to the global proteomic remodeling induced within the parasite, thereby evaluating TcNMT as a potential therapeutic vulnerability in T.cruzi.Recombinant TcNMT was expressed, …

Drylands are heterogeneous landscapes that provide important ecosystem services, but they are threatened by global change factors that may influence aspects of their diversity and function. Environmental factors may influence the three-dimensional physical characteristics of vegetation structure (structural diversity) across spatial scales in arid ecosystems. Prohibitin (PHB) is a highly conserved mitochondrial protein involved in cell cycle regulation, apoptosis, mitochondrial integrity, macrophage activation, cytokine production and many other critical cell functions. The prohibitin complex, formed by Prohibitin-1 (PHB1) and Prohibitin-2 (PHB2), functions as a mitochondrial membrane-associated receptor. Cancer, neurodegenerative disorders, and renal conditions were found to be linked to an irregular PHB expression. Emerging evidence has also highlighted a significant role for PHB in tuberculosis (TB), particularly through its interactions with Mycobacterium tuberculosis (Mtb) virulence determinants. Each year, an estimated 795,000 people in the U.S. suffer a stroke, with approximately 610,000 being first-time cases. Of these, 87% are ischemic strokes, while the remaining 13% are hemorrhagic. Current imaging methods, such as Computed Tomography (CT), Positron Emission Tomography (PET), and Magnetic Resonance Imaging (MRI), provide useful information into brain tissue properties; while each technique has its advantages, they remain expensive, non-portable, and often too slow for emergency bedside or in-ambulance use. Human activities are substantially altering global resource cycles with widespread implications for biogeochemistry and ecosystem functioning globally. Drylands, regions where precipitation is outweighed by water losses, are especially sensitive to these large shifts in resource cycles due to their inherently low and variable resource availability. Gregarious desert locusts produce stage-specific pheromones that facilitate cohesive behavior in juveniles and synchronize maturation and mating in sexually mature adults. During locust outbreaks, merging populations result in cross-stage interactions, yet their impact on locust biology remains poorly understood. This study tested the hypothesis that cross-stage interactions influence juvenile cohesion and physiological traits. Using behavioral assays and gas chromatography-mass spectrometry, we examined short- and long-term interactions between juvenile and adult desert locusts. Microsporidia MB, a vertically transmitted endosymbiont of Anopheles mosquitoes, shows strong potential as a malaria control agent due to its ability to inhibit Plasmodium development within the mosquito host. To support its deployment in malaria transmission reduction strategies, it is critical to understand how environmental factors, particularly temperature, influence its infection dynamics. In this study, we investigated the impact of four temperature regimes (22 °C, 27 °C, 32 °C, and 37 °C) on Microsporidia MB prevalence and infection intensity by rearing mosquito larvae under controlled laboratory conditions. Understanding the genetic underpinning and distribution of phenotypic variation within and between divergent groups is core towards shedding light into how populations diverge and adapt, as well as how hybridization breaks or builds on these scenarios; and thus, central to evolutionary biology. Background: Omega-3 polyunsaturated fatty acids (n-3 PUFAs) have demonstrated efficacy as adjunctive treatment for MDD. In fact, fewer studies assessed the prophylactic properties of n-3 PUFAs as monotherapy on the recurrence of MDD.

Aims: This study aimed to assess the prophylactic effect of n-3 PUFAs monotherapy against recurrent MDD.

Methods: We conducted a 6-month randomized controlled trial to assess the prophylactic effect of n-3 in preventing recurrent MDD. Tai Chi, a traditional Chinese martial art characterized by gentle, fluid movements and deep breathing, has gained increasing recognition for its cardiovascular health benefits. This study investigated the integration of Tai Chi into contemporary cardiovascular health practices, focusing on its physiological and psychological effects. The slow, controlled movements characteristic of Tai Chi contribute to enhanced cardiovascular fitness, decreased blood pressure, and improved vascular function, while simultaneously alleviating stress and fostering emotional well-being. Attention-deficit hyperactivity disorder (ADHD) is a common neurodevelopmental condition often accompanied by circadian rhythm disturbances, particularly delayed sleep phase. These involve superchiasmatic nucleus (SCN) dysregulation, altered melatonin secretion, and hypothalamic–pituitary–adrenal (HPA) axis activity, which may be exacerbated by artificial light exposure. Genetic studies further implicate circadian mechanisms, linking ADHD with polymorphisms in clock genes such as PER and CLOCK.

Nutraceuticals, particularly omega-3 polyunsaturated fatty acids (n-3 PUFAs), have been proposed as modulators of circadian rhythms. Melatonin, a hormone produced by the pineal gland, is known for regulating circadian rhythms and has emerging therapeutic potential in psychiatric disorders. This systematic review examines evidence from clinical and preclinical studies to assess melatonin’s efficacy in the following conditions: depression, anxiety, bipolar disorder, and schizophrenia. Beyond its role in circadian regulation, melatonin exhibits antioxidative, anti-inflammatory, and immunomodulatory properties, which intersect with nutrition, lifestyle medicine, and psychoneuroimmunology (PNI).

The review finds that melatonin improves sleep quality, restores circadian balance, and modulates stress-related neuroimmune pathways. It also supports neuroplasticity and reduces oxidative stress, contributing to resilience against psychosocial and environmental stressors. …

Background: Altered expression of microRNAs (miRNAs) is implicated in lung carcinogenesis, but little research has investigated the association of miRNA alterations with lung cancer stage or smoking status. To identify such alterations in lung adenocarcinoma, we conducted miRNA profiling.

Methods: Lung adenocarcinoma specimens and paired nonneoplastic lung tissues from 58 patients who had received no preoperative therapy and underwent tumor resection from 1991 to 2006 were collected from tumor tissue banks. Wheat stripe rust (Puccinia striiformis f. sp. tritici, Pst) poses a catastrophic threat to global food security. While MADS-box transcription factors regulate development and abiotic stress, their roles in plant-pathogen immunity remain enigmatic. In this study, we identified TaMADS2, a Pst-induced MADS-box gene, as a positive regulator of wheat resistance. Functional analyses demonstrated that TaMADS2 overexpression significantly enhanced Pst resistance, whereas its knockdown rendered wheat more susceptible. Further investigation revealed that TaMADS2 interacts with trichome birefringence-like protein 21 (TaTBL21) to activate glycerol kinase-like (TaGKL) expression. Freshwater planarians are an emerging model for toxicology and neuroscience because of their amenability to rapid behavioral screening and remarkable ability to regenerate a cephalized nervous system. As invertebrates, planarians can help reduce the use of vertebrates in research; however, laboratories typically maintain planarians on diets of homogenized organic beef or chicken liver, raising ethical concerns with feeding a vertebrate diet. Organic liver is difficult to obtain, and preparation methods vary, introducing intra- and interlaboratory variability. Background/objectives: MET overexpression is common in non-small cell lung cancer (NSCLC). The correlation between MET overexpression and immune checkpoint inhibitor (ICI) efficacy in NSCLC is underexplored.

Methods: In this retrospective observational cohort study, we curated a dataset of 279 stage IV NSCLC patients who received ICI treatment and had MET expression assessed by CLIA-certified immunohistochemistry (IHC) assay. MET expression was graded on a scale from 0 to 3+, with overexpression defined as 2+ or 3+. Clinicopathological features associated with MET expression were assessed using logistic regression. Organic material, such as biochar, can improve soil fertility and crop yield under water stress. A pot experiment was conducted to consider the effects of biochar on the pigment content, enzyme activity, physiological characteristics, and nutrient uptake of foxtail millet under water stress. Plants were tested under normal, mild, and severe irrigation regimes (100%, 75%, and 50% field capacity, respectively) using different biochar sources (licorice root biochar (LRB), cow manure biochar (CMB), cotton residue biochar (CRB), and municipal waste biochar (MWB)) and a control without fertilizer. Background and Aims

The bidirectional relationship between plants and their soils can be disrupted by the influence of ungulate grazers. Indeed, herbivores affect plant productivity and above-ground community structure, and plants affect soil health and below-ground community structure. However, the effects of herbivory can be subtle and challenging to address experimentally, even in the case of relatively large ungulate grazers. We attempt to resolve this paradox and experimentally test effects of ungulate grazers in two case studies on the Spanish Iberian Peninsula.

The safflowers (Carthamus tinctorius L. cv Olas) grown hydroponically under nickel (Ni) stress were examined in this study. T he effects of 0, 0.75, 1, 3, 5, 10, and 15 mM Ni on seed germination rate, protein, and pigment content of germinating seeds were investigated. Also, the toxicity of 0.75 mM Ni on plant growth, malondialdehyde (MDA) amount, and anatomy of 21-day-old safflower seedlings were studied. Ni adversely affected the germination rate, radicle length, protein, and pigment content of safflower seeds, particularly when higher than 3 mM. DNA triplet repeat expansion causes several primarly neurological disorders like Huntington's disease, myotonic dystrophy type 1, and fragile-X related disorders. There is general consensus that recognition of extrahelical extrusions or hairpin-loop structures (formed by strand slippage) by the DNA mismatch repair protein MutSβ leads to repeat expansion by a mutagenic process. By contrast, the FAN1 nuclease attenuates triplet repeat expansion, the molecular basis of which was explained by our recent finding that FAN1 nuclease cleaves and initiates removal of extrahelical extrusions. Despite being the standard-of-care treatment, neoadjuvant therapy (NAT) attains a complete response only in approximately half of the patients with triple negative breast cancer. Thus, methods to predict and optimize patient response to NAT are needed. Previously, we employed patient-specific MRI data to calibrate a biology-based mathematical model that describes cell movement, proliferation, and death due to drug at the tumor level and cell proliferation at an image voxel level. Driver mutations in IDH1 and IDH2 are initiating events in the evolution of chondrosarcoma and several other cancer types. Here, we present evidence that mutant IDH1 is recurrently lost in metastatic central chondrosarcoma. This may reflect either relaxed positive selection for the mutant IDH1 locus, or negative selection for the hypermethylation phenotype later in tumor evolution. This finding highlights the challenge for therapeutic intervention by mutant IDH1 inhibitors in chondrosarcoma.

This research tested the effect of the NewGait rehabilitative device in PD impaired individuals. Twenty individuals with PD participated in this study. Walking gait for three conditions were analyzed: Normal, wearing NewGait and Post-NewGait. In response to climate change, Earth’s increased global temperature has caused less snowfall and earlier snowmelt in spring. These changes can alter an organism’s phenology, or timing of life history events. Such shifts in phenology may lead to mismatches between interacting species, which are correlated with a decline in the populations of host plant specialists and at-risk species. The rare frosted elfin butterfly (Callophrys irus) is listed as impaired in 5 states, and critically imperiled in 14.
