Seagrass meadows play a key role in the global carbon cycle through storing carbon in their biomass and soils. However, the lack of global assessments of carbon stocks in seagrass biomass and net primary production (NPP) limits our understanding of their role in the carbon cycle. Here, we provide global estimates of biomass carbon stocks and NPP encompassing seagrasses with different life-history strategies, across bioregions and 66 countries. Transcription factors (TFs) function as precision switches that integrate signaling inputs, chromatin state, and protein homeostasis to control gene expression. This thesis unites three interconnected research themes: (1) mechanistic studies of the ubiquitin-proteasome system (UPS) in regulating eukaryotic TFs, focusing on polymerase-associated Factor 1 (Paf1) and TATA-box binding protein-associated factor 2 (Taf2); (2) identification of biomarker genes within odor-related TF networks; and (3) integration of published undergraduate research linking molecular regulation, bioinformatics, and synthetic biology. This thesis investigates the therapeutic potential of Avaren-Fc (AvFc), a novel lectin fusion protein that integrates the Fc region of human IgG1 with the engineered lectin Avaren, specifically targeting lymphoma. The study delineates the in vitro binding capabilities of Avaren-Fc with a human B-cell lymphoma cell line, a canine B-cell lymphoma cell line, healthy canine peripheral blood mononuclear cells (PBMCs), and healthy human PBMCs. AvFc was shown to exhibit significant binding to human lymphoma cells. Immune checkpoint blockade (ICB) therapies have revolutionized cancer treatment, but many patients still do not respond well. To improve therapeutic outcomes, chemotherapy is often combined with immunotherapy, as chemotherapy is known to promote immunogenicity. However, many patients develop resistance to this combination therapy, and the reasons behind it are not fully understood. In this study, we explored how platinum-based chemotherapy, specifically cisplatin, affects tumor signaling to create an immunosuppressive tumor microenvironment (TME). We found cisplatin triggers ataxia telangiectasia mutated (ATM)-mediated DNA damage signaling and induces higher expression of prostaglandin E₂ (PGE₂) via a cyclooxygenase-2 (COX-2) mediated pathway. Fear plays a crucial role in survival but can lead to psychological disorders when dysregulated. Fears can be innate due to evolutionary relevance or learned through associative processes, where predictive accuracy of threat is pivotal. Disruptions in fear neurocircuitry contribute to anxiety disorders, characterized by heightened activation and impaired learning. Fear conditioning and extinction paradigms based on classical Pavlovian conditioning have been used to study mechanisms underlying fear learning. Fetal Alcohol Spectrum Disorders (FASD) describes a large range of developmental defects due to prenatal alcohol exposure. It is characterized by neural and facial defects, including jaw hypoplasia. Many factors contribute to the susceptibility of ethanol-induced facial defects, including genetics, yet the genes involved, and the cell events they regulate, are poorly understood. Tropical forests are structured by complex vertical linkages and nutrient exchanges between canopy and understory habitats. These linkages include the downward flux of organisms, the movement of canopy fauna, and the recycling of nutrients through litter and organic matter deposition. This dissertation investigates arthropod rain and its role in connecting the canopy and forest floor processes in a lowland tropical forest in Panama. Chapter 2 quantifies “arthropod rain”, the fall of canopy arthropods into the understory. A subset of eukaryotic pre-tRNAs have introns that must be spliced out to become mature molecules capable of functioning in translation. This essential processing step is performed by tRNA splicing endonuclease (TSEN), a highly conserved heterotetramer with two catalytic and two structural subunits. In addition to this conserved function, archaeal TSEN also processes pre-rRNA and yeast TSEN cleaves some mRNAs that encode mitochondrial proteins to initiate mRNA degradation. Interestingly, single amino acid mutations in TSEN cause the human Mendelian disease pontocerebellar hypoplasia (PCH) by an unknown mechanism. Pancreatic Ductal Adenocarcinoma (PDAC) is the third leading cause of cancer-related deaths with a low 5-year relative survival rate of approximately 13.3% for all stages combined. We have previously shown that exercise can improve quality of life and enhance functional capacity among patients with PDAC. Exercise induces a variety of changes in the tumor microenvironment with beneficial effects in several tumor types. However, our understanding of the clinical effects of exercise and the mechanisms that mediate anti-tumor effects are limited in pancreatic cancer.

Energy drinks (EDs) are among the most consumed functional beverages, often marketed to enhance focus, alertness, and physical performance. Despite their popularity, the combined cardiovascular and autonomic effects of caffeine and sugar during exercise remain poorly understood. The purpose of this study was to examine the acute cardiovascular, metabolic, and autonomic responses to sugar-sweetened (ED-S), non-nutritive sweetened (ED-NNS), and placebo beverages during submaximal cycling in healthy young males.

Shiga toxin-producing Escherichia coli (STEC) represents an important group of zoonotic pathogens that pose ongoing risks to public health through foodborne outbreaks and environmental transmission. This project evaluated the prevalence and genetic distribution of stx1 and stx2 toxin genes in fecal samples collected from Illinois bison Bison bison) managed under either commercial or preservation conditions. A total of 320 fecal samples were screened for STEC presence by performing enrichment culturing and confirmed by Polymerase Chain Reaction (PCR) for stx1, stx2, and uidA, which confirmed E. coli identity. Herbicide-resistant (HR) Palmer amaranth (Amaranthus palmeri S. Watson) is regarded as one of the most prominent weeds within sugarbeet fields across the Central High Plains, with limited chemical control options. Although a few herbicides are available, their use is restrictive due to crop injury concerns and can only be applied at or after the 2–4 true leaf (TL) growth stage, which will provide no control for already emerged weeds. We report the results of two expeditions (2019 and 2022) to the Mbam Minkom Massif (Centre Region, Cameroon), focused on an assessment of the montane area's bat fauna. We describe bat species' richness and highlight the incredible diversity and ecological importance of this highly threatened ecosystem. Across both years, we captured a total of 36 bat species representing 21 genera and six families on Mbam Minkom Massif and in the surrounding lowland forest. Our findings include two new species records for Cameroon, and two additional records of the recently described bat, Pseudoromicia mbamminkom. We provide accounts for all captured species, …

Isthmohyla tica, the Tica Frog, is a critically endangered species endemic to Costa Rica and supposedly Panama that has only been seen in a few sites in recent decades. To aid in its conservation, we assessed the population status and microhabitat preferences of this frog in a section of the Danta stream at Rancho Alegre farm, San Ramón, Costa Rica. We conducted 18 nocturnal surveys and collected data on perch types, vegetation associations, stream characteristics, and environmental variables for each sighting of the Tica Frog. Plant mitochondrial genomes are complex when compared to most other eukaryotic mitochondrial genomes. They are large and variable in size, expand and rearrange frequently, abundant in non-tandem repeats, but maintain a very low synonymous substitution rate in the remaining coding regions. Many of these complexities can be explained by the increased use of double-strand break repair for DNA damage. To advance the understanding of mitochondrial genome dynamics as it relates to double-strand break repair, we performed a forward genetics screen to identify proteins involved in DNA repair in plant mitochondrial genomes. Dryland crop production in Nebraska's semi-arid High Plains faces challenges from fluctuating weather impacts of climate change, including extreme temperatures, short growing seasons, and recurring droughts. Proso millet is an alternative crop used to build a more robust three-year rotation in the region, rather than the traditional two-year wheat-fallow cycle. This dissertation provides a systems-level assessment of proso millet, a climate-resilient crop with great potential to support sustainable cropping systems in the US High Plains. The family Moraxellaceae is made up of a diverse group of 24 named species that fall into three phylogenetic groups. While most Moraxella spp. act as commensal respiratory and/or ocular flora of mammals, some are associated with disease. One of these diseases, infectious bovine keratoconjunctivitis (IBK), has significant welfare and economic impacts to both cattle and cattle producers. Moraxella bovis is the only experimentally determined cause of IBK, however, an association of Moraxella bovoculi with disease has been proposed. Human epidemiological research has shown the adverse health consequences of long-term accumulation of per- and polyfluoroalkyl substances (PFAS) in human tissue. Specifically, this review will examine the relationship between PFAS exposure, impaired immune system functioning, and an elevated risk of respiratory disease using both human and experimental data. Research on human populations indicates that children exposed to PFAS have lower levels of antibodies to vaccines; those children prenatally exposed to PFAS are more susceptible to upper respiratory infections; and that adult humans have a higher incidence of chronic obstructive pulmonary disease (COPD). Tallgrass prairies are among the most endangered ecosystems in North America, with Arkansas having lost over 99% of its native grasslands. This research evaluated the potential for using native tallgrass prairie species in Arkansas roadside vegetation. Two field experiments were conducted from 2022 to 2024. The first assessed seven native seed mixtures at three roadside locations (Clarksville, Huntsville, and Newport). Seed mixture composition significantly influenced establishment outcomes, with early-establishing forbs such as black-eyed Susan, partridge pea, and Illinois bundleflower performing well, though establishment varied by location. Background: The Harrison-Anderson-Mick (HAM) applicator is a high-dose-rate intraoperative radiotherapy (HDR-IORT) applicator used to position Ir-192 brachytherapy sources over surgically-accessed tumor volumes or post-resection tumor beds. Because of the lack of a 3D imaging system, dwell times are optimized pre-surgery by a TG-43-based treatment planning system (TPS) that assumes a perfectly flat applicator surface surrounded by an infinite water phantom. These assumed conditions are disparate from typical treatment conditions, especially in the pelvic regions, which often involve uneven patient surfaces and superficial irradiations with little to no backscatter material.

Microplastic pollution is a major threat to UK seagrass habitats, which provide essential ecosystem services by supporting biodiversity and economically valuable fish species. Seagrass beds, acting as long-term ‘sinks’ for microplastics due to their dense canopies face a high risk of degradation. This study investigated microplastic accumulation in the sediment of a mixed species (Zostera marina and Nanozostera noltei) seagrass bed in the Tamar Estuary, Southwest England, considering the effect of sediment particle size. Developmental plasticity enables organisms to adjust phenotypes to environmental cues, yet the transgenerational consequences of maternal effects remain unclear. Hypoxia, intensified by climate change and eutrophication, triggers taxon-specific plastic responses. Daphnia magna exhibits plasticity dependence, making it a critical model species for studying environmental responses. While maternal hypoxia (MH) influences generational plasticity, its long-term impacts on reproductive and physiological traits across generations remain insufficiently explored, limiting predictions of species resilience to environmental change. With marine environments facing growing threats and their connections to terrestrial environments often disregarded, understanding public perceptions of these connections is crucial for effective ocean conservation. This study aimed to assess public understanding of the interconnectedness of marine and terrestrial environments and its influence on ocean conservation efforts. To achieve this, two questionnaires (N=560) were distributed among over 18’s (N=301) and under 18’s (N=259) evaluating their knowledge on the connections between marine and terrestrial environments, engagement with conservation and views on education and responsibility. This research investigates the environmental conditions that drive hazardous wave overtopping using Dawlish, a vulnerable section of the UK’s railway, as a case study. Understanding these conditions is essential for improving knowledge, developing effective mitigation strategies, and enhancing the resilience of shoreline defences against future challenges. With rising sea levels and increasingly frequent and intense storms due to climate change, coastal communities and structures are at an increasing risk of overtopping occurring. This study investigates the coastal metocean conditions responsible for overtopping at coastal defences, using Delft3D to model these processes with Dawlish as the case study site. In Qingdao, China, harmful algal blooms (HABs) are a known hazard to coastal communities. Aerosolization is a mechanism by which HAB toxins can be transported from the sea surface into the air when wind speeds exceed 5 m/s. Previous studies have analysed the composition of aerosolized marine biotoxins (AMBs) in the breathable air surrounding Qingdao, but the role of the onshore winds in transporting them there would benefit from further research. Soil salinity is a major abiotic stressor limiting agricultural productivity. Many salt stress studies rely solely on sodium chloride (NaCl) solutions, despite not accurately representing natural saline conditions. This study investigated the stress response of Vicia faba to NaCl and marine aquarium salt (MAS) solutions to assess the impact of a more complex ionic composition on plant physiology. Plants were cultivated in a controlled greenhouse and treated with either NaCl, MAS, or rainwater (control) for up to 55 days.
