Mapping the entire transcriptome at single-cell resolution under its natural spatial context is essential for investigating the oncogenesis and progression of diseases. The recently emerged targeted in-situ technologies retain the spatial organization of cells at high resolution, although they remain limited in the number of genes that can be simultaneously measured. To overcome this obstacle, numerous computational methods have been developed to predict unmeasured gene expression in spatial transcriptomics data by leveraging scRNA-seq data. Immunological intratumor heterogeneity (ImTH) describes the variability in the types, spatial distribution, and functional states of immune cells within tumors. While evidence suggests that ImTH influences tumor progression and therapeutic response, few studies have provided a quantitative characterization of ImTH. Here, we present Scoring Immunological Intratumor Heterogeneity (ScImTH), a novel algorithm that quantifies ImTH by calculating the Shannon entropy of immune cell type proportions within the tumor microenvironment. Using bulk, single-cell, and spatial transcriptomic datasets, we show that reduced ScImTH scores are associated with unfavorable survival outcomes, tumor progression-related molecular and phenotypic features, immunosuppressive states, and resistance to immunotherapy across …

The importance of this study is to perform antibody discovery as a means to derive novel preventative and therapeutic antibody countermeasures against the Junín virus, to alleviate the potential risk of a global pandemic. The purpose of this methodological study was to develop a protocol that utilizes yeast surface display to discover antibodies, from naïve human antibody libraries, that can bind the Junín Virus antigen. The methodology of yeast surface display employs the galactose-inducible GAL1 promoter to synthesize and express antibody proteins bound to the Aga2p and Aga1p and anchored to the cell wall by glycosyl phosphatidylinositol motif (GPI). The subtidal zone is the area below the low tide line that supports a wide variety of organisms, including subtidal crustaceans, which are understudied on Sapelo Island, GA. To investigate the impact of wave action on biodiversity and develop baseline data, the biodiversity of subtidal crustaceans from two beaches was compared. It was hypothesized that the beach protected from wave action would have higher biodiversity than the beach exposed to wave action. Leydig cells are anatomically located in the testicular interstitial tissue, and their main function is to produce and secrete testosterone and indirectly support spermatogenesis. We previously reported that the combination treatment of cordycepin and radiation can effectively induce Leydig tumor cell apoptosis through cell cycle arrest, caspase activation, endoplasmic reticulum (ER) stress, reactive oxygen species (ROS) accumulation, and DNA damage. Gut microbiota has recently gained attention for its role in regulating multiple host pathways and contributing to disease developments. Fecal metabolomics using liquid chromatography—mass spectrometry (LC—MS) offers a promising approach to study gut microbial metabolites; however, it remains technically challenging due to the complex, heterogeneous nature of fecal samples and the lack of standardized protocols. This study aimed to establish a robust and reproducible untargeted fecal metabolomics workflow. Evidence had accumulated that serotonin (5-HT), a hormone and neurotransmitter, was not only involved in a variety of different physiological and central nervous system functions but also in the development and manifestation of psychiatric diseases. Magallana bilineata, a commercially important oyster species, yields a shelf-stable powder residue possessing bioactivities with unknown specific compounds. Carotenoids are key marine bioactive compounds, but their presence in oysters remains underexplored. The highly selective SGLT2 inhibitor (SGLT2i) is reported to have beneficial effects on diabetic cardiac hypertrophy; however, the molecular mechanisms underlying the cardioprotection of SGLT2i are not fully understood. Lung cancer is the leading cause of cancer-related deaths worldwide, driven by carcinogens such as tobacco-derived nitrosamines, dioxins, and urethane. This study evaluated the chemopreventive effects of two monoterpenes, α-pinene and D-limonene, using NNK/TCDD- and urethane-induced lung tumor models and A549 lung cancer cells. Both compounds significantly reduce tumor number and size, with enhanced effects when combined. Mechanistic studies revealed suppression of PRC1, β-catenin, and c-Myc, alongside activation of p53, Bax, and caspase-3, indicating inhibition of the PRC1—Wnt/β-catenin pathway and induction of apoptosis. These findings suggest α-pinene and D-limonene as safe, promising agents for lung cancer chemoprevention.

Excessive fructose intake leads to fatty liver and kidney dysfunction, with associated inflammation and metabolic disturbances. This study evaluated the effects of fish oil rich in ω-3 fatty acid on these conditions in rats fed a high- fructose diet. Male Sprague Dawley rats were divided into three groups: normal diet, high-fructose (HF) diet, and high- fructose diet with 5% fish oil supplementation. The experiment lasted for 21 weeks. Fish oil supplementation significantly reduced blood levels of total cholesterol, triglycerides, and advanced glycation end products (AGEs) in HF-fed rat. Cordycepin (3’-deoxyadenosine), a major bioactive component derived from fungi of the genus Cordyceps, has garnered significant attention in recent years for its potent antitumor properties. Drawing on literature indexed in the Web of Science Core Collection from 2004 to 2025, this study employs bibliometric tools―specifically CiteSpace and VOSviewer―to systematically examine developmental trends, research hotspots, and emerging frontiers in the field of cordycepin-related cancer research. Clerodendrum kelli (Lamiaceae), a new species from the montane forests of Carranglan, Nueva Ecija, Philippines, is described and illustrated. The species shares morphological features with Clerodendrum quadriloculare and C. mindorense, but differs in inflorescence structure, corolla tube length, and calyx shape and color. A lectotype for C. mindorense is also designated. Preliminary conservation assessment places C. kelli in the Critically Endangered category (CR B1ab(iii)+2ab(iii)). This study contributes to Philippine plant biodiversity knowledge and informs conservation strategies.

[Objective ] This study aims to investigate the effects of aeration on the microbial community structure and metabolic functional genes in pickles.[Method ] Amplicon sequencing was carried out for the 16S rRNA and ITS rRNA gene fragments from three groups (aerated group,sealed group,and the initial group ) within the same pickle system.The EasyAmplicon pipeline and PICRUSt 2 were employed to assess bacterial and fungal community diversity,composition,and functional gene variations.[Result ] Compared with the initial group,the sealed group showed only minor changes in microbial diversity,whereas the aerated group exhibited significant microbial divergence from the initial group.After aeration,the number of rare taxa …

[Objective ] This study aimed to investigate the feasibility of the fermentation process for L-lysine production by immobilized Corynebacterium glutamicum.[Method ] Firstly,the effects of different volume fractions of seed medium on the fermentation stability of immobilized Corynebacterium glutamicum were examined.Subsequently,the impacts of immobilization carriers with different specifications on the fermentation performance were analyzed.Finally,a comparative analysis was conducted on the differences between the fermentation processes with free and immobilized Corynebacterium glutamicum.[Result ] The addition of 5% volume fraction of seed medium adequately sustained the growth and metabolic activities of Corynebacterium glutamicum.Higher volume fractions diverted carbon sources towards bacterial growth,thereby limiting product …

[Objective ] This study aims to investigate the effects of different treatments on the postharvest preservation of grapes.[Method ] The impacts of 0.4 mg/L 24-epibrassinolide (EBR ),1×109 CFU/mL Metschnikowia pulcherrima,and their combination (YBR ) on postharvest storage of ‘Shine Muscat ’ grapes were evaluated.[Result ] Compared with the control (CK) group,EBR,Metschnikowia pulcherrima,and YBR treatments effectively reduced the water loss rate,maintained the firmness,and delayed the declines in protopectin and stem chlorophyll content of grape.In the later storage stage,YBR treatment delayed the decrease in flavonoid mass fraction,which was 12.95% higher than that of CK group.At the same time,YBR treatment reduced the mass …

Neuroinflammation has emerged as a critical modulator of cognitive function and neural circuit dynamics, yet the distinct contributions of acute and chronic neuroinflammation to brain function remain poorly defined. This dissertation examines how inflammation independently alters large-scale neural activity and behavior, focusing on interactions between the hippocampus (HPC) and anterior cingulate cortex (ACC), regions essential for memory and executive processes. Previous studies in the Gibbs lab have shown that starvation resistant Drosophila melanogaster flies have increased lipid content, lower metabolic rates, and poor physiological health. Historically, many evolutionary studies end at the phenotypic level, leaving the genomic basis largely unresolved. Using Evolve and Resequence (E&R) study, we can map genome-wide allele-frequency trajectories under selection, linking phenotypic shifts to their genomic underpinnings. This study utilized the E&R approach to identify enriched biological processes and pathways underlying starvation adaptation in Drosophila melanogaster.
