Introduction: Auto-segmentation of tumor volumes and organs at risk (OARs) is a critical step in cancer radiotherapy treatment planning, where rapid, precise adjustments to treatment plans are required to match the patient anatomy. Although auto-segmentation has been clinically accepted for most OARs, auto-segmentation of tumor volumes, particularly clinical target volumes (CTVs), remains a challenge. This difficulty arises because images alone are often insufficient to capture the necessary information for accurate delineation of microscopic tumor invasion invisible on the image itself.

Methods: We propose a deep learning-based medical image segmentation framework designed to mimic the clinical process of delineating CTVs and …

Despite the successful application of Imaging Reporting and Data Systems to improve the radiologic description and management of disease in many organs, one does not yet exist for the kidney. Instead, the radiologic approach to the kidney has focused on the Bosniak classification system, which is based on imaging characteristics for cystic renal masses, and detecting macroscopic fat within solid renal masses. Radiologically, cystic and solid renal masses are categorized and evaluated separately because of historical precedent, differences in appearance at imaging, and differences in biologic behavior. However, the World Health Organization classification of renal neoplasms does not support such …

Proteasome Augmentation Mitigates Age-Related Cognitive Decline In Mice, Danitra Parker, Kanisa Davidson, Pawel A Osmulski, Maria Gaczynska, Andrew M Pickering

Faculty, Staff and Student Publications

The aging brain experiences a significant decline in proteasome function. The proteasome is critical for many key neuronal functions including neuronal plasticity, and memory formation/retention. Treatment with proteasome inhibitors impairs these processes. Our study reveals a marked reduction in 20S and 26S proteasome activities in aged mice brains, including in the hippocampus, this is driven by reduced functionality of aged proteasome. The decline in proteasome activity is matched by a decline in 20S proteasome assembly. In contrast, 26S proteasome assembly was found to increase with age, though 26S proteasome activity was still found to decline. Our data suggests that age-related …

Fibroblast growth factor 21 (FGF21) is an endocrine FGF that plays a vital role in regulating essential metabolic pathways. FGF21 increases glucose uptake by cells, promotes fatty acid oxidation, reduces blood glucose levels, and alleviates metabolic diseases. However, detailed studies on its stability and biophysical characteristics have not been reported. Herein, we present the overexpression, biophysical characterization, and metabolic activity of a soluble recombinant FGF21 (rFGF21). The far-UV circular dichroism spectra of rFGF21 show a negative trough at 215 nm, indicating that the protein’s backbone predominantly adopts a β sheet conformation. rFGF21 shows intrinsic tyrosine fluorescence at 305 nm. Altered food landscapes contribute importantly to wildlife disease dynamics and may play a critical role in host heterogeneity in disease outcomes through changes in host diet composition. We explored the effects of dietary macronutrient composition on disease pathology and feeding behavior of canaries (Serinus canaria domestica) infected with Mycoplasma gallisepticum (MG). In the first experiment, we provided canaries with isocaloric diets composed of identical ingredients that varied in macronutrient content (high protein or high lipid) then MG- or sham-inoculated birds. In the second experiment, we offered both diets to canaries before and after MG or sham inoculation. In experiment one, …

Edge Feathering Across Forest-Meadow Ecotones Increases Light Heterogeneity And Understory Plant Diversity, Aruni Kadawatha, Jean H. Burns

Faculty Scholarship

Ecotones, the transitional zones between distinct habitats, are vital for ecosystem functioning and habitat diversity. Traditional management practices frequently create abrupt boundaries, leading to stressful conditions for organisms. To address this challenge, an underutilized land management technique called “edge feathering”, which involves gradual thinning of the canopy along the forest edge, has been introduced. This study, con- ducted at Holden Arboretum in Kirtland, Ohio, investigated the effects of edge feathering on light availability and understory plant diversity in edge feathered and control treatments. We calculated the coefficient of variation in light availability as light heterogeneity and plant diversity indices at …

Widespread use of genetically engineered maize targeting the corn rootworm complex (Diabrotica species) has raised concerns about insect resistance. Twelve years of university field trial and farm survey data from ten United States Corn Belt states indicate that maize hybrids expressing toxins derived from the bacterium Bacillus thuringiensis (Bt maize) exhibited declining protection from rootworm feeding with increased planting while pest pressures simultaneously decreased. The analysis revealed a tendency to overplant Bt maize, leading to substantial economic losses; this was particularly striking in eastern Corn Belt states. Our findings highlight the need to go beyond the “tragedy of the …

Current Challenges And Preliminary Morphological Reassessment Of The Genus Chaerilus Simon, 1877 In China (Scorpiones: Chaerilidae), Victoria Tang

Euscorpius

The genus Chaerilus Simon, 1877 in China is preliminarily revised on a morphological basis, derived from the available literature and newly examined specimens. The robustness of several commonly applied species-level diagnostic criteria are evaluated. Ten species are provisionally recognized for China, including one new morphospecies, Chaerilus herta sp. n., described from 9 females, 40 males, and 2 juvenile females collected in Mêdog County. The current study only reveals and resolves several most elementary issues in the taxonomy of Chinese Chaerilus. Further molecular investigations are warranted until topotypes of certain species become available.

Leveraging Arginine Catabolism To Treat Metabolic Complications, Yiming Zhang

Arts & Sciences Graduate Student Theses and Dissertations

Fasting and caloric restriction are effective to treat patients with pre-diabetes or metabolic dysfunction-associated steatotic liver disease (MASLD), previously known as non-alcoholic fatty liver disease (NAFLD), but intensive lifestyle changes are difficult for many to implement and sustain. Therefore, by identifying and studying fasting-mediated signals, we hope to elicit and convey the therapeutic effects of fasting against MASLD without actual physiological fasting. We discovered a key pathway in the liver through unbiased transcriptomic screening in fasted mice, a novel glucose fasting-activated effector: the amino acid hydrolase, arginase 2 (ARG2). Our previously published data demonstrates that forced hepatocyte-specific Arg2 overexpression is …

Protein Serine Kinase H1 (PSKH1) was recently identified as a crucial factor in kidney development and is overexpressed in prostate, lung, and kidney cancers. However, little is known about PSKH1 regulatory mechanisms, leading to its classification as a “dark” kinase. Here, we used biochemistry and mass spectrometry to define PSKH1’s consensus substrate motif, protein interactors, and how interactors, including Ca2+ sensor proteins, promote or suppress activity. Intriguingly, despite the absence of a canonical Calmodulin binding motif, Ca2+-Calmodulin activated PSKH1 while, in contrast, the ER-resident Ca2+ sensor of the Cab45, Reticulocalbin, Erc55, Calumenin (CREC) family, Reticulocalbin-3, suppressed PSKH1 catalytic activity. Monitoring And Modeling Legacy Tree Response To Restoration Treatments In The Klamath Mountains, Oregon, Usa, Isaac Joseph Ball

Dissertations and Theses

Climate change and its disruption of historical drought and wildfire patterns pose a substantial threat to forest health in the western United States. This threat is compounded by past management practices like fire suppression. The largest, oldest, and most ecologically valuable trees are particularly vulnerable, making their conservation a key objective of climate-adaptive forest management. Restoration treatments such as mechanical thinning have been shown to enhance tree growth and drought resilience in dry conifer forests, but long-term monitoring is necessary to understand their sustained effects. This study used dendroecological tree-ring analysis to investigate the effects of thinning on the growth, …

Background: Antiangiogenics combined with immune checkpoint blockade have become standard of care for recurrent endometrial cancer after standard platinum-based chemotherapy. To dissect mechanisms and define biomarkers associated with clinical outcomes to these combinations, we applied multidimensional immune monitoring to peripheral blood specimens collected from a randomized phase 2 trial of nivolumab with or without cabozantinib in 75 evaluable patients with recurrent endometrial cancer (NCI ETCTN 10104, NCT03367741). This trial demonstrated superiority of the combination to nivolumab alone.

Methods and results: Using Olink proteomics, mass cytometry, tumor antigen-specific ELISA, and whole exome tumor sequencing, we identified longitudinal immune signatures specific …
