Vesicular trafficking is a fundamental process essential to cell viability and dictates roles in both cellular and tissue function in multicellular organisms. To further understand various vesicular trafficking processes, our lab focuses on the moderators of these processes, Rab GTPases, using the model organism Dictyostelium discoideum. This dissertation focuses explicitly on the uncharacterized protein Rab32C, a homolog of human Rab32. While several Rab family members have been well defined, the functionality of Rab32C remains poorly understood. Climate change is reshaping environmental conditions across Southeast Asia (SEA) through rising temperatures, altered precipitation patterns, and increased droughts. These changes alter transmission dynamics of neglected tropical diseases (NTDs) by shifting spatial distribution, increasing larval development rates, faster pathogen growth development, and increasing transmission rates. NTDs are a group of preventable and treatable infections that disproportionately affect more than 1 billion people globally, specifically in tropical and subtropical regions. A subgroup of NTDs, parasitic helminth infections (PHIs) - particularly schistosomiasis, soil-transmitted helminths, and lymphatic filariasis remains a public health concern in the Southeast Asian region. Cultured meat is a promising alternative to traditional meat production techniques, but it requires optimization before it can be implemented on a large scale. Andisols are characterized by high phosphorus (P) retention, which often limits P availability for crops. This study aimed to analyze the independent and interactive effects of P fertilizer and organic matter (i.e. manure and rice straw compost) on P retention, total P, available P, P uptake, soil pH, and cabbage yield. The Sletr1-2 allele, which reduces ethylene sensitivity in tomatoes, offers a promising approach to extending fruit shelf life by slowing ripening-related metabolism. However, the trait’s success depends on compatibility with the recipient cultivar’s genetic background. This study evaluated fruit characteristics of F1 near-isogenic line (NIL) hybrids carrying Sletr1-2 in the commercial backgrounds ‘Costoluto’, ‘Oxheart’, and ‘Sart Roloise’. Growing strawberries in soilless substrates in the greenhouse relies on fertilizers and supplemental light to boost yield and fruit quality. The research evaluated the properties of the biofertilizer and its effectiveness on the yield of Japanese strawberry cultivars Nanatsuboshi and KS-75 under controlled greenhouse conditions, with and without light-emitting diodes (LED) as supplemental light. The cell viability and plant-growth-promoting properties of a liquid consortium biofertilizer consisting of nitrogen-fixing bacteria and phosphate-solubilizing microbes, were characterized. The development of this platform addresses the challenges associated with traditional tomato disease detection methods as well as the challenges of current machine learning and deep learning models in accurately detecting tomato diseases. Conventional methods of tomato disease diagnosis typically rely on expert evaluation but are time-consuming and susceptible to human mistakes. Incorrect diagnoses of certain diseases could lead to poor control measures, resulting in decreased agricultural productivity. About 13.4% of Americans aged 20-74 had obesity in 1960-1962 and the prevalence has steadily increased to 40.3% in 2021-2023--more than a 3-fold increase. Microbial uptake of trace metal cofactors such as copper (Cu), iron (Fe), and zinc (Zn) is governed by biogeochemical processes in hydrothermal systems where metal bioavailability varies widely. In situ microbial metal uptake rates were quantified by applying metal stable isotope probing (MSIP) in the Yellowstone hydrothermal system across diverse geochemical conditions in hydrothermal springs. The findings in these studies revealed that pH exerts a dominant influence on microbial copper uptake rate, also enhanced by increased Cu²⁺ ion availability and dissolved oxygen. Drought is becoming an increasingly dominant force shaping plant–pollinator interactions, yet we still know little about how nectar rewards respond to water limitation across species commonly used to support pollinators. We examined nectar volume and sucrose content in six flowering species relevant to California’s Mediterranean climate and quantified how drought alters these traits for four of those species. Under well-watered conditions, species differed in their nectar production, spanning more than two orders of magnitude. Perennial species such as Epilobium canum and Scrophularia californica produced the highest nectar volumes and sucrose amounts, suggesting that they contribute substantial carbohydrate resources when moisture …

Tea productivity and quality are largely determined by shoot structure, particularly the balance between pecco shoots (tender buds and young leaves) and banji shoots (older, less productive shoots). Conventional reliance on inorganic fertilizers often enhances overall growth but does not necessarily improve shoot structure, while biofertilizers alone provide limited nutrient availability. Digestion elevates metabolism through the heat increment of feeding (HIF) – the energy expended on mechanical and biochemical processes after eating. Quantifying this cost is essential for bioenergetic models that predict energy flow and prey requirements in populations. Using breath-by-breath respirometry, we measured oxygen consumption (V̇ O2 ) in eight common bottlenose dolphins (Tursiops truncatus) before and after feeding standardized meals (1659–2658 kcal of capelin and herring). Metabolic rate rose by ∼37% above resting levels, peaking 60 min after feeding before returning to baseline within 2 h. Background/Objectives: Traditional dietary monitoring methods such as 24 h recalls rely on self-report, leading to recall bias and underreporting. Similarly, dietary control approaches, including portion control and calorie restriction, depend on user accuracy and consistency. The opioid crisis in the United States has evolved from a medical response to pain into a nationwide public health emergency. The prescription opioids (POs) – associated cardiovascular adverse events (CVAEs) remain insufficiently studied, especially in relation to sex disparities. This dissertation investigates sex disparities in PO-associated cardiovascular adverse events (CVAEs) through an analytic framework integrating artificial-intelligence-assisted literature mining and real-world pharmacovigilance analytics. Studies were conducted in Oktibbeha and Clay Counties in Mississippi to determine the response of green antelopehorn milkweed and hemp dogbane to triclopyr and other auxin herbicides in non-cropland areas. These studies were conducted at four locations with well established populations for each species throughout 2020 to 2023, with applications repeated at each location the following year if populations persisted. Beef cattle routinely undergo a variety of stressors within the preconditioning and feedlot receiving phases which may impair animal performance and health. Trace mineral supplementation, either through dietary or injectable sources, is proposed to mitigate these effects through metabolic and immune system support. Two systematic reviews and meta-analyses were conducted to evaluate the efficacy of two different oral supplementation methods, organic (OTM) and inorganic (INR), and injectable trace mineral (ITM) solutions on average daily gain (ADG) and morbidity. Across 20 studies, OTM supplementation increased ADG (P = 0.01) when compared to INR, but did not affect morbidity (P = 0.92). …

This dissertation presents a comprehensive spatiotemporal analysis of M. ulcerans and mycolactone producing mycobacteria (MPM) ecology in the Americas and also reveals mycolactone's role in the environment as a microbial community regulator. Through comparative studies in French Guiana (endemic) and the southeastern United States (non-endemic), combined with experimental microcosm investigations, this research contributes to environmental pathogen ecology understanding. Apple peel color is one determining factor of apple quality and is a highly sought after trait by both growers and consumers. Historically plant growth regulators (PGRs) such as daminozide and auxins have been used to induce a greater amount of red color in the peel, but these products typically have negative production impacts and raise environmental concerns. We present a narrative review focusing on pulses’ geographical origin and distribution, their impact on human evolution and history, and their influence on human health. Pulses, including dry peas, beans, and lentils, are renowned for their richness in chemical antioxidants. Despite containing antinutrients, processing techniques preserve their health advantages. Epidemiological research has consistently demonstrated that the consumption of pulses is associated with favorable effects on metabolism. This evidence is further supported by molecular and clinical research, which has elucidated potential nutrigenomic mechanisms and effects on gut microbiota composition underlying their health benefits. Through-hikes on long distance backpacking routes have the potential to both improve physical fitness and pose health risks. PURPOSE: To quantify changes in cardiorespiratory fitness and body composition, while recording dietary intake and sleep habits in a sample of through-hikers. METHODS: Participants completed two lab visits consisting of a maximal graded treadmill test and body composition assessments. Food safety strategies are being developed in the hopes of combating foodborne pathogens and their risks. Previous techniques and technologies are struggling to meet the needs of the public, leading to the demand for new, innovative methods. Mild high-pressure processing (HPP) of 350 MPa was used in a synergetic combination with mild heat of 60°C and the antimicrobial carvacrol for the decontamination of non-typhoidal Salmonella serovars that resulted in a >5 log CFU/mL reduction. Chickpea proteins offer significant nutritional and functional benefits in food products; however, their utilization is often restricted by off-flavors, primarily for lipoxygenase (LOX) and lipase activity. This study aims to develop a novel extraction method by combining acid (pH 2.0) extraction with the conventional alkali (pH 9.0) method to inactivate enzymes and enhance protein quality. Acid-extracted proteins exhibit the highest purity (83.4%), albumin content, and structural flexibility (46.65% random coil). Both acid and sequential acid-alkali-extracted proteins significantly reduced LOX (0.75 and 0.88 U/g) and lipase (0.10 and 0.08 U/g) activity.
