Nuclear And Plasma Membrane Properties Of Macrophage Heterokaryons And Hybrids, Siamon Gordon Jan 1971

Nuclear And Plasma Membrane Properties Of Macrophage Heterokaryons And Hybrids, Siamon Gordon

Student Theses and Dissertations

Mouse peritoneal macrophages, which do not synthesize DNA in vitro, were fused with a strain of mouse melanoma cells which proliferates rapidly in vitro. The plasma membrane of the macrophage has specific receptors which enable the cell to ingest antibody-coated sheep red cells and is also rich in a divalent cation dependent adenosine triphosphatase (ATPase) activity. The melanoma cell lacks these macrophage membrane markers. High yields of mouse macrophage-melanoma cell heterokaryons and macrophage-macrophage homokaryons were obtained through the Sendai virus-induced fusion of cells spread on a glass surface. After fusion there was a striking reorganization of cellular architecture by means …

Hormone Induced Magnesium Transfer In Fat Cell Plasma Membranes: A Mechanism For Effecting Changes In Cellular Metabolism, Donald Allison Elliott Jan 1971

Hormone Induced Magnesium Transfer In Fat Cell Plasma Membranes: A Mechanism For Effecting Changes In Cellular Metabolism, Donald Allison Elliott

Student Theses and Dissertations

A new method was developed to measure magnesium transfer across the plasma membrane. This technique made use of the ligand, 8-hydroxyquinoline-5-sulfonic acid (8-OH-Q) which fluoresces in the presence of magnesium and was used to demonstrate epinephrine stimulation of magnesium uptake. The ligand was incorporated into plasma membrane vesicles prepared from rat epididymal fat cells. In the presence of calcium and magnesium but not in the presence of calcium alone, epinephrine and other lipolytic hormones stimulated an increase in fluorescence which was equivalent to an increase in intravesicular magnesium of 1.2 mM. The influx rate of magnesium entering the intravesicular space …

Survival Of The Oyster Crassostrea Virginica (Gmelin) In The Laboratory Under The Effects Of Oil Drilling Fluids Spilled In The Laguna De Tamiahua, Mexico, Jorge Cabrera Jan 1971

Survival Of The Oyster Crassostrea Virginica (Gmelin) In The Laboratory Under The Effects Of Oil Drilling Fluids Spilled In The Laguna De Tamiahua, Mexico, Jorge Cabrera

Gulf and Caribbean Research

In 1965, 970.12 m3 of oil drilling fluid were spilled in the Laguna de Tamiahua, Mexico. Laboratory experiments were carried out to determine possible effects of this upon the oyster Crassostrea virginica. It was found that drilling fluid reduced the survival of oysters to a significant degree in concentrations above 200 ppm. At turbidities between 200 and 500 ppm, there was 50% survival on the seventh day. Tanino in concentrations between 90 and 170 ppm had a drastic effect upon survival which was 50% between the fourth and fifth days. Bentonita in 110 to 190 ppm resulted in …

Responses Of Rumen Microflora To Whey Products Added To High-Grain Low-Roughage Rations, Virgil L. Metzger Jan 1971

Responses Of Rumen Microflora To Whey Products Added To High-Grain Low-Roughage Rations, Virgil L. Metzger

Electronic Theses and Dissertations

Many factors present in livestock feeds affect the numbers and types of protozoa and bacteria in the rumen. A small particle size of feed and a fast rate of passage of the feed through the ruminant decreases protozoal numbers. Full feeding of a concentrate ration and an acid pH in the rumen also tend to reduce protozoa numbers. Bacteria numbers tend to increase as the protozoa numbers decrease. Knowledge of rumen microflora population changes on different rations is lacking. It is known that restricted-roughage rations will generally cause lower protozoal numbers. Since whey is an available source of lactose and …

A Study Of The Endogenous Electron Donors For Nitrogen Fixation By Cell-Free Extracts Of Klebsiella Pneumoniae M5al (Aerobacter Aerogenes M5al), Elisabeth Yi-Jan Wu Jan 1971

A Study Of The Endogenous Electron Donors For Nitrogen Fixation By Cell-Free Extracts Of Klebsiella Pneumoniae M5al (Aerobacter Aerogenes M5al), Elisabeth Yi-Jan Wu

Electronic Theses and Dissertations

Biological nitrogen fixation, the mechanism of the enzymatic conversion of the inert nitrogen gas into amino acids in the cytoplasm of the microorganism has puzzled many scientists for decades. The recent discovery of some free-living nitrogen-fixing bacteria has encouraged the investigation of this mechanism. A big step taken in the rapid elucidation of this mechanism was the first reproducible cell-free nitrogen-fixing system from Clostridium pasteurianum successfully obtained by Carnahan et al. (9) in 1960. In the following decade, reproducible cell-free systems were demonstrated with the strict aerobe, Azotobacter vinelandii (4), facultative anaerobic Bacillus polymyxa (16) and Klebsiella pneumoniae (31) and …

Evaluation Of Orally Administered Sodium D-Thyroxine As A Teratogen In New Zealand White Rabbits, Gary A. Thibodeau

Electronic Theses and Dissertations

Sodium D-thyroxine was evaluated for possible embryotoxic and/or teratogenic effects following oral administration in New Zealand White rabbits. All animals utilized were alert, active and apparently disease-free virgin does weighing three and one-half to four kilograms. Rabbits were individually housed in stainless steel batteries and maintained on a commercially available palletized diet and mineral supplement. Feed and water were provided ad libitum in animal quarters air conditioned to minimize environmental temperature fluctuations. Following a 14-day post-delivery observation period does were bred to New Zealand White bucks and randomly allocated to one of two experimental groups or the control group each …

An Ecological Study Of Coleoptera Succession In Bovine Manure With Emphasis On Natural Enemies Of The Face Fly (Musca Autumnalis De Geer) In Eastcentral South Dakota, Howard Kessler Jan 1971

An Ecological Study Of Coleoptera Succession In Bovine Manure With Emphasis On Natural Enemies Of The Face Fly (Musca Autumnalis De Geer) In Eastcentral South Dakota, Howard Kessler

Electronic Theses and Dissertations

The frequency and succession patterns of coleopterous species in cattle dung were determined for two localities in eastcentral South Dakota during 1969 and 1970. Thirty-nine species of beetles were screened as potential predators of the face fly, Musca autumnalis De Geer, a pest of cattle and horses; and 1 Coleoptera, 1 Hymenoptera and 1 nematode were screened as parasites of the same host. Succession studies of insects inhabiting bovine manure revealed that four families of Coleoptera were recovered from the manure and, in descending order of abundance, were Hydrophilidae, Staphylinidae, Scarabaeidae, and Histeridae. The hydrophilids were first to invade freshly …

Pseudomonas As Causative Agents Of Fluorescence In Anguispira Kochi (Pfeiffer) Mucus, John Michael Baum

Masters Theses

The mucus from land snail specimens of Anguispira kochi collected from Coles and Edgar counties, Illinois, emits a blue fluorescence when exposed to ultra-violet light. Pseudomonas, a genus of bacteria which produces fluorescent pigments, has been isolated from the mucus of these snails. The object of the study was to compare the fluorescence of the mucus from individuals of A. kochi with the fluorescence of the pigments produced on a synthetic medium by pseudomonads isolated from the mucus of these snails. An effort was made to establish that Pseudomonas is the causative agent of fluorescence in the mucus of …

The Biological Activity Of Phytotoxic Extracts From Corynebacterium Sepedonicum, Doran W. Pearson Jan 1971

The Biological Activity Of Phytotoxic Extracts From Corynebacterium Sepedonicum, Doran W. Pearson

Open Access Master's Theses (through 2010)

Isolation of a phytotoxin from the ring rot pathogen of potatoes, Corynebacterium sepedonicum, had previously been reported by Strobel (21). The first phase in our study was to investigate more extensively the general nature of this compound. The phytotoxin could possibly be a specific compound associated only with the ring rot pathogen. However, the possibility that the substance could be obtained from sources other than the bacterium had not been completely eliminated. In an attempt to answer this question, three extracts were purified and compared. First, compounds were isolated from a broth culture of bacteria in the manner described …

The Nature Of Salmonella Phage P3 Modification And Restriction By Growth In Escherichia Coli, Clark A. Pearson Jan 1971

The Nature Of Salmonella Phage P3 Modification And Restriction By Growth In Escherichia Coli, Clark A. Pearson

Loma Linda University Electronic Theses, Dissertations & Projects

The Salmonella potsdam group B bacteriophage P3, which grows in the rough variant strain of S. potsdam (SP2-R) and E. coli KCS11, KW117, and C+/L with similar efficiencies, was restricted by SP2-R after a passage though E. coli. The entrance of restricted E. coli-grown phage DNA into nonpermissive SP2-R cells was confirmed by the finding that after adsorption of restricted 32P-labeled P3-C1·C+/L to SP2-R and removal of the adsorbed phage heads from cells, 80.3% of the labeled DNA activity sedimented with the cells. Thus restricted P3 DNA …

Changes In Escherichia Coli Associated With Acquired Tolerance For Quaternary Ammonium Compounds, R. B. Maxcy, N. P. Tiwari, P. R. Soprey Jan 1971

Changes In Escherichia Coli Associated With Acquired Tolerance For Quaternary Ammonium Compounds, R. B. Maxcy, N. P. Tiwari, P. R. Soprey

Department of Food Science and Technology: Faculty Publications

Acquired tolerance for a quaternary ammonium compound produced a tolerance for a similar compound. Tolerance was associated with the structure and the extent of adsorption of the compound. Morphological changes and resistance to disruption by pressure and by sonic treatment accompanied the development of tolerance. An otherwise weakened culture evolved with the acquisition of tolerance. The maximum obtainable viable population density of tolerant cells in growth medium was approximately 5 %, of that obtained in the parent culture. Tolerant cultures died off more rapidly in the original growth medium as well as when washed cell suspensions were stored at 5 …

Antigenic Relationships In Seven Strains Of Tetrahymena, W.E. Engelhard, B.W. Mccashland Jan 1971

Antigenic Relationships In Seven Strains Of Tetrahymena, W.E. Engelhard, B.W. Mccashland

Papers in Microbiology

Tetrahymena, along with other genera of the family Frontoniidae, have been the subject of many scientific investigations concerning problems in protozoan physiology. Unfortunately in many instances, difficulty arises when attempting to identify properly the form that was used in the particular study. Until rather recently, relatively little had been accomplished by way of a comprehensive study of the interrelated genera, and of these very few have dealt with taxonomic problems serologically.

Study Of Control Of Public Health Problems Using Irradiation, R. B. Maxcy, N . P. Tiwari, C. C. Anagnostis

Department of Food Science and Technology: Faculty Publications

Red meat, as exemplified by ground beef and pork, is subject to numerous opportunities for contamination and harborage of pathogenic organisms with the present commercial system of processing and distribution. An exploration was undertaken therefore to determine the usefulness of gamma irradiation for public health protection when applied to red meat. This work indicated that retail products are commonly heavily contaminated. The contamination can be reduced somewhat through central processing as shown by the data obtained from ground beef. Considerable numbers of the contaminants can be destroyed by irradiation at a 68 Krad level.
This work indicates that commercial ground …

Characterization Of Deoxyribonucleic Acid From Actinomycetes, Lynn William Enquist Jan 1971

Characterization Of Deoxyribonucleic Acid From Actinomycetes, Lynn William Enquist

Theses and Dissertations

Deoxyribonucleic acid (DNA) analyses were used to assess on a molecular level, the relationships among representatives of the genera Streptomyces, Nocardia and Mycobacterium. The methods developed in this study have been used for routine analysis of DNA from a large number of actinomycetes and have given reliable and reproducible data.

DNA isolated from various actinomycetes was characterized by buoyant density determinations in CsCl from which the mole fraction guanine plus cytosine (GC) content was calculated. All the streptomycete DNA preparations studied had buoyant densities in the range of 1.7287 to 1.7312 g cm-3 which corresponded to GC compositions of …

Influence Of Temporary Chemical Immobilization Of Boar Spermatozoa Upon In Vitro Survival, W. L. Singleton, T. D. Rich Nov 1970

Influence Of Temporary Chemical Immobilization Of Boar Spermatozoa Upon In Vitro Survival, W. L. Singleton, T. D. Rich

South Dakota Swine Field Day Proceedings and Research Reports, 1970

The use of artificial insemination (AI) in swine has been restricted because of the inability to store semen. It is usually recommended that for AI use boar semen not be stored more than 48 hours. Obviously, the goal is to develop a technique whereby boar semen could be stored indefinitely such as is the case with bull semen. However, until such a technique is developed, a method of storing boar semen for a few days would be of value for promoting AI in swine.
