Synthesis Of 6-Aminopenicillanic Acid-Protein Conjugates For Development Of Enzyme Immunoassay For B-Lactam Antibiotics, Alice Ann Heth May 1984

Synthesis Of 6-Aminopenicillanic Acid-Protein Conjugates For Development Of Enzyme Immunoassay For B-Lactam Antibiotics, Alice Ann Heth

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

An enzyme immunoassay specific for several β-lactam antibiotics rather than individual antibiotics was investigated. The goal to develop an enzyme immunoassay for analysis of a whole class of compounds at one time is different than the goal of most enzyme immunoassays which desire specificity for drugs or hormone levels. Detection of the presence of all β-lactam antibiotics is wanted and identification of specific antibiotics is not needed.

6-Aminopenicillanic acid, the common structural moiety of β-lactam antibiotics was used tin this investigation. Methods of preparation of 6-aminopencillanic acid conjugates and antibodies needed for enzyme immunoassays have been developed. 6-Aminopenicillanic acid was …

Comparison Of Several Forms Of Equations For Predicting Cheddar Cheese Yield From Milk Composition, Craig A. Moore May 1984

Comparison Of Several Forms Of Equations For Predicting Cheddar Cheese Yield From Milk Composition, Craig A. Moore

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

This study was conducted to evaluate several forms of equations for predicting Cheddar cheese yields based on the fat and protein content of milk and moisture content of cheese. Production and quality control data from a Cheddar cheese plant for one entire year was used. This included the pounds of milk that went into each vat of cheese, yield of cheese from each vat, cheese moisture from each vat, and fat and protein percentages of the milk.

Seven models were derived to predict the yield of Cheddar cheese. The seven models were statistically fitted to the data by applying the …

A Lactic Culture Stimulant Blend From Kluyveromyces Fragilis And Whey, Steven L. Wright

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

A buffered growth medium was developed that sustained a significantly greater concentration of various strains of Streptococcus lactis and Streptococcus cremoris than did milk. The active buffering ingredients of this medium were magnesium hydroxide, ammonium and sodium salts of phosphate and citrate. This medium was entitled Lactic Culture Buffered Growth medium (LABGRO) and possessed about 8 times the buffering strength between pH 6.6 and 5.1.

Significant differences in growth and acid production rates were noted among Prt+ and Prt- lactic strains evaluated. Also noted were significant differences in cellular acid production rates. The two Prt- strains having the slowest growth …

Photoaffinity Labeling Of The Antimycin Binding Site In Rhodopseudomonas Sphaeroides, Emily Wilson May 1984

Photoaffinity Labeling Of The Antimycin Binding Site In Rhodopseudomonas Sphaeroides, Emily Wilson

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The purpose of this study was to identify the site of interaction of antimycin with the ubiquinone-cytochrome b-c1 oxidoreductase in the photosynthetic bacteria, Rhodopseudomonas sphaeroides. To accomplish this goal, three areas of research were undertaken: the synthesis of a radiolabeled, photoaffinity analog of antimycin, identification of the inhibitory characteristics of this analog, and the photoaffinity labeling of the antimycin binding site. All three areas were accomplished.

The major finding of this study was the identification of an 11,000 dalton polypeptide as the predominantly labeled protein. Although this polypeptide was not exclusively labeled, it was consistently labeled and showed competition …

Effect Of Physiological Age On Radiation Resistance Of Some Bacteria That Are Highly Radiation Resistant, Lewis Keller, R. Burt Maxcy May 1984

Effect Of Physiological Age On Radiation Resistance Of Some Bacteria That Are Highly Radiation Resistant, Lewis Keller, R. Burt Maxcy

Department of Food Science and Technology: Faculty Publications

Physiological age-dependent variation in radiation resistance was studied for three bacteria that are highly radiation resistant: Micrococcus radiodurans, Micrococcus sp. isolate C-3, and Moraxella sp. isolate 4. Stationary-phase cultures of M. radiodurans and isolate C-3 were much more resistant to gamma radiation than were log-phase cultures. This pattern of relative resistance was reversed for isolate 4. Resistance of isolate 4 to UV light was also greater during log phase, although heat resistance and NaCl tolerance after heat stress were greater during stationary phase. Radiation-induced injury of isolate 4 compared with injury of Escherichia coli B suggested that the injury process, …

Dynamics Of Arid-Land, Perennial Plant Populations With An Examination Of Potential Causal Agents, Henry George Gardiner May 1984

Dynamics Of Arid-Land, Perennial Plant Populations With An Examination Of Potential Causal Agents, Henry George Gardiner

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Rigorous statistical examination of the population dynamics of a number of long-lived perennial plant species permitted an interpretation of changes in those populations and the establishment of potentially important causal agents. Survival and recruitment of six species from the mulga-zone rangelands of Western Australia varied across three climatically different periods following the removal of domestic livestock.

Two species' populations (Eragrostis xerophila and Maireana glomerifolia) had their rates of increase reduced by kangaroo and euro grazing. Both survival and recruitment of Eragrostis xerophila were reduced by this grazing while Maireana glomerifolia suffered reduced recruitment during all periods. These negative …

Effect Of Casein/Fat Ratio On Milk Fat Recovery In Cheddar Cheese, Nana A. Yiadom-Farkye May 1984

Effect Of Casein/Fat Ratio On Milk Fat Recovery In Cheddar Cheese, Nana A. Yiadom-Farkye

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Cheddar cheese was made by the traditional 4.5-h method from three experimental lots of milk, each standardized to casein/fat ratios of approximately 0.64, 0.67 and 0.70. The effect of casein/fat ratio on milk fat recovery was determined. The effects of milk composition on curd firmness at cutting, cheese composition and resulting yield of cheese were evaluated. Correlations between milk constituents and various cheese components were obtained. Milk fat recovery was unaffected by casein/fat ratios within the limits of 0.64 and 0.71. Average milk fat recovery was 91.58 ± 1.73%. Cheese yield was a function of milk protein, milk fat and …

Effects Of Folic Acid Or Zinc Malnutrition On Rotaviral Infection In A Murine Model, John Douglas Morrey May 1984

Effects Of Folic Acid Or Zinc Malnutrition On Rotaviral Infection In A Murine Model, John Douglas Morrey

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The influences of dietary deficiencies of folic acid or zinc on rotaviral disease infant mice were studied. Preliminary studies indicated that bovine and simian rotaviruses, but not porcine rotavirus, caused diarrhea in infant mice. Bovine and porcine rotaviruses were not, however, sufficiently infectious to replace murine rotavirus in studies utilizing the murine model. It was also determined that murine rotavirus purified by a cesium chloride gradient was highly infectious and useful for subsequent studies on nutritional influences of rotaviral disease. In dietary studies, female Swiss Webster mice were fed diets containing deficient, moderately deficient, or adequate levels of folic acid …

Rhizobium Phaseoli Symbiotic Mutants With Transposon Tn5 Insertions, K. Dale Noel, Araceli Sanchez, Leonor Fernandez, Jan Leemans, Miguel A. Cevallos

Biological Sciences Faculty Research and Publications

Rhizobium phaseoli CFN42 DNA was mutated by random insertion of Tn5 from suicide plasmid pJB4JI to obtain independently arising strains that were defective in symbiosis with Phaseolus vulgaris but grew normally outside the plant. When these mutants were incubated with the plant, one did not initiate visible nodule tissue (Nod-), seven led to slow nodule development (Ndv), and two led to superficially normal early nodule development but lacked symbiotic nitrogenase activity (Sna-). The Nod- mutant lacked the large transmissible indigenous plasmid pCFN42d that has homology to Klebsiella pneumoniae nitrogenase (nif) genes. The other mutants had normal plasmid content. In …

Activation Of 2-Aminoanthracene By Cell-Free Microbial Extracts, Thomas Hamill Mar 1984

Activation Of 2-Aminoanthracene By Cell-Free Microbial Extracts, Thomas Hamill

Masters Theses & Specialist Projects

The activation of the precarcinogen 2-aminoanthracene was investigated employing cell-free extracts of Escherichia coil, Pseudomonas aeruginosa, Streptococcus faecalis, Bacteroides fragilis, Clostridium perfringens and the eukaryote Candida albicans. A modification of the Salmonella typhimurium/mammalian microsomal mutagenicity assay was employed to determine activation. Of the species tested, only cell-free extracts of Escherichia coli and Bacteroides fragilis failed to induce the reversion of the Salmonella typhimurium his- mutants TA-98 and/or TA-100 to his+ revertants.

Separation of the cytosol and membrane pellet enzymes was completed. Of the organisms tested only the membrane pellet enzymes of Clostridium perfringens showed activation of …

Financial Exigency: Need It Affect The Quality Of Biology Curricula?, Robert G. Thomson, Peter Abramoff

Biological Sciences Faculty Research and Publications

Declining enrollments and financial restraints require that science departments seek ways to meet academic commitments within the framework of reduced budgets and faculty resources without sacrificing quality programs. The following describes our evaluation of the role of the laboratory in the undergraduate biology curriculum and the positive effects achieved on our academic, financial, and faculty resources by separating labs from lecture courses and reducing the number of labs required for majors and nonmajors. Several years ago we experienced increased enrollments coupled with only modest increases in funds to deliver our undergraduate instructional programs. To resolve this problem we developed a …

Aeromonas Hydrophila And Motile Aeromonad Septicemias Of Fish, Rocco C. Cipriano, G. L. Bullock, S. W. Pyle Jan 1984

Aeromonas Hydrophila And Motile Aeromonad Septicemias Of Fish, Rocco C. Cipriano, G. L. Bullock, S. W. Pyle

United States Fish and Wildlife Service: Publications

Aeromonas hydrophila and other motile aeromonads are among the most common bacteria in freshwater habitats throughout the world, and are frequently associated with severe disease among cultured and feral fishes. Determinations of the etiology of diseases involving aeromonad infections has been complicated by the genetic, biochemical, and antigenic heterogeneity of members of this group. Consequently, motile aeromonads consist of a complex of disease organisms that are associated with bacterial hemorrhagic septicemias in fishes. From descriptions of fish diseases in the early scientific literature, Otte (1963) speculated that septicemic infections in fish caused by motile aeromonads were common throughout Europe during …

Recent Developments In Numerical Electron Optics, Erwin K. Kasper Jan 1984

Recent Developments In Numerical Electron Optics, Erwin K. Kasper

Scanning Electron Microscopy

The familiar methods for the numerical calculation of fields in electron optical devices are outlined briefly. for the solution of self- adjoint elliptic differential equations in orthogonal curvilinear coordinate systems a favourable ninepoint discretization is worked out which can be applied favourably e.g. to spherical mesh grids. The field calculation in magnetic deflection systems by means of an integral equation method is also highly advantageous. The methods for the field calculation can be still more improved by means of suitable hybrid procedures.

A second and shorter contribution is concerned with ray tracing and aberrations. Some favourable numerically stable new forms …

Progress In The Theory Of Electron-Beam Deflection, E. F. Ritz Jr. Jan 1984

Progress In The Theory Of Electron-Beam Deflection, E. F. Ritz Jr.

Scanning Electron Microscopy

Analysis, simulation, and design of electron-beam-deflection systems are reviewed in light of the current state of theoretical understanding. A brief review of the physical principles is followed by a detailed discussion of electrostatic, magnetostatic, mixed-field, traveling-wave, and scan-expansion systems. Each methodology is examined from a triple perspective: calculation of electromagnetic fields, calculation of electron trajectories, and calculation of the ensemble of trajectories forming the beam. Applications discussed include deflectors for television displays, lithography, scanning microscopes, and CRT oscillography. Developments of the last ten years are stressed, thereby supplementing and updating the author's previous review on this subject.

In field calculation, …

Displays: Market And Technologies, Steve Blazo Jan 1984

Displays: Market And Technologies, Steve Blazo

Scanning Electron Microscopy

Displays provide the essential human interface to virtually all electronics instrumentation. The market is large with new applications appearing every year; sometimes with profound impact. Digital watches with liquid crystal displays appeared in the early 1970s and have virtually wiped out the mechanical timepiece industry. Personal computers with cathode ray tube (CRT) displays are proliferating with diverse applications in the industry and in the home. Work stations with high-resolution color displays are changing the way architects, draftsmen, and IC designers perform their job. The CRT is the dominant technology in today's market, and will, no doubt, continue to be for …

Extracellular Enzyme Production By Rhizopus And Mucor Species On Solid Media, D Thompson, Broderick Eribo Jan 1984

Extracellular Enzyme Production By Rhizopus And Mucor Species On Solid Media, D Thompson, Broderick Eribo

Department of Biology Faculty Publications

Solid media were employed to determine the presence and absence of extracellular enzyme production by two genera of fruit-rot fungi, Rhizopus and Mucor. The results of this investigation revealed that phosphatase was released into the cultural medium by all the fungi examined; however, only R. oryzae, R. tritici, M. mucedo, and M. piriformis showed the possibility of being high producers of the enzyme. Protease, urease, ribonuclease, pectate lyase, and polygalacturonase, at varying levels of activity, were detected, in the majority of the fungi, in the cultural medium.
