Axicabtagene ciloleucel (axi-cel) is an autologous anti-CD19 chimeric antigen receptor (CAR) T-cell therapy approved for relapsed/refractory (R/R) follicular lymphoma (FL). Here, we report updated clinical outcomes from ZUMA-5 in 159 enrolled patients with R/R indolent non-Hodgkin lymphoma (iNHL; 127 with FL and 31 with marginal zone lymphoma) after a median follow-up of 64.6 months. Patients underwent leukapheresis and received lymphodepleting chemotherapy and axi-cel (2 × 106 CAR T cells/kg). The overall response rate was 90% (75% complete response rate). Triple-negative breast cancer (TNBC) is a heterogeneous disease with variable response to neoadjuvant systemic therapy (NAST). Patients with pathologic complete response (pCR) following NAST have improved survival. Our goal was to establish readily accessible imaging biomarker to identify which TNBC patients will have pCR. Building on prior favorable results in the literature, we hypothesized that manually measured tumor volume changes at DCE-MRI may predict pCR early during NAST. The rapid digital transformation of the food-away-from-home (FAFH) sector has redefined how consumers access and select meals. Digital delivery platforms such as Uber Eats, Deliveroo, and Meituan have become influential market intermediaries, shaping the nutritional landscape of food environments and raising concerns about their healthfulness. This study reviews empirical evidence on policy-based and industry-led interventions aimed at improving dietary outcomes in digital FAFH environments. Following PRISMA guidelines and using machine-learning–assisted screening (ASReview), 37 peer-reviewed studies were identified across four domains: nutritional labeling, fiscal measures, food-assistance programs, and digital nudges or decision-support tools. Bruton tyrosine kinase inhibitors (BTKis) and cell therapy have successfully been used to treat mantle cell lymphoma (MCL). However, therapy resistance inevitably emerges. Cancer cells can progressively develop stable resistance by traversing through a transient drug-tolerant persister (DTP) state. The mechanisms enabling DTP cells to reversibly adapt to therapies and evolve to acquire heterogeneity remain poorly understood, and characterizing DTP cells in MCL continues to pose a challenge for clinic translation. International Journal of Exercise Science 18(8): 1393-1409, 2025. https://doi.org/10.70252/GEHB5425 Sedentary behaviors contribute to law enforcement officer fitness declines over a career. An officer’s motivation (desire to exercise) and self-efficacy (belief in ability to exercise) could affect whether they exercise and predict resulting fitness. Data from 60 officers (48 men, 12 women) were analyzed. Motivation (amotivation, external, introjected, identified, integrated, and intrinsic regulation) was measured via the Behavioral Regulation in Exercise Questionnaire. A 6-item questionnaire assessed coping (confidence in exercising under challenging conditions) and scheduling (confidence in scheduling exercise) self-efficacy. This research aimed to evaluate the potential of using plantain peels as a raw material for producing bioethanol with the help of Saccharomyces cerevisiae. The study utilized a four-factor Box-Behnken design (BBD) and response surface methodology (RSM) to optimize the fermentation conditions. The factors considered for optimization were substrate concentration (1-4 g), pH (5-7), temperature (30-45°C), and fermentation time (24-96 hours). Through this optimization process, the study found that the optimal conditions for bioethanol production were 4 g substrate concentration, pH 6, 45°C temperature, and 60 hours of fermentation time. Conjunctival melanoma (CJM) is a rare and aggressive malignancy. Mainly attributed to its rarity, the genomic features of CJM have not been well characterized. In the present study, we sequenced the exomes of primary and metastatic CJM tumors with the aim to unravel their genomic landscape. The results of the study suggest that CJM is a molecularly distinct melanoma subtype with mixed genotype and targetable mutations. Notably, there were features overlapping with both mucosal, and mainly cutaneous melanoma. We assessed the social learning benefits provided by online panel discussions, where topics and questions on forestry issues were collected from the audience, but the discussions were limited to the people on the panel. We also examined the influence of different panelists including experts, extension professionals and peers. Findings indicate that the panels advanced understanding of issues, but also generated a variety of social benefits (e.g., validation, self-awareness, empathy, passion) that supported views about sustainable forestry and stewardship. Panels that involve peers and extension professionals were slightly better at generating a productive social learning environment, compared to panels containing experts.

Educational farm tours offer learning opportunities that cannot be duplicated in a classroom setting. Developing a consistent farm tour evaluation tool is paramount to quantifying individual Extension farm tours while also providing a common platform to measure outcomes and impacts nationally and over time. An IRB-approved farm tour evaluation tool was developed to capture agricultural awareness and used to evaluate the 2022 National Association of County Agricultural Agents Annual Meeting/Professional Improvement Conference (NACAA AM/PIC) tours. The evaluation tool attempts to quantify farm tour outcomes and impacts and demonstrate the value of agriculture to decision makers, consumers, and community members.

This study explores the lives and deaths of the Szekler people recovered from a burial mound in the Transylvanian village of Városfalva. The Transylvanian plateau is located in present day Romania, and is protected on the east and the south by the Carpathian Mountains. The Szekler people arrived in this area around 1000 CE, and were a nomadic group originating in the Asian Steppes region who later intermarried with various migratory groups as they moved across the Eurasian plains. Filth-breeding flies pose significant health risks and costs in the equine industry. Although an arsenal of management strategies exists, integrated pest management (IPM) utilizing biological control (BC) offers cost and labor efficient pest control for equine facilities. Yet, little data exists on IPM adoption across the equine industry in the United States. This quantitative study examines the knowledge, perceptions, and adoption of IPM of those involved in the equine industry. We evaluated Extension professionals' attitudes and beliefs from Midwestern "1862" land-grant universities around three themes: prioritizing climate programming, farmers' perceptions, and the role of Extension leadership in building capacity to integrate climate change into programs. Using key informant interviews and a two-hour virtual workshop, we developed 'coordinate planes' to illustrate the tensions and contradictions among the three themes, with participants expressing highly diverse viewpoints across the region. Results identify the need for Extension leadership to support multi-state programs and shared resources that provide consistent guidance and expectations around climate change programming while recognizing the intrinsic state context and values.

Many jobs in agriculture have been deemed too hazardous for children younger than 16 years. A waiver allowing 14- and 15- year-old youth to work on farms requires safety course certification. [Redacted] Extension has partnered with a NIOSH funded Agricultural Safety Center to provide an annual tractor and equipment safety course across [Redacted]. To gauge impact, a survey addressed questions regarding the program's long-term impact, perceived value, injuries, close calls, recommendations for improvement and adoption and implementation of safety practices. The results have led to improved programming. The findings provide a roadmap to establish effective tractor and equipment certification programs.

Neuroscience offers language and evidence that can strengthen interdisciplinary communication, support advocacy in biomedically dominated settings, and validate relational practices long recognized in social work, including the impacts of early adversity, attachment, and healing through connection. At the same time, an epistemological tension underlies the profession’s unease with neuroscience when brain-based narratives are used to individualize structural harm or justify surveillance of marginalized families and communities. BACKGROUND: Innovations in protein engineering offer promising solutions for redesigning allergenic proteins to minimize adverse reactions in sensitive individuals. Earlier models for predicting allergenicity have relied on the knowledge of physicochemical properties and sequence homology to assess the potential risk. However, to better understand the allergenic proteins’ sequence features, we need a novel sequence-based deep learning model for predicting allergenicity.

Liver cancer has a high incidence and mortality rate globally, particularly in patients requiring intensive care unit (ICU) admission. Early prediction of in-hospital mortality for these patients is crucial, yet lacking reliable tools. This study aims to develop and evaluate machine learning (ML) models for predicting in-hospital mortality in critically ill liver cancer patients admitted to the ICU. This retrospective study used data from the MIMIC-III and MIMIC-IV databases, including 862 patients from MIMIC-III (training cohort) and 692 patients from MIMIC-IV (validation cohort). Cancer-associated fibroblasts (CAFs) in the tumor microenvironment (TME) have garnered significant research attention in the last decade. As key stromal cells of the TME, studies have explored them as a potential target for controlling cancer. Using high-throughput technologies like single-cell RNA sequencing coupled with proteomics, the classification of different CAF subgroups reveals a complex system that varies by cancer type. Background: Only a subset of polymerase epsilon (POLE) mutations is associated with hypermutant phenotype; we hypothesized that only loss-of-proofreading (LOP) POLE mutations are associated with favorable immunotherapy response.

Methods: This retrospective cohort study included a pan-cancer cohort of 69,223 patients from cBioPortal and a cohort of patients with 41 POLE mutant metastatic colorectal (CRC) treated with immunotherapy at the MD Anderson Cancer Center between January 2017 and May 2023. We evaluated prognosis according to POLE mutation functionality.

Renal medullary carcinoma (RMC) is a rare but highly aggressive kidney cancer that resists conventional therapies. To identify therapeutic targets, this study employs histopathologic, genomic, and transcriptomic profiling of 25 RMC samples. TROP2, EPCAM, CLDN6, and CDH6 are significantly overexpressed compared with other renal and solid tumors. Pathway analyses indicate Hippo pathway upregulation and a tumor microenvironment rich in fibroblasts and neutrophils. We subsequently explore treatment of four heavily pretreated patients, all with high TROP2 expression, using sacituzumab govitecan, a TROP2-targeted antibody-drug conjugate. Photo-oxidation of spilled crude oil generates polar compounds that can heighten toxicity to marine fish embryos. We investigated the effects of photo-oxidized crude oil on early life stages of Gulf killifish (Fundulus grandis) by exposing embryos under flow-through conditions to water-accommodated fractions of four oil treatments: fresh oil unexposed to either sunlight or weathering referred to as dark control (DC), a UV-light oxidized whole oil (LT), the polar fraction extracted from LT (Pol) and the remnants after extracting the Pol from LT (NP). Dr. James Van Etten was awarded the UNL Outstanding Research and Creative Activities Award in 1992. He was born in Virginia but grew up Peoria, Illinois. Something in his youth must have sparked an interest in science because he earned his bachelor’s degree in biology from Carleton College in 1960. Carleton is a well-regarded liberal arts institution in Northfield, Minn., about 400 miles from Peoria. Next, Jim earned a master’s and doctorate at the University of Illinois in Champaign-Urbana, both in plant pathology. Sepsis is characterized by the over-production of pro-inflammatory cytokines. Cecal ligation and puncture (CLP) is a well-accepted model for recreating sepsis-induced renal injury in mice. The current study investigates how citronellol, a naturally occurring substance with a variety of biological characteristics, can prevent acute kidney inflammation brought on by CLP. In the CLP mouse model, citronellol was administered orally at doses of 50 and 100 mg/kg. Serum levels of creatinine and urea were used as markers of renal function, and the Murine Sepsis Score (MSS) was used to assess the severity of sepsis. According to our findings, CLP caused a …

Background: Oral stents may reduce toxicities during radiation therapy for head and neck cancer (HNC). Customized 3D-printed oral stents offer faster production and achieve comparable patient-reported outcomes to conventionally fabricated stents. However, their design process remains time-consuming, lacks standardization, and relies heavily on skilled technicians. We hypothesized that semi-automating the design process for 3D-printed, mouth-opening, tongue-depressing (MOTD) stents could standardize the design workflow and decrease design time.

Methods: Using oral stent design principles established over decades by oral oncologists, we created a customized computer program (Autostent) using MATLAB to semi-automate the design process of MOTD stents. We subsequently compared Autostent …

B lymphocytes play major adaptive immune roles, producing antibodies and driving T cell responses. However, how immunometabolism networks support B cell activation and differentiation in response to distinct receptor stimuli remains incompletely understood. To gain insights, we systematically investigated acute primary human B cell transcriptional, translational, and metabolomic responses to B cell receptor (BCR), TLR9, CD40-ligand (CD40L), IL-4, or combinations thereof. Periodontitis and type 2 diabetes mellitus are associated with each other in a bidirectional relationship. The mechanism by which diabetes affects periodontitis is understood to be due to the inflammatory effects of hyperglycemia (high blood glucose levels) and a hospitable environment for sugar-consuming bacteria. Periodontitis is known to be associated with higher levels of glycated hemoglobin (HbA1c), a long-term measurement of blood glucose control, and treatment of periodontitis reduces HbA1c. This dissertation presents an integrated research framework that bridges nanotechnology, green chemistry, and sustainable agriculture, aiming to address two critical global challenges: enhancing plant growth under resource-limited conditions and valorizing agricultural waste for bioactive compound extraction. The study is divided into three major projects that together highlight the innovative application of magnesium oxide-coated halloysite nanotubes (MgO-HNTs) and the development of environmentally conscious extraction methods for high-value phytochemicals.
