Type 2 diabetes mellitus (T2DM), characterized by insulin resistance, is closely associated with Alzheimer's disease (AD). Cerebrovascular dysfunction is manifested in both T2DM and AD, and is often considered as a pathological link between the two diseases. Insulin signaling regulates critical functions of the blood-brain barrier (BBB), and endothelial insulin resistance could lead to BBB dysfunction, aggravating AD pathology. However, insulin signaling is intrinsically dynamic and involves interactions among numerous molecular mediators. Hence, a mechanistic systems biology model is needed to understand how insulin regulates BBB physiology and the consequences of its impairment in T2DM and AD. In this study, …

Diabetes mellitus is a metabolic disorder that claims 1.5 million lives annually. Opuntia ficus-indica, commonly known as the prickly pear, has been commonly used in traditional folk medicine for its hypoglycemic properties. It has been claimed that it has antimicrobial and antioxidant properties as well. The plant is native to Mexico and is commonly found in the middle east and India and folk medicine from all of these areas have been using prickly pear medicinally for years. What Women Intercollegiate Coaches Say: Decision-Making Factors When Navigating Career Choices, Courtney Boucher, Nicole M. Lavoi
International Journal of Physical Activity and Health
To address misconceptions about the lack of women in sport coaching, factors influencing intercollegiate women coaches' career decisions, including their choices to leave positions, apply for new ones, and accept or deny job offers were explored. Utilizing a multi-level analysis underpinned by critical feminist theory, a comprehensive understanding of the systemic and structural barriers women coaches faced, as well as the strategies they employed to navigate their careers were illuminated. Key findings revealed that unequal pay, lack of administrative support, insufficient resources, and pervasive gender biases significantly impacted women's career choices. Biological nitrogen fixation is the conversion of dinitrogen (N2) gas into bioavailable nitrogen by microorganisms with consequences for primary production, ecosystem function, and global climate. Here we present a compiled dataset of 4793 nitrogen fixation (N2-fixation) rates measured in the water column and benthos of inland and coastal systems via the acetylene reduction assay, 15N2 labeling, or N2/Ar technique. While the data are distributed across seven continents, most observations (88%) are from the northern hemisphere. 15N2 labeling accounted for 67% of water column measurements, while the acetylene reduction assay accounted for 81% of benthic N2-fixation observations. Evidence of worship and religion in the form of architectural remains is rather scanty before the shift in human behavior, from hunter-gatherers to sedentary communities, and the emergence of permanent settlements. Communal and ritual structures first appeared in the Natufian culture (12,500–9400 BCE) and expanded during the Neolithic period (9400–5300 BCE), alongside the treatment of burials linked to ritual related to ancestor worship. The excavations at Motza’s Final PPNB (7100–6700 BCE) layer exposed many non-residential structures, some presumably used for worship and ritual. Some burials, including the one interpreted here as a shaman burial, included objects of symbolic importance. Cultic …

Legionella pneumophila, the causative agent of Legionnaires' disease, utilizes a Type IV secretion system (T4SS) to transport effector proteins into host cells. This bacterium is said to have over 300 effector proteins. Among these, the Ceg10 effector protein had remained uncharacterized, despite evidence suggesting the importance of bacterial effector proteins in the host-pathogen interaction. This study aimed to perform a structural and functional characterization of Ceg10 through transcriptomic, bioinformatics, and molecular analyses. Transcriptomic profiling of HEK 293T cells expressing Ceg10 revealed significant differential gene expression, with approximately 20 upregulated genes, including leucine-rich repeat, as well as other downregulated genes. …

This research focuses on the development and characterization of a biobased photocurable resin synthesized from soybean oil for use in stereolithography (SLA) 3D printing of microfluidic devices. The study explores a sustainable alternative to petroleum-based resins by performing an acrylation reaction on epoxidized soybean oil (ESO) to produce acrylated epoxidized soybean oil (AESO), which was then formulated into a photocurable resin with the addition of a photo initiator. The fabrication process involved designing microfluidic molds in SolidWorks and printing them using an Elegoo Mars 4 SLA printer under various exposure and layer thickness parameters. Once considered metabolic waste produced during exercise, lactate is now known to have diverse roles in biology. Muscle-derived lactate can travel through the bloodstream and be taken up by other tissues and i) oxidized, ii) converted to glucose via gluconeogenesis, or iii) bind to its receptor to trigger molecular signaling cascades. Lactate has been described as a “myokine” capable of inter-organ crosstalk and compelling evidence demonstrates lactate’s role as a signalling molecule in pathways related to the regulation of appetite. In my thesis, I examine how various forms of news media from the 1900s to present day in Ohlone territory have framed a particular hegemonic ideology around fire that matches early colonial journal entries – an ideology that is fueled by fear, sensationalism, and exclusion and directly commits violence against the Ohlone people. I investigate this time period because this is when modern news corporations, like The New York Times, rose into prominence. Industrial hemp (IH) establishment has many challenges including: poor emergence, low stand establishment, and variable plant development in direct seeded hemp. In floral hemp production, quality transplant production is a challenge with seedlings often becoming root bound and etiolated. In both direct seeded and transplant hemp, the relationship between root vigor and vegetative growth has not been studied and may prove insightful for managing emergence and establishment challenges. In vitro plant regeneration, particularly through organogenesis and somatic embryogenesis, is essential for plant transformation and crop improvement, especially in recalcitrant species like sunflower. In this study, we explore the genetic and molecular mechanisms underlying regeneration in Helianthus eggertii, a wild sunflower species with high regeneration potential. Global gene expression profiling, conducted using Oxford Nanopore sequencing across multiple time points and genotypes, identified sixty-three core differentially expressed genes (DEGs) critical to regeneration. These DEGs are associated with critical biological processes such as cell proliferation, transcriptional regulation, and photosynthesis. INTRODUCTION: Integrating scientific research across multiple disciplines to advance breakthroughs is at the heart of clinical-translational science (CTS); among competencies that have been identified as essential for progress, skillful communication is critical. Few tools are available to address the social dynamics of the multidimensional diversity characteristics of CTS. We created the "Building a Diverse Biomedical Workforce Through Communication Across Difference (CAD)" workshop intervention. Based on principles of intercultural communication, CAD taught novel situationally-based communication skills to dyads of near-peer mentors and their undergraduate mentees. Vitamin C (ascorbic acid) is a vital micronutrient recognized for its antioxidant capacity and essential bodily functions, such as collagen formation, neurotransmitter activity, and regulation of the immune system. This study explores how vitamin C can protect and potentially heal cells under oxidative stress and inflammation, using A549 cells, a human lung epithelial cell line, as the model system. Background: Melanoma arises from the uncontrolled multiplication of melanocytes, posing an escalating global health concern. Despite early detection and surgical removal being crucial, metastatic melanoma poses treatment challenges, with limited options like chemotherapy and immunotherapy. All-trans retinoic acid (ATRA), a natural metabolite of vitamin A, has shown promise in melanoma treatment by inducing differentiation, apoptosis, growth arrest, and immune modulation. However, ATRA treatment alone can lead to resistance and relapse. This article explores the transformative potential of fibre arts—such as quilting, weaving, and embroidery—as tools for personal growth, neuroplasticity, and well-being. Drawing on neuroscience and psychology, it demonstrates how engaging with fibre arts stimulates brain regions associated with problem-solving, sensory integration, and adaptability, fostering neural growth and enhancing cognitive flexibility. This thesis investigates the therapeutic potential of nanoformulated antiretroviral therapy (NF cART) and its implications for HIV-associated chronic inflammation and neurobehavioral outcomes. In the first study, NF cART demonstrated enhanced efficacy in modulating inflammasome activation and reducing viral reservoirs in gut-associated lymphoid tissue (GALT) and the small intestine of HIV-infected humanized mice. Key markers of inflammation (NLRP3, Caspase-1, and IL-1β) and viral replication (LTR and p24 levels) were significantly suppressed with NF cART compared to conventional therapy, though residual LTR expression persisted in the small intestine. Chondrostereum purpureum is a circumboreal polypore with high potential as a native fungal biocontrol for weedy, deciduous tree species in the Northern Hemisphere. This thesis describes the development of three multi-year studies designed to evaluate the effectiveness of C. purpureum as a management tool for invasive buckthorn (Rhamnus cathartica and Frangula alnus) in Upper Michigan, in a manner that tests a broader range of application methods beyond girdling or cut-stump treatments. Chronic alcohol use leads to metabolic dysfunction in adipose tissue. The underlying mechanisms and the contribution of alcohol-induced adipose tissue dysfunction to systemic metabolic dysregulation are not well understood. In our previous studies, we found that chronic alcohol feeding induces mesenteric lymphatic leakage, perilymphatic adipose tissue (PLAT) inflammation, and local insulin resistance in rats. The goal of this study was to further explore the link between alcohol-induced lymphatic leakage and PLAT immunometabolic dysregulation, locally and systemically, using in vivo and ex vivo approaches. Enhancing employee well-being is a primary goal for enterprise sustainable development. Although seafarers represent a major human resource in the maritime industry, the distinctive demands of their profession often compel them to compromise on certain elements in fulfilling their basic psychological needs, thereby contributing to mental health challenges. The dynamic cellular microenvironment, made up of resident cells and various macromolecules, differs markedly across tissues in the body. The multidirectional signals that cells receive from this microenvironment, along with the signaling they send back, heavily influence their physiology and behavior. Recent important findings have further validated this notion as the mammary microenvironment has been shown to suppress tumor progression by redirecting cancer cells to adopt a normal mammary epithelial progenitor fate in vivo. Despite the potential of oral immunotherapy against food allergy, adverse reactions and loss of desensitization hinder its clinical uptake. Dysbiosis of the gut microbiota is implicated in the increasing prevalence of food allergy, which will need to be regulated to enable for an effective oral immunotherapy against food allergy. Here we report an inulin gel formulated with an allergen that normalizes the dysregulated ileal microbiota and metabolites in allergic mice, establishes allergen-specific oral tolerance and achieves robust oral immunotherapy efficacy with sustained unresponsiveness in food allergy models. The weakness in students’ scientific argumentation, particularly in providing evidence-based arguments, has become a significant concern in education. This study aims to assess students’ scientific argumentation skills specifically on the topic of static fluids. The survey was conducted with 78 twelfth-grade students from private high schools in East Java, Indonesia, who had previously studied this topic. A scientific argumentation test instrument, consisting of an essay format with five questions, was employed to collect data. The data were analyzed using descriptive statistics, followed by a classification based on static fluid subtopics and the level of scientific argumentation skill. The results indicated …

Invasive species harm natural and managed ecosystems. Awareness and management of these species depends on effective education and outreach. Traditional common names, including those with geographic references, for many invasive pests may perpetuate slanderous terms or stigmatize people from that place. To create more inclusive invasive species educational materials, the University of Minnesota Extension’s Invasive Species Community of Practice developed guidelines for selection of common names. Suggested names were shared with others involved in invasive species communications, leading to broader adoption. These guidelines may be useful to others who struggle to find descriptive, non-alienating common names for invasive species.

Ferroptosis has been recognized as a unique cell death modality driven by excessive lipid peroxidation and unbalanced cellular metabolism. In this study, we established a protein interaction landscape for ferroptosis pathways through proteomic analyses, and identified choline/ethanolamine phosphotransferase 1 (CEPT1) as a lysophosphatidylcholine acyltransferase 3 (LPCAT3)-interacting protein that regulates LPCAT3 protein stability. In contrast to its known role in promoting phospholipid synthesis, we showed that CEPT1 suppresses ferroptosis potentially by interacting with phospholipases and breaking down certain pro-ferroptotic polyunsaturated fatty acid (PUFA)-containing phospholipids. Together, our study reveals a previously unrecognized role of CEPT1 in suppressing ferroptosis.
