Investigating The Immunomodulatory Effects Of Cdk8/19 Inhibitor Senexin631 In A Her2-Positive Breast Cancer Model, Stephen O'Rourke
Theses and Dissertations
HER2-positive (HER2⁺) breast cancer is aggressive and often develops resistance to anti-HER2 therapies. CDK8 and CDK19, transcription-associated cyclin-dependent kinases (tCDKs) in the Mediator complex, are dysregulated in HER2⁺ breast cancer, correlate with poor survival, and support transcriptional escape driving therapeutic resistance. Previous studies Previous studies in immune-deficient models demonstrated that CDK8/19 inhibition (CDK8/19i) reduces pro-tumor macrophages (M2Φs), but its role in immune-competent tumors and innate immune–mediated tumor control remains unclear. Since HER2⁺ tumors are often immunogenic and anti-HER2 therapies rely on innate immune activation, reprogramming immunosuppressive M2Φ toward an M1Φ-like phenotype could enhance antitumor immunity. We hypothesized that CDK8/19i modulates …
Novel Approaches To Combat Neuro-Degradation: Investigations Of Two Supplemental Strategies Aimed At Improving Warfighter Performance, Blaine Lints
Theses and Dissertations
This dissertation examines the efficacy of two distinct nutritional supplementation strategies for improving physical and/or cognitive performance under stress. The first study investigated the effects of 150 mg caffeine plus 150 mg theacrine ingestion in tactically trained individuals. Results indicated that the combination of caffeine and theacrine was associated with significant improvements in reaction time, accuracy, and measures of ocular control following high-intensity interval (HII) exercise. The HII exercise protocol consisted of ten repetitions of 60s high-intensity intervals (≥90% maximal oxygen uptake [VO2max]) followed by 120s low-intensity intervals (~40% VO2max) on a treadmill. These benefits were …

- Current evidence suggests that liana (woody vine) competition with trees could be threatening the carbon sink by reducing carbon uptake and storage in tropical forests. Previous studies investigating forest demography in liana removal experiments have, however, assumed similar allometries for trees, regardless of the amount of lianas they support. Yet, recent observations suggest that liana load may alter tree shape and structure, including height, which could result in an underestimation of the liana effects on forest carbon stocks and tree growth.
- In this study, we used terrestrial laser scanning (TLS) in a liana removal experiment in Gigante Peninsula, Panama, collected …

Objective: Macrophage accumulation in metabolically active tissues during obesity is common in both animals and humans, but the lipid signaling mechanisms that trigger macrophage inflammation remain unclear. This study investigates the role of Ces1d, an unconventional lipase, in regulating macrophage inflammation under nutritional stress.
Methods: A myeloid-specific Ces1d knockout (LysM-Cre-Ces1d floxed/floxed, KO) mouse model was used for the studies. For in vitro tests, bone marrow-derived macrophages (BMDMs) from control (Ces1d floxed/floxed, WT) and KO mice were assessed for migration, polarization, and activation. For in vivo experiments, WT and KO mice were induced to obesity via a high-fat diet (HFD) and …

Understanding disease progression and sophisticated tumor ecosystems is imperative for investigating tumorigenesis mechanisms and developing novel prevention strategies. Here, we dissected heterogeneous microenvironments during malignant transitions by leveraging data from 1396 samples spanning 13 major tissues. Within transitional stem-like subpopulations highly enriched in precancers and cancers, we identified 30 recurring cellular states strongly linked to malignancy, including hypoxia and epithelial senescence, revealing a high degree of plasticity in epithelial stem cells. By characterizing dynamics in stem-cell crosstalk with the microenvironment along the pseudotime axis, we found differential roles of ANXA1 at different stages of tumor development. In precancerous stages, reduced …

Objective: Periventricular nodular heterotopia (PVNH) is a malformation of cortical development with high rates of epilepsy. The extent to which nodules participate in normal cerebral functions in addition to pathological processes is unclear. The authors assessed the functional utility (i.e., cognitive impacts) of surgically ablated epileptogenic PVNH tissue via comprehensive neuropsychological testing.
Methods: The sample included 32 patients with PVNH who underwent presurgical neuropsychological evaluation and a subsample of 16 patients who underwent MR-guided laser interstitial thermal therapy (MRgLITT) of nodules, with postsurgical neuropsychological testing. In 11 patients in whom intracranial recordings were performed, reading and naming tasks were tested …

Background: Complex Regional Pain Syndrome (CRPS) is a challenging and often disabling condition marked by persistent pain, most commonly in a limb following injury or surgery. It presents with a wide array of symptoms, including intense pain, swelling, alterations in skin color and temperature, motor dysfunction, and trophic changes such as skin and tissue atrophy. While the precise cause of CRPS is not fully understood, it is thought to stem from abnormal nervous system activity, leading to heightened pain sensitivity and inflammatory responses. A thorough understanding of CRPS is essential for accurate diagnosis, effective treatment, and enhancing patients' quality of …

Vapour pressure deficit (VPD) and blue light (BL) are import signals for stomatal regulation and highly relevant during cultivation of economically important crops such as cucumber in controlled environments. However, the information about combined effects of VPD and BL and the regulation of abscisic acid (ABA) in this respect, is limited and seems to vary between species. This study investigated how moderate (1.12 kPa) or low (0.28 kPa) VPD combined with 5% or 30% BL affect the foliar ABA metabolism, transpiration rate, nutrient uptake, and growth in well-watered cucumber plants (Cucumis sativus ‘Quatro’). Additional BL, regardless of VPD, resulted in …

Multi-modal spatial omics data are invaluable for exploring complex cellular behaviors in diseases from both morphological and molecular perspectives. Current analytical methods primarily focus on clustering and classification, and do not adequately examine the relationship between cell morphology and molecular dynamics. Here, we present MorphLink, a framework designed to systematically identify disease-related morphological-molecular interplays. MorphLink has been evaluated across a wide array of datasets, showcasing its effectiveness in extracting and linking interpretable morphological features with various molecular measurements in spatial omics analyses. These linkages provide a transparent view of cellular behavior heterogeneity within tissue regions with similar cell type compositions, …

Significance: This review investigates how radiation therapy (RT) increases the risk of delayed cardiovascular disease (CVD) in cancer survivors. Understanding the mechanisms underlying radiation-induced CVD is essential for developing targeted therapies to mitigate these effects and improve long-term outcomes for patients with cancer.
Recent Advances: Recent studies have primarily focused on metabolic alterations induced by irradiation in various cancer cell types. However, there remains a significant knowledge gap regarding the role of chronic metabolic alterations in normal cells, particularly vascular cells, in the progression of CVD after RT.
Critical Issues: This review centers on RT-induced metabolic alterations in vascular cells …

Contributions Of Citizen Science Data To Butterflies Of The Lower Rio Grande Valley, Akane Jazmyn Benitez
Theses and Dissertations
Citizen science-provided insect data inherently exhibits variability in accuracy. In the Lower Rio Grande Valley (LRGV), butterfly occurrence reporting is predominantly reliant on citizen science contributions. My research compared fieldwork conducted monthly over a year to occurrence records submitted by visitors at the National Butterfly Center and in Mission, Hidalgo County, Texas. I hypothesized that butterfly occurrences at the Center would be maximized during the fall and minimized during winter and summer months. My research findings revealed that butterfly taxa richness is contingent upon the following factors: 1) the impact of citizen science submitting reports, and 2) monthly butterfly activity …

Kernza® is the trademarked name of the perennial crop intermediate wheatgrass (Thinopyrum intermedium). In addition to its potential for dual-use as a grain and forage crop, Kernza is marketed to provide needed environmental services by increasing crop biodiversity, sequestering soil carbon, and reducing nitrate leaching. However, more work is needed in cultivar selection and crop management to increase total grain yields and prevent long-term grain yield decline. Furthermore, research in quantifying and understanding Kernza’s ability to affect soil properties and water dynamics is limited.
Two field experiments were conducted at the Eastern Nebraska Research Extension and Education …

Lavandula angustifolia Mill., lavender, is an aromatic plant in the Lamiaceae family. Lavender, which is native to the Mediterranean region but cultivated throughout the world, is an important economic plant. Several studies have investigated two aspects of this aromatic plant: (1) which pollinators, particularly bees, pollinate lavender, and (2) the composition of lavender essential oil. However, little research has been conducted to investigate how pollination affects either the yield or phytochemistry of lavender. The current study, which was conducted in North America, investigates which bee species visit lavender and how pollination affects plant chemistry, specifically the essential oil produced by …

Subcellular targeting of signaling enzymes influences where and when various modes of intracellular communication operate. Macromolecular complexes of signal transduction and signal termination elements favor reversible control of repetitive processes. This includes adrenergic stimulation of excitation–contraction coupling in the heart. Long isoforms of A-kinase anchoring protein 18 (AKAP18γ and δ) modulate this process via regulation of calcium uptake into the sarcoplasmic reticulum through the Ca2+ATPase 2a (SERCA2a). AKAP18 proximity-proteomic screening in cardiomyocytes identifies networks for protein kinase A (PKA) and ubiquitin-specific proteinases (USPs). A 2′phosphoesterase domain on AKAP18 interfaces with the USP4 isoform at the Z bands of sarcomeres. PKA …

Background and purpose: Accurate delineation of orodental structures on radiotherapy computed tomography (CT) images is essential for dosimetric assessment and dental decisions. We propose a deep-learning (DL) auto-segmentation framework for individual teeth and mandible/maxilla sub-volumes aligned with the ClinRad osteoradionecrosis staging system.
Materials and methods: Mandible and maxilla sub-volumes were manually defined on simulation CT images from 60 clinical cases, differentiating alveolar from basal regions; teeth were labelled individually. For each task, a DL segmentation model was independently trained. A Swin UNETR-based model was used for mandible sub-volumes. For smaller structures (e.g., teeth and maxilla sub-volumes) a two-stage model first …

Cellular Indexing of Transcriptomes and Epitopes by Sequencing (CITE-seq) enables paired measurement of surface protein and mRNA expression in single cells using antibodies conjugated to oligonucleotide tags. Due to the high copy number of surface protein molecules, sequencing antibody-derived tags (ADTs) allows for robust protein detection, improving cell-type identification. However, variability in antibody staining leads to batch effects in the ADT expression, obscuring biological variation, reducing interpretability, and obstructing cross-study analyses. Here, we present ADTnorm, a normalization and integration method designed explicitly for ADT abundance. Benchmarking against 14 existing scaling and normalization methods, we show that ADTnorm accurately aligns populations …

Circulating blood platelets are responsible for maintaining hemostasis. They are released into blood vessels from mature megakaryocytes. Although several transcription factors have been reported to orchestrate the transcriptional programs required for platelet production, how chromatin regulators control these processes is still poorly understood. MLL3 and MLL4 are the main lysine methyltransferases responsible for the deposition of H3K4me1 histone marks at enhancers. MLL3 and MLL4 typically form complexes with other co-factors, such as PTIP. Recently, we showed that loss of PTIP leads to decreased platelet numbers in mice. Here, we find that, although MLL3/4 double deficiency does not alter megakaryopoiesis and …

The treatment landscape of non-small-cell lung cancer (NSCLC) is evolving rapidly, driven by advances in the development of targeted agents and immunotherapies. Despite this progress, some patients have suboptimal responses to treatment, highlighting the need for new therapeutic strategies. In the past decade, the important role of the tumour microenvironment (TME) in NSCLC progression, metastatic dissemination and response to treatment has become increasingly evident. Understanding the complexity of the TME and its interactions with NSCLC can propel efforts to improve current treatment modalities, overcome resistance and develop new treatments, which will ultimately improve the outcomes of patients. Memantine is an US Food and Drug Administration (FDA) approved drug that is thought to selectively inhibit NMDA-subtype of ionotropic glutamate receptors (NMDARs). NMDARs enable calcium influx into neurons and are critical for normal brain function. However, increasing evidence shows that calcium influx in neurological diseases is augmented by calcium-permeable AMPA-subtype ionotropic glutamate receptors (AMPARs). Here, we demonstrate that these calcium-permeable AMPARs (CP-AMPARs) are inhibited by memantine. Electrophysiology unveils that memantine inhibition of CP-AMPARs is dependent on their calcium permeability and the presence of their neuronal auxiliary subunit transmembrane AMPAR regulatory proteins (TARPs). Through cryo-electron microscopy we elucidate that memantine …

Advanced practitioners (APs), including nurse practitioners (NPs), physician assistants (PAs), clinical nurse specialists, and pharmacists, are pivotal in health care. Despite their critical role, record numbers of APs are leaving the profession, primarily due to poor work-life balance, which contributes to burnout and high turnover rates. This article explores the impact of administrative time on AP job satisfaction and retention, drawing from recent surveys and case studies. Findings indicate that dedicated administrative time significantly enhances job satisfaction and reduces burnout, with data showing that APs with administrative time are less likely to leave their roles. The Advanced Practitioner Society for …

BAU Journal - Science and Technology
Afforestation is a promising strategy for improving soil quality and enhancing carbon sequestration, contributing to climate change mitigation efforts. This study assessed the impact of afforestation on soil parameters in two afforested areas and one natural forest, compared with adjacent non-forested sites in Rashaya al-Wadi, Lebanon. Soil samples (0–30 cm depth) were analyzed for texture, pH, electrical conductivity (EC), soil organic matter (SOM), soil organic carbon (SOC), organic nitrogen (ON), mineral nitrogen (MN), and available phosphorus (AP) using ISO and standardized protocols at the Lebanese Agricultural Research Institute (LARI). The findings indicate that afforestation significantly increased SOM, SOC, and ON, …

The present study provides the characterization of the phytochemical content and the antibacterial activity of ethanolic extracts from the leaves (LM) and stems (SM) of Micromeria barbata (M.barbata) against Pseudomonas aeruginosa . Important functional groups were determined by analyzing the FTIR spectra of LM and SM. The phytochemical profiles were analyzed by GC-MS, and these characterized the chemicals according to retention periods and peak regions. The binding affinities of discovered compounds with the P. aeruginosa LasR and PqsD proteins were evaluated using molecular docking approach. Assays for biofilm formation, MIC, MBC, and agar well diffusion were used to assess the …
Machine Learning Applications For Berth Allocation Optimization In Major Red Sea Ports: A Multi-Port Analysis, Mohammed H. Alshareef
Machine Learning Applications For Berth Allocation Optimization In Major Red Sea Ports: A Multi-Port Analysis, Mohammed H. Alshareef
Journal of King Abdulaziz University: Marine Science
Efficient berth allocation remains a critical challenge in Red Sea port operations, particularly given the diverse operational capabilities across the region's ports. The research utilized historical operational data from 147,892 berth allocation instances across these ports spanning 2019--2023. A deep learning model was developed using four dense layers with ReLU activation functions, incorporating vessel characteristics, weather patterns, and operational constraints. The model's performance was validated through comparison with traditional allocation methods using real-time port operation data. Port selection criteria included geographical distribution along the Red Sea, varying levels of technological adoption, and willingness to share operational data for research purposes. …

From Differentiation To Disease: Cyclin C Directly Coordinates Transcription And Mitochondrial Dynamics, Alicia Campbell
Theses and Dissertations
Organelle crosstalk is essential for coordinating cellular responses to various external cues, including stress and differentiation. Two key organelles, the nucleus and mitochondria, engage in extensive crosstalk to manage responses to a range of environmental and developmental signals. Complex processes such as stem cell differentiation and neuronal development rely on organelle crosstalk. Both processes integrate changes in gene expression with cytosolic and organellar remodeling. However, it remains unclear how these two complex processes are coordinated to achieve the desired outcome. This project concentrates on cyclin C (CCNC), a component of the Mediator Kinase Module (MKM), and its role in directly …
