Tedera (Bituminaria bituminosa (L.) C.H. Stirt.) is a Mediterranean drought-tolerant species that shows potential as a forage perennial legume for Mediterranean livestock systems. This three-year study investigated the phenology of the newly developed variety of tedera (Lanza) in response to an annual 24% rainfall reduction and competition compared to alfalfa (Medicago sativa L.) in a typical Mediterranean environment of the Iberian Peninsula. Tedera showed early phenology for the reproductive stages from inflorescence emergence to ripening compared to alfalfa, with a long flowering period from early April to mid-May, overlapping with mature fruits. In general, tedera responded to a …

Given the importance of protected areas for biodiversity, the growth of visitation to many areas has raised concerns about the effects of humans on wildlife. In 2020, the COVID-19 pandemic led to temporary closure of national parks in the United States, offering a pseudonatural experiment to tease apart the effects of permanent infrastructure and transient human presence on animals. We compiled GPS tracking data from 229 individuals of 10 mammal species in 14 parks and used third-order hierarchical resource selection functions to evaluate the influence of the human footprint on animal space use in 2019 and 2020. Averaged across all …

Control of tsetse flies constitutes a cornerstone of trypanosomiasis control and elimination efforts in Africa. The use of eco-friendly odor-based bait technologies has been identified as a safer method for control of tsetse flies. These technologies are significantly augmented by development of effective repellents that reduce contact between trypanosome-infected tsetse flies and their vertebrate hosts. Waterbuck Repellent Compounds (WRC) and Novel Repellent Blend (NRB) are recently developed tsetse fly repellent formulations. Information on relative efficacy of these formulations against major tsetse fly vectors of trypanosomiasis in Kenya is limited. Such information can inform choices of repellent technology for optimal control …

Recent studies have highlighted the importance of mitochondria in NP cells and articular chondrocyte health. Since the understanding of mechanisms governing mitochondrial dynamics in these tissues is lacking, we investigated the role of OPA1, a mitochondrial fusion protein, in their homeostasis. OPA1 knockdown in NP cells altered mitochondrial size and cristae shape and increased the oxygen consumption rate. OPA1 governed the morphology of multiple organelles, including peroxisomes, early endosomes and cis-Golgi and loss resulted in the dysregulation of autophagy. Metabolic profiling and

Bioenergy Crop Production: Implications For Grassland Bird Communities In Southwestern Nebraska, Grace E. Schuster Jul 2025

Bioenergy Crop Production: Implications For Grassland Bird Communities In Southwestern Nebraska, Grace E. Schuster

School of Natural Resources: Dissertations, Theses, and Student Research

Biofuel and bioenergy systems are components of most climate stabilization pathways to reduce greenhouse gas emissions and limit global warming. Currently, corn (Zea mays) is the predominant feedstock used for bioenergy production in the United States. However, widespread production of this monoculture crop has resulted in many negative environmental impacts. The most notorious impact has been the loss of grassland habitat due to agriculture expansion which has had detrimental effects on wildlife that depend on grassland habitat. One such group, grassland birds, has faced steeper, more consistent, and more widespread declines than any other avian guild. Therefore, strategies …

Overcoming Maternal Antibody Interference In Swine Influenza: Advancing Dna Vaccine Strateies For Enhanced Immune Protection In Piglets, Danh Cong Lai Jul 2025

Overcoming Maternal Antibody Interference In Swine Influenza: Advancing Dna Vaccine Strateies For Enhanced Immune Protection In Piglets, Danh Cong Lai

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Maternally derived antibodies (MDAs) play a vital role in protecting neonates from infectious diseases, but their presence at the time of vaccination can interfere with vaccine-induced immune responses, thereby reducing vaccine effectiveness. MDA interference has been documented in pigs vaccinated with whole inactivated virus (WIV) vaccines against swine influenza A virus (IAV). In this thesis, we evaluated the efficacy of a lipid nanoparticle encapsulated DNA (LNP-DNA) vaccine against swine IAV in the presence and absence of MDAs, comparing it to a WIV vaccine. In MDA-negative piglets, both the LNP-DNA and WIV vaccines induced strong immune responses and effectively prevented the …

Identification Of Novel Components Of The Retinal Determination Gene Network In Drosophila Cell Lines, Robert Drewell, Jacqueline Dresch Jul 2025

Identification Of Novel Components Of The Retinal Determination Gene Network In Drosophila Cell Lines, Robert Drewell, Jacqueline Dresch

Biology

The retinal determination genetic network controls the development of the visual system in all seeing animals through the molecular regulation of cells to adopt an eye tissue fate. The compound eye of the fruit fly, Drosophila melanogaster, is an excellent model system to study the complex mechanisms within the network that regulate specification of cellular identity during embryogenesis. In Drosophila, the two Pax6 paralogues, eyeless and twin of eyeless, sit at the very pinnacle of the network and their expression early in development activates critical downstream components of the retinal determination pathway. In this study, we investigate the expression of …

With limited land and water resources available it has become critical to maximize output for agricultural products while maintaining sustainable farming practices. Ricefish co-culture can be used to maximize limited land and water resources for production of rice and fish concurrently and reduce the need of chemical inputs. While rice-fish co-culture has been practiced in Asia for centuries, very few studies have examined the feasibility and economic impact of rice fish co-culture in the U.S.A. Two studies were conducted in Stuttgart, Arkansas, U.S.A. from 2023-2024 to investigate the suitability of raising Koi carp in flooded rice paddies with a sushi …

Investigating The Role Of Msbdnf – Trkb Signaling In The Maintenance Of Mitochondrial Populations Through The Bnip3 Mitophagy Pathway, Jack A. Horton Jul 2025

Investigating The Role Of Msbdnf – Trkb Signaling In The Maintenance Of Mitochondrial Populations Through The Bnip3 Mitophagy Pathway, Jack A. Horton

All NMU Master's Theses

Maintenance of mitochondrial transport and clearance at the pre- and post-synaptic terminal is an essential function of neuromuscular junctions to meet their high energy demands. Damaged or dysfunctional mitochondria can put stress on these synaptic junctions, ultimately leading to the degradation of the synapse and subsequent cell death. Dysfunctional mitochondria are also often present in neuromuscular diseases such as ALS. One possible mechanism by which neurons may eliminate dysfunctional mitochondria is through the muscle-synthesized BDNF (msBDNF) /TrkB-mediated Bcl-2 nineteen-kD interacting protein (BNIP3) mitophagy pathway, although the relationship is not well-defined. BNIP3 localizes to the mitochondria and triggers a loss of …

Elucidating The Role Of Mir-10a/B In The Wound Healing Process In Larval Zebrafish, Jacquelyn E. Jacobs

All NMU Master's Theses

This study explores the regulatory role of microRNA-10 (miR-10) in larval zebrafish caudal fin regeneration, a model with broad implications for the future of regenerative medicine. Building on prior research from the lab, which indicated that miR-10 knockdown impairs regeneration, we verified this finding through rescue experiments, assessed its influence on healing projections, and examined its role in the innate immune response during the wound healing process. To address these questions, we employed a multi-faceted approach, including timed matings, microinjections, immunohistochemistry, fixed sample microscopy, and live imaging. Our results show that the experimental introduction of exogenous miR-10 can rescue the …

Contributions Of Citizen Science Data To Butterflies Of The Lower Rio Grande Valley, Akane Jazmyn Benitez Jul 2025

Contributions Of Citizen Science Data To Butterflies Of The Lower Rio Grande Valley, Akane Jazmyn Benitez

Theses and Dissertations

Citizen science-provided insect data inherently exhibits variability in accuracy. In the Lower Rio Grande Valley (LRGV), butterfly occurrence reporting is predominantly reliant on citizen science contributions. My research compared fieldwork conducted monthly over a year to occurrence records submitted by visitors at the National Butterfly Center and in Mission, Hidalgo County, Texas. I hypothesized that butterfly occurrences at the Center would be maximized during the fall and minimized during winter and summer months. My research findings revealed that butterfly taxa richness is contingent upon the following factors: 1) the impact of citizen science submitting reports, and 2) monthly butterfly activity …

Oxytocin As A Possible Vegf Inhibitor For Treatment Of Diabetic Retinopathy, Daira Yepez Jul 2025

Oxytocin As A Possible Vegf Inhibitor For Treatment Of Diabetic Retinopathy, Daira Yepez

Theses and Dissertations

Diabetic retinopathy (DR) is a major complication of diabetes and a leading cause of vision loss largely driven by the overexpression of vascular endothelial growth factor (VEGF), which promotes neovascularization and vascular permeability in the retina. Current treatments, primarily anti-VEGF injections, inhibit the effects of VEGF once it has been secreted but can be ineffective in a certain subset of patients. Recent studies suggest that oxytocin (OXT), a neuropeptide mainly known for its roles in reproduction and social bonding, may downregulate VEGF expression. This study aimed to investigate whether OXT influences VEGF secretion in the human retinal pigment epithelial (RPE) …

The Diabetes Prevention Program And Its Outcomes Study: Niddk's Journey Into The Prevention Of Type 2 Diabetes And Its Public Health Impact, Jill P Crandall, Dana Dabelea, William C Knowler, David M Nathan, Marinella Temprosa, Dpp Research Group

Children’s Nutrition Research Center Staff Publications

The current-day epidemic of type 2 diabetes, largely driven by increased adiposity and reduced physical activity in the setting of genetic susceptibility, is a major public health challenge. The National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) presciently proposed the Diabetes Prevention Program (DPP), a multicenter randomized clinical trial, designed by investigators in conjunction with NIDDK staff and initiated in 1996. The primary goal of DPP was to determine whether an intensive lifestyle intervention (ILS) or metformin in comparison with placebo would reduce the development of diabetes in a high-risk population with prediabetes. After mean 2.8 years, ILS …

To Pause With A Cliffhanger Or A Temporary Closure? The Differential Impact Of Serial Versus Episodic Narratives On Children's Physical Activity Behaviors, Amy Shirong Lu, Melanie C Green, Caio Victor Sousa, Jungyun Hwang, I-Min Lee, Debbe Thompson, Tom Baranowski Jul 2025

To Pause With A Cliffhanger Or A Temporary Closure? The Differential Impact Of Serial Versus Episodic Narratives On Children's Physical Activity Behaviors, Amy Shirong Lu, Melanie C Green, Caio Victor Sousa, Jungyun Hwang, I-Min Lee, Debbe Thompson, Tom Baranowski

Children’s Nutrition Research Center Staff Publications

Research has supported the effectiveness of narratives for promoting health behavior, but different narrative presentation formats (serial vs. episodic) have seldom been compared. Suspense theories suggest that serial narratives, which do not provide a full resolution at the end of an episode, may create higher motivation for continued engagement with a story. Forty-four 8 to 12-year-old children were randomly assigned to watch an animation series designed for an existing active video game in which the plot was delivered either continuously across multiple episodes (serial) or in multiple yet relatively independent self-contained episodes (episodic). Controlling for social desirability, children who watched …

Biology Faculty Works

Background: Cardiac neural crest cells (cNCCs) are critical for heart development, and their disruption can result in congenital heart defects. Serotonin (5-HT) signaling, specifically via 5-HT2B and 5-HT2C receptors, regulates diverse physiological processes, including neural crest migration. This study investigates how modulation of 5-HT2B and 5-HT2C receptor activity impacts cNCC migration and the development of their derivatives, with relevance to serotonergic drug safety during pregnancy.

Methods

Chicken embryos at HH8 were treated with 50 μL of 20 μM 1-Methylpsilocin (1-MP), an inverse agonist of 5-HT2B and agonist of 5-HT2C, and collected at HH14 to assess cNCC migration. Embryos were pre-treated …

Malheur NWR, one of the oldest (established 1908) and largest refuges in the system, is one of those with significant habitat for mammals. In addition to the refuge, we have included Harney County, …

Investigating The Immunomodulatory Effects Of Cdk8/19 Inhibitor Senexin631 In A Her2-Positive Breast Cancer Model, Stephen O'Rourke Jul 2025

Investigating The Immunomodulatory Effects Of Cdk8/19 Inhibitor Senexin631 In A Her2-Positive Breast Cancer Model, Stephen O'Rourke

Theses and Dissertations

HER2-positive (HER2⁺) breast cancer is aggressive and often develops resistance to anti-HER2 therapies. CDK8 and CDK19, transcription-associated cyclin-dependent kinases (tCDKs) in the Mediator complex, are dysregulated in HER2⁺ breast cancer, correlate with poor survival, and support transcriptional escape driving therapeutic resistance. Previous studies Previous studies in immune-deficient models demonstrated that CDK8/19 inhibition (CDK8/19i) reduces pro-tumor macrophages (M2Φs), but its role in immune-competent tumors and innate immune–mediated tumor control remains unclear. Since HER2⁺ tumors are often immunogenic and anti-HER2 therapies rely on innate immune activation, reprogramming immunosuppressive M2Φ toward an M1Φ-like phenotype could enhance antitumor immunity. We hypothesized that CDK8/19i modulates …

Subcellular targeting of signaling enzymes influences where and when various modes of intracellular communication operate. Macromolecular complexes of signal transduction and signal termination elements favor reversible control of repetitive processes. This includes adrenergic stimulation of excitation–contraction coupling in the heart. Long isoforms of A-kinase anchoring protein 18 (AKAP18γ and δ) modulate this process via regulation of calcium uptake into the sarcoplasmic reticulum through the Ca2+ATPase 2a (SERCA2a). AKAP18 proximity-proteomic screening in cardiomyocytes identifies networks for protein kinase A (PKA) and ubiquitin-specific proteinases (USPs). A 2′phosphoesterase domain on AKAP18 interfaces with the USP4 isoform at the Z bands of sarcomeres. PKA …
