Metformin is the most commonly prescribed antidiabetes drug, yet its precise mechanism of action remains controversial. Previous studies have suggested that metformin acts peripherally by reducing hepatic glucose output and altering gut functions. Here, we report a neural mechanism via the small guanosine triphosphatase Ras-related protein 1 (Rap1). Mice with forebrain-specific Rap1 knockout exhibited resistance to the antidiabetic effects of low-dose metformin while remaining sensitive to other antidiabetic agents. Centrally administered metformin inhibited brain Rap1 and reduced hyperglycemia. Conversely, forced activation of brain Rap1 increased glycemia and abolished the glycemic effect of metformin. Development of acquired therapeutic resistance limits the efficacy of cancer treatments and accounts for therapeutic failure in most patients. How resistance arises, varies across cancer types and differs depending on therapeutic modalities is incompletely understood. Novel strategies that address and overcome the various and complex resistance mechanisms necessitate a deep understanding of the underlying dynamics. We are at a crucial time when innovative technologies applied to patient-relevant tumour models have the potential to bridge the gap between fundamental research into mechanisms and timing of acquired resistance and clinical applications that translate these findings into actionable strategies to extend therapy efficacy. …

The nervous system has a pivotal role in cancer biology, and pathological investigations have linked intratumoural nerve density to metastasis1. However, the precise impact of cancer-associated neurons and the communication channels at the nerve–cancer interface remain poorly understood. Previous cancer denervation models in rodents and humans have highlighted robust cancer dependency on nerves, but the underlying mechanisms that drive nerve-mediated cancer aggressivity remain unknown2,3. Here we show that cancer-associated neurons enhance cancer metabolic plasticity by transferring mitochondria to cancer cells. Breast cancer denervation and nerve–cancer coculture models confirmed that neurons significantly improve tumour energetics. …

Background & aims: Metastatic cancers arise from a decades-long succession of increasingly virulent precursor lesions, each of which represents prospective targets for therapeutic intervention. This evolutionary process has been particularly vivid in esophageal adenocarcinoma (EAC), as this cancer and associated precursor lesions, including Barrett's esophagus (BE), low-grade dysplasia (LGD), and high-grade dysplasia (HGD), coexist in an accessible, 2-dimensional pattern in esophageal mucosa. Given the durability of these precursor lesions, it is likely that they, like EAC, rely on stem cells for their regenerative growth. Ecosystem services are vital to human welfare and society, but often are overlooked when it comes to planning and setting policy. By neglecting to incorporate the benefits received from natural resources, policy makers may unknowingly be making decisions that harm the long term sustainability and use of these services. Small Molecule Inhibitor 3i-1000 Dosed During Bouts Of Anoxia Increases Survivability Of Austrofundulus Limnaeus Embryos, Connor Kime

The annual killifish Austrofundulus limnaeus possesses exceptional tolerance to prolonged anoxia, offering a unique vertebrate model for investigating cardiac resilience and survival under metabolic stress. This study tested whether inhibition of the GATA4–NKX2-5 transcriptional axis with the small molecule 3i-1000 enhances post-anoxic survival. WS40 embryos were exposed to 50 µM 3i-1000 during anoxia and early aerobic recovery. Outcomes were assessed across two independent experimental cohorts. Treated embryos exhibited higher survivability during reoxygenation: in the October cohort, survival increased from 76.0% in vehicle controls (n = 25) to 91.7% in treated embryos (n = 24); in the November cohort, survival improved …

Reptiles and amphibians are commonly used indicator species in wetlands because of their increased sensitivity to environmental conditions and use of both aquatic and terrestrial habitats. Herpetofauna can reflect wetland ecological functionality through their community composition and responsiveness to environmental stressors. This study evaluated reptile and amphibian communities in natural and ACEP-WRE wetlands within the Red River Watershed of northeastern Texas and Northwestern Louisiana to assess whether restored wetlands support herpetofaunal assemblages compares to their natural counterparts. Turkey hemorrhagic enteritis virus (THEV) is an adenovirus (AdV) that causes hemorrhagic enteritis (HE) in turkeys, a debilitating disease affecting predominantly 6-12-week-old turkey poults characterized by immunosuppression (IMS) and intestinal lesions leading to bloody diarrhea. Although a naturally-occurring avirulent THEV strain has been adopted as live vaccine and has proven effective at preventing HE in turkeys, it still retains its immunosuppressive ability. Thus, vaccinated birds are rendered more susceptible to opportunistic infections and death than unvaccinated cohorts, leading to substantial economic losses. The tumor immune microenvironment (TIME) critically influences cancer progression and determining therapeutic response. As immunotherapies continue to improve and reshape cancer treatments, there is a need for tools that can more efficiently and accurately assess and monitor the TIME. Extracellular vesicles (EVs), especially small EVs (sEVs), are lipid-bound nanoparticles that carry proteins, DNA, and RNA reflective of their cell of origin, offering a novel, non-invasive biomarker of the TIME. They act as settlement cues for coral larvae, but this interaction is often species-specific, as CCA species can be either inducers or inhibitors to larval settlement and metamorphosis. Bird infestations in industrial areas can cause infrastructure damage, contamination, and health risks. Despite these impacts, they are rarely addressed in environmental management strategies. At the Kaltim Industrial Estate (KIE) in Bontang, East Kalimantan, synanthropic bird species that thrive in human-altered environments have become persistent nuisances, particularly because of structural opportunities for roosting and altered ecological conditions. Anthropogenic activities such as logging and converting mangrove forests into plantations have degraded coastal ecosystems and reduced their ecological stability. Silvofishery is a restoration system that balances the ecological and economic functions of mangroves. [Excerpt] Interspecific interactions are relatively rare among coinhabiting Orthoptera. Most records are of overzealous males courting females not of their species or, for that matter, inanimate objects. Less frequent are reports of two signalling males interacting, yet these are of interest. Calling songs are used to distinguish species (Alexander 1956). The possibility then exists that the "interference" or, more correctly, the competition for broadcast space that occurs when two closely related species with similar songs interact, may have been influential in shaping aspects of song delivery or pattern.

Leaf protein concentrate extracted from tree legume Leucaena leucocephala cultivar Cunningham (LLP) was stored under humid tropical conditions (24-28°C and 84-92% rh) in the form of wet cake during a 10 day period. Samples of LLP were taken every second day for chemical analyses and protein quality assays. The effect of storage on dried LLP was also assessed against two control proteins: peanut meal (PNM) and fish meal (FM), either free of aflatoxins or infected by the aflatoxigenic fungus Aspergi_llus flavus to the level 280 mcg aflatoxin B1 equivalent/kg. Ensiling of forages is a common practice in temperate areas, but is not used extensively in tropical countries. Preservation of excess forage during rainy seasons would provide a more even distribution of feed during the year. Experiments were conducted to investigate fermentation characteristics, and digestibility and palatability of Guinea-'A' (Panicum maximum, ecotype A) silage by sheep. Guinea-'A' grass was harvested after 2- and 3-week growth periods, and ensiled chopped or unchopped in 210 liter metal drums, double lined with polyethylene. The work discusses anaerobic fermentation of juice and press cakes of plant green mass in order to obtain stabilized feeds upon minimum energy expenses. We have studied the conversion of carbohydrates into acids, protein coagulation and proteolysis. There has been constructed a new technology "TPF-1". The work deals also with the stability and biological effectivity of the fermented juice and coagulate. Reliable, inexpensive and rapid methods of determing in vitro dry matter digestibility (DMD) are necessary to evalute animal production potential of a given herbage. The objective of this research was to determine the relationship between DMD measured by a procedure using a potent cellulase solution (from Trichoderma reese1) and by a procedure using rumen liquor. Herbage from 22 species was assayed. Correlations between in vitro DMD determined by the cellulase procedure and rumen liquor procedure ranged from 0.24 to 0.99. An experiment determined the effect of infestation by bent grass on the productivity of pastures grazed by sheep in Victoria. The 5-year study of liveweights and wool production from sheep grazing bent grass and treated pasture demonstrated that the presence of bent grass can reduce the productivity from sheep grazing such pastures by up to 63%. Thorough, shallow cultivation (5-7 cm deep) during summer, followed by resowing of more desirable species, produced the greatest increase in animal production. Herbage and animal production of a short grass pasture established by unplowed sowing were investigated for seven years under continuous grazing. Herbage production and consumption on the pasture were estimated by using cages. Moreover, light intensity was measured with the new portable solarimeter devised by the authors for pastures. Standing crop was kept at 200-300 g dry matter (DM)/m2 throughout the grazing season, 32-78% of which was below 3 cm from the ground surface. The mountain grassland of the Greater Xing'an Mountains are divided into two sorts, nine groups, fifteen types. It is suggested that mountain grasslands are sub-climax for its character of the climate and artificial succession. This classification and the rules of community dynamics will help us to know the grasslands of this area systematically. It will provide a scientific basis for the rational exploiting and utilizing the resources of the forest and grasslands, and determining the way of the agricultural production on it.

In a southern Chilean humid ecosystem, the productive potentiality of Chusquea coleu and Ch. quila was studied for animal utilization. During 2 years was seasonally recorded crude protein, neutro detergent fibre and dry matter in vitro digestibility. Also was analized the production of leaves and sprigs, and culms (utilizable and not by livestock, respectively). It was recorded a remarkable seasonality in dry matter production as well as in quality charateristics in both species.
