Cleavage and polyadenylation specific factor 6 (Cpsf6) translocation is frequently reported in APLL, which is Acute Promyelocytic Leukemia (APL)-like acute myeloid leukemia (AML) and other types of blood diseases associated with abnormal hematopoiesis. CPSF6 is a subunit of the Cleavage Factor I mammalian (CFIm) complex, which regulates post-transcriptional alternative polyadenylation (APA) of pre-mRNAs. Previous research indicated that Cpsf6 plays an essential role in alternative polyadenylation, development, and tumorigenesis. However, the role of Cpsf6 in hematopoiesis is unknown. Sand tigers Carcharias taurus are mackerel sharks with a disjunct global distribution in tropical and temperate seas. In the northwest Atlantic, sand tigers migrate along the coast of the eastern United States from the Gulf of Maine to Florida and occupy structurally complex benthic habitats. The long-term, underwater camera installation, SharkCam, at Frying Pan Tower, off Cape Fear, North Carolina, USA, sits atop an expanse of hard bottom reef frequented by sand tigers. The citizen-science platform Spot A Shark USA (SAS USA) uses images of sand tigers to identify and track individuals. This study utilized image encounters from SAS USA …

During the Last Glacial Maximum (LGM), the Great Plains in Nebraska consisted of wooded parklands and grasslands, whereas spruce (Picea) forests, parklands, and steppe grasslands dominated east of the Missouri River. I analyzed bulk carbon (δ13C), oxygen (δ18O), and strontium (87Sr/86Sr) isotopes from tooth enamel in isolated mammoth (Mammuthus) molars to compare diet and movement patterns in two late Pleistocene assemblages: one in Nebraska (n = 24), and one in western Iowa and Missouri (n = 17). This dissertation evaluates the performance of three on-farm swine carcass disposal methods: whole carcass composting (WCC), ground carcass composting (GCC), and shallow burial with carbon (SBC), focusing on biosecurity factors and groundwater contamination risks. Foreign animal diseases (FADs), such as African swine fever and classical swine fever, pose severe economic and animal well-being risks if introduced to the United States. With confirmation of an FAD, swine movement will be halted, creating an urgent need for practical, biosecure, and environmentally responsible on-farm disposal strategies for swine carcasses. Cardiovascular diseases are one of the leading causes of mortality worldwide in which hypertension is a risk factor. Present pharmacological interventions require long-term adherence to the therapy and are often associated with undesirable side effects. Phosphatidylcholine (PtdCho) is a prominent structural building block of cell and organelle membranes in animal, plant, and fungal cells and some bacteria. Due to its abundant nature in the plasma membrane and intracellular organelle membranes in eukaryotes, the biosynthesis of PtdCho must remain regulated to preserve the correct membrane lipid composition. Dysregulations in PtdCho metabolism can lead to mitochondrial dysfunction, cell death, and the development of diseases including cancer, atherosclerosis, and hepatic diseases. In this thesis, we report the results of four aptamers obtained from an aptamer selection process called the systematic evolution of ligands by exponential enrichment (SELEX) with affinity to Neuropeptide Y (NPY). NPY is a highly abundant peptide transmitter with widespread distribution throughout the body which affects various physiological functions. These functions include energy homeostasis, appetite regulation, stress response, and anxiety regulation. The aryl hydrocarbon receptor (AHR) is a ligand activated transcription factor that is essential in regulating development, immune homeostasis, and drug and xenobiotic metabolism. Activation of the AHR by specific ligands results in its translocation to the nucleus where it induces a myriad of gene products, including cytochrome P450 (CYP) drug metabolizing enzymes (e.g., CYP1A1) and other components of the chemical defensome, as well as inflammatory control. Interestingly, the AHR can bind a wide variety of endogenous and xenobiotic ligands which are able to induce unique downstream effects dependent on cell type, inflammatory context, and ligand stability. Glycine functions as an inhibitory neurotransmitter in the central nervous system (CNS),and it is mainly involved in autonomic functions. The levels of extracellular glycine are regulated by two membrane transporters, glycine transporter 1 and glycine transporter 2 (GlyT1 and GlyT2, respectively). These transporters differ in function and localization within the CNS. For many years, studies suggested that only GlyT2 served as a neuronal marker; however, GlyT1 has been localized in neurons in recent studies. Artificial reefs are deployed to enhance marine habitats and fisheries, yet their role in supporting secondary production and their vulnerability to climate stressors remain understudied. This dissertation investigates the trophic ecology and climate sensitivity of benthic invertebrates at artificial reefs in the northern Gulf of Mexico. Chapter 1 assessed reef food webs using stable isotopes from fishes, invertebrates, and primary producers across six reefs and two seasons. Mixing models showed that most fishes rely on benthic reef carbon, while suspension feeders consume plankton and sediment microphytobenthos. Isotopic niche analysis revealed functional diversity and seasonal shifts in δ¹³C and δ³⁴S, though …

The term soil health has gained popularity in agriculture due to prolonged droughts, rising food costs, and the growing public concern for sustainable and organic practices. Soil health practices can offer solutions to all of these pressures. This thesis provides Utah's farmers and ranchers with quantifiable data on implementing soil health practices in Utah's unique climate. Vibrio cholerae’s AB5 enterotoxin, cholera toxin (CT), has a homopentameric, non-toxic B subunit (CTB). CTB is a well-known vaccine adjuvant used for its ability to create a strong immune response, however more recent studies have shown its ability to also behave in an anti-inflammatory manner, performing as a protective agent in a variety of different inflammatory diseases. Why do people who experience the same environmental and social conditions vary so much in their resource transfer behavior? This dissertation addresses that question through examination of behavioral variation in subsistence horticulturalists in the dry tropical forest bufer conservation area of Gorongosa National Park in Mozambique. In this region of moderate and acute food shortages at both household and regional levels, individuals face similar challenges of addressing social and resource uncertainty yet respond with marked behavioral variation that cannot be explained solely by diferences in wealth, kinship, or reciprocity patterns.

Mule deer populations have been declining over the past couple of decades in many parts of the western United States. Often times, predators are blamed for the decreases in deer populations. Livestock depredation is one of the oldest and most widespread forms of human-wildlife conflict. The coyote (Canis latrans), a midsized canid, represents a significant proportion of domestic sheep depredations in the United States, costing livestock producers an estimated 24.6 million dollars annually. Management strategies for coyote-sheep depredation vary widely in their effectiveness and in their level of public support. Non-invasive fecal sampling is essential for wildlife gut microbiome (GMB) research, yet field conditions often limit preservation options. I compared gut bacterial communities in paired fecal samples preserved in stabilization tubes and air-dried in paper bags collected from anesthetized African lions (Panthera leo) and spotted hyenas (Crocuta crocuta) in Etosha National Park, Namibia. Additional opportunistic samples from carnivore kill sites were also analyzed. No significant differences in alpha or beta diversity were detected between preserved and dried samples, although bacterial beta diversity differed between preserved and opportunistic samples (PERMANOVA). Scleroderma/Systemic Sclerosis (SSc) is a multifaceted autoimmune disorder affecting approximately 300,000 people in the United States. Main features include vasculitis, immune dysfunction (innate and adaptive), autoantibody production, perivascular leukocyte infiltration, and extensive tissue fibrosis. The University of California, Davis (UCD) SSc chicken line was shown to exhibit all clinical and biological manifestations of human SSc. One early and distinctive clinical sign in both humans and UCD-SSc chickens is Raynaud’s phenomenon (RP), characterized by decreased blood flow in extremities and skin lesions. In the fall of 2024, the Food Safety and Inspection Service (FSIS) updated its guidelines for substantiating animal-raising and environment-related claims used on meat and poultry products. The updates aim to address numerous comments and petitions from industry stakeholders by improving guidance and strengthening the documentation recommended for substantiation. While the updates included both minor and substantial revisions, the most notable addition was the strong emphasis on the inclusion of a third-party certification in the substantiating documents. The COVID-19 pandemic exposed vulnerabilities in the U.S. food supply chain, particularly in the beef sector, where processing bottlenecks and labor shortages disrupted meat production and the market. In response, several policies were implemented to strengthen food system resilience, and significant funding was allocated to expand independent meat processing capacity nationwide. However, the long-term viability of expanding regional processing capacity remains uncertain, notably in Arkansas, a net exporter of live cattle. This study evaluates whether consumer demand supports the expansion of local beef processing in Arkansas. This thesis investigates the factors associated with loan defaults in the USDA's Direct Farm Ownership (FO) loan program, emphasizing factors that affect how long it takes to default. This study uses a Cox Proportional Hazards model on a dataset of more than 47,000 loan records originated during 2011-2020 to identify loan characteristics, financial factors, and geographic settings related to the likelihood and timing of delinquency, which is defined as being 90 days past due. Craniofacial malformations lie at the heart of Fetal Alcohol Spectrum Disorders (FASD). While there is growing evidence for a genetic component to FASD, little is known of the cellular mechanisms underlying these ethanol-sensitive loci in facial development. Bone Morphogenetic Protein (Bmp) signaling pathway dependent pouch formation is a key mechanism in facial development. We have previously shown that multiple Bmp mutants are sensitized to ethanol-induced facial defects. However, ethanol does not directly impact Bmp signaling. Salivary glands and the saliva they secrete are important for digestion, chewing, and maintaining oral health. Tumors are dynamic ecosystems that evolve under selective pressures, shaping their ability to either elicit or evade immune responses. In pancreatic ductal adenocarcinoma (PDAC), where immunotherapy has yet to become an effective treatment option, we investigated the role of intratumoral heterogeneity in influencing immune interactions. Using orthotopic clonal replica tumors, we tracked clonal dynamics in response to anti-PD1 therapy and found that while treatment had limited impact on overall tumor volume, it induced profound shifts in clonal composition. Circadian rhythms affect almost every biological process from hormone secretion, gene expression, immune regulation, and metabolism. Studies show that disruption to these internal rhythms through irregular sleep, artificial sleep, and irregular sleep can significantly increase risk for chronic disease such as diabetes, cardiovascular disorders, neurodegeneration, and cancer. Saliva presents a viable alternative for capturing biomarkers found through blood or cerebrospinal fluid, offering substantial utility for diagnostic and chronobiological profiling. Fluoride has long been praised for its role in preventing dental caries, primarily through its effects on enamel remineralization and antibacterial effects. However, growing evidence from molecular and cellular studies suggests that excessive fluoride exposure, especially during tooth development, can lead to dental fluorosis and systemic toxicity. This paper examines the biological mechanisms underlying fluoride-induced dental fluorosis, with a focus on stress responses in ameloblasts, oxidative damage, impaired autophagy, and disrupted protein processing. Cocoa farmers in Ghana face numerous challenges that undermine their incomes and living standards, including high labor costs, fluctuating cocoa prices, high cost and limited access to inputs, lack of credit, illegal mining, weather risks, small farm sizes, outdated farming and drying technologies, and inadequate extension services. These issues reduce productivity, lower incomes, and perpetuate poverty. This study examined farmers’ perceptions about the importance of these challenges and assessed possible differences across regions and farm households’ socioeconomic status.
