An experiment was carried out to investigate the effects of herbage mass and height on cattle ingestive behavior and herbage intake, when grazing Asiatic bluestem (Bothriocloa spp.) swards. Three levels of herbage mass were maintained under continuous grazing, corresponding to 1.8, 2.0 and 4.4 t dry matter/ha. Estimates of diet in vitro organic matter digestibility, rate of biting, grazing time and intake per bite were used to calculate digestible organic matter intake (DOMI). Differences in herbage mass led to significant differences in grazing time, intake per bite and DOMI but not in diet digestibility or rate of biting. The …

The relationship between pasture sward structure and animal intake of digestible dry matter is not well understood. To aid in developing an understanding of the plant-animal interface, flaccidgrass (Pennisetum flaccidum Griseb.) and switchgrass (Panicum virgatum L.) swards were harvested and stratified by cutting them into 5-cm layers. Strate began 5-cm above the soil and continued to the extended height of the sward. The strate were separated into leaf blade, stem, and dead material. The aim of this study is to improve understanding of sward responses to grazing manipulation so that objective guidelines for management may be provided. Rates of growth and senescence per tiller, tiller population densities and the photosynthetic efficiency of the sward canopy were measured at intervals on Lolium perenne dominated swards under contrasting management regimes involving steady-state and controlled fluctuations in sward conditions. Grazing and behavioral trials were run over a 4-yr period to investigate use of warm season (WS) range grasses in management systems for beef cattle on marginal hill land. Grasses tested were switchgrass (Panicum virgatum L.), big bluestem (Andropogon gerardii Vitman), Caucasian bluestem [Bothriochloa caucasica (Trin.) C.E. Hubb.], indiangrass [Sorghastrum nutans (L.) Nash] and Plains bluestem [B. ischaemum (L.) Keng var. ischaemum]. Two experiments were carried out to examine the effects of mixed stocking with cattle and sheep on aspects of herbage production and utilization from sown swards. On plots grazed down over three days both sheep and cattle tended to concentrate their grazing activity progressively on areas of sward previously grazed by the opposite species. Cattle were shown to be much less tolerant of cattle dung than of sheep dung, but sheep were relatively tolerant of both sheep and cattle dung. Animal productions were compared on two pastures, i.e. PW pasture (perennial ryegrass-white clover dominant pasture) and PO pasture (perennial ryegrass-orchardgrass dominant pasture), through precursor-follower system using feeder steers and beef cows. The experiment was carried out in 1983 and 1984. After close of grazing feeder steers were fattened for 6 or 7 months, then the carcass characteristics were examined. The response to N-fertilizer of animal output and of herbage production was compared. There appeared to be no difference in the responses to applied nitrogen between intensive continuous grazing and weekly cutting in the range of 300 to 500 kg N/ha/yr. It is hypothesized that the response to applied nitrogen in a continuously grazed sward is mainly determined by the intensity of grazing and that the benefit from nitrogen circulation through excreta has little meaning for the pasture as a whole.

Grazing intensity models have traditionally related animal production per head and per ha to stocking rate. The value of such models is limited by the omission of pasture information. In this paper the relationships between average daily gain, grazing days and standing herbage are used to develop models relating animal production per ha to standing herbage. These models can also be extended by relating changes in their parameters to additional quantifiable variables, or by using them in economic models to express profit per ha in terms of standing herbage.

Integration of this physiological research with studies of forage consumption on the same pastures is proving a valuable inter- disciplinary approach to defining management options for maximising pasture and/or animal productivity. The field of single-cell biology is growing rapidly, generating large amounts of data from a variety of species, disease conditions, tissues, and organs. Coordinated efforts such as CZI CELLxGENE, HuBMAP, Broad Institute Single Cell Portal, and DISCO allow researchers to access large volumes of curated datasets, including more than just scRNA-seq data. These resources have created an opportunity to build and expand the computational biology ecosystem to develop tools necessary for data reuse and for extracting novel biological insights. We highlight achievements made so far, areas where further development is needed, and specific challenges that need to be overcome.

The study assessed milk production performance of local cows, milk handling practices, physicochemical properties, and milk production constraints in Kucha District, Southern Ethiopia. A purposive and random sampling technique was employed to select 138 respondents for the interview. For physicochemical property assessment, milk samples were collected from 90 interviewed households. Majorities (76.7%) of the respondents were males, and the average family size was 6.82±0.25. Mean daily milk yield (DMY) of 1.80±0.07, 1.94±0.06, and 1.52±0.09 liters was recorded for local cows, respectively, in early, mid, and late lactation stages. Elaeagnus pungens Thunb is a significant source of flavonoids with biological function. This study aims to develop orthogonal tests and alkali alcohol extraction methods for extracting flavonoids from leaves of E. pungens. The levels of factors in the orthogonal test were determined by pre-experiment, and the physiological efficacy was verified by the in vivo test. The results indicated that the optimal extraction yield was 4.07% when the solid-to-liquid ratio was 1:40, the concentration of ethanol was 60%, the extraction temperature was 80℃, and the extraction time was five h. Lysosomes are essential organelles for cellular homeostasis and signaling, with dysfunction linked to neurological disorders, lysosomal storage diseases, and cancer. While proteomics has advanced our understanding of lysosomal composition, the structural characterization of lysosomal membrane proteins in their native environment remains a significant challenge. Here, we developed a cryo electron tomography workflow to visualize lysosomal membrane proteins within intact, native lysosomal membranes. We isolated endolysosomes by independently targeting two lysosomal membrane proteins, transient receptor potential mucolipin 1 and transmembrane protein 192, enriching organelles that exhibited the expected morphology and proteomic composition of the endolysosomal system. Schisandra chinensis polysaccharides (SCP) are isolated from Schisandra chinensis with a mean molecular weight of 32.401 kDa. Previous studies have demonstrated that Schisandra chinensis extract effectively prevented colitis and modulated gut microbiota in dextran sulfate sodium (DSS)-induced mice. In this study, 3% DSS was used to induce acute ulcerative colitis in mice, and then the SCP with different doses (50 mg/kg, 100 mg/kg, 200 mg/kg) and 5- aminosalicylic acid (5-ASA) were used to treat mice by oral gavage for one week. SNP paternity testing panels offer high-resolution capabilities for determining livestock paternity, thereby enhancing the reliability of genetic verification and ensuring pedigree record accuracy. In this study, we aimed to develop a SNP-based paternity test panel utilizing a 50K SNP chip for paternity testing and pedigree verification in the Eşme sheep breed. Additionally, the study sought to validate the paternity test panel. Based on data from 26 rams and 524 lambs of the Eşme sheep breed, the results indicated that the 50K SNP chip used demonstrated a very high genotyping rate, with the majority of its 53,738 SNPs exhibiting significant polymorphism. …

This study evaluated the effects of varying water pH levels, adjusted with organic (formic acid) and inorganic (sodium hydrogen sulfate) acidifiers, on the growth performance, blood biochemistry, carcass yield, and meat quality of broiler chickens. The presence of substandard and counterfeit veterinary medicinal products represents a significant concern from a One Health perspective, given the potential consequences for human and animal health, as well as the environment. In this study, an isocratic flow liquid chromatography method was developed for the simultaneous determination of thymol, coumaphos, amitraz, tau-fluvalinate, and flumethrin in bee health products used for varroosis control. Injury is common in wild animals, yet some species possess an adaptive advantage: regeneration. Enallagma vesperum, a species of damselfly, exhibits this regenerative ability, though its persistence and effectiveness across developmental stages remain poorly understood. While previous studies indicate that regeneration ceases following adult emergence, it is unclear whether this ability also diminishes prior to the final larval molt. This study investigated the effectiveness of limb regeneration in E. vesperum across early and late larval instars. [Objective ] The basic characteristics of L-malic acid and the application of L-malic acid in orange juice were studied.[Method ] The basic characteristic differences of L-malic acid,citric acid,and DL-malic acid were compared in terms of thermal stability,pH stability,and taste quality changes of electronic tongue.Subsequently,the three sour agent were added into orange juice.The effects of L-malic acid on the stability and volatile flavor of orange juice were analyzed based on the differences in centrifugal sedimentation rate,pH,electrical conductivity,particle size,Zeta potential,and volatile components.[Result ] L-Malic acid had good stability in the ranges of 40~121 ℃ and pH 2.0~7.0.L-Malic acid differed markedly from citric …

[Objective ] This study aims to employ synthetic biology and metabolic engineering techniques to engineer Pichia pastoris (P.pastoris ) for the efficient heterologous synthesis of the UV resistant compound gadusol.[Method ] On the basis of a bioinformatic analysis and codon optimization of the genes involved in the gadusol biosynthetic pathway in Danio rerio,the gadusol fermentation pathway was constructed in P.pastoris via strategies such as promoter engineering,metabolic engineering,and fermentation optimization.[Result ] A high-purity gadusol standard was prepared through optimized extraction and chromatographic separation.The gadusol yield in the recombinant strain P.pastoris HME reached 105 mg/L.By screening of gene expression promoters and optimization …

[Objective ] This study aims to improve the utilization of maize resources and address the shortage of feed resources.[Method ] The intestinal colonization capacity,tolerance,and antibacterial properties of three Lactobacillus strains,Lactobacillus paracasei ZB- 1 (ZB-1),L.paracasei DY- 2 (DY-2),and L.rhamnosus DY- 4 (DY-4) were evaluated.The most suitable strain for fermenting cornhusks was identified,and its fermentation process was optimized.[Result ] ZB-1 exhibited higher intestinal colonization ability and antibacterial properties than DY- 2 and DY- 4.The antibacterial mechanism of ZB- 1 was likely driven by the action of organic acids in an acidic environment and certain protein-based antimicrobial substances.The fermentation conditions for ZB- 1 …

Plant transformation efficiency is highly dependent on species, individual genotypes, and tissue types. In maize, immature embryos are regularly used for transformation. The process relies heavily on callus development, as it is intricately associated with somatic embryogenesis and subsequent plant regeneration, both of which directly affect transformation efficiency. Immature embryos of the segregation progeny derived from the two inbred parents, a transformation-amenable line A188 and a recalcitrant line B73, can be cultured to form two primary callus types: Type I and Type II. Northern hardwood forests of the Lake Superior region face a series of novel disturbance pressures including canopy dieback. Previous studies have linked regional sugar-maple (Acer saccharum) canopy dieback to introduced earthworms, which may have coinciding impacts on the ground-layer plant community. Dieback–earthworm interactions may lead to important longer-term changes in forest structure and function, but these relationships but have not been characterized. We sampled ground-layer plant communities in five national forest units in Michigan, Wisconsin, and Minnesota in 2010, and again just over a decade later in 2021. Patients with non–small cell lung cancer (NSCLC) with loss of the tumor suppressor gene STK11 are resistant to immune checkpoint therapies like anti–PD-1. In this study, we conducted an in vivo CRISPR screen that identified histone deacetylase 1 as a target to reverse anti–PD-1 resistance driven by loss of STK11 and developed TNG260, a potent small-molecule inhibitor of the CoREST complex with selectivity exceeding previously generated inhibitors in this class in preclinical studies. Treatment with TNG260 led to increased expression of immunomodulatory genes in STK11-deficient cancer cells.
