In December 2016, a panel of experts in microbiology, nutrition and clinical research was convened by the International Scientific Association for Probiotics and Prebiotics to review the definition and scope of prebiotics. Consistent with the original embodiment of prebiotics, but aware of the latest scientific and clinical developments, the panel updated the definition of a prebiotic: a substrate that is selectively utilized by host microorganisms conferring a health benefit. This definition expands the concept of prebiotics to possibly include non-carbohydrate substances, applications to body sites other than the gastrointestinal tract, and diverse categories other than food. Small bowel transplantation (SBT) can be a life-saving medical procedure. However, these recipients experience high risk of bloodstream infections caused by Candida. This research aims to characterize the SBT recipient gut microbiota over time following transplantation and investigate the epidemiology of candidemia in seven pediatric patients. Candida species from the recipients’ ileum and bloodstream were identified by internal transcribed spacer sequence and distinguished to strain by multilocus sequence typing and randomly amplified polymorphic DNA. Antifungal susceptibility of bloodstream isolates was determined against nine antifungals. Twenty-two ileostomy samples harbored at least one Candida species. Aims: The objective of this study was to determine if the faecal microbiome has an association with Escherichia coli O157:H7 prevalence and enumeration.

Methods and Results: Pyrosequencing analysis of faecal microbiome was performed from feedlot cattle fed one of three diets: (i) 94 heifers fed low concentrate (LC) diet, (ii) 142 steers fed moderate concentrate (MC) diet, and (iii) 132 steers fed high concentrate (HC) diet. A total of 322 585 OTUs were calculated from 2,411,122 high-quality sequences obtained from 368 faecal samples. Scope: The Soybean (Glycine max) leghemoglobin c2 (LegHb) gene was introduced into Pichia pastoris yeast for sustainable production of a heme-carrying protein, for organoleptic use in plant-based meat. The potential allergenicity and toxicity of LegHb and 17 Pichia host-proteins each representing ≥ 1% of total protein in production batches are evaluated by literature review, bioinformatics sequence comparisons to known allergens or toxins, and in vitro pepsin digestion.

Methods and results: Literature searches found no evidence of allergenicity or toxicity for these proteins. There are no significant sequence matches of LegHb to known allergens or toxins. Eleven Pichia proteins have modest …

Mucispirillum schaedleri is an abundant inhabitant of the intestinal mucus layer of rodents and other animals and has been suggested to be a pathobiont, a commensal that plays a role in disease. In order to gain insights into its lifestyle, we analyzed the genome and transcriptome of M. schaedleri ASF 457 and performed physiological experiments to test traits predicted by its genome. Although described as a mucus inhabitant, M. schaedleri has limited capacity for degrading host-derived mucosal glycans and other complex polysaccharides. Background: Eliciting doses (EDs) of allergenic foods can be defined by the distribution of threshold doses for subjects within a specific population. The ED05 is the dose that elicits a reaction in 5% of allergic subjects. The predicted ED05 for peanut is 1.5 mg of peanut protein (6 mg of whole peanut).

Objective: We sought to validate the predicted peanut ED05 (1.5 mg) with a novel single-dose challenge.

Changes in the gastrointestinal microbial community are frequently associated with chronic diseases such as Inflammatory Bowel Diseases. However, understanding the relationship of any individual taxon within the community to host physiology is made complex due to the diversity and individuality of the gut microbiota. Defined microbial communities such as the Altered Schaedler Flora (ASF) help alleviate the challenges of a diverse microbiota by allowing one to interrogate the relationship between individual bacterial species and host responses. Controlled studies on the effect of exercise on intestinal uptake of protein are scarce and underlying mechanisms largely unclear. We studied the uptake of the major allergen Ara h 6 following peanut consumption in an exercise model and compared this with changes in markers of intestinal permeability and integrity. Ten overnight-fasted healthy non-allergic men (n = 4) and women (n = 6) (23 +/- 4 years) ingested 100 g of peanuts together with a lactulose/rhamnose (L/R) solution, followed by rest or by 60 min cycling at 70% of their maximal workload. Wheat (Triticum aestivum L.) is the most widely cultivated crop in the world and contributes about 20% of the total dietary calories and proteins globally. Unique properties of doughs formed from wheat flour make it feasible to produce a range of food, including bread. Loaf volume is the most perceptible indicator for breadmaking quality. Effects Of High Intensity Ultrasound Frequency And High-Speed Agitation On Fat Crystallization, Roberta Silva, Juhee Lee, V. Gibon, Silvana Martini

The objective of this research was to examine the effect of ultrasound frequency and high-speed agitation on lipid crystallization. Interesterified soybean oil was crystallized at 44 °C without and with the application of high intensity ultrasound (HIU—20 and 40 kHz) or with high-speed agitation (6000 and 24,000 rpm). Two tip amplitudes (24 and 108 µm) and three pulse durations were evaluated (5, 10, and 15 s) for the acoustic frequencies tested. Sonication at 20 kHz of frequency significantly reduced crystal size, increased (p < 0.05) elasticity (435.9 ± 173.3–80,218 ± 15,384 Pa) and SFC (0.2 ± 0.0–4.5 ± 0.4%). No significant difference was observed in the crystallization behavior of these samples when sonicated at different amplitudes for 5 and 10 s. The crystallization behavior was significantly delayed (p < 0.05) in samples sonicated using 108 µm amplitude for 15 s. Larger crystals were formed in samples sonicated at 40 kHz compared to those obtained with 20 kHz and lower SFC (3.7 ± 0.0%) and elasticity (3943 ± 1459 Pa) values were obtained. High-speed agitation at 24,000 rpm increased SFC (5.5 ± 0.1%) and crystallized area and decreased the elasticity (42,602 ± 11,775 Pa) compared to the samples sonicated at 20 kHz.

Physical blends (PB) of high oleic sunflower oil and tristearin with 20 and 30% stearic acid and their interesterified (IE) products where 20 and 30% of the fatty acids are stearic acid at the sn-2 position crystallized without and with application of high intensity ultrasound (HIU). IE samples were crystallized at supercooling temperatures (ΔT) of 12, 9, 6, and 3 °C while PB were crystallized at ΔT = 12 °C. HIU induced crystallization in PB samples, but not in the IE ones. Mixing is a ubiquitous unit operation within the food industry and employed not only to combine multiple ingredients, but also to control reactions and modify the structure of foods. Failure of the mixing process may result in poor quality, organoleptic or functional attributes of foods along with product safety concerns. The industry is increasingly adopting automation and advanced control strategies such as Process Analytical Technology. Sensors can provide both insights into the complex mechanisms of mixing along with effective control. Here we discuss some recent advances in the control of food mixing operations.

The aim of this work was to investigate the efficacy of dielectric barrier discharge atmospheric cold plasma (DBD ACP) against bacteria associated with grains quality and safety. ACP inactivation efficacy was tested against biofilms formed by different strains of E. coli, Bacillus and Lactobacillus in grain model media and against B. atrophaeus endospores either in grain media or attached on abiotic surfaces. Effects were dependent on bacterial strain, media composition and mode of ACP exposure. ACP treatment for 5min reduced E. coli spp., B. subtilis and Lactobacillus spp. biofilms by >3 log10, whereas insignificant reductions were achieved for B. atrophaeus. …

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาผลของวัตถุเจือปนประเภทคาร์โบไฮเดรตต่อสมบัติของเพสต์และเจลสตาร์ชข้าวเจ้า วัตถุเจือปนที่ใช้ในงานวิจัยนี้แบ่งเป็น 2 กลุ่ม กลุ่มแรกได้แก่ น้ำตาลและน้ำตาลแอลกอฮอล์ (กลูโคส ฟรุกโทส และซอร์บิทอล) และกลุ่มที่สองได้แก่ ออลิโกแซ็กคาไรด์และพอลิแซ็กคาไรด์ที่มีสมบัติเป็นพรีไบโอติก (พอลิเด็กซ์โทรส ฟรุกโทออลิโกแซ็กคาไรด์ และอินูลิน) โดยเติมวัตถุเจือปนในปริมาณ 5, 10, 20, และ 30% โดยน้ำหนักของสตาร์ช จากการติดตามพฤติกรรมการเกิดเพสต์ของสตาร์ชข้าวพบว่าเมื่อความเข้มข้นของน้ำตาลหรือน้ำตาลแอลกอฮอล์เพิ่มขึ้น อุณหภูมิการเกิดเพสต์มีแนวโน้มเพิ่มสูงขึ้น ในขณะที่ความหนืดสูงสุดมีแนวโน้มลดลง นอกจากนี้ความหนืดเบรกดาวน์และความหนืดเซ็ตแบ็กก็มีแนวโน้มลดลงด้วย ในด้านสมบัติทางความร้อนพบว่าอุณหภูมิและเอนทาลปีของการเกิดเจลาทิไนเซชันมีแนวโน้มเพิ่มสูงขึ้นตามความเข้มข้นของน้ำตาลหรือน้ำตาลแอลกอฮอล์ที่เพิ่มขึ้น โดยกลูโคสและซอร์บิทอลทำให้อุณหภูมิและเอนทาลปีของการเกิดเจลาทิไนเซชันของสตาร์ชข้าวเพิ่มขึ้นได้สูงสุด ในแง่การเกิดรีโทรเกรเดชัน พบว่าเอนทาลปีของการหลอมผลึกแอมิโลเพกทินของตัวอย่างที่เติมกลูโคสและซอร์บิทอลมีค่าต่ำกว่าตัวอย่างที่เติมฟรุกโทสที่ความเข้มข้นเท่ากัน ซึ่งชี้ให้เห็นว่ากลูโคสและซอร์บิทอลมีประสิทธิภาพในการชะลอการเกิดรีโทรเกรเดชันได้ดีกว่าฟรุกโทส นอกจากนี้จากการติดตามการเปลี่ยนแปลงของปริมาณของส่วนเสมือนของแข็งพบว่ามีแนวโน้มเพิ่มสูงขึ้นตามระยะเวลาการเก็บรักษาที่เพิ่มขึ้นในทุกตัวอย่าง โดยตัวอย่างควบคุมมีอัตราการเพิ่มขึ้นของส่วนเสมือนของแข็งสูงกว่าเจลสตาร์ชที่เติมน้ำตาลและน้ำตาลแอลกอฮอล์โดยเฉพาะอย่างยิ่งในช่วง 14 วันแรกของการเก็บรักษา ในแง่ความสามารถในการเคลื่อนที่ของน้ำในเจลสตาร์ชพบว่ามีแนวโน้มลดลงเมื่อระยะเวลาการเก็บรักษาเพิ่มขึ้น ซึ่งอาจเป็นผลมาจากน้ำถูกจับไว้โดยองค์ประกอบอื่นภายในระบบ เช่น การเกิดไฮเดรชันกับผลึกสตาร์ช โดยตัวอย่างควบคุมมีการลดลงของ 1H NMR T1 และ T2 สูงที่สุด สำหรับผลของการเติมพรีไบโอติกพบว่าการเติมพรีไบโอติกมีผลสำคัญต่อพฤติกรรมการเกิดเพสต์ของสตาร์ชข้าว โดยเมื่อความเข้มข้นของพรีไบโอติกเพิ่มสูงขึ้น อุณหภูมิการเกิดเพสต์มีแนวโน้มเพิ่มสูงขึ้น ในขณะที่ความหนืดสูงสุดมีแนวโน้มลดลง เมื่อเปรียบเทียบที่ความเข้มข้นเท่ากัน ความหนืดสูงสุดของตัวอย่างที่เติมอินูลินมีค่าสูงกว่าตัวอย่างที่เติม พอลิเด็กซ์โทรสหรือฟรุกโทออลิโกแซ็กคาไรด์ เมื่อความเข้มข้นของพรีไบโอติกเพิ่มขึ้นพบว่าความหนืดเบรกดาวน์และความหนืดเซ็ตแบ็กมีแนวโน้มลดลง สำหรับสมบัติทางความร้อนพบว่าอุณหภูมิของการเกิดเจลาทิไนเซชันเพิ่มสูงขึ้นตามความเข้มข้นของพรีไบโอติกที่เพิ่มขึ้น ในขณะที่เอนทาลปีในการเกิดเจลาทิไนเซชันลดลงเล็กน้อย การเติมพรีไบโอติกสามารถชะลอการเกิดรีโทรเกรเดชันของสตาร์ชข้าว โดยตัวอย่างที่เติมอินูลินมีเอนทาลปีของการหลอมผลึกแอมิโลเพกทินต่ำกว่าตัวอย่างที่เติมพอลิเด็กซ์โทรสและฟรุกโทออลิโกแซ็กคาไรด์ที่ความเข้มข้นเท่ากัน ซึ่งสอดคล้องกับการเปลี่ยนแปลงของปริมาณส่วนเสมือนของแข็ง ที่พบว่าเจลสตาร์ชที่เติมพรีไบโอติกมีการเพิ่มขึ้นของส่วนเสมือนของแข็งในอัตราที่ต่ำกว่าตัวอย่างควบคุม ในด้านความสามารถในการเคลื่อนที่ของน้ำ การเติมพรีไบโอติกทำให้ 1H NMR T1 และ T2 ลดลงในอัตราที่ช้ากว่าเมื่อเทียบกับตัวอย่างควบคุม โดยสรุปพบว่าวัตถุเจือปนคาร์โบไฮเดรตที่ใช้ในงานวิจัยนี้ทำให้สตาร์ชเกิดเจลาทิไนเซชันได้ยากขึ้น ในขณะเดียวกันก็ทำให้สตาร์ชแกรนูลทนทานต่อความร้อนและแรงเฉือนมากขึ้น รวมทั้งช่วยชะลอการเกิดรีโทรเกรเดชัน

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาผลของชนิดและความเข้มข้นของตัวออกซิไดส์ รวมทั้งผลของอุณหภูมิและระยะเวลาการบ่มแผ่นฟิล์มด้วยความร้อนต่อสมบัติของฟิล์มไข่ขาว ในขั้นตอนแรกเป็นการศึกษาผลของชนิดและความเข้มข้นของตัวออกซิไดส์ ตัวออกซิไดส์ที่ใช้ในการศึกษามี 3 ชนิด ได้แก่ ไฮโดรเจนเพอร์ออกไซด์ โซเดียมเพอร์ไอโอเดต และโซเดียมไฮโปคลอไรต์ แปรความเข้มข้นของตัวออกซิไดส์เป็น 3 ระดับ ได้แก่ 0.02, 0.04 และ 0.06 โมลาร์ พบว่าความต้านทานแรงดึงขาดและการยืดตัวถึงจุดขาดของฟิล์มที่เติมตัวออกซิไดส์สูงกว่าตัวอย่างควบคุมอย่างมีนัยสำคัญ (p≤0.05) โดยทั่วไปพบว่าฟิล์มที่เติมโซเดียมเพอร์ไอโอเดตมีความต้านทานแรงดึงขาดและการยืดตัวถึงจุดขาดสูงกว่าฟิล์มที่เติมไฮโดรเจนเพอร์ออกไซด์และฟิล์มที่เติมโซเดียมไฮโปคลอไรต์ ฟิล์มที่เติมโซเดียมเพอร์ไอโอเดตเข้มข้น 0.06 โมลาร์มีความต้านทานแรงดึงขาด (2.73 เมกะพาสคาล) และการยืดตัวถึงจุดขาด (63.89%) สูงสุด โดยมีค่าเป็น 425% และ 326% ของตัวอย่างควบคุม ตามลำดับ การเชื่อมข้ามโปรตีนด้วยพันธะไดซัลไฟด์สามารถยืนยันได้โดยใช้เทคนิครามานสเปกโทรสโกปี การเติมตัวออกซิไดส์ยังมีผลต่อสมบัติเชิงแสง โดยทำให้ฟิล์มที่ได้มีความโปร่งใสลดลง นอกจากนี้พบว่าฟิล์มที่เติมโซเดียมเพอร์ไอโอเดตและฟิล์มที่เติมโซเดียมไฮโปคลอไรต์มีสีเหลืองและสีส้มเหลืองตามลำดับ ในขณะที่ฟิล์มที่เติมไฮโดรเจนเพอร์ออกไซด์มีสีใกล้เคียงกับตัวอย่างควบคุม การเติมตัวออกซิไดส์ทำให้ผิวฟิล์มมีสมบัติความไม่ชอบน้ำเพิ่มขึ้น ในขณะที่ความสามารถในการละลายน้ำและสภาพให้ซึมผ่านได้ของไอน้ำมีค่าลดลง ในขั้นตอนที่สองเป็นการศึกษาผลของการบ่มแผ่นฟิล์มด้วยความร้อน โดยฟิล์มที่คัดเลือกมาศึกษาได้แก่ฟิล์มที่เติมไฮโดรเจนเพอร์ออกไซด์เข้มข้น 0.06 โมลาร์ แปรอุณหภูมิการบ่มเป็น 4 ระดับ ได้แก่ 40, 60, 70 และ 80 องศาเซลเซียส และแปรระยะเวลาการบ่มเป็น 3 ระดับ ได้แก่ 24, 48 และ 72 ชั่วโมง พบว่าฟิล์มที่บ่มด้วยความร้อนมีความต้านทานแรงดึงขาดสูงกว่าและการยืดตัวถึงจุดขาดต่ำกว่าตัวอย่างควบคุมซึ่งไม่ได้บ่มอย่างมีนัยสำคัญ (p≤0.05) เมื่ออุณหภูมิและระยะเวลาการบ่มเพิ่มขึ้น ความต้านทานแรงดึงขาดมีค่าเพิ่มขึ้น (p≤0.05) แต่การยืดตัวถึงจุดขาดมีค่าไม่เปลี่ยนแปลง (p>0.05) ตัวอย่างฟิล์มที่บ่มที่อุณหภูมิ 80 องศาเซลเซียส เป็นระยะเวลา 72 ชั่วโมง มีความต้านทานแรงดึงขาดสูงสุด (9.55 เมกะพาสคาล) ซึ่งเป็น 683% ของตัวอย่างควบคุม การเชื่อมข้ามโปรตีนด้วยพันธะไดซัลไฟด์สามารถยืนยันได้โดยใช้เทคนิครามานสเปกโทรสโกปี ในด้านสมบัติเชิงแสง พบว่าการบ่มด้วยความร้อนมีผลต่อค่าสีค่อนข้างน้อย แม้ว่ามุมของสีตัวอย่างฟิล์มมีค่าแตกต่างกัน แต่ความเข้มสีที่มีค่าต่ำมากทำให้ฟิล์มทุกตัวอย่างปรากฏเป็นสีเทาเมื่อดูด้วยตาเปล่า การบ่มด้วยความร้อนทำให้ผิวฟิล์มมีสมบัติความไม่ชอบน้ำเพิ่มขึ้น และสามารถผลิตฟิล์มที่มีผิวไม่ชอบน้ำ (มีมุมสัมผัสระหว่างหยดน้ำกับผิวฟิล์มสูงกว่า 90 องศา) ได้โดยบ่มที่อุณหภูมิสูงเป็นระยะเวลานาน นอกจากนี้ยังพบว่าฟิล์มที่บ่มด้วยความร้อนมีความสามารถในการละลายน้ำและสภาพให้ซึมผ่านได้ของไอน้ำลดต่ำลง

Brochothrix thermosphacta is a predominant spoilage microorganism in meat and its control in processing environments is important to maintain meat product quality. Atmospheric cold plasma is of interest for control of pathogenic and spoilage microorganisms in foods. This study ascertained the potential of dielectric barrier discharge atmospheric cold plasma (DBD-ACP) for control of B. thermosphacta in response to key parameters such as treatment time, voltage level, interactions with media composition and post treatment storage conditions. Camellia sinensis teas, and tisanes derived from herbs or fruit, are rich in polyphenolic, antioxidant compounds. This study compared the total phenolic content (TPC), total flavonoid content (TFC), ferric reducing antioxidant power (FRAP), DPPH radical scavenging capacity, and caffeine content of teas (black, green, white, chamomile, and mixed berry/hibiscus) over a range of infusion times (0.5–10 mins) at 90°C. Two antimicrobial coatings, namely Sodium octanoate and Auranta FV (a commercial antimicrobial composed of bioflavonoids, citric, malic, lactic, and caprylic acids) were used. These two antimicrobials were surface coated onto the inner polyethylene layer of cold plasma treated polyamide films using beef gelatin as a carrier and coating polymer. This packaging material was then used to vacuum pack beef sub-primal cuts and stored at 4 °C. A control was prepared using the non-coated commercial laminate and the same vacuum packaged sub-primal beef cuts. During storage, microbial and quality assessments were carried out. Sodium octanoate treated packages significantly (p < 0.05) reduced microbial counts for all bacteria tested with an increase of 7 and 14 days, respectively compared to control samples. No significant effect on pH was observed with any treatment. The results suggested that these food grade antimicrobials have the potential to be used in antimicrobial active packaging applications for beef products.

This study evaluates the efficiency of multipoint near‐infrared spectroscopy (NIRS) to predict the fat and moisture content of minced beef samples both in at‐line and on‐line modes. Additionally, it aims at identifying the obstacles that can be encountered in the path of performing in‐line monitoring. Near‐infrared (NIR) reflectance spectra of minced beef samples were collected using an NIR spectrophotometer, employing a Fabry‐Perot interferometer. The effects of ultrasound irradiation were studied on the phenolic compounds, color and hydroxyl radical scavenging ability of black rice wine. The results showed that the total content of phenolic, flavonoids, anthocyanins and the scavenging capacity of hydroxyl radical for the black rice wine decreased after ultrasonic treatment, while the brightness (L*), redness (a*) and yellowness (b*) of wine color had some certain increasing. [Purpose] The different influencing factors are investigated on producing AGEs by Lysine in Maillard reaction, and the different between single amino acid (Lysine, or Argine) -reducing and mixed Lys- Arg -reducing sugar system are compared. [Method] Fluorescence spectrometry (λex / λem =370 / 440 nm) is used to detect the amount of AGEs, the factors of sort and concentration of sugar, metal ion, temperature, pH and genistein concentration in Maillard reaction and proportion of lysine and arginine in Lys/Arg-reducing sugar system was discussed. Effect Of Wheat Gluten Hydrolysate On The Recovery Of Intestinal Mucositis In Methotrexate Treated Rats, Yan Bin, Liu Liya, Wang Yanzhou, Zhou Xianrong, Zhou Sumei

To investigate the effect of wheat gluten hydrolysate(WGH) rich in glutamine prepared through the enzymatic hydrolysis by trysin on small intestinal mucosal by methotrexate induced mucositis model in rats. The results showed that the WGH can significantly increase intestinal mucosa weight, villus height, crypt depth, protein and DNA content, decrease diamine oxidase level in plasma and increase glutathione content of small intestine mucosa. The WGH can efficiently alleviate the intestinal mucosal injury in rats.

Effect Of Processing Conditions On The Functional Properties Of Antioxidant Peptides From Pseudosciaena Crocea, Zhang Ningning, Zheng Baodong, Zhang Chong, Li Zhiyu

The functional properties of antioxidant peptides from Pseudosciaena crocea under different processing conditions were studied, including the solubility, water absorption, retention properties, fat absorption, apparent viscosity, foaming and emulsifying properties. The results showed that the isoelectric point of the antioxidant peptides was pH 4.0, and at this point, the peptides possessed the lowest solubility and emulsifying activity, while the highest emulsion stability and foaming ability and stability. Moreover, it was also found that the antioxidant peptides showed higher water and fat absorptions, fluidity and lower retention properties.
