An Iterative & Analytical Hybrid Deep Learning Framework For Enhanced Brain Tumor Detection And Segmentation In Mri Video Scans, Swati V. Sakhare
Mansoura Engineering Journal
The critical need for accurate and robust brain tumor detection in MRI video scans is for early diagnosis and treatment planning operations. However, the existing methods suffer from several limitations, such as not being able to capture spatio-temporal features, handling class imbalances, and generalizing across datasets with diverse characteristics. To address these challenges, I propose a hybrid deep learning framework that integrates multiple advanced techniques to enhance the detection and segmentation of brain tumors. My approach starts by using the 3D CNN-LSTM (ResNet3D-LSTM) to extract spatio-temporal feature, which utilizes ResNet3D for spatial detail extraction and LSTM for temporal coherence across …

Powering The Machine, Draining The Planet: Whether U.S. Environmental Law Is Equipped To Regulate The Energy And Water Demands Of Ai Data Centers, Michael Marcu
Journal of Earth and Life Science
Artificial intelligence (AI) data centers have become one of the United States' fastest-growing and least-regulated sources of environmental stress. In 2024 alone, U.S. data centers consumed 183 terawatt-hours (TWh) of electricity more than the entire nation of Pakistan and consumed an estimated 17 billion gallons of water (IEA, 2025; Berkeley Lab, 2024). By 2030, electricity demand from these facilities is projected to reach 426 TWh, a 133% increase in six years (Pew Research Center, 2025). This paper examines whether the existing U.S. environmental regulatory framework put by the National Environmental Policy Act (NEPA), the Clean Water Act (CWA), and the …

The vagina is a fibromuscular tube-shaped organ that plays critical roles in menstruation, pregnancy, and female sexual health. Vaginal tissue constituents, including cells and extracellular matrix components, contribute to tissue structure, function, and prevention of injury and pathology. However, much microstructural function remains unknown, including how the fiber-cell and cell–cell interactions influence macromechanical properties. A deeper understanding of these interactions will provide critical information needed to reduce and prevent vaginal pathologies. Our objective for this work is to design a novel tissue-mimicking biomaterial for vaginal tissue engineering, and characterize its biological and mechanical performance in the vaginal microenvironment. We successfully …

Noise degrades both EEG and gait signals, and classical IIR filters (Butterworth, Chebyshev, elliptic) involve trade-offs between passband flatness, ripple, and roll-off. This study compared a novel exponential "Reza" filter with these designs for neural and locomotor data. We analyzed an open-source mobile brain-body imaging dataset with EEG and gait data from 49 healthy adults (EEG: 256-channel, 512 Hz; IMUs: six APDM Opals, 128 Hz). EEG channels were grand-averaged and band-pass filtered at 0.5-50 Hz, while IMU axes were averaged and band-pass filtered at 0.5-5 Hz. The outcomes were signal-to-noise ratio SNR (dB) and band-integrated Welch PSD (EEG:0.5-50 Hz; IMU:0.5-5 …

Engineering Plga Nanoparticle Size To Modulate Immune Responses After Spinal Cord Injury, Daniel J. Kolpek
University of Kentucky Doctoral Dissertations
Spinal cord injury is a severe and debilitating condition that often results in lifelong complications, including paralysis. Many of these chronic outcomes are driven by a sustained inflammatory response at the injury site, which limits tissue regeneration and functional recovery. As a result, therapeutic strategies that modulate this immune response have gained significant interest. Polymer based nanoparticle therapies have emerged as a promising approach due to their ability to interact with and influence immune cell behavior. Importantly, the physicochemical properties of nanoparticles can be precisely tuned to optimize these effects. Among these properties, size has been shown to influence key …

Computer Science and Engineering Dissertations
Hematoxylin and eosin (H&E) staining remains central to cancer diagnosis, providing morphological information essential for pathological assessment. Immunohistochemistry (IHC) and newer multiplexed imaging technologies complement H&E by revealing molecular information critical for accurate tumor subtyping and treatment decisions. In practice, however, H&E and IHC are obtained from different consecutive sections that are not spatially aligned, comprehensive multiplexed panels are expensive and tissue-consumptive, and not all stains are available at every clinical site, limiting comprehensive molecular profiling and the full diagnostic potential of these technologies in clinical practice. This dissertation addresses these gaps through three complementary generative deep learning studies in …

The expansion of adeno-associated virus (AAV)-based therapeutics has increased the demand for efficient clarification. In this study, a functionalized deconstructed depth filtration platform was developed to dissect how membrane–virus charge, modulated by pH, influences separation. Three charge regimes, positive (pH 4), neutral (pH 6), and negative (pH 8) between the membranes and AAVs, were examined. The flat-sheet system, composed of sequential 1.2, 0.8, 0.45, and 0.1 µm membranes, enabled the first layer-by-layer analysis of depth filtration behavior. This revealed that Stage 1 and Stage 4 dominate clarification. Remarkably, substantial DNA capture (> 75%) was observed even in the largest-pore stage, …

Coronary artery disease (CAD), one of the leading causes of mortality worldwide, necessitates effective risk assessment strategies, with coronary artery calcium (CAC) scoring via computed tomography (CT) being a key method for prevention. Traditional methods, primarily based on UNET architectures implemented on pre-built models, face challenges like the scarcity of annotated CT scans containing CAC and imbalanced datasets, leading to reduced performance in segmentation and scoring tasks. In this study, we address these limitations by introducing DINO-LG, a novel label-guided extension of DINO (self-distillation with no labels) that incorporates targeted augmentation on annotated calcified regions during self-supervised pre-training. Our three-stage …

Most tissue optical spectroscopy platforms use a fiber probe for light delivery and collection, while the inconsistent probe-sample contact could induce significant distortions in the measured optical signals, which consequently bring analysis errors. Moreover, it will be practically difficult to use a fiber probe for measurements in some cases such as oral cancer investigations using small animal models. To address the critical challenge, we report a portable, lens-based, optical spectroscopy device capable of quantifying key vascular and metabolic parameters in vivo without probe-sample contact. We combined lenses based diffuse reflectance and fluorescence spectroscopy into one portable platform to enable multi-parametric …

The hydrothermal liquefaction (HTL) process offers an energetic advantage over pyrolysis because it does not require prior drying of the biomass feedstock. However, there are significant challenges in simultaneously estimating both the yields and characteristics of products from the HTL of biomass with theoretical support. This study developed a unique element-based kinetic model to predict the yields, higher heating values, and fuel characteristics of solid residue and heavy bio-oil, based on the temperature, residence time, solid loading, and elemental composition (C, H, N, and O) of corn stover. Furthermore, the model predicted the weights of dissolved carbon and nitrogen in …

Bone regeneration is a multistage and tightly regulated process driven by coordinated interactions between the immune and skeletal systems. While inflammation is essential for initiating repair, its dysregulation contributes to delayed union, nonunion, and impaired healing commonly seen in trauma, chronic disease, and aging. Traditional interventions, including autografts and inert biomaterials, provide structural support but fail to engage or modulate the immune microenvironment that governs successful regeneration. Emerging research in osteoimmunology has revealed the centrality of immune cell populations such as macrophages, T regulatory cells, dendritic cells, and myeloid-derived suppressor cells in directing osteogenesis, angiogenesis, and the resolution of inflammation. …

The marine aquarium trade (MAT) is a significant global industry harvesting millions of wild-caught, live coral reef fishes for public and private aquaria markets in the United States and Europe annually, while supporting fisher livelihoods in the Indo-Pacific. This diverse and species-rich trade is considered data-limited, creating barriers to quantifying the current and future socio-ecological sustainability of the fishery. We present a revised and expanded productivity–susceptibility analysis (PSA) that serves as a holistic risk assessment to estimate the vulnerability of marine aquarium fish to overfishing. Our global analysis includes 306 species that are actively in trade. Improvements to the PSA …

Sulfhydryl group (SH) plays important roles in reactions with various functional groups, especially in biologically active systems. Photoswitchable diarylethene (DAE) derivatives have demonstrated applications in many research fields. Among the many classes of diarylethene derivatives, thiol derivatives have not been extensively explored. In this study, we systematically synthesized and characterized a series of bis-thiol functionalized dithienylethene derivatives. The thiol groups in this series are attached directly to the thiophene, or through a phenyl or methylene linker. Each series incorporated both dithienyl cyclopentene and hexafluorocyclopentene bridges and total six thiol substituted diarylethenes were synthesized. Among the six sulfhydryl DAE derivatives, four …

Antifreeze peptides inhibit ice crystal growth and recrystallization, and are promising components of cryoprotective formulations for cell, tissue, and food preservation, as well as anti-icing surface coatings. However, the discovery of new antifreeze peptides has been hindered by their sequence diversity and the limited scalability of experimental screening. In this study, we identify novel antifreeze peptide candidates from a microbiome-derived sequence library using ensemble machine learning and molecular dynamics (MD) simulations. We developed an ensemble classifier composed of 10 adapter-tuned protein-language models and a random forest meta-learner. After training on a curated dataset of 73 766 sequences, we applied this …

Glioblastoma (GBM) remains the most prevalent and lethal invasive brain tumor, denoted by a median survival of 12-15 months despite standard procedures such as safe surgical resection, radiotherapy, and chemotherapy. This limited effectiveness largely arises from the infiltrative nature of GBM cells, which interact with the brain’s extracellular matrix (ECM) by migrating along blood vessels and axonal pathways. The ECM contains the interstitial matrix (IM) and the
basement membrane (BM) made up of proteins and glycans which interact with the malignant cells. While research has mainly been focused on understanding how GBM cells interact with IM components, our knowledge about …
