Microbially Induced Calcite Precipitation (MICP) has emerged as a promising technique for soil stabilization, traditionally involving the preparation and mixing of treatment solutions with the soil. However, this thesis explores a novel approach of applying MICP by directly mixing dry chemical compounds into the soil and subsequently adding water. This alternative method offers potential advantages in terms of convenience, ease of implementation, and cost savings.
The objective of this research is to investigate the applicability of dry mixing protocols for MICP in soil stabilization. Through comprehensive experimentation, three different dry protocols were developed and applied to five different soils. The …

The application of olivine to coastal areas is a proposed method of removing carbon dioxide from the atmosphere for climate change mitigation. Olivine is abundant and dissolves relatively fast, but it contains trace amounts of nickel (Ni), chromium (Cr), and cobalt (Co) which may be toxic to marine biota in cases of widespread coastal olivine spreading. Prior studies have suggested that Ni concentrations in marine sediments and/or the overlying water column due to olivine dissolution could be a limiting factor for its carbon capture potential. This dissertation makes significant contributions to the field of ophthalmology, addressing the segmentation of retinal layers and the diagnosis of diabetic retinopathy (DR). The first contribution is a novel 3D segmentation approach that leverages the patientspecific anatomy of retinal layers. This approach demonstrates superior accuracy in segmenting all retinal layers from a 3D retinal image compared to current state-of-the-art methods. It also offers enhanced speed, enabling potential clinical applications. The proposed segmentation approach holds great potential for supporting surgical planning and guidance in retinal procedures such as retinal detachment repair or macular hole closure. In Colombia, ethnic communities have traditionally been responsible stewards of natural resources. They recognize the importance of these resources for their livelihood, as well as their ancestral and cultural heritage. El Tiple, a rural Afro-Colombian community, has been affected by the incursion of private corporations that promoted the expansion of sugarcane monocrops in their territory. Identifying disease biomarkers and detecting hazardous, explosive, flammable, and polluting gases and chemicals with extremely sensitive and selective sensor devices remains a challenging and time-consuming research challenge. Due to their exceptional characteristics, semiconducting metal oxides (SMOxs) have received a lot of attention in terms of the development of various types of sensors in recent years. The key performance indicators of SMOx-based sensors are their sensitivity, selectivity, recovery time, and steady response over time. DNA sensing is critical in various applications such as the early diagnosis of diseases and the investigation of forensic evidence, food processing, agriculture, environmental protection, etc. As a wide-bandgap semiconductor with excellent chemical, physical, electrical, and biocompatible properties, silicon carbide (SiC) is a promising material for DNA sensors. In recent years, a variety of SiC-based DNA-sensing technologies have been reported, such as nanoparticles and quantum dots, nanowires, nanopillars, and nanowire-based field-effect-transistors, etc. This article aims to provide a review of SiC-based DNA sensing technologies, their functions, and testing results.

The purpose of this experiment is to explore the inflammatory effects of focused ultrasound paired with intravenously delivered microbubbles when used in the lumbar spinal cord. There are currently very few pharmaceuticals that can successfully enter the blood spinal cord barrier (BSCB). This technique used at the lumbar enlargement might allow targeted treatment of pathologies involving the spinal cord.
To test the possible inflammatory response of this technique, an animal experiment was performed. 15 animals were given a T12 laminectomy and allowed to fully recover. Once recovered, all animals received FUS+MB at L2 and were euthanized in groups of five …

Cancer progression is linked to the emergence of aberrant mechanical signaling in the tumor microenvironment. Modulation of extrinsic signals, such as ECM stiffness and composition, have been thoroughly explored. However, the development of solid stresses within the tumor remains poorly understood. To address this, we have developed a microfluidic platform that generates deformable alginate microbeads that allow for the quantification of compressive stresses generated within a growing glioblastoma (GBM) tumorsphere. PDMS microfluidic devices were fabricated via SU-8 mold with channels ranging from 10µm-40µm in diameter. Car-following models traditionally focus on vehicle kinematics or dynamics parameters without considering human psychology. However, recent research has attempted to incorporate psychology into car-following models. Despite these efforts, there are limitations in understanding the psychological triggers that influence individual drivers' responses. To address this gap, we propose a new car-following model based on the Theory of Planned Behavior (TPB), which incorporates a human element by utilizing concepts of psychology. Two-dimension (2D) nanomaterials have gained popularity for the last few decades due to their excellent mechanical, electrical and thermal properties. These unique properties of 2D nanomaterials can be exploited in various applications specially in sensor, energy, and separation devices. In this study, the sensing and energy generation performance of PVDF/PAni fiber mat systems made by the forcespinning method with and without graphene coating. The graphene-coated nanocomposites show an average output voltage of 75 mV (peak-to-peak) which is 300% higher compared to bare fiber mats and an output current of 24 mA (peak-to-peak) by gentle finger pressing. Phosphatase of regenerating liver 3 (PRL-3) is associated with cancer metastasis and has been shown to interact with the cyclin and CBS domain divalent metal cation transport mediator (CNNM) family of proteins to regulate the intracellular concentration of magnesium and other divalent metals. Despite PRL-3’s importance in cancer, factors that regulate PRL-3’s phosphatase activity and its interactions with CNNM proteins remain unknown. Here, we show that divalent metal ions, including magnesium, calcium, and manganese, have no impact on PRL-3’s structure, stability, phosphatase activity, or CNNM binding capacity, indicating that PRL-3 does not act as a metal sensor, despite its interaction …

The Toxin Complex (Tc) superfamily consists of toxin translocases that contribute to the targeting, delivery, and cytotoxicity of certain pathogenic Gram-negative bacteria. Membrane receptor targeting is driven by the A-subunit (TcA), which comprises IgG-like receptor binding domains (RBDs) at the surface. To better understand XptA2, an insect specific TcA secreted by the symbiont X. nematophilus from the intestine of entomopathogenic nematodes, we determined structures by X-ray crystallography and cryo-EM. Contrary to a previous report, XptA2 is pentameric. Capture of atmospheric carbon by freshwater algae is being proposed in Clemson University’s outdoor algal carbon capture system, formerly described as the Partitioned Aquaculture System (Drapcho and Brune, 2000). Several algal species have been present in the system, depending on cultivation conditions. However, mechanical harvesting of the algae has proven difficult because of the cost of chemical flocculants, the energy cost of filtration due to small particle size of algae, and the relatively dilute algal cell concentrations in the system. Polylactic acid (PLA) is a biopolymer made from renewable resources such as sugar and corn. PLA filament is a popular material used in Fused Deposition Modeling (FDM) 3D-printing. While this material has many advantages, all the failed parts, support structures, rafts, nozzle tests, and the many prototype iterations during the 3D-printing process contribute to the plastic pollution and release of greenhouse gases. Although PLA is biodegradable, it can take years to degrade in landfills. Instead of throwing away PLA waste and buying new filaments, PLA can be recycled. Introduction: The Great Plains ecosystem offers an opportunity for young people to gain knowledge about intricate systems through practical learning. The spread of woody plants into grasslands, known as the “Green Glacier,” poses a risk to biodiversity and animal production. Although effective management strategies exist, some land managers are reluctant to use them. Roughness is an important parameter in rock engineering. Sine wave-based simulated wavy profiles were used to investigate systematic changes of amplitude and frequency on roughness indices. Profiles were 1000 mm in length with amplitude ranging from 0-10 mm and frequency ranging from 0-10 Hz. The roughness algorithms used were Root Mean Square (RMS), Energy, Sinuosity, Z2, Mean Absolute Angle (MAA), and Modified Divider a-Value. Roughness indices were visualized as contour plots. A brain computer interface (BCI) is a system which connects neural signals to a computer system. They have been used for controlling systems including robotics, on-screen computer control such as mouse movement, typing, and synthesizing audio signals. Invasive, or implanted, systems are often long-term medical solutions, or used for research where very clear signal is required. Non-invasive systems usually rely on exterior signals gathered through a headset using one or more electrode sensors. Colorectal cancer (CRC) is the third leading cause of cancer-related deaths among both men and women worldwide, a statistic that can be improved with early detection. Increased understanding of the tumor landscape has identified surface antigens that can be utilized to distinguish normal and cancerous tissue lining the colonic lumen. Use of antibodies conjugated to nano/micromaterials offers a means to detect these antigens, however, the heterogeneous nature of tumors makes it unlikely that antibodies against a single antigen will have sufficient sensitivity and specificity to detect all tumors. Triple negative breast cancer (TNBC) is the most resilient form of breast cancer, being one of the leading causes of death for women and making up 7% of all cancer deaths. Photodynamic therapy (PDT) offers a minimally invasive solution to TNBC as a passive-targeting treatment that reduces the need for other well established yet harsh treatments that can be taxing on the patient. Decentralized wastewater treatment exists primarily in suburban and rural areas where centralized treatment is not an option. Traditional on-site treatment systems usually include the use of a septic tank and a drain field. This orthodox method works well when implemented into environments that are accommodating for the infiltration of effluents, and thus allowing for soil treatment. Nanopore sensing has been successfully used to characterize biological molecules with single-molecule resolution based on the resistive pulse sensing approach. However, its use in nanoparticle characterization has been constrained by the need to tailor the nanopore aperture size to the size of the analyte, precluding the analysis of heterogeneous samples. Additionally, nanopore sensors often require the use of high salt concentrations to improve the signal-to-noise ratio, which further limits their ability to study a wide range of nanoparticles that are unstable at high ionic strength. To continue space operations with the increasing space debris, accurate characterization of fragment fly-out properties from hypervelocity impacts is essential. However, with limited realistic experimentation and the need for data, available static arena test data, collected utilizing a novel stereoscopic imaging technique, is the primary dataset for this paper. This research leverages machine learning methodologies to predict fragmentation characteristics using combined data from this imaging technique and simulations, produced considering dynamic impact conditions. This dissertation explores development and characterization of membrane chromatography tools for downstream purification of therapeutic biomolecules. Convective technologies, particularly membrane chromatography, are emerging in the field of bioseparations as an alternative to resin chromatography due to their improved throughput capability. Lignin is an abundant biopolymer with significant promise due to its aromaticity. It has been targeted as a replacement for a number of petroleum-based products including adhesives, coatings, polyurethane foams, activated carbon, and carbon fibers. However, commercially available bulk lignins are too polydisperse, and contain too many residual metals from the pulping process that are detrimental to the properties of the final product.

This dissertation explores the impact of the classroom environment on undergraduate engineering students by integrating three manuscripts using the process-person-context-time (PPCT) model within Bioecological Systems Theory. Each manuscript focused on students’ sense of belonging, motivation, and academic confidence. The study confirms prior research suggesting a link between students' perceptions of the environment and their sense of belonging, motivation, and academic confidence. The findings highlight the complex nature of student and classroom environment relationships throughout their college experience.
