Lentiviral vectors derived from human immunodeficiency virus type I are widely used to deliver functional gene copies to mammalian cells for research and gene therapies. Post-transcriptional splicing of lentiviral vector transgene in transduced host and transfected producer cells presents barriers to widespread application of lentiviral vector-based therapies. The present study examined effects of indole derivative compound IDC16 on splicing of lentiviral vector transcripts in producer cells and corresponding yield of infectious lentiviral vectors. Amyloid-beta (Aβ42) aggregates are characteristic Alzheimer’s disease signatures, but probing how their nanoscale architectures influence their growth and decay remains challenging using current technologies. Here, we apply time-lapse single-molecule orientation-localization microscopy (SMOLM) to measure the orientations and rotational “wobble” of Nile blue (NB) molecules transiently binding to Aβ42 fibrils. We correlate fibril architectures measured by SMOLM with their growth and decay over the course of 5 to 20 min visualized by single-molecule localization microscopy (SMLM). We discover that stable Aβ42 fibrils tend to be well-ordered and signified by well-aligned NB orientations and small wobble. SMOLM also shows that increasing order …

Total knee arthroplasty (TKA) is one of the most commonly performed orthopedic surgeries, with nearly one million performed in 2020 in the United States alone. Changing patient demographics, predominately indicated by increases in younger, more active, and more obese patients undergoing TKA, poses a challenge to orthopedic surgeons as these factors present a greater risk of long-term complications. This research developed methods for culturing self-assembling capillaries in an injectable gel as a potential method for vascularizing tissue-on-a-chip models to mimic physiological drug delivery. Additionally, a mathematical model was developed as a tool for understanding nutrient delivery and comparison of potential delivery systems. Organs-on-a-chip provide novel platforms for studying biology and physiology in 3D, allow exploration of tissue engineering on a manageable scale, and serve as models for drug screening and drug-delivery testing.

Creating a sustainable, ethical, and affordable substitute for animal blood in balloon catheter testing for Deep Vein Thrombosis (DVT) is important to take into consideration. This research utilizes agarose and dextran to develop hydrogel-based artificial blood clots. These hydro gels replicate the mechanical characteristics of human thrombi across acute, subacute, and chronic stages through their elastic and viscous moduli (G' and G") and phase angles. This project addresses extant testing protocols, including temperature regulation, agarose processing, and well plate size, and suggests efficient approaches to encounter these problems.

Patients with chronic kidney disease (CKD) have lost kidney function. 800,000 Americans have end-stage kidney disease resulting in one of the leading causes of mortality cross-listed with diabetes and heart disease. Boundary conditions (BCs) is one pivotal factor influencing the accuracy of hemodynamic predictions on intracranial aneurysms (IAs) using computational fluid dynamics (CFD) modeling. Unfortunately, a standard procedure to secure accurate BCs for hemodynamic modeling does not exist. To bridge such a knowledge gap, two representative patient-specific IA models (Case-I and Case-II) were reconstructed and their blood flow velocity waveforms in the internal carotid artery (ICA) were measured by ultrasonic techniques and modeled by discrete Fourier transform (DFT). Dear Editor, this letter is concerned with prescribed-time Nash equilibrium (PTNE) seeking problem in a pursuit-evasion game (PEG) involving agents with second-order dynamics. In order to achieve the prior given and user-defined convergence time for the PEG, a PTNE seeking algorithm has been developed to facilitate collaboration among multiple pursuers for capturing the evader without the need for any global information. Then, it is theoretically proved that the prescribed-time convergence of the designed algorithm for achieving Nash equilibrium of PEG. Eventually, the effectiveness of the PTNE method was validated by numerical simulation results.

Understanding higher level cognitive processes is a central problem in neuroscience. The Neuroidal model provides a useful framework for posing these problems in a computer science context. There has been significant recent work trying to understand memory capacity in the Neuroidal model but this work was done assuming that the network of neurons was an Erdos-Renyi random graph. However the network of neurons in the brain has been shown to exhibit small-world properties, which are not present in Erdos-Renyi graphs. This methodology called the Importance Hierarchy method, provides a comprehensive framework for decomposing the interactions of aerospace systems, focusing on identifying and evaluating critical components and interactions.

In vitro assays are essential for studying cellular biology, but traditional monolayer cultures fail to replicate the complex three-dimensional (3D) interactions of cells in living organisms. 3D culture systems offer a more accurate reflection of the cellular microenvironment. However, 3D cultures require robust and unique methods of characterization.

Methods

The goal of this study was to create a 3D spheroid model using cancer cells and macrophages, and to demonstrate a custom image analysis program to assess structural and metabolic changes across spheroid microregions.

The objective of this project was to convert a Sarastro 2000 confocal laser scanning microscope (CLSM) into a system capable of far-field two-photon excitation (TPE) imaging for the use of the PSU Biology department. TPE microscopy operates on the ability of fluorophores to accept two photons each with half the energy of a desired transition in a single quantum event via a virtual energy state and then emit a higher energy photon upon relaxation. Background: Acoustically activatable perfluoropropane droplets (PD) can be formulated from commercially available microbubble preparations. Diagnostic transthoracic ultrasound frequencies have resulted in acoustic activation (AA) predominately within myocardial infarct zones (IZ). Objective: We hypothesized that the AA area following acute coronary ischemia/reperfusion (I/R) would selectively enhance the developing scar zone, and target bioeffects specifically to this region. Methods: We administered intravenous PD in 36 rats and 20 pigs at various stages of myocardial scar formation (30 minutes, 1 day, and 7 days post I/R) to determine what effect infarct age had on the AA within the IZ.
