Construction Of A Derivative Of Agrobacterium Tumefaciens C58 That Does Not Mutate To Tetracycline Resistance, Zhao-Qing Luo, Thomas E. Clemente, Stephen K. Farrand

Agrobacterium tumefaciens C58 mutates to tetracycline resistance at high frequency, complicating the use of many broad-host-range cloning and binary vectors that code for resistance to this antibiotic as the selection marker. Such mutations are associated with a resistant gene unit, tetC58, that is present in the genome of this strain. By deleting the tetC58 locus, we constructed NTL4, a derivative of C58 that no longer mutates to tetracycline resistance. The deletion had no detectable effect on genetic or physiological traits of NTL4 or on the ability of this strain to transform plants.

Hybridization of complementary oligonucleotides mediated by a cationic surfactant at the water/hexane interface leads to hydrophobic, double-helical DNA which may be readily phase transferred to the organic phase and cast into thin films on solid substrates.

Presents a novel approach to sharing electronic health-care records that leverages the Internet and the World Wide Web, developed as part of two European Commission-funded projects, Synapses and SynEx. The approach provides an integrated view of patient data from heterogeneous, distributed information systems and presents it to users electronically. Synapses and SynEx illustrate a generic approach in applying Internet technologies for viewing shared records, integrated with existing health computing environments. Prototypes have been validated in a variety of clinical domains and health-care settings

The self-organizing map (SOM) is naturally unsupervised learning, but if a class label is known, it can be used as the classifier. In a SOM classifier, each neuron is assigned a class label based on the maximum class frequency and classified by a nearest neighbor strategy. The drawback when using this strategy is that each pattern is treated by equal importance in counting class frequency regardless of its typicalness. For this reason, the fuzzy class membership can be used instead of crisp class frequency and this fuzzy membership-label neuron provides another perspective of a feature map. The Integrative Experience was developed to help the college students consider the relationship of science and technology with contemporary society. Junior and senior students enrolled in the course designed WebQuests for middle school students. The WebQuests involved the integration of an environmental issue, problem, or technology and societal effects. Crystallins are a family of water-soluble proteins that constitute up to 90% of the water-soluble proteins in mammalian eye lenses. We present in this paper an alternative purification method for these proteins using polyethylene glycol/dextran aqueous two-phase extraction. Under the appropriate conditions, we were able to recover the γ-crystallin fraction essentially free of the remaining proteins. High concentrations of salt at a neutral pH maximize the recovery of γ-crystallins in the top phase and minimize the contamination by the other proteins present in the lenses. The temporal dynamics of electroporation of cells subjected to ultrashort voltage pulses are studied based on a coupled scheme involving the Laplace, Nernst-Plank, and Smoluchowski equations. A pore radius dependent energy barrier for ionic transport, accounts for cellular variations. It is shown that a finite time delay exists in pore formation, and leads to a transient overshoot of the transmembrane potential Vmem beyond 1.0 V. Pore resealing is shown to consist of an initial fast process, a 10−4s delay, followed by a much slower closing at a time constant of about 10 −1s. This establishes a …

An extensive literature exists on the estimation of the model parameters of time delayed processes. The objective of this paper is to review the main B-polynomial approaches to the delay estimation problem. More specifically, two algorithms are discussed and compared theoretically and in simulation. The algorithms involve various procedures to assess how the model relates to the observed data, to a priori knowledge, and to the algorithms’ intended use. Simulation results indicate that both the algorithms can suitably identify the model parameters, including the time delay. The research was conducted by the Utah Water Research Laboratory with funding provided by the Utah Division of Water Resources through Project RP 110080, March 1998. This paper reviews the problem of translating signals into symbols preserving maximally the information contained in the signal time structure. In this context, we motivate the use of nonconvergent dynamics for the signal to symbol translator. We then describe a biologically realistic model of the olfactory system proposed by Walter Freeman that has locally stable dynamics but is globally chaotic. We show how we can discretize Freemans model using digital signal processing techniques, providing an alternative to the more conventional Runge-Kutta integration. "Microarray technology is increasingly used as a means of high throughput analysis of human, non-human and plant genomes. Manual methods of array production have inherent imperfections and variations of the quality of output data derived from these arrays. A dynamic programming-based approach is presented to validate the quality of such data prior to insertion into evolving databases in order to assign reliably, genetic aberrations to bio-medical functions. Background:

During routine follow-up of patients treated with a three-piece stainless-steel modular femoral nail, osteolysis and periosteal reaction around the modular junctions of some of the nails were noted on radiographs. The purpose of this study was to evaluate the prevalence, etiology, and clinical relevance of these radiographic findings.

Methods:

Forty-four femoral fractures or nonunions in forty-two patients were treated with a modular stainless-steel femoral intramedullary nail. Seventeen nails were excluded, leaving twenty-seven intramedullary nails in twenty-seven patients for this study. All patients had had a femoral diaphyseal fracture; nineteen had had an acute fracture and eight, a nonunion. These …

This research explores four experiments of adaptive host-based intrusion detection (ID) techniques in an attempt to develop systems that can detect novel exploits. The technique considered to have the most potential is adaptive critic designs (ACDs) because of their utilization of reinforcement learning, which allows learning exploits that are difficult to pinpoint in sensor data. Preliminary results of ID using an ACD, an Elman recurrent neural network, and a statistical anomaly detection technique demonstrate an ability to learn to distinguish between clean and exploit data. Past mining activities have left a legacy of water quality problems in the Tri-State Mining District. Contemporary environmental risks stem from the release of lead and zinc from mine tailings previously dispersed far downstream in alluvial deposits as well as abandoned tailings piles. This study examines mining-related heavy metal contamination in the Chat Creek Watershed. Chat Creek drains the area near Aurora, Missouri on the eastern edge of the Tri-State District where mining occurred from 1886 to about 1930. The present paper presents a thermodynamic approach to assess the quality of human-thermal environment interaction and quantify thermal comfort. The approach involves development of entropy generation term by applying second law of thermodynamics to the combined human-environment system. The entropy generation term combines both human thermal physiological responses and thermal environmental variables to provide an objective measure of thermal comfort. In this research project a Sequencing Batch Reactor (SBR) plant was tested for performance in terms of Biochemical Oxygen Demand (BOD), Chemical Oxygen Demand (COD), and Suspended Solids (SS), under different temporal conditions. In addition, the removal of biological nutrients, namely ammoniacal nitrogen, nitrate nitrogen and phosphorous in the form of phosphate, from the sewage, with anaerobic and aerobic operations using SBR was studied.
