Principles Of Biology In Environmental Engineering: Molecular Biology-Based Identification Of Microorganisms, Daniel B. Oerther

Civil, Architectural and Environmental Engineering Faculty Research & Creative Works

The various aspects of a course on use of technology from molecular biology to screen microorganism in environmental samples and to monitor microbial biocatalysts in reactors, is discussed. The objective of the course is to teach students to adapt techniques from molecular biology to address important issues in environmental engineering. In case of methanogens, design considerations in environmental engineering include stabilization of sewage sludge through anaerobic digestion and wastewater treatment. The hands-on format encourages students to develop scientific questions, learn appropriate methodology, conduct careful experimentation, and draw conclusions worthy of presentation.

The objective of the proposed work is to acquire knowledge needed for the development and deployment of manufacturing processes utilizing the enormous technological potential of water ice. Material removal by blasting with ice media such as particles, pellets and slugs was investigated. The ice media was accelerated by entrainment in a fluid stream (air, steam, liquid water, supercritical C02), impact of rotating blades, fluid expansion, etc. The ice-airjet has to replace sand blasting and the ice-waterjet has to replace the abrasive waterjet. Cardiac hypertrophy is the remodeling or enlargement of the heart exhibited by individuals suffering from heart disease. Cardiac hypertrophy involves two mechanisms, mechanical stresses and humoral changes in the cardiac tissue. One such humoral change involves the Renin Angiotensin System (RAS). The primary fimction of RAS is maintenance of hemodynamics of the body mediated through an effector protein, Angiotensin 11. However, RAS has been shown to play a significant role in the pathophysiology of the heart; contributing to hypertension, cardiac hypertrophy and myocardial infarction. A new image processing method is described for measuring the 3-D coordinates of a complex, biological surface. One of the problems in stereo vision is known as the accuracy-precision tradeoff problem. This thesis proposes a new method that promises to solve this problem. To do so, two issues are addressed. First, stereo vision instrumentation methods are described. This instrumentation includes a camera system as well as camera calibration, rectification, matching and triangulation. Second, the approach employs an array of cameras that allow accurate computation of the depth map of a surface by propagation of correspondences through pair-wise camera views.

It is generally accepted that spasticity results from changes in the excitability of the stretch reflex. This change lowers the threshold of the motoneurons of the spinal cord where the integration of a signal from velocity/position sensors is processed and then fed back to the contracting muscle (alphaextrafusal and gamma-intrafusal fibers). The stretch reflex depends on the initial length of the muscle, the stretch velocity and voluntary activity. The exact sequence of the triggering events remains unknown, is poorly understood and as a result is controversial. The clinical classification scales are mainly subjective and by definition, inaccurate.

We report a DNA microarray-based method for genome-wide monitoring of competitively grown transformants to identify genes whose overexpression confers a specific cellular phenotype. Whereas transcriptional profiling identifies differentially expressed genes that are correlated with particular aspects of the cellular phenotype, this functional genomics approach determines genes that result in a specific physiology. This parallel gene-trait mapping method consists of transforming a strain with a genomic library, enriching the cell population in transformants containing the trait conferring gene(s), and finally using DNA microarrays to simultaneously isolate and identify the enriched gene inserts. In this paper, a two-dimensional transient radiation transport algorithm is developed to analyze short pulse laser transport through a tissue medium having tumors and inhomogeneities imbedded in it. Short pulse probing techniques have distinct advantages over conventional very large pulse width or cw lasers primarily due to the additional information conveyed about the tissue interior by the temporal variation of the observed signal. The distinct feature is the multiple scattering induced temporal signatures that persists for time periods greater than the duration of the source pulse and is a function of the source pulse width, the scattering/absorbing properties and nature …

To determine the long-term success of the recommended Yucca Mountain high-level nuclear waste repository, studies of bacterial colonization and biofilm development are needed. Bacteria involved in microbially-influenced corrosion and degradation are known to form biofilms with the potential to impact the integrity of repository packaging and structural materials. Temperature and humidity are environmental factors that can greatly affect biofilm formation. Therefore, it is necessary to determine the temperature and humidity conditions that affect biofilm formation. This thesis was designed to detect the type of spatial ability most effected by auditory stimulation, as described in the Mozart effect. Previous research has shown enhancement of performance on the paper-folding and cutting subtest of the Stanford-Binet Intelligence Scale IV with failures to replicate in research using other measures. Seventy-five college students enrolled in introductory psychology classes were exposed to one of three types of auditory stimulation, followed by two measures of spatial ability. This study evaluated the potential of using the Surface Energy Balance Algorithm for Land (SEBAL) as a means for estimating evapotranspiration (ET) for local and regional scales in Southern Idaho. The original SEBAL model was refined during this study to provide better estimation of ET in agricultural areas and to make more reliable estimates of ET from other surfaces as well, including mountainous terrain. The manufacture of edible/biodegradable films or coatings can potentially add value to soy protein. This study was conducted to determine the effect of sodium dodecyl sulfate (SDS) on selected physical properties of glycerin-plasticized soy protein isolate (SPI) films. This research focused on the novel use of hollow fiber membranes for gas delivery in biological dentrification using hydrogen-oxidizing bacteria. Autotrophic denitrification is a biological process that reduces nitrate to nitrogen gas using an inorganic carbon source. Hydrogen gas is an electron donor and nitrate is the electron acceptor in the reaction. The specific research objectives were to:

  • develop a mixed acclimated culture of hydrogen-oxidizing bacteria;
  • evaluate biodegradation kinetics of the acclimated culture;
  • evaluate hydrogen transfer characteristics of hollow fiber membrane modules;

and

  • demonstrate technical feasibility of a continuous bioreactor-membrane system for denitrification.

Niobium cavities are important parts of the integrated NC/SC high-power linacs. Over the years, researchers in several countries have tested various cavity shapes. They concluded that elliptically shaped cells are the most appropriate shape for superconducting cavities. The need for very clean surfaces led to the use of a buffered chemical polishing procedure for surface cleaning to get good performance of the cavities. This proposal discusses the second phase of research in the second year of the project.

The purpose of this collaborative research project involving the University of Nevada, Las Vegas (UNLV) and Idaho State University (ISU) is to evaluate the feasibility of determining residual stresses of welded (after pre-straining) engineering materials using a new nondestructive technique based on positron annihilation spectroscopy. The proposed technique is to use γ-rays 2 from a small MeV electron Linac to generate positrons inside the sample via pair production. This method can be used for materials characterization and investigation of defects in thick samples that could not be accomplished by conventional positron techniques or other nondestructive methods. The marron industry is of considerable importance to the Department of Fisheries, being one of the older aquaculture industries in Western Australia, and one that accounts for around half the aquaculture licence holders in the State. While it has not grown as quickly as many hoped in the past, in 2000/01, some 52.2 tonnes of marron were produced, worth $1.18 million at the farm gate.

Cardiovascular disease is the leading cause of death in North America. Cardiac infarction caused by thrombus plaque rupture can often lead to sudden death. Arterial stenosis caused by atherosclerosis is an important precursor leading to thrombus formation. Shear stress, abnormal lipid metabolism, and subendothelial layer exposure are the major contributors to arterial thrombus formation. Platelet activation by the above factors, followed by adhesion is the basic sequence in thrombus formation.

This study investigated the relationship between gender and competence as perceived by pilots. Scenarios were utilized that depicted an airline captain successfully landing an airplane amidst adverse conditions. Scenarios varied only by the gender of the Captain: male, female, or unspecified by variance of pilot name. Perceived effectiveness, competence, avoidance, blame, and attribution as depicted by the Captain's performance in the scenario were assessed by 30 male and 30 female certified flight instructors. Results showed that female flight instructors' ratings of avoidance ability and attribution for success differed from male flight instructors.
