Assembly permits the integration of different materials and manufacturing processes to increase system functionality. It is an essential step in the fabrication of useful systems across size scales from buildings to molecules. However, at the microscale, traditional “grasp and release” assembly methods and chemically inspired self-assembly processes are less effective due to many scaling effects. A task overload condition often leads to high stress for an operator, causing performance degradation and possibly disastrous consequences. Just as dangerous, with automated flight systems, an operator may experience a task underload condition (during the en-route flight phase, for example), becoming easily bored and finding it difficult to maintain sustained attention. When an unexpected event occurs, either internal or external to the automated system, the disengaged operator may neglect, misunderstand, or respond slowly/inappropriately to the situation. Methamphetamine (Meth) is a widely abused stimulant and its users are at increased risk for multiple infectious diseases. To determine the impact of meth on the immune system, we utilized a murine model that simulates the process of meth consumption in a typical addict. Our phenotypic analysis of leukocytes from this dose escalation model revealed that meth affected key immune subsets. Meth administration led to a decrease in abundance of natural killer (NK) cells and the remaining NK cells possessed a phenotype suggesting reduced responsiveness. Dendritic cells (DCs) and Gr-1(high) monocytes/macrophages were also decreased in abundance while Gr-1(low) monocytes/macrophages appear …

Neuropeptidases are responsible for degradation of signaling peptides in the central nervous system and periphery. Some neuropeptidases have also been shown to play a role as part of the cell’s hydrolytic machinery responsible for breaking down proteins and peptides into amino acids, and these enzymes therefore influence small peptide availability for antigen presentation. A better understanding of how neuropeptidases recognize their substrates could lead to therapeutics that modulate the activity of these important enzymes. Alternatively, re-engineering these enzymes to selectively hydrolyze undesirable peptides could make them attractive as therapeutics themselves. The carotid bodies are small sensory organs located along the bifurcation of the carotid arteries. They detect changes in blood gases and relay this information to the brain to allow for initiation of appropriate respiratory and cardiovascular responses. A decrease in oxygen (hypoxia) sensed by the carotid body results in an increase in firing of the carotid sinus nerve and ultimately a change in one's breathing pattern. Insulin resistance and kidney disease are intimately linked and both independently tied to dysregulation of the renin angiotensin system (RAS). The purpose of this research was to better investigate Angiotensin Converting Enzyme (ACE) 2 as a possible motif in the pathophysiology of these two disease processes. The project was divided into two parts. Part 1 investigated ACE2 KO mice as a model of albuminuria. Part 2 investigated how the lack of ACE2 enzyme affected glucose homeostasis and subsequent albuminuria.

Organophosphorous compounds (OPs) are a class of serine esterase inhibitors that have widespread application as pesticides, veterinary pharmaceuticals and chemical warfare agents. Environmental contamination is ubiquitous. The threat of exposure is a concern for both military and civilian populations. Acute inhibition of acetylcholinesterase by OPs triggers a cholinergic crisis that results in muscle flaccidity, paralysis, convulsions and death. At low doses OPs can alter neuronal differentiation, cell signaling, behavior and cognition through unknown mechanisms.
