Microbes live in complex and continually changing environments. Rapid shifts in nutrient availability are a common challenge for microbes, and cause changes in intracellular metabolite levels. Microbial response to dynamic environments requires coordination of multiple levels of cellular machinery including gene expression and metabolite concentrations. This coordination is achieved through metabolic control systems, which sense metabolite concentrations and direct cellular activity in response. Several reoccurring control architectures are found throughout diverse metabolic systems, which suggests underlying evolutionary advantages for using these control systems to coordinate metabolism. One common, yet understudied, control architecture is the positive feedback metabolite uptake loop, which …

Microbes naturally coexist in complex, multi-strain communities that are valuable assets for their host. Commensal and probiotic microbes prevent pathogen colonization, reduce the frequency and severity of various ailments, provide essential nutrients, and offer various additional benefits. Understanding the dynamics of and tailoring microbial communities to provide additional beneficial functions is a primary focus of researchers in medicine and agriculture. To date, consortia have primarily been manipulated by supplementing the communities with microbes that were engineered in vitro or by introducing stimuli that alter the metabolism or composition of the community. This dissertation describes a “smart dining table” that can detect and measure consumption events. This work is motivated by the growing problem of obesity, which is a global problem and an epidemic in the United States and Europe. Chapter 1 gives a background on the economic burden of obesity and its comorbidities. Arsenic is a ubiquitous trace metalloid. However, increasing concentrations of arsenic in soil and water is a problem the world faces due to inputs from anthropogenic sources such as mining, milling, agriculture, and coal ash. One method of remediation for these arsenic-contaminated sites is bioleaching. While it is most often used to extract metals from raw ore, it is a viable and environmentally friendly method to remove the same metals from soil. Various fractionation processes like kraft, sulfite, and organosolv, usually need to be carried out in expensive reactors due to high temperatures (e.g., 160-260 °C) and high pressures (e.g., 5-50 atm). During fractionation, conventional chemicals such as mineral acids (e.g., sulfonic acid and hydrochloric acid), aliphatic organic acids (e.g., formic acid and oxalic acid) and alkalis (e.g., sodium hydroxide and potassium hydroxide) are typically used as catalysts, which could cause structural alterations or even losses to the fractionated components. Poly(3-hydroxybutyrate) (PHB) is one of the most broadly studied members of the polyhydroxyalkanoates (PHA) family. It is a fully biodegradable thermoplastic produced by microbial fermentation having properties similar to polypropylene. PHB has a market price that is three times higher than the cost of synthetic plastics. Substrate cost makes up 50 to 60 % of that cost; hence, researchers have been exploring various cheap natural resources with high carbohydrate content to be used as a substrate for PHB production.

In the present study, Zea mays seedlings grown under nano Cu(OH)2 (nCu), bulk Cu(OH)2 (bCu), and ionic CuSO4 (iCu) compound exposure were harvested after six days. The nutritional profile was determined to be significantly disrupted in the roots by 1000 ppm bCu treatment, resulting in a 58.7% reduction in potassium compared to the control. In the shoots, a significant decrease of manganese was observed for 10 and 1000 ppm iCu treatments with 55.7% and 64.2% reductions, respectively. The overall protein content and catalase (CAT) enzymatic activity, however, remained unaffected in either roots or shoots, while an absence of polyphenol oxidase …

Nanoparticles (NPs) are particles less than 100 nm (~4 millionths of an inch) in a direction. NPs, due to their small size, are used in a variety of products, such as silver (Ag) NPs as an antimicrobial in clothes. Copper Oxide (CuO) NPs are used in electronics as semiconductors and other fields as antimicrobials and purposefully or accidentally end up in the environment. Copper (Cu) is a necessary nutrient for plants, but at higher amounts is toxic to plants and beneficial soil microbes.

The headgear design research community has focused on developing accurate computational and experimental analysis techniques to better assess the ability of headgear technology to attenuate impacts and protect athletes from TBI. Despite efforts to innovate the headgear system, minimal progress has been made to innovate the faceguard. Deep learning has proved to be successful for many computer vision and natural language processing applications. In this dissertation, three studies have been conducted to show the efficacy of deep learning models for computer vision and natural language processing. In the first study, an efficient deep learning model was proposed for seagrass scar detection in multispectral images which produced robust, accurate scars mappings. Adolescent Idiopathic Scoliosis, a three-dimensional deformity of the thoracolumbar spine, affects approximately 1-3% of patients ages 10-18. Surgical correction and treatment of the spinal column is a costly and high-risk task that is consistently complicated by factors such as patient-specific spinal deformities, curve flexibility, and surgeon experience. The following dissertation utilizes finite element analysis to develop a cost-effective, building-block approach by which surgical procedures and kinematic evaluations may be investigated. Harmful Algal Blooms (HABs) are an increasing concern not only in Nebraska, but across the world. Increasing global temperatures in conjunction with overuse of synthetic fertilizers are leading causes to the increasing frequency and concern of HABs. In the present work, the effectiveness of simulated bank filtration (BF), as an economical water treatment option, to remove cyanotoxins under different levels of nutrients (nitrates and phosphates) was investigated. Emissions of carbon dioxide, methane, and nitrous oxide – potent greenhouse gases - from stored beef feedlot manure are a significant concern relative to climate change. Research on methane reduction strategies for enteric emissions has identified the application of organic additives, such as bromoform and linseed oil, to ruminant diets as potential solutions for reducing enteric emissions and pathogenic bacteria in excreted manure. The main goal of this study is to develop a paste formulation containing a mixture of ginger, lemon and mint powder, as well as a combination of legumes and cereals with high nutritional value. This will expand the range of pasta. The task of achieving high quality and economic efficiency is solved by improving a number of technological processes and applying innovative methods. The effectiveness of using innovative methods of freeze-drying and microwave drying is shown in comparison with the process of drying instant pasta by frying them in vegetable oil.

Coda wave interferometry (CWI) has been used to measure the relative wave-velocity change (dV/V) caused by small changes in materials. This study uses the stretching processing technique which has been used for CWI analysis to investigate velocity changes of direct longitudinal (P) wave, direct shear (S) wave, and coda wave in concrete by choosing different time windows of ultrasonic signals. It is found that the obtained wave-velocity change depends on the time window position, because the relative contribution of P wave and S wave is different in each signal window.
