Osteoarthritis is a joint disease characterized by the breakdown of articular cartilage. The field of tissue engineering is interested in developing methods to produce biological alternatives to current orthopedic procedures. Lubricin is a molecule which is important in the proper lubrication of articular cartilage. It is a challenge in the field of tissue engineering to produce cartilage with sufficient lubricin expression. Developing a reporter cell for lubricin allowed for a more efficient investigation of the conditions which may influence its expression. In January 2020, the Government of Belize enacted an Implementation Strategy and Action Plan to phase-out single-use plastics and to transition to products like bioplastics. This work investigated the environmental effects of using alternative waste management techniques to manage polylactic acid biopolymer (PLA) waste by using life cycle assessment (LCA). The following treatment options were compared: landfill, landfill expansion, cogeneration, and anaerobic digestion. The landfill and landfill expansion processes both had a global warming potential of 0.01 kg of CO2 eq. per kg of PLA waste managed compared to the cogeneration and anaerobic digestion processes -0.03 and -0.06 kg …

3D cell culture and microfluidics both represent powerful tools for replicating critical components of the cell microenvironment; however, challenges involved in integration of the two and compatibility with standard tissue culture protocols still represent a steep barrier to widespread adoption. Here we demonstrate the use of engineered surface roughness in the form of microfluidic channels to integrate 3D cell-laden hydrogels and microfluidic fluid delivery. When a liquid hydrogel precursor solution is pipetted onto a surface containing open microfluidic channels, the solid/liquid/air interface becomes pinned at sharp edges such that the hydrogel forms the “fourth wall” of the channels upon solidification. …

Personalized Medicine - from a one-size fits all to a tailored approach is the future in pharmaceutical therapeutics and medical applications. The aim of this study is demonstrate additive manufacturing in two personalized medicine applications: 1. Patient- customized solid dose medicines, and 2. Patient-customized wound care device. The aim of the first application is to develop an inexpensive and accurate method to design and fabricate patient-customized tablets. Alfalfa is a popularly grown crop because of its value as a nutritious feed source for livestock. The efficient production of an alfalfa crop relies on the monitoring of certain parameters, like height, quality, and yield. Traditionally, producers have used manual measurements of alfalfa plant height to estimate the nutritive quality and yield of a growing alfalfa crop. Manual measurements of plant height are often labor intensive and provide low resolution data that is not acceptable for full field scale assessment of growing alfalfa. The In-pond Raceway (IPR) is a novel option for production aquaculture, depending on water moving devices to provide constant flow. Device failure may result in catastrophic fish loss, requiring power backup systems to mitigate risk in case of power outages. Because these systems must be dependable and many suitable locations are remote, off-grid solar photovoltaic (PV) systems with battery storage have been considered since they eliminate need for utility power. Such systems can be hard to size and expensive. Metal 3-D printing has been relegated to high-cost proprietary high-resolution systems and low-resolution low-cost metal inert gas (MIG) systems. In order to provide a path to high-resolution, low-cost, metal 3-D printing, this manuscript proposes a new open source metal 3-D printer design based around a low-cost tungsten inert gas (TIG) welder coupled to a commercial open source self replicating rapid prototyper. Optimal printing parameters for the machine are acquired using a novel computational intelligence software. Systemic Lupus Erythematosus (SLE) is a chronic autoimmune disease that primarily affects women. The etiology is not known yet, but genetics and environmental factors trigger the disease. These epigenetic changes modify gene expression and contribute to the development of the disease. It is known that T cells from patients with lupus overexpress genes that may contribute to the onset and progression of the disease. Dr. Gorelik has shown that OGT (O-linked N-acetyl-Glucosamine Transferase), which is an X-linked gene, is overexpressed in women with SLE. OGT adds b-N-acetylglucosamine (O-GlcNAc) to serine and threonine residues of proteins. …

Childhood nutrition establishes consumption norms that affect an individual’s health over the course of their lives. However, early nutrition interventions to establish such norms are uncommon owing to the various inefficiencies associated with current methods of measuring childhood nutrition. Here, we present an IoT measuring device, called the SensiTray, which accurately tracks mealtime intake in a child-friendly and cost-effective fashion. Principal technologies underlying the SensiTray (including mass-sensing technologies, microcontrollers) are identified and analyzed, along with other design choices. Biofilm infections in wounds seriously delay the healing process, and methicillin-resistant Staphylococcus aureus is a major cause of wound infections. In addition to inactivating micro-organisms, low-temperature gas plasma can restore the sensitivity of pathogenic microbes to antibiotics. However, the combined treatment has not been applied to infectious diseases. In this study, low-temperature gas plasma treatment promoted the effects of different antibiotics on the reduction of S. aureus biofilms in vitro. Low-temperature gas plasma combined with rifampicin also effectively reduced the S. aureus cells in biofilms in the murine wound infection model. The modern era holds vast modalities in human data utilization. Within Biocybersecurity (BCS), categories of biological information, especially medical information transmitted online, can be viewed as pathways to destabilize organizations. Therefore, analysis of how the public, along with medical providers, process such data, and the methods by which false information, particularly propaganda, can be used to upset the flow of verified information to populations of medical professionals, is important for maintenance of public health. Herein, we discuss some interplay of BCS within the scope of propaganda and considerations for navigating the field.

This dissertation uses multi-dimensional modeling for environmental impact assessment at intersections of the Food-Energy-Water (FEW) Nexus, including life cycle assessment (LCA) modeling for quantification of environmental impacts due to household FEW consumption, a linear regression framework for quantification of water-use impacts of marginal electricity generation, and a multi-objective optimization model to assess monetization of water withdrawals for electricity generation and impacts to water stress due to electricity dispatch schemes. Huntington’s disease (HD) has classically been categorized as a neurodegenerative disorder. However, the expression of the disease-causing mutated huntingtin gene in skeletal muscle may contribute to the symptoms of HD, namely those that involve involuntary muscle contraction. In the R6/2 transgenic mouse model of HD, we previously observed ion channel defects that could contribute to involuntary muscle contraction. Here, in R6/2 muscle we investigated the consequence of these ion channel defects on action potentials (APs), the first step in excitation-contraction (EC) coupling. We found that the ion channel defects were associated with depolarizing the baseline membrane potential during AP trains. …

Globally, dairy farming is a $700 billion industry, with more than 9 million dairy cows in the United States alone. Depriving cows of required activities such as sleep has been shown to negatively impact reproductive efficiency, decrease the volume of milk produced, and increase the risk of culling. Overcrowded herds can decrease individual animal health, demanding the need for automatic behavior detection that would provide insight into their state of health.

Pectin is a structural anionic heteropolysaccharide and abundantly found in the cell wall of terrestrial trees and plants. It exhibits several advantageous properties such as non-toxicity, cheap, biodegradable, biocompatible, abundant, flexible, etc. Functional groups like carboxylic acid and hydroxyl make pectin suitable to be covalently bonded with other biomolecules and proteins. The land application of animal manure can introduce manure microbiome and resistome to croplands where food crops are grown. The objective of this study was to characterize the microbiome and resistome on and in the leaves of lettuce grown in manured soil and identify the main transmission routes of microbes and antibiotic resistance genes (ARGs) from soil to the episphere and endosphere of lettuce. Shotgun metagenomic results show that manure application significantly altered the composition of the microbiome and resistome of surface soil. Mechanical loading is essential for maintaining bone tissue. Reduced mechanical loading has been shown to have a negative effect on bone, and can result in the development of disuse osteoporosis. Disuse models of muscle inactivity and immobilization, like the Botox model used in this study, result in changes in the bone microarchitecture, the mechanisms behind which are not fully understood. In a previous four-week Botox disuse study, skeletally mature 20- week-old rats experienced degradation of intracortical bone, increased vascular porosity, and decreased osteocyte lacunar density in the tibiae. Low back pain is the most common cause of disability in the world and is often caused by degeneration or injury of the intervertebral disc (IVD). The IVD is a complex, fibrocartilaginous tissue that allows for the wide range of spinal mobility. Disc degeneration is a progressive condition believed to begin in the central, gelatinous nucleus pulposus (NP) region of the tissue, for which there are few preventative therapies. It is difficult for developers, researchers, and consumers to compare results among emerging wearable technology without using a uniform set of standards. This study evaluated the accuracy of commercially available wearable technology heart rate (HR) monitors using the Consumer Technology Association (CTA) standards. Participants (N = 23) simultaneously wore a Polar chest strap (criterion measure), Jabra Elite earbuds, Scosche Rhythm 24 armband, Apple Watch 4, and Garmin Forerunner 735 XT during sitting, activities of daily living, walking, jogging, running, and cycling, totaling 57 min of monitored activity. Excessive algal growth in freshwater lakes can negatively impact ecosystems, recreation, and human health. Though algae are a natural part of freshwater ecosystems, elevated nutrient loading from anthropogenic and natural sources can lead to algal blooms. Staff technical challenges and resistance to change can affect technology return on investment, which concerns telemedicine program leaders. To develop well-planned and effective policies and programs for reducing avoidable food waste, it is important to quantify the actual food waste level in particular settings and assess relationships among consumers’ awareness, attitudes, and behaviors. Recognizing these considerations, this paper measured avoidable food waste generated by university students living in dormitory buildings and identified its underlying causes in the case of Kanchanaburi campus, Mahidol University, Thailand. The study applied a food waste composition survey 18 times between January and May 2019 while administering questionnaires in October 2019 to the dormitory students. We evaluated the use of attenuated total reflectance infrared spectroscopy for simultaneous in situ quantification of the nutritional composition of liquid food stuffs in the industrial kitchen context. Different methodologies were compared, including dry and wet acquisition along with instrument parameters and measurement times of 4 and 60 s. The most effective technique was 1-minute measurement, with prediction errors of 2.6, 0.7, 1.0, 2.2, 0.8, 2.4 g/100 mL and 150 Kcal, for carbohydrates, proteins, fat, sugars, saturated fat, water and energy values, respectively.The 4-second method resulted in larger errors but was more applicable for inline measurements. The company adopted bulk-purchasing to packaging inventory to realize the economy of scales. Because of the lacks of purchasing standard and monitoring, this inventory policy led to the overstock, deadstock, and obsolete packaging inventories as well as the fluctuation of inventory turnover rate.
