While reinforcement learning has been a vital component in artificial intelligence and machine learning, there exist many open questions about its implementations and how to improve them, in both minds and machines. Among these are i) the contribution of non-neuronal cell types to reinforcement learning, and ii) information-seeking behavior during reinforcement learning. In this thesis, we studied these main topics pertaining to reinforcement learning. Engineering practices are increasingly adopting sustainable methods, with Leadership in Energy and Environmental Design (LEED) being a widely recognized standard. LEED ensures environmental health and efficiency in building projects. This project focuses on renovating an existing building and adding a warehouse for the PENCIL Foundation, a local non-profit. The design includes structural, mechanical, and HVAC drawings along with stormwater and indoor water use evaluations, and EPSC (Erosion Prevention and Sediment Control) plans. The rising demand for sustainable and resilient energy systems has accelerated the transformation from traditional power grids toward smart grids integrated with renewable energy networks. This thesis explores the integration of solar energy sources into smart grids to replace conventional fossil-fuel-based generation. A simulation model is developed using MATLAB/Simulink to evaluate the operational performance, stability, and adaptability of the proposed smart grid architecture. Key challenges, including intermittency, synchronization, and bidirectional power flows, are identified, and corresponding solutions, such as energy storage systems, are proposed to address these issues. Crash prediction models are essential for evaluating traffic safety by analyzing crash occurrence, frequency, or severity. Recently, machine learning techniques have gained prominence in statistical regression modeling and data analysis. This study assesses the effectiveness of machine learning in predicting crashes on Egyptian rural multi-lane divided roads using limited regional data. Supervised machine learning ensemble techniques were applied to predict crash-prone segments (classification) and estimate the total number of crashes per segment (regression). A comparative analysis aims to identify the most suitable method. Visualization literacy assessments shape how we understand people's ability to interpret data, yet most existing instruments embed Western datasets and assumptions that limit their relevance for global audiences. This thesis argues that because data is personal, assessments must also be culturally grounded. We introduce a unified framework for adapting the Mini-VLAT into 22 regionally responsive short-form assessments, each retaining the structure of the original test while incorporating datasets and scenarios tailored to specific regions around the world. The ATP-sensitive potassium (KATP) channel is a critical metabolic sensor in skeletal muscle, yet its definitive molecular composition and functional roles remain contested. In this study, I conclusively demonstrate that the Kir6.2/SUR2 complex forms sarcolemmal KATP channel in mouse fast-twitch muscle. Genetic knock-out (KO) of Kir6.2 (Kcnj11−/−) or SUR2 (Abcc9−/−) resulted in a similar phenotype with increased fatigue resistance and a pathological rise in unstimulated resting tension at high-frequency stimulation. In contrast, SUR1 (Abcc8−/−) KO was ineffective. Furthermore, a CRISPR/Cas9 knock-in mouse model of a human SUR2A truncation variant (KCGV/KCGV) recapitulated the abnormal force accumulation, demonstrating that even loss-of-function of …

Split-thickness skin grafts are widely used to treat chronic wounds. Procedure design requires surgeons to predict how much a patch of the patient's own skin expands when it is meshed with rows of slits and stretched over a larger wound area. Accurate prediction of graft expansion remains a challenge, with current models overestimating the actual expansion, leading to suboptimal outcomes. Inspired by the principles of mechanical metamaterials, we developed a model that distinguishes between the kinematic rearrangement of structural elements and their stretching, providing a more accurate prediction of skin graft expansion. ATP-sensitive potassium (KATP) channels, composed of the SUR1 and Kir6.2 subunits encoded by the ABCC8 and KCNJ11 genes, are critical regulators of membrane excitability and insulin secretion in pancreatic β-cells. Gain-of-function (GOF) mutations in these genes cause neonatal diabetes mellitus through impaired insulin secretion and persistent hyperglycemia, whereas loss-of-function (LOF) mutations lead to congenital hyperinsulinism (CHI) with hypoglycemia due to β-cell hyperexcitability. I have addressed a paradoxical form of maturity-onset diabetes of the young (MODY) arising from KATP mutations, in which patients transition from CHI to glucose intolerance later in life. Integrating nephelometers are key instruments for quantifying aerosol light scattering, a fundamental parameter governing the direct radiative effect of aerosols on climate and visibility, and for interpreting satellite observations of aerosol optical depth. The AirPhoton integrating nephelometer quantifies aerosol light scattering by measuring total scattering from the sample air and subtracting a clean-air reference that represents Rayleigh scattering, instrument wall scattering, and detector background noise. Gradient biomaterials that exhibit spatially varying physical, chemical, or biological properties can be used in various applications such as tissue engineering, organoid development, mechanobiology, and spatially controlled drug delivery. However, current fabrication methods often suffer from limited gradient precision, restricted material compatibility, and poor reproducibility. Here, we introduced gradient regulation via acoustofluidic dynamic engineering (GRADE), a programmable system to generate high-fidelity gradient biomaterials across different material systems. By incorporating focused interdigital transducers with the pulsed surface acoustic wave actuation, GRADE can achieve tunable and directional acoustic streaming (0 to 22 millimeters per second), which allows accurate regulation of the gradient …

Calcium phosphate cements (CPCs) and biomaterials, such as mesoporous bioactive glass (MBG), are critical for bone tissue engineering. This study aimed to 3D-print CPC scaffolds modified with MBG to enhance their osteogenic potential and regenerative ability. MBG powder was synthesized and characterized using transmission electron microscopy (TEM), X-ray diffraction (XRD), and nitrogen adsorption-desorption techniques. A commercial CPC ink (hydroxyapatite/alpha-tricalcium phosphate) was mixed with 5% MBG (w/w; CPC/MBG), and, after rheological assessment, the mixture was used to obtain scaffolds via 3D printing. Harmful algal blooms (HABs) are a serious and growing risk to waterbodies around the world. Excessive algal growth, especially when it involves cyanobacteria, impairs the functioning of aquatic ecosystems and threatens the health of humans and animals using the water. There is currently great interest in developing effective prevention and mitigation strategies for these blooms, particularly in ecologically and economically vital freshwater lakes. Such development, however, requires detailed understanding of HAB drivers and dynamics, which can be highly complex and difficult to characterize. Optical emission spectroscopy (OES) measurements of the reentry shock layer can provide invaluable information about the dynamic aerothermal chemistry during atmospheric reentry and yield critical data needed to improve fluid dynamics modeling of reentry systems. On 28 February 2025, the Air Force Research Laboratory successfully executed the Optical Sensing of Plasmas in the ReEntry Environment (OSPREE) experiment using Varda Space Industries’ W-2 hypersonic test-bed vehicle, obtaining in situ shock-layer radiative emission measurements during reentry from an altitude of 105 km down through 50 km, encompassing a range of Mach numbers between 29.5 and 15.0. The importance of this study is to perform antibody discovery as a means to derive novel preventative and therapeutic antibody countermeasures against the Junín virus, to alleviate the potential risk of a global pandemic. The purpose of this methodological study was to develop a protocol that utilizes yeast surface display to discover antibodies, from naïve human antibody libraries, that can bind the Junín Virus antigen. The increasing demand for sustainable practices in the metal casting industry has driven the use of recycled aluminum alloys such as ADC12. However, the addition of aluminum scrap tends to increase oxide inclusions, which reduce melt fluidity and compromise casting quality. This study utilizes the Inclusion and Fluidity Measurement Instrument (IFMI), equipped with mullite ceramic filters, to assess the melt quality of ADC12 aluminum alloy containing 30% scrap. Fluidity and inclusion characteristics were evaluated at five pouring temperatures (660°C, 680°C, 700°C, 720°C, and 740°C). This research investigates the production decline and cost increase in the X&Y oil and gas fields from 2022 to 2023. Production fell by 34%, while production costs per barrel rose by 79%. To address these issues, a series of optimization processes are proposed. These aim to restructure and enhance the production facilities to reduce current production costs. The optimizations include reducing pressure at the PPP Platform in 2024. This study investigates the potential of ceramic membranes derived from areca nut fiber as a cost-effective and environmentally sustainable material for the removal of iron (Fe) and manganese (Mn) from groundwater. Two types of membranes were fabricated: one without activation and one chemically activated using 10% sodium hydroxide (NaOH). The morphological and elemental characteristics of both membranes were analyzed using Scanning Electron Microscopy (SEM) and Energy Dispersive X-ray (EDX) mapping. The concentrations of Fe and Mn before and after treatment were measured using Atomic Absorption Spectroscopy (AAS). Gamma spectroscopy is a technique used for the detection and identification of nuclear materials. Gamma-ray emission by radio nuclides is detected using scintillation detectors or semiconductor detectors such as NaI and CZT. Robot-based gamma probes enable radiation measurements in large areas and hard-to-reach cluttered hazardous environments, which significantly reduces radiation risks to personnel. Every second, human skin processes over one million sensory signals while maintaining properties such as electrical conductivity, mechanical adaptability, and regenerative capability that surpass all synthetic materials. Contemporary bioelectronic devices prove inadequate when contacting skin surfaces due to poor adhesion and electrical contact issues that prevent effective sensing. This research explores early college science majors’ conceptions of the Mojave Desert ecosystem as a complex system. A multi-level, explanatory sequential mixed methods design was employed to study college science majors’ understanding of the Mojave Desert ecosystem to better inform science pedagogy. The first level, the quantitative phase of the study, consisted of developing a mathematical representation of twelve ecosystem variables present within the Mojave Desert. The second level, the qualitative phase of the study, consisted of phenomenological semi-structured interviews with 23 early college science majors. This work applies the Lyapunov method to identify instabilities and compute the growth rate of a linear time-varying system. The linear system studied describes cold fresh water on top of hot salty water with a periodically time-varying background shear flow. A time-dependent weighting matrix is employed to construct a Lyapunov function candidate. The resulting linear matrix inequalities are discretized in time using the forward Euler method. As the number of temporal discretization points increases, the growth rate predicted by the Lyapunov method or Floquet theory, used for comparison, will converge to the same value obtained from numerical simulations. The Fitness for Service (FFS) analysis is performed as a quantitative assessment to evaluate the integrity condition of a pipeline. Essentially, FFS assessment helps determine whether equipment components can operate safely despite existing deficiencies. This evaluation is carried out using the Finite Element Method (FEM). In the case of an underwater pipeline that experiences leak due to anchor pull at the flange connection, it undergoes plastic deformation and is lifted approximately 1 meter. Despite achieving over 90% accuracy on medical benchmarks, recent studies show physicians cannot effectively leverage language models to improve clinical reasoning. Current benchmarks test isolated factual recall, but clinical practice requires hierarchical navigation through diagnostic categories—starting broad and narrowing systematically from chest pain to cardiovascular pathology to myocardial infarction to specific STEMI types. Existing evaluations cannot measure whether models preserve this taxonomic structure essential for clinical reasoning.

This study developed and validated a hybrid ISO 31000:2018 Kosovo-compliant risk assessment framework tailored for construction site managers operating within transitional economies. The research employed audits of 20 ISO-certified contractors in Pristina, Kosovo, along with field observations, interviews, and quantitative risk matrices. The findings indicate a critical concentration of risk, with 70% of firms classified as "High" or “Very High” risk. The most prevalent hazards identified were working at height without collective protection, unprotected electrical systems, and manual handling of heavy materials. Regression analysis revealed that firms with a designated safety champion achieved significantly lower risk scores, whereas those with …

Deep borehole disposal is an emerging concept which involves disposing of radioactive
waste in boreholes several kilometers deep then backfilling to minimize radionuclide
transport and distribute weight. Unfortunately, little documentation exists on borehole
safety case development, especially for pre-closure operations. This study aims to close
this gap by proposing a graded approach matrix based on consequence, maturity, and
complexity. Two case studies are used to demonstrate this application: Hanford CsCl
